Birth of healthy female twins after preimplantation genetic diagnosis of cystic fibrosis combined with gender determination.

Two healthy sisters with a familial history of mental retardation were referred to our centre for preimplantation genetic diagnosis (PGD). Their two brothers showed severe mental retardation. The molecular basis for their disorder could not be identified, but one of the sisters and the mother presented a highly skewed pattern of X-inactivation reinforcing the likelihood of an X-linked mode of inheritance. Both sisters requested PGD to avoid the abortion of potentially affected male fetuses. PGD for sex by fluorescent in-situ hybridization was carried out for the first sister and resulted in th

Linkage analysis for the diagnosis of autosomal dominant polycystic kidney disease, and for the determination of genetic heterogeneity in Italian families.

Sixty-eight individuals from six Italian families in which autosomal dominant polycystic kidney disease (ADPKD) is segregating, were typed in DNA polymorphisms linked to the PKD1 locus on chromosome 16. A total of ten probes were used: 3' HVR, HMJ1, EKMDA, GGG1, 26-6, VK5B, 218EP6, 24.1, CRI090, and 41.1. Zmax was 4.502 at theta = 0.082 between ADPKD and 3'HVR, and 4.382, 1.947, and 1.576 between ADPKD and GGG1, 26.6, and 218EP6, respectively, at theta = 0.0. No clear evidence of genetic heterogeneity was found. Multipoint analyses were consistent with linkage to PKD1. Twenty-nine diagnoses an

Cloning of the adenylate cyclase genetic determinant of Bordetella pertussis and its expression in Escherichia coli and B. pertussis.

A recombinant plasmid, pRMB1, identified from a gene library of B. pertussis, restored adenylate cyclase (AC) and haemolysin (HLY) activities to B. pertussis BP348 (a Tn5-insertion mutant deficient in both these activities). B. pertussis BP348 was considerably less virulent than wild type strains of B. pertussis when 3-week-old mice were challenged intranasally; possession of pRMB1 restored virulence. Neither AC nor HLY activities were expressed in E. coli harbouring pRMB1. However, expression of calmodulin-responsive AC was obtained in E. coli when restriction fragments of pRMB1 were subclone

High fetal hemoglobin production in sickle cell anemia in the eastern province of Saudi Arabia is genetically determined.

Homozygous sickle cell disease in the eastern province of Saudi Arabia is clinically mild. Circulating fetal hemoglobin levels of 16.0 +/- 7.4% were found in these anemic patients, but only 1.09 +/- 0.97% in their sickle trait parents. To determine whether these sickle cell anemia patients inherit an increased capacity to synthesize fetal hemoglobin, a radioimmunoassay of fetal and adult hemoglobin was performed on erythroid progenitor (BFU-E)-derived erythroblasts from Saudi Arabian sickle cell patients and their parents. Mean fetal hemoglobin content per BFU-E-derived erythroblast from Saudi

Genetics of resistance to African trypanosomes: role of the H-2 locus in determining resistance to infection with Trypanosoma rhodesiense.

Susceptibility and resistance to Trypanosoma rhodesiense infections in inbred and H-2 congenic strains of mice were studied. Mean survival times and patterns of parasitemia were examined. C3HeB/FeJ mice were highly susceptible; CBA/J, A/J, and BALB/cByJ mice displayed an intermediate level of susceptibility; whereas C57BL/10 mice were highly resistant. H-2 congenic strains with the BL/10 background resembled the BL/10 parental type, thereby suggesting that the major histocompatibility complex does not play a major role in regulating resistance and susceptibility to infection with T. rhodesiens

Natural, genetically determined resistance toward influenza virus in hemopoietic mouse chimeras. Role of mononuclear phagocytes.

Radiation chimeras produced by crosswise transfers of bone-marrow cell among histocompatible mice susceptible, or genetically resistant, to lethal challenge by a number of myxoviruses were used to test whether macrophage resistance (as assessed in vitro) and resistance of the animal (as measured in vivo), both previously shown to be brought about by the gene Mx, were causally related. 49 chimeras were tested individually, both of resistance of their macrophages to in vitro challenge with M-TUR (a strain of avian influenza virus A/Turkey/England/63 adapted to grow in cultured mouse peritoneal m

Plasma testosterone heterogeneity reveals genetic and demographic determinants of donor health and red blood cell quality.

Testosterone is a key determinant of red blood cell (RBC) biology and susceptibility to hemolysis; however, its contribution to donor-dependent variation in RBC quality and transfusion effectiveness remains unclear. Using data from 2,357 male participants in the NHLBI REDS-III RBC-Omics study, we quantified plasma total testosterone, free testosterone (bioavailable form), and androstenedione and evaluated their associations with donor characteristics, RBC hemolysis, and recipient post-transfusion hemoglobin increments, a clinical measure of RBC transfusion effectiveness. Subphysiological testo

Genetic determinants of serum creatinine and cystatin C independent of kidney function generate bias in filtration markers.

