Icariin suppresses glycolysis in prostate cancer by upregulating ALKBH5 to mediate EARS2 m6A demethylation.

Prostate cancer (PCa) is among the most frequently occurring cancers in men. Its occurrence and progression are closely related to metabolic reprogramming, with abnormally active glycolysis being a notable feature sustaining rapid proliferation. Recent years have seen growing interest in the regulatory role of epigenetic modifications, particularly N⁶-methyladenosine (m6A) RNA methylation. AlkB homolog 5 (ALKBH5), as an m6A demethylase, may influence glycolysis in PCa by regulating target genes, offering new directions for mechanistic studies and therapy. The impact of different concentrations

Epigenetic mechanism of LncRNA KCNQ1OT1 in high-glucose-induced podocyte injury via m6A methylation modification.

Podocyte injury is a hallmark of diabetic nephropathy associated with proteinuria and renal dysfunction. This study elucidated the role of long non-coding RNA KCNQ1 overlapping transcript 1 (LncRNA KCNQ1OT1) in high-glucose (HG)-induced podocyte injury. In HG-treated podocytes, KCNQ1OT1, WTAP, and MAPK6 were quantified. After KCNQ1OT1 interference, cell viability, apoptosis, inflammation, and oxidative stress were measured. Subcellular localization of KCNQ1OT1 was determined by nuclear-cytoplasmic fractionation and RNA FISH. Binding of MLL1 to KCNQ1OT1 was validated via RNA pull-down and RIP.

METTL3 depletion mitigates hypoxic pulmonary arterial smooth muscle cell over-proliferation via YTHDF2-dependent regulation of FOXO1 mRNA N6-methyladenosine modification: a potential mechanism of hypoxic pulmonary hypertension.

AIMS: To determine the role of METTL3-mediated m6A modification in hypoxic pulmonary arterial smooth muscle cells (PASMCs) over-proliferation and hypoxic pulmonary hypertension (HPH). MATERIALS AND METHODS: Human PAH tissues, smooth muscle-specific Mettl3 knockout mice, HPH models, scRNA-seq, hPASMCs culture, MeRIP-qPCR, RIP-qPCR, luciferase assay, and functional assays. KEY FINDINGS: METTL3 was upregulated in HPH PASMCs. Mettl3 depletion alleviated HPH and suppressed PASMC proliferation via FOXO1 upregulation. METTL3 reduced m6A on FOXO1 3'UTR, decreased YTHDF2 binding, and stabilized FOXO1

Dysregulated m6A via compensatory arginine methylation primes premalignancy in metabolic dysfunction-associated steatotic liver disease.

Hepatocellular carcinoma (HCC) originates from premalignant disease-associated hepatocytes (daHeps) that emerge during the progression of metabolic dysfunction-associated steatotic liver disease (MASLD) to metabolic dysfunction-associated steatohepatitis (MASH). As daHeps are compensatorily primed by metabolic stress, we reproduced the accelerated progression of MASLD-associated HCC in mice by phenocopying the decreased expression of a metabolic regulator, protein arginine methyltransferase 1 (PRMT1). In Prmt1 liver-specific knockout (LKO), m6A-mediated changes in mRNA stability reprogram the

LINE1 RNA demethylation sensitizes cancer cells to PARPi through global chromatin remodeling.

BACKGROUND: Repression of DNA damage repair by PARP inhibitors (PARPi) has shown great efficacy in cancer treatment. However, therapy resistance remains a significant clinical challenge. RESULTS: Here, we demonstrate that the N6-methyladenosine (m6A) modification of long interspersed nucleotide element-1 (LINE1, or L1) facilitates DNA damage repair in cancer cells, whereas removal of L1 m6A modifications enhances the sensitivity of both BRCA-wild-type and BRCA-mutant cells to PARPi. Specifically, following olaparib treatment, METTL3 accumulates on chromatin at non-damage sites, which results

FTO-mediated NR1D1 m6A demethylation contributes to hyperandrogenemia during continuous light-induced circadian disruption.

