The role of dendritic cells in the development of chronic tonsillar disease in children.

Thirty-one children, 2.6-12.2 years (mean = 7.9 +/- 5.0) were divided into four clinical categories: normal controls (n = 7), recurrent tonsillitis (n = 8), idiopathic tonsillar hyperplasia (n = 9) and recurrent tonsillitis with hyperplasia (n = 7). Immunohistopathologic studies were performed to determine the presence and location of HLA-DR, S-100 positive antigen presenting dendritic cells (DCs) in six microanatomic compartments: surface and crypt epithelium and submucosae, extrafollicular areas and lymphoid follicles. The results were analyzed in the context of the clinical history, bacteri

Assessment of epidermal dendritic cell markers and T-lymphocytes in psoriasis.

Epidermal dendritic and T-cell counts have been performed in lesional and non-lesional skin from 35 psoriatic patients. The aims were to investigate absolute changes and interrelationships between these cellular elements in psoriasis and to explain apparent discrepancies between these results and reports in the literature. In non-lesional skin, the most frequently expressed dendritic cell marker was CD1a. HLA-DR+ and alpha-mannosidase+ dendritic cells were approximately 50 per cent and S100+ cells were 25 per cent as frequent. T-lymphocytes were rare, CD4+ cells predominating. In lesional psor

Identification of macrophages and dendritic cells in the osteopetrotic (op/op) mouse.

We used a panel of monoclonal antibodies and immunocytochemistry to identify macrophages and dendritic cells in mice that are deficient in macrophage colony stimulating factor (M-CSF or CSF-1) because of the recessive osteopetrotic (op/op) mutation. Prior work had shown that osteopetrosis is associated with a lack of osteoclasts, phagocytic cells required for remodelling in bone. Additional macrophage populations proved to be very M-CSF dependent. op/op mice had few and sometimes no peritoneal cavity phagocytes, splenic marginal zone metallophils, and lymph node subcapsular sinus macrophages.

The distribution and functional properties of dendritic cells in patients with seronegative arthritis.

Dendritic cells (DC), potent antigen-presenting cells, are known to be increased in numbers in inflammatory lesions in rheumatoid arthritis and juvenile chronic arthritis. In this study, patients with seronegative arthritis were studied; the distribution and functional properties of DC enriched low density cells (LDC) from peripheral blood (PB) and synovial fluid (SF) were compared. The composition of LDC from both sources was similar, comprising approximately 30% DC, 60% monocytes with few T lymphocytes. SF was significantly enriched for LDC compared with paired peripheral blood (P less than

[Immunohistochemical characterization of S100 proteinpositive dendritic cells in gastric mucosa].

Immunohistochemical staining was performed by the ABC method using antibody against S100 protein for the identification of dendritic cells (DC) in the gastric mucosa, including gastric ulcer (n = 19), chronic atrophic gastritis (n = 28), gastric carcinoma (n = 65), fetal gastric mucosa (n = 11) and normal controls (n = 15). Lower numbers of DC may indicate an antigen-deficient state in normal and fetal gastric mucosa, while the diverse densities of DC in pathological conditions may reflect the differences in antigenic quantities and qualities and in individual immune responses. The close assoc

Activity-dependent accumulation of calcium in Purkinje cell dendritic spines.

The calcium content of synapses of parallel fibers on Purkinje cell dendritic spines was determined by electron probe x-ray microanalysis of freeze-dried cryosections from directly frozen slices of mouse cerebellar cortex. In fresh slices frozen within 20-30 sec of excision, calcium concentrations ranging from 0.8 to 18.6 mmol/kg of dry weight were measured in cisterns of smooth endoplasmic reticulum within Purkinje cell dendritic spines. The average calcium content of spine cisterns in rapidly excised slices (6.7 +/- 0.6 mmol/kg of dry weight +/- SEM) was higher than the average calcium conte

Dendritic cells in cutaneous, lymph node and pulmonary lesions of sarcoidosis.

To determine whether dendritic cells (DC) are a consistent feature of lesions of sarcoidosis, we have used monoclonal antibodies to identify the HLA-DR-expressing populations of cells in cryostat sections of 15 lymph node, pulmonary and cutaneous lesions. The commonest HLA-DR positive cells in granulomas were epithelioid and giant cells, although lymphocytes within granulomas and tissue macrophages around them were also positive. Dendritic cells with Langerhans cells (NA1/34+ = OKT6+) and interdigitating cell (RFD1+) phenotype were consistently associated with granulomas only in skin lesions.

