Atypical hemolytic uremic syndrome (aHUS): making the diagnosis.

Atypical hemolytic uremic syndrome (aHUS) is a major thrombotic microangiopathy (TMA). A TMA is recognized by the laboratory signs of microangiopathic hemolysis, as indicated by schistocytes, elevated lactate dehydrogenase, low haptoglobin, and low hemoglobin, plus thrombocytopenia and accompanying signs and symptoms of organ system involvement. aHUS results from chronic, uncontrolled activity of the alternative complement pathway. In most patients, this defect is related to a genetic deficiency in one or more soluble and/or membrane-bound complement regulatory proteins. Complement factor H is

ADAMTS13--more than just TMA and TTP.

ADAMTS13 (a disintegrin-like and metalloprotease with thrombospondin type-1 motifs 13) has been shown to be of major pathophysiological importance for thrombotic microangiopathy (TMA) in the setting of thrombocytic thrombocytopenic purpura (TTP) when either lacking (inherited TTP) or if antibodies against ADAMTS13 are present (acquired TTP). A potential pathophysiological role of ADAMTS13 has also been postulated in other diseases i.e. myocardial infarction, atrial fibrillation and diabetic angiopathy. Recent data provides evidence, however, for a completely different role of ADAMTS13 in vascu

Influence of ADAM28 on biological characteristics of human dental follicle cells.

OBJECTIVES: The aim of this study was to investigate the effects of a disintegrin and metalloproteinase 28 (ADAM28) on the biological characteristics of human dental follicle cells (HDFCs) and possible action mechanism. METHODS: Eukaryotic expression plasmid containing ADAM28 coding region and ADAM28 antisense oligodeoxynucleotides (AS-ODN) with FITC labelling were constructed and synthesised by gene clone and recombination. Then we respectively transfected them into HDFCs by Lipofectamine 2000 system and detected their effects on proliferation, differentiation and apoptosis of HDFCs by MTT a

Bothrops insularis venomics: a proteomic analysis supported by transcriptomic-generated sequence data.

A joint transcriptomic and proteomic approach employing two-dimensional electrophoresis, liquid chromatography and mass spectrometry was carried out to identify peptides and proteins expressed by the venom gland of the snake Bothrops insularis, an endemic species of Queimada Grande Island, Brazil. Four protein families were mainly represented in processed spots, namely metalloproteinase, serine proteinase, phospholipase A(2) and lectin. Other represented families were growth factors, the developmental protein G10, a disintegrin and putative novel bradykinin-potentiating peptides. The enzymes w

A Novel KGD-Based αIIbβ3 Antagonist Prevents Arterial Thrombosis While Preserving Hemostasis and Avoiding Thrombocytopenia.

Current αIIbβ3 antagonists are potent antithrombotic agents, their clinical use is limited by the risk of life-threatening bleeding. Emerging evidence has highlighted key mechanistic differences between thrombosis and hemostasis, opening avenues for safer antithrombotic strategies. Targeting integrin αIIbβ3 outside-in signaling has been proposed to mitigate bleeding risk; however, the short half-life of peptide-based therapeutics remains a major challenge. In this study, we developed an optimized αIIbβ3 antagonist, KGDRR-a recombinant mutant protein derived from snake venom disintegrin, incorp

ADAM17 as a proteolytic rheostat linking inflammation, tissue repair, and failure of inflammatory resolution in liver injury and fibrosis.

A disintegrin and metalloproteinase 17 (ADAM17) is a membrane-bound sheddase that regulates inflammatory signaling, tissue repair, and fibrotic remodeling by converting cell-surface proteins into soluble forms. Rather than viewing ADAM17 simply as a proinflammatory enzyme, this review frames it as a proteolytic rheostat that dynamically adjusts the balance between membrane-bound and soluble molecular states in the injured liver. Within this framework, apparently opposing effects of ADAM17 can be understood as substrate-, cell-, and disease-stage-dependent consequences of ectodomain shedding. W

Blood biomarkers of frailty and cognition: A scoping review.

