Mechanisms of glacial-to-future atmospheric CO2 effects on plant immunity.

The impacts of rising atmospheric CO2 concentrations on plant disease have received increasing attention, but with little consensus emerging on the direct mechanisms by which CO2 shapes plant immunity. Furthermore, the impact of sub-ambient CO2 concentrations, which plants have experienced repeatedly over the past 800 000 yr, has been largely overlooked. A combination of gene expression analysis, phenotypic characterisation of mutants and mass spectrometry-based metabolic profiling was used to determine development-independent effects of sub-ambient CO2 (saCO2 ) and elevated CO2 (eCO2 ) on Ara

Abscisic Acid-Cytokinin Antagonism Modulates Resistance Against Pseudomonas syringae in Tobacco.

Phytohormones are known as essential regulators of plant defenses, with ethylene, jasmonic acid, and salicylic acid as the central immunity backbone, while other phytohormones have been demonstrated to interact with this. Only recently, a function of the classic phytohormone cytokinin in plant immunity has been described in Arabidopsis, rice, and tobacco. Although interactions of cytokinins with salicylic acid and auxin have been indicated, the complete network of cytokinin interactions with other immunity-relevant phytohormones is not yet understood. Therefore, we studied the interaction of k

Jasmonate signaling in the field, part II: insect-guided characterization of genetic variations in jasmonate-dependent defenses of transgenic and natural Nicotiana attenuata populations.

The introduction of genetically modified plants into natural habitats represents a valuable means to determine organismic level functions of a gene and its effects on a plant's interaction with other organisms. Nicotiana attenuata, a wild tobacco species native of the southwestern USA that grows in the immediate postfire environment, is one of the important host plants for herbivore populations recolonizing recently burned habitats in the Great Basin Desert. Here, we provide detailed guidelines for the analysis, under field conditions, of jasmonate-dependent defense and its impact on the plant

Wounding systemically activates a mitogen-activated protein kinase in forage and turf grasses.

Forage and turf grasses are continually cut and grazed by livestock, however very little is known concerning the perception or molecular responses to wounding. Mechanical wounding rapidly activated a 46 kDa and a 44 kDa mitogen-activated protein kinase (MAPK) in six different grass species. In the model grass species Lolium temulentum, the 46 kDa MAPK was rapidly activated within 5 min of wounding both locally and systemically in an adjacent unwounded tiller. This indicates that wounding generates a rapidly propagated long-distance signal that activates a MAPK in the distal portions of the pla

Targeting pandemic influenza: a primer on influenza antivirals and drug resistance.

The emergence of the 2009 H1N1 pandemic influenza A virus, as well as constant antigenic drift of seasonal influenza, underscores the remarkable versatility of this virus in adapting to the human population. While vaccines are the principal public health defence against influenza, rapid vaccine development can be a daunting task. Antiviral drugs offer the promise of inhibiting influenza regardless of its genetic variations. However, the rapid rise of resistance to several antivirals has highlighted the need for developing novel therapeutics with reduced drug resistance potential. In this revie

[Current anti-influenza virus chemotherapy].

The environment surrounding influenza is changing in recent years. In the spring of 2009, pandemic (H1N1) 2009 occurred in Mexico, and became epidemic on a global scale thereafter. Therefore, control of influenza is very important all over the world. Now in Japan, four specific anti-influenza antiviral drugs are available: amantadine, oseltamivir, zanamivir and peramivir. Pandemic (H1N1) 2009 are amantadine-resistant viruses, thus is not recommended for use. Oseltamivir is most commonly used in Japan, however, we have to pay attention to oseltamivir-resistant influenza virus. Almost no zanamiv

Jasmonates and its mimics differentially elicit systemic defence responses in Nicotiana attenuata.

Coronalon (6-ethyl indanoyl isoleucine), a synthetic jasmonate mimic, is known to regulate levels of transcripts and secondary metabolites that are commonly elicited by methyl jasmonate (MeJA) in a variety of plants. The ability of coronalon and its derivative (In-L-Ile-Me) to elicit MeJA-activated transcriptional and defence responses [nicotine and trypsin proteinase inhibitors (TPIs)] was compared in treated and systemic untreated tissues of wild-type (WT) and NaLOX3-silenced Nicotiana attenuata plants which are unable to activate either local or systemic defence responses. Coronalon and its

The effects of acamprosate and neramexane on cue-induced reinstatement of ethanol-seeking behavior in rat.

This study examines, for the first time, the effects of acamprosate and the non-competitive N-methyl-D-aspartate (NMDA) receptor antagonist neramexane on ethanol-seeking induced by alcohol-related environmental stimuli in an animal model of relapse. Wistar rats were trained to operantly self-administer ethanol (10% w/v) or water on a fixed-ratio 1 schedule in a 30-min daily session. Ethanol availability was signaled by an olfactory discriminative stimulus of orange extract (S+). In addition, each lever press was accompanied by a 5-s illumination of the operant chamber's house light (CS+). Wate

Contact allergens modulate the expression of MHC class II molecules on murine epidermal Langerhans cells by endocytotic mechanisms.