N6-methyladenosine (m6A) is a key epigenetic modification closely associated with reproductive physiology; however, its involvement in circadian-related ovarian dysfunction remains unclear. Female Sprague Dawley rats were exposed continuously to light for 10 weeks (LL group) to induce circadian desynchrony, with controls maintained under standard cycles (DL group). Transcriptomic analysis was performed to identify markedly dysregulated pathways, which were validated via qRT‒PCR, immunoblotting, and KGN cell models. Continuous light exposure induced PCOS-like phenotypes in female rats, includin

Trdn-as directs m6A-dependent transcriptional termination for accurate triadin isoform switching, preventing aberrant dyads and cardiomyopathy.

Heart failure is a leading cause of mortality, and impaired cardiac excitation-contraction coupling represents a potentially fatal trigger for myocardial dysfunction. Long non-coding RNAs (lncRNAs) can contribute to cardiomyopathy, but comprehensive mechanistic insights remain elusive. We demonstrate that reduction of the lncRNA TRDN-AS in human cardiomyopathy or abrogating it in human iPSC-derived cardiomyocytes and mice causes a switch of cardiac TRDN/TRISK32 to skeletal muscle TRDN/TRISK95. Transcription of Trdn-as in cis is essential for stalling RNA Pol II at the 3' end of the cardiac Trd

Global landscape and emerging trends in programmable mRNA-based therapeutics for myocardial infarction: a multi-database bibliometric analysis.

BACKGROUND: Myocardial infarction (MI) remains a leading cause of mortality worldwide. Although current reperfusion-based interventions have significantly improved clinical outcomes, they are insufficient to restore lost cardiomyocytes or repair irreversible myocardial injury. Programmable messenger RNA (mRNA)-based therapeutics have emerged as a cutting-edge strategy for cardiac regenerative repair owing to their advantages of transient and controllable protein expression, high design flexibility, and avoidance of genomic integration. However, the global research landscape, collaborative netw

Enhancing targeted strategies for cancer immunotherapy by elucidating mRNA processing mechanisms.

Immune checkpoint inhibitors have transformed the landscape of cancer therapy; however, the challenge that most patients do not achieve durable benefits urgently necessitates the development of new strategies that extend beyond mere T-cell activation. mRNA processing-comprising alternative splicing, RNA modifications, and RNA editing-establishes a dynamically regulated connection between the intrinsic characteristics of tumors and anti-tumor immunity. This review systematically summarizes how mechanistic insights into these processes can be translated into concrete approaches that enhance the

Prevalence of N6-methyladenosine (m6A) in mycoplasma mRNA:Epitranscriptomic regulation in minimal genomes.

Messenger RNA N6 methyladenosine (m6A) modification has been considered as the main post-transcriptional modification of eukaryotic mRNA; however, its role in the regulation of prokaryotic mRNA transcription remains unclear. The N6 methyladenosine (m6A) modifications in prokaryotic mRNA has been found in Pseudomonas aeruginosa and Escherichia coli so far. In this study, ultra-high-pressure liquid chromatography coupled with triple quadrupole tandem mass spectroscopy (UHPLC-QQQ-MS/MS) was used to calculate the m6A/A ratio in multiple mRNA from a wide range of mycoplasma species, representing as

Analysis of mRNA m6A modification and mRNA expression profiles in middle ear cholesteatoma.

Introduction: Middle ear cholesteatoma is characterized by the hyperproliferation of keratinocytes. In recent decades, N6-methyladenosine (m6A) modification has been shown to play an essential role in the pathogenesis of many proliferative diseases. However, neither the m6A modification profile nor its potential role in the pathogenesis of middle ear cholesteatoma has currently been investigated. Therefore, this study aimed to explore m6A modification patterns in middle ear cholesteatoma. Materials and methods: An m6A mRNA epitranscriptomic microarray analysis was performed to analyze m6A modi

Increased METTL1 expression in B cell subsets correlates with disease activity and immunological features in SLE.