Frailty increasingly is recognized as a factor that modifies the relationship between disease biomarkers, including neuropathology, and dementia expression. The mechanisms underlying the relationship between frailty and dementia remain unclear, but blood biomarkers can offer insight into these mechanisms. We completed a scoping review of research examining the associations between blood biomarkers, frailty, and cognition. Three online databases were searched to identify original research examining blood biomarkers in the context of frailty and/or cognitive decline that accounted for the other

Involvement of ADAM12 in TGF-β1-Induced Proliferation of Rheumatoid Arthritis Synovial Fibroblasts.

A disintegrin and metalloproteinase 12 (ADAM12) is known to be involved in chondrocyte proliferation and is upregulated in the synovial tissue of osteoarthritis (OA). However, the underlying mechanisms of ADAM12 on rheumatoid arthritis (RA) synovial cell proliferation remain unknown. Here, we investigated the role of ADAM12 in the proliferation of RA synovial fibroblasts (RASFs). The expression and localization of ADAM12 in RA synovial tissues were examined by immunohistochemistry and compared with OA and healthy control (HC) synovial tissues. The effect of inflammatory cytokines (TNF-α, TGF-β

Unbiased plasma proteomics reveals hemolysis during ADAMTS13 relapse in patients with immune TTP.

Thrombotic thrombocytopenic purpura (TTP) is a thrombotic microangiopathy that is caused by a deficiency of a disintegrin and metalloprotease with thrombospondin type 1 repeats, member 13 (ADAMTS13). Most patients with TTP have immune-mediated TTP (iTTP), for which the loss of activity results from the development of autoantibodies directed toward ADAMTS13. Preemptive treatment of iTTP with rituximab when ADAMTS13 activity drops to ∼20% (ie, an "ADAMTS13 relapse") can effectively prevent clinical relapse. Nevertheless, patients still undergo prolonged periods of low ADAMTS13 activity. We used

Contributions of ADAM12 via HB-EGF/EGFR signaling cascades to EMT and cancer progression in pancreas.

Pancreatic cancer (PC) is among the most aggressive malignancies, with a five-year survival rate of <7% in China. The substantial stromal component and activation of epithelial-mesenchymal transition (EMT) in PC contribute to drug resistance and poor outcomes. Between January 2017 and December 2020, 62 pancreatic specimens were obtained via surgical resection from two hospitals. In alignment with the GEPIA database, the expression levels of a disintegrin and metalloproteinase 12 (ADAM12) and heparin-binding epidermal growth factor (HB-EGF) were significantly elevated in 43 PC tissues compared

Involvement of ADAM17-Klotho Crosstalk in High Glucose-Induced Alterations of Podocyte Function.

Microalbuminuria is the earliest clinical abnormality in diabetic kidney disease. High glucose (HG) concentrations are associated with the induction of oxidative stress in podocytes, leading to disruption of the glomerular filtration barrier. Our recent study revealed a significant decrease in the membrane-bound fraction of Klotho in podocytes that were cultured under HG conditions. Given that disintegrin and metalloproteinase 17 (ADAM17) is responsible for the shedding of Klotho from the cell membrane, the present study investigated the impact of HG on the interplay between ADAM17 and Klotho

ADAM17 knockdown is more effective than AT1R blocker in alleviating diabetic cardiomyopathy through downregulating the RhoA-ROCK1 pathway.

Diabetic cardiomyopathy (DCM) is a serious complication of diabetes that contributes to increased mortality. A-disintegrin and metalloproteinase 17 (ADAM17), a novel member of the renin-angiotensin-aldosterone system (RAAS), has been shown to exert a protective effect against cardiac fibrosis, but the exact molecular mechanisms remain unclear. This study aims to elucidate the role of ADAM17 in the pathogenesis of DCM and its interaction with the RhoA/ROCK1 signaling pathway, focusing on cardiac fibroblast-to-myofibroblast transition and cardiac remodeling. AAV9-mediated gene silencing was used

Systemic Lupus Erythematosus-Associated Thrombotic Thrombocytopenic Purpura: A Case Report.