S-100-protein-positive dendritic cells in nodular vasculitis.

We investigated the distribution of S-100-protein-positive dendritic cells in 20 cases of nodular vasculitis (erythema induratum Bazin). The cells were identified by immunohistochemical demonstration in paraffin sections. S-100-protein-positive dendritic cells were present in all cases and comprised up to 11% of the infiltrating cells in lymphohistiocytic areas (mean 3.7%). Because of the dendritic shape and the association with lymphocytes the cells are considered to represent T-zone histiocytes. Our findings suggest a possible role of delayed-type hypersensitivity in nodular vasculitis.

Human synovial dendritic cells. Direct observation of transition to fibroblasts.

A subpopulation of human synovial cells develop a stellate morphology during in vitro culture. These cells, called stellate cells or dendritic cells, appear capable of collagenase production. In other tissues, particularly mouse spleen and human peripheral blood, cells with a similar shape express immune region associated (Ia) antigens, stimulate a primary mixed leukocyte reaction, and present antigen to lymphocytes. To characterize synovial dendritic cells, we observed changes in their morphology by time-lapse videomicroscopy. Parallel cultures of cells from the same specimens were observed p

Wafer-Scale Dendritic Nanoporous Platinum Films for Neural Electrodes.

Two-dimensional (2D) metallic nanosheets are promising for neural implants, but their application is hindered by limited lateral dimensions, poor mechanical compliance, and insufficient electrochemical stability. Here, we report a wafer-scale wet-chemical synthesis of an ultrathin, intrinsically porous dendritic platinum (Pt) film (den-2DPtwaf) that can be transferred onto flexible substrates for robust neural interfacing. Confined Pt growth on a layered double hydroxide (LDH)-modified Si wafer produces a wafer-sized, 6-nm-thick film that is vertically conductive but laterally insulating. As a

Dendritic interaction of timescales in afterdepolarization potentials and nonmonotonic spike-adding.

Depolarizations that occur after action potentials, known as afterdepolarization potentials or ADPs, are important for neuronal excitability … ief step current and AMPA current input. Additionally, dendritic size and passive properties of the dendrites were found to be key modulators of ADP amplitude. However, increasing magnitudes of NMDA current input resulted in nonmonotonic spike-adding in a manner dependent on dendritic Ca2+ influx and Ca2+ activated K+ currents, which was found to arise from …

Dysfunctional synaptic competition at dendritic spines in Fragile X syndrome.

Dendritic spines are highly dynamic structures whose morphology and lifespan are modified in response to synaptic efficacy changes. Structural modifications following activity support the long-term encoding of information and could allow for the remodeling of neural circuits. Long-term depression (LTD) is a key mechanism for synaptic weight regulation, yet its structural correlates - particularly for long-lasting, protein synthesis dependent forms - remain poorly understood. Furthermore, in humans, this type of plasticity is often disrupted in neurodevelopmental disorders, correlating with cog

Matrix Viscoelasticity Regulates Dendritic Cell Migration and Immune Priming.

The tumor microenvironment shapes immune surveillance through its mechanical properties, yet the role of matrix viscoelasticity remains unclear. Here, we used a tunable collagen system that models human tissue viscoelasticity to define how matrix relaxation directs dendritic cell (DC) behavior. Slow-relaxing, elastic networks restrict actomyosin-driven remodeling, limiting DC motility and reducing DC-T cell encounters and activation. Blocking DC migration in fast-relaxing matrices recapitulated key aspects of the impaired T cell priming seen in elastic networks, identifying migration as a mech

CD5+ dendritic cell robots mediated in situ immunocyte activation.

Immune cell microbots with self-propelling and navigating capabilities have become an exciting field of research. Isolation, activation, and adoptive transfer of cytotoxic immune cells in vitro are some of the most common technical methods. However, the ex situ activation and adoptive transfer of cytotoxic immune cells carry the risk of systemic inflammatory responses and functional exhaustion. To address this bottleneck, we report a CD5+ dendritic cell microbot (CD5+DC robot) constructed by engineering natural CD5+ dendritic cells to phagocytose magnetic nanoparticles coated with tumor cell m
