Commonly prescribed medicines antagonise anti-MRSA antibiotics and select for resistance.

Many commonly prescribed non-antibiotic medicines have off-target antimicrobial activity, yet their impact on antibiotic efficacy remains poorly understood. In this study, we investigated eight widely used UK prescription medicines and identified simvastatin, amlodipine and fluoxetine as growth inhibitory towards methicillin-resistant Staphylococcus aureus. These drugs disrupt bacterial membranes, with amlodipine and fluoxetine also triggering stress responses linked to cell wall and membrane damage. Further mechanistic analysis using transposon mutant screening revealed that simvastatin impai

The impact of treatment strategies on the epidemiological dynamics of plasmid-conferred antibiotic resistance.

The issue of antibiotic resistance is a critical concern for public health, prompting numerous investigations into the impact of treatment strategies on preventing or slowing down the emergence of resistance. While existing studies have predominantly focused on chromosomal resistance mutations, the consequences of often clinically more relevant plasmid-conferred resistance remain insufficiently explored. To address this gap, we conducted three extensive in vitro experiments utilizing a liquid-handling platform. These experiments evaluated the efficacy of five distinct treatment strategies usin

Transcontinental spread and evolution of Mycobacterium tuberculosis W148 European/Russian clade toward extensively drug resistant tuberculosis.

Transmission-driven multi-/extensively drug resistant (M/XDR) tuberculosis (TB) is the largest single contributor to human mortality due to antimicrobial resistance. A few major clades of the Mycobacterium tuberculosis complex belonging to lineage 2, responsible for high prevalence of MDR-TB in Eurasia, show outstanding transnational distributions. Here, we determined factors underlying the emergence and epidemic spread of the W148 clade by genome sequencing and Bayesian demogenetic analyses of 720 isolates from 23 countries. We dated a common ancestor around 1963 and identified two successive

The antibiotic resistance crisis, with a focus on the United States.

Beginning with the discovery of penicillin by Alexander Fleming in the late 1920s, antibiotics have revolutionized the field of medicine. They have saved millions of lives each year, alleviated pain and suffering, and have even been used prophylactically for the prevention of infectious diseases. However, we have now reached a crisis where many antibiotics are no longer effective against even the simplest infections. Such infections often result in an increased number of hospitalizations, more treatment failures and the persistence of drug-resistant pathogens. Of particular concern are organis

A hospital-based epidemiological study of the genes coding for carbapenemase production among carbapenem resistant Acinetobacter isolates in India.

BACKGROUND: Worldwide, Acinetobacter species is a multidrug-resistant (MDR) nosocomial pathogen capable of acquiring or upregulating resistant determinants thereby effecting drug efficacy. Resistance to Carbapenems causes high morbidity and mortality. The study aims to know prevalence of carbapenemase producing genes in Acinetobacter spp., and understand the local epidemiology of carbapenem-resistant Acinetobacter spp. in a tertiary care hospital in India. METHODS: Carbapenem-resistant Acinetobacter isolates from clinical samples were analysed to identify the resistance mechanism to carbapene

Microbe Profile: Candidozyma auris: an emergent and resilient yeast and new antifungal strategies.

Candidozyma auris is an emerging opportunistic yeast that is important because of its multidrug resistance, persistence in healthcare environments and ability to cause outbreaks. Since its discovery in 2009 in Japan, it has rapidly spread worldwide and is now recognized as a major global public health threat and was recognized by the World Health Organization (WHO) in 2022 as a critical priority fungal pathogen. Distinct phylogeographic clades demonstrate simultaneous emergence on different continents, suggesting ecological or environmental triggers. Clinical management is complicated by frequ

Giving Antibiotics a Second Chance: Evolutionary Trade-Offs and Phage-Driven Restoration of Antibiotic Susceptibility.

Antimicrobial resistance poses a critical and escalating threat to global public health, driven by the widespread and often unjustified use of antibiotics and the rapid dissemination of resistance determinants. With the antibiotic discovery pipeline largely depleted, alternative and complementary strategies are urgently needed to preserve the effectiveness of existing antimicrobials. Bacteriophages-viruses that specifically infect bacteria-have re-emerged as promising tools not only for direct bacterial eradication but also for reshaping bacterial evolutionary trajectories. This review examine

Convergent Architecture of the Acinetobacter baumannii Resistome: A Co-Occurrence Network Analysis of 20,739 Genomes Under the One Health Framework.