Systemic lupus erythematosus-associated thrombotic thrombocytopenic purpura (SLE-TTP) is a rare but life-threatening condition that requires prompt recognition and treatment. We report a case of a patient with systemic lupus erythematosus (SLE) who presented with encephalopathy and was subsequently diagnosed with thrombotic thrombocytopenic purpura (TTP) based on ADAMTS13 (a disintegrin and metalloproteinase with thrombospondin motifs 13) deficiency. The patient was successfully treated with plasmapheresis. A 42-year-old woman with a history of SLE presented with febrile encephalopathy and was

The protease ADAMTS5 controls ovarian cancer cell invasion, downstream of Rab25.

Ovarian cancer is the 3rd most common gynaecological malignancy worldwide, with a 5-year survival rate of < 30% in the presence of metastasis. Metastatic progression is characterised by extensive remodelling of the extracellular matrix, primarily mediated by secreted proteases, including members of the 'a disintegrin and metalloprotease with thrombospondin motif' (ADAMTS) family. In particular, ADAMTS5 has been reported to be upregulated in ovarian malignant tumours compared to borderline and benign lesions, suggesting it might play a role in metastatic progression. Furthermore, it has been su

ADAMTS2: More than a procollagen N-proteinase.

A disintegrin and metalloproteinase with thrombospondin motifs 2 (ADAMTS2) is a member of the ADAMTS zinc metalloproteinase family, best known for its role as a procollagen I N-proteinase in the maturation of fibrillar collagens. Biallelic defects in the ADAMTS2 gene, resulting in a loss of ADAMTS2 enzyme activity and consequent retention of N-propeptides in type I procollagen molecules, lead to the rare monogenic disease Ehlers-Danlos syndrome dermatosparaxis type (dEDS) in humans, and dermatosparaxis in animals, conditions that are hallmarked by extreme fragility of the skin and other soft c

Sesame oil downregulates the expression of ADAMTS-4 in high-fat diet-induced atherosclerosis.

Atherosclerosis is a chronic inflammatory disease forming plaques in medium and large-sized arteries. ADAMTS-4 (a disintegrin and metalloproteinase with thrombospondin motifs-4) is an extracellular-matrix remodelling enzyme involved in the degradation of versican in the arterial wall. Recent reports indicated that increased expression of ADAMTS-4 is associated with plaque progression and vulnerability. Bioactive components of dietary oil, like sesame oil, are reported to have anti-inflammatory and antioxidant properties. Here, we studied the effect of sesame oil on regulating ADAMTS-4 in high-

Association Between Plasma ADAMTS-7 Levels and Diastolic Dysfunction in Patients with Type 2 Diabetes Mellitus.

A disintegrin and metalloproteinase with thrombospondin motifs-7 (ADAMTS-7) belongs to the family of metalloproteinases that contributes to tissue homeostasis during morphogenesis and reproduction. These metalloproteinases regulate various cell functions such as cell proliferation, are important regulators in tissue regeneration, and play a role in vascular remodelling, which is involved in atherosclerosis development. Despite the well-established association between ADAMTS-7 and atherosclerotic disease, data regarding the metalloproteinase's association with LV function remain scarce. The aim

ADAM9 promotes type I interferon-mediated innate immunity during encephalomyocarditis virus infection.

Viral myocarditis, an inflammatory disease of the heart, causes significant morbidity and mortality. Type I interferon (IFN)-mediated antiviral responses protect against myocarditis, but the mechanisms are poorly understood. We previously identified A Disintegrin And Metalloproteinase domain 9 (ADAM9) as an important factor in viral pathogenesis. ADAM9 is implicated in a range of human diseases, including inflammatory diseases; however, its role in viral infection is unknown. Here, we demonstrate that mice lacking ADAM9 are more susceptible to encephalomyocarditis virus (EMCV)-induced death an

An ADAM10 Exosite Inhibitor Is Efficacious in an In Vivo Collagen-Induced Arthritis Model.

Rheumatoid arthritis is a systemic autoimmune inflammatory disease that affects millions of people worldwide. There are multiple disease-modifying anti-rheumatic drugs available; however, many patients do not respond to any treatment. A disintegrin and metalloproteinase 10 has been suggested as a potential new target for RA due to its role in the release of multiple pro- and anti-inflammatory factors from cell surfaces. In the present study, we determined the pharmacokinetic parameters and in vivo efficacy of a compound CID3117694 from a novel class of non-zinc-binding inhibitors. Oral bioavai