MHC class II molecules play an important role during the sensitization phase of allergic contact dermatitis. To study the influence of contact allergens on the expression of these molecules by murine epidermal Langerhans cells (LC), we performed a flow-cytofluorometric analysis of the Ia-antigen expression after in vivo application of contact allergens. A distinct decrease in the Ia-antigen expression of the entire LC population was noticed 3 h after in vivo application of the contact allergen 2,4-dinitrofluorobenzene (DNFB). This decrease was transient and balanced 24 h after in vivo applicat

Machine learning-guided multi-omics suggests iron-dependent hormonal signaling drives root morphological plasticity in wheat under temperature stress.

Root plasticity is crucial for crop survival under climate change. However, the coordinated regulatory network between metabolic disturbances and hormonal signaling that drives morphological adaptation under temperature fluctuations remains unclear. This study integrated machine learning-driven multi-omics analysis, in situ histochemical localization, and pharmacological validation to decipher the root adaptation strategies of wheat under temperature gradients. Wheat roots exhibited convergent morphological plasticity under temperature stress. However, this convergence was associated with dist

Temporal transcriptional networks coordinate ABA-GA-JA crosstalk during cold-induced modulation of seed dormancy and germination in wheat (Triticum aestivum L.).

Using a combination of genome-wide transcriptome, targeted hormonome and pharmacological approaches, this study showed that temporally resolved transcriptional regulatory networks integrating hormonal crosstalk underlie cold-induced modulation of seed dormancy and germination in wheat. Gene homoeologues (GHs) differentially expressed between cold-treated and untreated dormant seeds imbibed at optimal temperature (OT, 22°C) as well as between the same seed samples imbibed in cold and at OT represent distinct biological processes (BPs), indicating the role of coordinated activation/repression sp

Nitric Oxide and Phytohormone Crosstalk Mediates Soybean Growth and Defence Against Aphis glycines.

Aphid infestation, particularly by Aphis glycines, poses a major threat to soybean (Glycine max) production, necessitating sustainable defence strategies. This study evaluated the role of nitric oxide (NO) donor S-nitrosocysteine (CySNO) in enhancing soybean resistance to aphids through hormonal crosstalk. A dose optimisation assay revealed that 0.1 mM CySNO significantly improved seed germination, shoot and root growth, while higher concentrations inhibited development. Greenhouse trials were then conducted by applying 0.1 mM CySNO directly to the roots, followed by foliar applications of sal

HIV proviral transcription and infectivity are enhanced by neddylation.

A spreading infection can quickly restart from the persistent reservoir of cells harboring HIV proviruses if antiretroviral therapy (ART) is stopped. HIV transcription can also be increased by latency-reversing agents (LRAs) during ART. Neddylation is a post-translational modification that activates Cullin-RING ligases (CRLs), which ubiquitinate several proteins that regulate HIV transcription and infectivity, marking these regulators for proteasomal degradation. We studied how inhibiting neddylation affects HIV gene expression ex vivo after three LRAs: TNFα, PMA and ionomycin, and JQ1. In pro

Novel hexahydrocyclopentapyrrol-indolemethanimine as highly potent α-glucosidase inhibitors; design, biological evaluation, and computational insights.

A series of novel hexahydrocyclopentapyrrol-indolemethanimine derivatives were synthesized and evaluated for their α-glucosidase inhibitory activity. All compounds demonstrated promising inhibition with IC50 values in the range of 1.05 to 95.60 μM, which compounds 9i, 9l, and 9o demonstrated the best potential anti-α-glucosidase activity (IC50 of 1.05, 14.45, and 17.13 μM, respectively), surpassing the efficacy of the positive control, acarbose (IC50 = 750.0 ± 2.0 μM). The kinetic studies showed that the most potent derivative (compound 9i) inhibits competitively, which suggests its direct int

Multi-omics reveals DEAD-box RNA helicase 20 as key protein enhancing wilt resistance in transgenic chickpea.

AtNPR1 expression strengthens Fusarium wilt resistance in chickpea by activating SAR. Multi-omics analyses suggest CaDEAD-box20 as a candidate gene contributing to resistance through possible interaction with AtNPR1. Traditional breeding for broad-spectrum disease resistance in crops is often slow and resource-intensive, whereas genetic engineering provides a more precise and efficient alternative. To enhance resistance in chickpea (Cicer arietinum) against Fusarium oxysporum f. sp. ciceris, the causal agent of Fusarium wilt, we introduced the Arabidopsis NPR1 (AtNPR1) gene to activate systemi
