Reprogramming Disulfide Reduction in Endoplasmic Reticulum Uncouples Immune Evasion of Pancreatic Cancer.

The modulation of immune checkpoint activity exerts profound impacts on tumor immunotherapy. However, the interfere of mature immune checkpoints encounter efficacy challenges in solid tumors, which underlies a critical barrier in clinical translation of multiple potential targets. Herein, we propose a biochemical immune modulation strategy for immunosuppression reversal by blocking the mature of CD47 within the endoplasmic reticulum (ER). Clinically used radionuclides iodine-131 (131I) is reengineered to encapsulate within microporous barium titanate nanoparticles, forming an immunoactive nano

A disulfide bond sculpts the CTNIP4 phytocytokine fold for recognition by the receptor kinase HSL3.

Precise ligand recognition by closely related leucine-rich repeat receptor kinases (LRR-RKs) is essential for plants to coordinate immunity, development and environmental adaptation. Here we show how the LRR-RK HSL3/NUT specifically recognizes the folded, disulfide-stabilized CTNIP4/SCREW2 phytocytokine in Arabidopsis. Quantitative binding assays define a minimal CTNIP4 region required for high-affinity HSL3 interaction and signalling activation. A 2.12-Å crystal structure of the HSL3-CTNIP4 complex reveals a unique C-terminal receptor pocket that accommodates the peptide's cyclic architecture

Comparison Between Ultrasonic and Thermal Exfoliation for Molybdenum Disulfide Nanoparticles-A Multifunctional Study.

The transition metal dichalcogenides (MX2, TMD) are a family of two-dimensional compounds, and MoS2 is perhaps the most representative material among them. MoS2, in its bulk form, is semiconductive, with an indirect band gap of around 1.2-1.3 eV. Monolayer MoS2, nevertheless, exhibits a direct band gap of 1.8-1.9 eV. For zero-dimensional MoS2 (quantum dots), the band gap would be even wider. These changes are mainly due to quantized energy levels arising from geometric confinement. Quantum effects directly yield distinct optoelectronic properties, such as photoluminescence, which are absent in

Influence of the intramolecular disulfide Cys46-Cys55 bridge on the interaction of human neuroglobin with SDS.

Human neuroglobin (hNgb) is a globin involved in the protection of neurons and retinal cells which features an intramolecular disulfide bond between Cys46 and Cys55 under oxidative conditions. Here, conformational changes and oxidative degradation of hNgb wt and its C46AC55A mutant, lacking the disulfide bridge, were investigated in the presence of sodium dodecyl sulfate (SDS) by electronic absorption spectroscopy, intrinsic fluorescence emission and circular dichroism. Both proteins are found to undergo multiple SDS-induced conformational changes resulting in the formation of two non-native h

Treatment with the Antimicrobial Product Diallyl Disulfide Is Associated with Major Changes to Soil Microbiota.

Natural products derived from Allium spp., such as garlic oil, garlic powder, and diallyl disulfide (DADS), are strong elicitors of sclerotia germination in the fungus Sclerotium cepivorum (syn. Stromatinia cepivora), the causal agent of Allium white rot. However, these compounds can also have broad antimicrobial activity against a wide range of bacteria, oomycetes, and other fungi when they are applied to soil. The objective of this study was to determine the potential impacts that DADS application has on soil microbial communities. DADS was applied to two soil types and incubated under aerob

Assessment of Thiol/Disulfide Homeostasis and Its Association With Clinical Manifestations in Behcet's Disease.

Behcet's disease (BD) is a multisystemic inflammatory vasculitis characterized by recurrent episodes of exacerbation and remission, with unpredictable duration and system involvement in each episode. Although previous studies have shown the role of oxidative stress in the pathogenesis of BD, the relationship between the clinical features of Behcet's disease and oxidative stress has not been fully studied to date. This study aims to assess the parameters of dynamic thiol disulfide homeostasis (DTDH) and their relationship with different clinical characteristics of disease in BD patients. Fifty

Characterization of three novel dimethyl disulfide degrading bacteria and their potential degradation pathways.

Dimethyl disulfide (DMDS) is an odor compound characterized by the lowest olfactory threshold and high toxicity. It is indispensable to explore the bacteria with high resistance and degradation efficiency to DMDS. Acinetobacter lwoffii, Pseudomonas mendocina, and Myroides odoratus were isolated from kitchen waste. After 6 days of individual treatment, the removal rates were 34.22 %, 40.95 %, and 41.94 % respectively. The DMDS metabolic pathways based on metagenomic assays were discovered to be incomplete due to the insufficient annotation of some key genes in the current database. Following 3

Selenium Disulfide from Sustainable Resources: An Example of "Redneck" Chemistry with a Pinch of Salt.

Selenium disulfide (often referred to as SeS2) encompasses a family of mixed selenium-sulfide eight-membered rings, traditionally used as an anti-dandruff agent in shampoos. SeS2 can be produced by reacting hydrogen sulfide (H2S) with selenite (SeO32-) under acidic conditions. This chemistry is also possible with natural spring waters that are rich in H2S, thus providing an avenue for the more sustainable, green production of high-quality SeS2 particles from an abundant natural source. The orange material obtained this way consists of small globules with a diameter in the range of 1.1 to 1.2 µ

Assignment of Disulfide Bonds in HNTX-XXI by Double-Enzymatic Digestion and Edman Degradation.

