New phenolic derivatives and bioactive constituents from Castanopsis lamontii Hance: enzyme inhibitory activities and structure-activity insights.

Phytochemical investigation of the mature leaves of Castanopsis lamontii, an underexplored traditional folk medicine, led to the isolation of two new phenolic derivatives, lamontin B (1) and 6-O-(5-O-methylgalloyl)-D-glucopyranose (2) and eleven known compounds. Their structures were elucidated through extensive spectroscopic analysis. Bioactivity assays revealed that kaempferol (10), quercetin (11), and castanopsinin E (13) exhibited potent α-glucosidase inhibitory activities (IC50: 0.03-0.15 mM), which were significantly more active than the positive control acarbose (IC50: 1.5 mM). Self-sufficient nanoparticles with dual-enzyme activity trigger radical storms and activate cascade-amplified antitumor immunologic responses.

Radiotherapy (RT) can potentially induce systemic immune responses by initiating immunogenic cell death (ICD) of tumor cells. However, RT-induced antitumor immunologic responses are sporadic and insufficient against cancer metastases. Herein, we construct multifunctional self-sufficient nanoparticles (MARS) with dual-enzyme activity (GOx and peroxidase-like) to trigger radical storms and activate the cascade-amplified systemic immune responses to suppress both local tumors and metastatic relapse. In addition to limiting the Warburg effect to actualize starvation therapy, MARS catalyzes glucose

Influences of potassium solubilizing bacteria and K-feldspar on enzyme activities and metabolic activities of the bacterial communities in kiwifruit planting soil.

A pot experiment was conducted with kiwifruit planting soil to evaluate the impacts of potassium solubilizing bacteria (KSB) and K-feldspar on the soil nutrient levels, enzyme activities, and microecological environment. The effects were investigated of three inoculation treatments (T1: K-feldspar, T2: KSB, and T3: KSB with K-feldspar) and a non-inoculation treatment (CK) on the enzyme activities and the metabolic activities of the bacterial communities in kiwifruit rhizosphere soil. The results showed that the total nitrogen, available phosphorus, available potassium, and organic matter conte

Chronic Administration of S-Allylcysteine Activates Nrf2 Factor and Enhances the Activity of Antioxidant Enzymes in the Striatum, Frontal Cortex and Hippocampus.

Oxidative stress plays an important role in neurodegenerative diseases and aging. The cellular defense mechanisms to deal with oxidative damage involve the activation of transcription factor related to NF-E2 (Nrf2), which enhances the transcription of antioxidant and phase II enzyme genes. S-allylcysteine (SAC) is an antioxidant with neuroprotective properties, and the main organosulfur compound in aged garlic extract. The ability of SAC to activate the Nrf2 factor has been previously reported in hepatic cells; however this effect has not been studied in normal brain. In order to determine if

Molecular changes in mitochondrial respiratory activity and metabolic enzyme activity in muscle of four pig breeds with distinct metabolic types.

Skeletal muscles are metabolically active and have market value in meat-producing farm animals. A better understanding of biological pathways affecting energy metabolism in skeletal muscle could advance the science of skeletal muscle. In this study, comparative pathway-focused gene expression profiling in conjunction with muscle fiber typing were analyzed in skeletal muscles from Duroc, Pietrain, and Duroc-Pietrain crossbred pigs. Each breed type displayed a distinct muscle fiber-type composition. Mitochondrial respiratory activity and glycolytic and oxidative enzyme activities were comparable

Inhibitory effect of 5β-pregnane-3α,20β-diol on transcriptional activity and enzyme activity of human bilirubin UDP-glucuronosyltransferase.

Prolonged unconjugated hyperbilirubinemia in infants associated with breast milk feeding is a common pediatric problem known as breast milk jaundice (BMJ). A polymorphic mutation (G71R) of bilirubin UDP-glucuronosyltransferase (UGT1A1) is a known cause of BMJ on the infantile side, but the responsible components of breast milk are not currently known. We analyzed the inhibitory effect of 5β-pregnane-3α,20β-diol (pregnanediol) on transcriptional activity and enzyme activity of UGT1A1. To this end, we constructed two types of expression vectors. One type comprised vectors including the upstream

Expression and activation of the steroidogenic enzyme CYP11A1 is associated with IL-13 production in T cells from peanut allergic children.