INTRODUCTION: Non-GFR determinants of filtration markers reduce the accuracy of estimated GFR (eGFR) and genetic factors may explain part of this reduced performance. Here, we identify genetic variants associated with serum levels of creatinine or cystatin C (but not GFR), compute their effect, and investigate candidate genes. METHODS: We used data from the UK Biobank (approximately 470,000 people) and the CKD-EPI creatinine (eGFRcr) and cystatin C (eGFRcys) equations. Using genome-wide association studies (GWAS), we defined strict criteria to classify loci as creatinine-specific or cystatin

Analysis of whole genome sequencing and plasma metabolomics unveil genetic determinants and clinical implications for human health.

Blood metabolites are critical biomarkers linking genetic variation to diverse health outcomes, exhibiting significant variability across individuals. Here, we analyze and integrate whole-genome sequencing with plasma metabolomics to investigate the genetic architecture of 313 metabolic biomarkers in up to 199,138 UK Biobank participants. Through single-variant analyses, we identify 36,105 independent signals, with 22.20% being novel. Furthermore, we pinpoint 12,361 putative causal variant-trait associations, demonstrating enhanced causal signal discovery and improved fine-mapping resolution c

Genetic Determinants of Peanut-Specific IgG4 Levels in the Context of Sustained Oral Peanut Exposure in the LEAP Study.

In the Learning Early About Peanut Allergy (LEAP) study, participants in the peanut consumption group, at 60 months of age, had higher levels of peanut-specific IgG4 (psIgG4), a biomarker of immune modulation, compared to those in the peanut-avoidance group. We investigated the genetic determinants of psIgG4 among participants who consumed peanuts. Using whole-genome sequencing data, we performed a genome-wide association study (GWAS) for psIgG4 in the LEAP peanut consumption group participants (N = 267). We generated a cumulative genetic score from the identified loci and evaluated its associ

Genetic and clinical determinants of tacrolimus accumulation in early liver transplantation: evidence for a combined genotype-antibiotic effect.

BACKGROUND: Tacrolimus is a first-line immunosuppressant essential for preventing graft rejection after liver transplantation, but its narrow therapeutic index and extensive interindividual pharmacokinetic variability complicate optimal dosing during the early postoperative phase. METHODS: The pharmacokinetics of tacrolimus were analyzed in liver transplant recipients from longitudinal trough concentrations collected within the first 30 days of administration using non-compartmental models. Associations between pharmacokinetic indices and covariates were assessed using Spearman's correlation,

Massive Genetic Diversity Loss Within Asian Elephant Subspecies: New Insights for Determining Conservation Priorities.

In this study, we provide a comprehensive genomic analysis of population structure, genetic diversity, and load across the four Asian elephant (Elephas maximus) subspecies using whole-genome sequencing data of 81 wild-born individuals, with most of them currently living under human care. Our results reveal striking contrast in inbreeding: the Bornean subspecies (E.m. borneensis) exhibits the highest level of inbreeding, with ∼65% of its genome in runs of homozygosity (ROH), and the lowest genome-wide heterozygosity (∼46 heterozygous sites per 100 kb). The Sumatran subspecies (E.m. sumatranus)

Genetic Determinants of Microvascular Complications of Type 1 Diabetes Mellitus: New Data from a Replication Study.

Background: Diabetic retinopathy (DR) and chronic kidney disease (CKD) are among the leading causes of disability in individuals with type 1 diabetes mellitus (T1DM). However, the genetic architecture of these complications remains poorly understood. Genome-wide studies demonstrate significant interpopulation heterogeneity, while candidate gene studies yield conflicting results due to limited power. Independent replication of previously obtained results in separate cohorts, with appropriate intergroup comparisons, is essential for identifying the most significant biomarkers of microvascular co

African-specific genetic loci determine iron status and risk of severe malaria and bacteremia in African children.

Iron is essential for both humans and pathogens, yet its genetic regulation remains understudied in African populations. Here, we report genome-wide association studies of six iron-related biomarkers in 3928 children from five sites across Africa, with replication in 2868 African American adults and investigate associations with severe malaria and bacteremia. We identify previously unreported loci at genome-wide significance, for transferrin at GTF3C5, and for hepcidin at CHCHD7/SDR16C5. Variants tagging the DUP4 haplotype, encoding the Dantu blood group (rs552439837) are associated with solub