HNTX-XXI, a peptide toxin derived from the venom of the spider Ornithoctonus hainana, comprises a 64-amino-acid protein architecture that notably incorporates eight cysteine residues positioned at positions 2, 10, 14, 16, 17, 23, 36, and 63. The close spatial proximity of Cys16 and Cys17 poses a challenge in resolving their disulfide bridge configurations using standard methodologies. In this study, we introduce an innovative and highly efficient approach for delineating disulfide pairings in peptides containing adjacent cysteines. Our methodology integrates a two-step proteolytic digestion st

N-Heterocyclic Carbene-Derived Carbon Disulfide Radical Ligands for Palladium Diradicals.

N-heterocyclic carbenes (NHCs) are recognized for their ability to stabilize various main group radicals; however, NHC-derived, sulfur-based radicals remain rare. In this study, we successfully synthesized and characterized a series of palladium diradical complexes that featured new sulfur-based radical ligands from NHC-carbon disulfide adducts. Spectroscopic and computational characterizations of the palladium complexes confirmed the open-shell singlet ground state, which resulted from the antiferromagnetic coupling of two unpaired electrons on each ligand. Proton nuclear magnetic resonance r

Glutathione depletion results in S-nitrosylation of protein disulfide isomerase in neuroblastoma cells.

AIMS: Protein disulfide isomerase (PDI) is an essential enzyme involved in oxidative protein folding. PDI is S-nitrosylated in the brains of Alzheimer's disease patients, and S-nitrosylated PDI is considered one of main causes of Alzheimer's disease. However, the mechanisms underlying PDI S-nitrosylation have not yet been elucidated. Because glutathione (GSH) depletion is a pathological feature of Alzheimer's disease, we investigated the effect of GSH depletion on the S-nitrosylation level of PDI. MAIN METHODS: SH-SY5Y cells, which is a human derived neuroblastoma cells, were used in this stu

Cyclin D-CDK4 Disulfide Bond Attenuates Pulmonary Vascular Cell Proliferation.

BACKGROUND: Pulmonary hypertension (PH) is a chronic vascular disease characterized, among other abnormalities, by hyperproliferative smooth muscle cells and a perturbed cellular redox and metabolic balance. Oxidants induce cell cycle arrest to halt proliferation; however, little is known about the redox-regulated effector proteins that mediate these processes. Here, we report a novel kinase-inhibitory disulfide bond in cyclin D-CDK4 (cyclin-dependent kinase 4) and investigate its role in cell proliferation and PH. METHODS: Oxidative modifications of cyclin D-CDK4 were detected in human pulmo

Thiol/disulfide homeostasis as a new oxidative stress marker in patients with Fabry disease.

This is the first study to show both dynamic thiol-disulfide balance and oxidative stress levels in patients with Fabry disease (FD). This prospective study consists of 30 FD patients and 30 healthy controls. Thiol and disulfide values of the study groups were evaluated using a new, cost-effective and fully automatic colorimetric method. A total of 60 subjects were included in the study. A statistically significant difference was found between the patient and control groups for native and total thiol levels (p < 0.001). In addition, disulfide levels were significantly higher in FD patients com

Excess copper catalyzes protein disulfide bond formation in the bacterial periplasm but not in the cytoplasm.

Copper avidly binds thiols and is redox active, and it follows that one element of copper toxicity may be the generation of undesirable disulfide bonds in proteins. In the present study, copper oxidized the model thiol N-acetylcysteine in vitro. Alkaline phosphatase (AP) requires disulfide bonds for activity, and copper activated reduced AP both in vitro and when it was expressed in the periplasm of mutants lacking their native disulfide-generating system. However, AP was not activated when it was expressed in the cytoplasm of copper-overloaded cells. Similarly, this copper stress failed to ac

Protein disulfide isomerase modulation of TRPV1 controls heat hyperalgesia in chronic pain.

Protein disulfide isomerase (PDI) plays a key role in maintaining cellular homeostasis by mediating protein folding via catalyzing disulfide bond formation, breakage, and rearrangement in the endoplasmic reticulum. Increasing evidence suggests that PDI can be a potential treatment target for several diseases. However, the function of PDI in the peripheral sensory nervous system is unclear. Here we report the expression and secretion of PDI from primary sensory neurons is upregulated in inflammatory and neuropathic pain models. Deletion of PDI in nociceptive DRG neurons results in a reduction i

The disulfide catalyst QSOX1 maintains the colon mucosal barrier by regulating Golgi glycosyltransferases.

Mucus is made of enormous mucin glycoproteins that polymerize by disulfide crosslinking in the Golgi apparatus. QSOX1 is a catalyst of disul … gut mucus glycome of the QSOX1 knockout mice, leading to the discovery that QSOX1 forms regulatory disulfides in Golgi glycosyltransferases. Sialylation defects in the colon are known to cause colitis in humans. Here we show that QSOX1 redox control of sialylation is essential for maintaining mucosal function.

0D/2D mixed-dimensional perovskite constructed by thiol- and disulfide-containing ligands.

Reduced-dimensional (RD) perovskites have shown attractive chemical and physical properties for optoelectronic applications. Incorporating large organic ligands enables infinite tunability in the components and structures. Theoretically, it is feasible to apply multiple types of organic ligands in a single RD crystal to achieve multiple-dimensional perovskites. However, the coexistence of different organic ligands commonly introduces competing crystal growths that inhibit the formation of a more complex crystal structure. Herein, we report a case of mixed-dimensional (MD) perovskite single cry

Concerted and sequential three-body fragmentation of deep-core-ionized carbon disulfide.

Momentum vector correlation is a powerful tool to study molecular dissociation. We have studied the three-body fragmentation of carbon disulfide after sulfur 1s photoionization by means of momentum imaging techniques. Concerted and sequential pathways are disentangled in three-body fragmentation using adapted analysis strategies. In particular, we introduce various data visualization schemes that are proved to be particularly efficient to determine dissociation dynamics.