Activation of the steroidogenic enzyme CYP11A1 was shown to be necessary for the development of peanut-induced intestinal anaphylaxis and IL-13 production in allergic mice. We determined if levels of CYP11A1 in peripheral blood T cells from peanut-allergic (PA) children compared to non-allergic controls were increased and if levels correlated to IL-13 production and oral challenge outcomes to peanut. CYP11A1 mRNA and protein levels were significantly increased in activated CD4+ T cells from PA patients. In parallel, IL-13 production was significantly increased; IFNγ levels were not different b

A critical tryptophan and Ca2+ in activation and catalysis of TPPI, the enzyme deficient in classic late-infantile neuronal ceroid lipofuscinosis.

BACKGROUND: Tripeptidyl aminopeptidase I (TPPI) is a crucial lysosomal enzyme that is deficient in the fatal neurodegenerative disorder called classic late-infantile neuronal ceroid lipofuscinosis (LINCL). It is involved in the catabolism of proteins in the lysosomes. Recent X-ray crystallographic studies have provided insights into the structural/functional aspects of TPPI catalysis, and indicated presence of an octahedrally coordinated Ca(2+). METHODOLOGY: Purified precursor and mature TPPI were used to study inhibition by NBS and EDTA using biochemical and immunological approaches. Site-di

Enzyme kinetic characterization of the smooth muscle myosin phosphorylating system: activation by calcium and calmodulin and possible inhibitory mechanisms of antagonists.

A native-like smooth muscle filamentous myosin system was characterized from an enzyme kinetic point of view. The system contains endogenous myosin light chain kinase (MLCKase) and calmodulin (CM) (A. Sobieszek, J. Muscle Res. Cell Motil. 11 (1990) 114-124) and is, therefore, well suited for testing the action of CM-antagonists or other inhibitory compounds. However, this has not been done due to its complexity. The characterization of the system includes: (1) derivation of a relationship for rate of myosin phosphorylation in terms of total CM, free Ca2+ and total MLCKase concentrations, which

Post-synthetic infiltration of enzymes in hierarchical ZIF-8 for enhanced stability and activity retention.

Herein, we report that the widely used one-pot in situ encapsulation of enzymes in ZIF-8 can inhibit enzyme activity due to precursor-induced toxicity. However, post-synthetic infiltration of the enzyme into hierarchical ZIF-8 prevents this deactivation, yielding superior enzyme stability and activity retention for efficient biocatalytic applications.

Oxidative stress and antioxidant enzyme activities in elite female Turkish cross-country skiers.

BACKGROUND: Oxidative stress–related processes contribute to exercise physiology through redox-sensitive signaling, yet when oxidant production exceeds antioxidant buffering, oxidative modifications may be reflected in circulating biomarkers. Evidence in elite female endurance athletes remains limited, particularly in cross-country skiers, and interpretation should be restricted to biomarker-level descriptions within the constraints of cross-sectional designs. METHODS: This cross-sectional comparative study included 17 elite female cross-country skiers and 17 age- and BMI-matched sedentary wom

Supporting activities of cognate redox partners for sterol-metabolizing P450 enzymes in Mycobacterium neoaurum.

Steroids with anti-inflammatory, anti-allergic, endocrine-regulating, and other pharmaceutical activities represent the second most widely used class of drugs worldwide, following antibiotics. Their industrial production primarily relies on Mycobacteria-mediated biotransformation of sterols into key intermediates, followed by chemical or enzymatic modifications. While the sterol metabolic pathways in Mycobacteria have been intensively studied, the identification and functional characterization of key enzymes, particularly cytochrome P450 enzymes (CYPs or P450s) and their cognate redox partners

Rigid-Flexible Layered Immobilization Enables Precise Confinement and Dynamic Activation of Small Enzymes.

