Four stereoisomers of 4-acetoxy-2-hexyltetrahydrofuran and their sulfur analogues were synthesized. Odor evaluation showed that configuration governs properties: in the acetoxy series, (2R,4S) had the lowest thresholds (89 μg/L in water, 9.1 μg/L in air) with a peach-like sweet odor, while (2S,4R) had the highest (3.3 × 103 μg/L, 7.7 × 102 μg/L) with earthy notes; only (2R)-configured isomers exhibited the characteristic sweet peach note. Plant-derived smoke - butenolide, called at present karrikin 1 (KAR1) is known as an important inductor of seed germination and seedling growth. In this study, tricyclic butenolides were synthesized and their effects on germination of dormant and non-dormant Avena fatua caryopses were compared, as were also their effects versus those of KAR1 on seedling growth. A mechanistic study of the hydrogenation reaction of levulinic acid (LA) to 2-methyltetrahydrofuyran (MTHF) was performed using three different solvents under reactive H2 and inert N2 atmospheres. AIM: To identify conditions that induce an increase in the sensitivity of highly Halaven (HAL)-resistant cancer cells compared to sensitive cells.
MATERIALS AND METHODS: We observed that drug-resistant KBV20C cells are highly resistant to HAL compared to other antimitotic drugs. The concentration required to treat KBV20C cells was almost 500-fold higher than that used to treat sensitive parent KB cells. The copper-catalyzed asymmetric addition of 2-silyloxyfurans to cyclic unsaturated oxo esters is reported. The reaction proceeds with excellent diastereocontrol (usually dr 99:1) and modest to high enantioselectivity, depending on the nature of the ester group and the substitution of the cyclic oxo ester. We have shown that these substrates can be transformed into a variety of building blocks bearing a γ-butenolide or γ-lactone connected to a cycloalkane or cycoalkene moiety.

The asymmetric total synthesis of pyranicin (1) is reported. The butenolide ring was constructed via an asymmetric alkylation/ring-closing metathesis strategy. The three stereocenters in the left-hand tetrahydropyran ring were installed by sequential chiral auxiliary-mediated aldol reactions. Closure of the tetrahydropyran and fusion of the alkyl backbone were affected via a sequential ring-closing metathesis-cross-metathesis strategy.

Calix[4]pyrrole (1), calix[2]m-benzo[4]pyrrole (2), 10alpha,20beta- and 10alpha,20alpha- bis(4-nitrophenyl)-calix[4]pyrroles 3 and 4, respectively, were found to exhibit various organocatalytic activities in the diastereoselective vinylogous addition reaction of 2-trimethylsilyloxyfuran (TMSOF, 7) to aldehydes. The gamma-hydroxybutenolide products are obtained in fairly good yields and with moderate diastereoselectivity. The structures of the catalysts, as well as the reaction conditions, strongly influence the efficiency of the reaction.

Density functional theory using the B3LYP hybrid functional has been employed to study the formation of [Cu(II)(TPA(H))(O2-)]+ and [Cu(II)(TPA(MeO))(O2-)]+ (TPA = tris(2-pyridylmethyl)amine) in two different solvents, THF and EtCN. Two related butenolides have been isolated from a culture broth of a strain of the fungus, Acremonium sp. Although the lactone acid (3) is a known compound, the structure of the lactone diacid (4), deduced by spectroscopic analysis, is assigned for the first time. This compound appears to be identical to a partly characterised metabolite isolated, along with 3, from Chaetomium indicum in 1953.