OBJECTIVE: Systemic lupus erythematosus (SLE) is a complex autoimmune disease characterized by aberrant B cell activation, excessive plasma cell differentiation, and pathogenic autoantibody production. Epitranscriptomic modifications have emerged as important regulators of immune cell function. Among these, N7-methylguanosine (m7G) modification, catalyzed by methyltransferase-like 1 (METTL1), has been implicated in immune regulation and autoimmune diseases. However, the expression pattern of METTL1 across peripheral B cell subsets and its clinical relevance in SLE remain incompletely character

Tamoxifen differentially modulates endometrial hyperplasia via wild-type and mutant p53 regulation of the ALKBH5-REG1A axis.

INTRODUCTION: Tamoxifen is a cornerstone of endocrine therapy for estrogen receptor-positive breast cancer; however, its partial estrogen agonist activity in the endometrium predisposes patients to hyperplasia and, in some cases, malignant transformation. The molecular mechanisms underlying this tissue-specific adverse effect remain incompletely understood. METHODS: We employed immortalized human endometrial epithelial cells to investigate the role of p53 in tamoxifen-induced proliferation. Cells were genetically manipulated to express wild-type (WT) or mutant p53 (R248Q), and ALKBH5 or REG1A

Characterization of m6A modifiers and RNA modifications in uterine fibroids.

Uterine leiomyoma or fibroids are the most common prevalent noncancerous tumors of the uterine muscle layer. Common symptoms associated with fibroids include pelvic pain, heavy menstrual bleeding, anemia, and pelvic pressure. These tumors are a leading cause of gynecological care but lack long-term therapy as the origin and development of fibroids are not well understood. Several next-generation sequencing technologies have been performed to identify the underlying genetic and epigenetic basis of fibroids. However, there remains a systemic gap in our understanding of molecular and biological p

m6A RNA Methylation Is Increased in Tumour Invasive Regions and Influences Invasive Capability and Chemotherapeutic Sensitivity in Adult Glioblastoma.

Adult glioblastoma multiforme (GBM) is the most common primary malignant brain tumour caused by multiple molecular factors. N6-methyl-adenosine (m6A) is an abundant RNA modification that governs cellular RNA metabolism. We hypothesise that changes in m6A-modified RNA and regulatory machinery such as the writer proteins, Methyltransferase 3 (METTL3) and WT1-associating protein (WTAP), the demethyltransferase protein, and Alpha-ketoglutarate dependent dioxygenase (FTO), are driving factors of GBM development and treatment resistance. Here, we investigated m6A-RNA spatial and quantitative abundan

METTL14-mediated m6A modification of CXCL9 mRNA regulates macrophage CS polarization to suppress OSCC growth and stemness.

Oral squamous cell carcinoma (OSCC), the most prevalent histological subtype of oral cancer, is characterized by an immunosuppressive tumor microenvironment (TME) predominantly modulated by tumor-associated macrophages (TAMs). The CXCL9/SPP1 ratio (CS ratio) has been validated as a more reliable prognostic biomarker than the conventional M1/M2 classification. Although methyltransferase-like 14 (METTL14) participates in TAM polarization, its specific role in regulating the CS balance in OSCC remains elusive. This investigation elucidates the mechanism by which METTL14 modulates TAM CS polarizat

FTO-modulated m6A demethylation and upregulation of KIF11 mRNA promotes retinal microvascular dysfunction in diabetic retinopathy.

BACKGROUND: The N6-methyladenosine (m6A) demethylation of mRNAs is critical for the progression of diabetic retinopathy (DR). Fat mass and obesity-associated protein (FTO) has been reported to be overexpressed in DR and is considered to be an important epitranscriptomic regulator (m6A eraser) in retinal angiogenesis. The principle of Kinesin family member 11 (KIF11) gene in DR is unclear. METHODS: Methylated RNA immunoprecipitation sequencing (MeRIP-Seq) was used to screen abnormal m6A modification in vitreous body samples of proliferative DR (PDR) patients.