Small enzymes (10-50 kDa) encounter challenges of unstable enzyme-carrier interactions and low activity retention during immobilization, with existing strategies lacking specificity. Unlike previous industrial enzyme-focused studies, a rigid-flexible layered immobilization strategy integrating networked metal-organic frameworks (MOFs) into flexible hydrogels is proposed. This innovative strategy stabilizes enzyme conformation, strengthens enzyme-matrix interactions, and prevents enzyme leakage via a dual-pore system. This synergistic system, composed of MOF micropores (50-150 nm, confinement)

Designing ligands for tuning the active site of novel artificial vanadium-based enzymes for sulfoxidation.

Artificial metalloenzymes, which are formed by inserting inorganic complexes into biological scaffolds, have attracted interest as a potential biotechnological alternative of natural enzymes. In this study, vanadium complexes were incorporated as artificial active sites for sulfoxidation catalysis within NikA, a bacterial Ni(II) import protein. Two of the designed metal complexes, namely VOL1 and VOL3, which are based on either an amino alcohol or an amino acid ligand, respectively, were spectroscopically and structurally characterized, demonstrating transformation of a VOL3 into a VO(L3)2 spe

Comparative Analysis of Carbohydrate-Active Enzyme (CAZyme) Diversity Across Diverse Algal Lineages.

Algae are characterized by lineage-specific storage and structural carbohydrates; however, the metabolic pathways governing these compounds remain largely uncharacterized despite their high commercial value in the food, pharmaceutical, and nutraceutical sectors. This study addresses this knowledge gap through a comprehensive genome-wide analysis of Carbohydrate-Active Enzymes (CAZymes) across green, red, and brown algae. By utilizing functional homology searches, the CAZyme repertoires of these lineages were identified. Systematic comparative profiling revealed both distinct lineage-specific "

Cu-HEPES Nanosheets with Dual Enzyme-Mimetic Activities for Colorimetric Detection of 2,4-Dichlorophenol.

A convenient one-pot strategy was adopted to fabricate Cu-4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid (HEPES) nanosheets through the coordination of HEPES with copper ions under alkaline conditions, and the obtained nanosheets possessed dual laccase-mimetic and oxidase-mimetic activities for phenolic pollutant analysis and show potential for dye decolorization/removal. The Cu-HEPES nanosheets displayed significant dual enzyme-mimetic activities, effectively catalyzing the oxidation conversion for phenolic compounds, specifically 2,4-dichlorophenol (2,4-DCP), to yield quinone imine produ

Enzyme-Mimetic Hydrogel Balancing Antibacterial Activity and Cytoprotection for Corneal Regeneration in Bacterial Keratitis.

Current bacterial keratitis treatments, ranging from topical antibiotics to sutured corneal grafts, fail to concurrently address infection control, inflammation modulation, and tissue regeneration. To reconcile effective antimicrobial action with host-tissue protection, we developed an injectable bioinspired composite matrix that integrated photoresponsive collagen/hyaluronic acid-nitrobenzene conjugate (CHB) hydrogel with multifunctional carbon dots (CDs) derived from curcumin and chitosan (CC-CDs). Upon injection, the CHB hydrogel forms a transparent adhesive interface in situ, while the CC-

Reconstructing protein conformations to activate follicular enzyme-nutrient reservoirs for reversing follicle aging.

Hair regeneration depends on a reservoir of follicular enzymes that regulate the microenvironment and provide metabolic support. However, in degenerative alopecia, this enzymatic reservoir is disrupted, impairing microenvironmental homeostasis. Here, through bioinformatics and molecular dynamics analyses, we revealed that several Zn2+/Cu2+-dependent proteases (e.g., LOX and SOD1) were downregulated and exhibit aberrant conformations and activities in degenerative alopecia, further weakening microenvironmental support. To address this, we developed a Zn2+/Cu2+-doped decellularized extracellular

Enzyme-activated molecular MRI for specific delineation of the ischemic penumbra in acute ischemic stroke.

Ischemic stroke, the second leading cause of death and a primary source of severe disability in adults worldwide, is associated with high morbidity and mortality rates that correlate closely with the extent of neuronal damage. A critical strategy for improving patient outcomes is the early identification and rescue of the ischemic penumbra, a region at risk but potentially salvageable. However, accurately identifying and visualizing this penumbra poses a significant challenge in treatment and is crucial for predicting patient prognosis. Conventional magnetic resonance imaging (MRI) techniques