Acinetobacter baumannii is a critical priority ESKAPE pathogen whose resistome co-occurrence architecture, temporal dynamics, and One Health distribution remain poorly characterized. A total of 20,739 genomes (2000-2025; NCBI, PubMLST, BV-BRC) were annotated using CARD-RGI v6.0.5, AMRFinderPlus v4.2.7, and ResFinder v4.7.2. A co-occurrence network was constructed using Jaccard filtering (≥10%; >0.30) with Bayesian bootstrap and Louvain. Temporal trends were assessed by linear regression with Benjamini-Hochberg (2003-2024), and One Health compartments were assessed using balanced PERMANOVA (Bra

Collateral sensitivity and cross-resistance in six species of bacteria exposed to six classes of antibiotics.

De novo resistance can develop in bacteria as a result of exposure to sublethal concentrations of antibiotics. Once the strain has become resistant to an initial antibiotic, this can cause cross-resistance or collateral sensitivity to a second antimicrobial. Specific collateral sensitivity is rarely conserved across species because the mechanisms triggered in different microorganisms to resist antibiotics are often different. In this study, we explored which collateral sensitivity or cross-resistance networks are present in six species of bacteria with induced de novo resistance. These six spe

Pre- and postantibiotic epoch: The historical spread of antimicrobial resistance.

Plasmids are now the primary vectors of antimicrobial resistance, but our understanding of how human industrialization of antibiotics influenced their evolution is limited by a paucity of data predating the antibiotic era (PAE). By investigating plasmids from clinically relevant bacteria sampled and isolated between 1917 and 1954 and comparing them with modern plasmids, we have captured more than 100 years of evolution. We show that although virtually all PAE plasmids were devoid of resistance genes and most never acquired them, a minority evolved to drive the global spread of resistance to fi

Bathing seawater and sand as reservoirs of clinically relevant and antimicrobial resistant Klebsiella pneumoniae strains.

Klebsiella pneumoniae is included in the ESKAPE list of bacteria, which are currently considered the greatest threat to human health. It is widely distributed in the environment, including waters, soils, and plants. According to the One Health concept, it is essential to enhance our understanding of the distribution and genetic characteristics of this human pathogen in aquatic ecosystems, particularly in environments with frequent human contact, such as beaches and seawater used for recreational purposes. The aim of this study is to investigate whether bathing seawater and sand could serve as

Tuberculosis drug development; fluoroquinolone structural tailoring.

Tuberculosis (TB) is a contagious infectious disease caused by the bacillus Mycobacterium tuberculosis (Mtb). It is transmitted through small particles in the air (<5 µm) expelled by active tuberculosis patients; when inhaled by a new host, they can potentially cause infection. Nowadays, TB is still the major cause of morbidity and mortality by a single infectious agent, this is further exacerbated by the worldwide emergence of multidrug-resistant strains of Mtb. Thus, effective methods of diagnosis, prophylaxis, and new pharmacological therapies must be carried out in order to control this di

The role of bacterial extracellular vesicles in promoting antibiotic resistance.

The burgeoning proliferation of infections attributed to multidrug-resistant (MDR) bacterial pathogens is profoundly undermining conventional chemotherapeutic modalities, portending a grave menace to global public health. The propagation of drug resistance among bacteria is fundamentally facilitated by bacterial interactions, with extracellular vesicles (EVs) assuming a critical role in interbacterial communication. Here, we briefly delineate the methodologies for isolation, extraction, and characterization of EVs from both Gram-negative and Gram-positive bacterial origins. We further investig

[Impact, and treatment possibility of chemotherapy resistance in cancer].

Chemotherapy resistance in tumours is due to complex processes and is responsible for about half of all cancer deaths. In my thesis, I have investigated multiple different resistance mechanisms, most in depth the effect of multidrug resistance (MDR) caused by expression and function of P-glycoprotein (Pgp), and the MDR-selective compounds (such as NSC297366) effectively targeting it. The mechanism was investigated using cell models with different Pgp expression. Seeking the mechanism of action of the MDR-selective NSC297366, we showed that the intracellular iron-binding chelator molecule is ab

Whole-Genome Analysis of Acinetobacter baumannii Strain AB43 Containing a Type I-Fb CRISPR-Cas System: Insights into the Relationship with Drug Resistance.

The CRISPR-Cas system is a bacterial and archaea adaptive immune system and is a newly recognized mechanism for controlling antibiotic resistance gene transfer. Acinetobacter baumannii (A. baumannii) is an important organism responsible for a variety of nosocomial infections. A. baumannii infections have become problematic worldwide because of the resistance of A. baumannii to multiple antibiotics. Thus, it is clinically significant to explore the relationship between the CRISPR-Cas system and drug resistance in A. baumannii. This study aimed to analyze the genomic characteristics of the A. ba
