The role of lipid-lowering drugs in urologic and male reproductive cancers: a Mendelian randomization study.

BACKGROUND: Urologic and male reproductive system tumors are common in older men and frequently coexist with cardiovascular and metabolic disorders, suggesting potential shared biological pathways. Although lipid-lowering therapies are widely used for cardiovascular prevention, their long-term associations with these cancers remain incompletely understood. We therefore applied a drug-target Mendelian randomization (MR) framework to evaluate the associations between genetically proxied lipid-lowering targets and the risks of urologic and male reproductive cancers. METHODS: We performed a drug-

Genome- and Exome-Wide Identification of Common-to-Rare Variants Associated with Middle Ear Cholesteatoma.

HYPOTHESIS: To investigate the genetic susceptibility of middle ear cholesteatoma (MEC) and construct an MEC risk prediction model by integrating genetic risk with clinical factors. BACKGROUND: MEC represents a relatively rare disorder that is associated with high morbidity, whereas its genetic etiology remains poorly understood. METHODS: Using genetic data from the UK Biobank (UKB), we performed both genome-wide association study (GWAS) and exome-wide association study (ExWAS) involving 702 MEC patients and 491,503 controls. Gene-based and gene set-based association studies were then perfor

Demarcation of anxiety and fear: Evidence from behavioral genetics.

Anxiety and fear are emotions often intertwined in response to aversive stimuli, complicating efforts to differentiate them and understand their distinct consequences. This study explores the common genetic and environmental factors contributing to the co-occurrence of anxiety disorders and dimensions of the revised Reinforcement Sensitivity Theory (rRST). A sample of 356 monozygotic (22.5 % males; M = 25.73, SD = 8.3) and 386 dizygotic (33.9 % males; M = 24.21, SD = 8.33) twins from the Serbian Twin Advanced Registry was analyzed. Causal association between celiac disease and inflammatory bowel disease: A two-sample bidirectional Mendelian randomization study.

BACKGROUND: An epidemiological link between celiac disease (CeD) and inflammatory bowel disease (IBD) has been well established recently. In this study, Mendelian randomization (MR) analysis was performed employing pooled data of publicly available genome-wide association studies (GWAS) to determine the causal relationship between CeD and IBD, encompassing ulcerative colitis (UC) and Crohn's disease (CD). METHODS: Dataset of CeD was acquired from GWAS for 12,041 cases and 12,228 controls. A GWAS of more than 86,000 patients and controls was used to identify genetic variations underlying IBD.

Immune Cells and Polymyalgia Rheumatica: Insights from a Bidirectional Mendelian Randomization Analysis.

INTRODUCTION: Polymyalgia Rheumatica (PMR) is an inflammatory disease of older adults in which immune dysregulation is thought to play a central role. However, the specific circulating immune cell phenotypes linked to PMR remain incompletely defined. METHODS: We performed a two-sample, bidirectional Mendelian Randomization (MR) study using Genome-Wide Association Study (GWAS) summary statistics from European- ancestry cohorts. In total, 731 immune cell traits were analyzed. Forward MR assessed associations between genetic liability to PMR and immune phenotypes, and reverse MR tested whether g

Antibody-mediated immune responses and inflammatory bowel disease: insights from genetic correlation and Mendelian randomization.

INTRODUCTION: Observational studies suggest a complex relationship between infectious diseases and inflammatory bowel disease (IBD), yet the underlying mechanisms remain unclear. This study examined the genetic correlations and causal associations between antibody-mediated immune responses (targeting 46 infectious pathogens) and IBD, using linkage disequilibrium score regression (LDSC) and Mendelian randomization (MR) to inform early prevention and personalized treatment. MATERIAL AND METHODS: Data from the UK Biobank (8,735 samples) were used to identify independent SNPs associated with anti

Missing a beat: assessment of circadian rhythm abnormalities in bipolar disorder in the genomic era.

Circadian rhythm abnormalities have been recognized as a central feature of bipolar disorder (BD) but a coherent biological explanation for them remains lacking. Using genetic mutation of 'clock genes', robust animal models of mania and depression have been developed that elucidate key aspects of circadian rhythms and the circadian clock-mood connection. However, translation of this knowledge into humans remains incomplete. In recent years, very large genome-wide association studies (GWAS) have been conducted and the genetic underpinnings of BD are beginning to emerge. However, these genetic s

Delayed Diagnoses of SGCE Myoclonus-Dystonia.

BACKGROUND: Myoclonus-dystonia due to SGCE mutations (OMIM: 159900) most commonly presents during childhood with mainly upper body myoclonus, and mild dystonia affecting the neck and arms. CASE REPORTS: Herein, we report patients misdiagnosed during childhood with Tourette syndrome and dyskinetic cerebral palsy, and, during adulthood, found to harbor SGCE frameshift mutations. DISCUSSION: Myoclonus-dystonia may be underdiagnosed due to phenotypic misclassification during childhood. SGCE mutations should be included in the differential diagnosis of childhood movement disorders that ostensibly

Genetically Proxied Leukocyte Telomere Length and Epigenetic Age Acceleration in Relation to Healthspan: A Mendelian Randomization Study.

BACKGROUND: Leukocyte telomere length (LTL) and epigenetic age acceleration (EAA) are widely studied biomarkers of biological aging, but their potential roles in healthspan remain unclear. We evaluated whether genetically proxied LTL and EAA show evidence of potential effects on healthspan. METHODS: We conducted a two-sample Mendelian randomization study. Genetic instruments for LTL and four EAA biomarkers were obtained from published genome-wide association studies, including up to 472,174 individuals for LTL and approximately 35,000 individuals for each EAA biomarker. Summary statistics for

Mitochondrial dysfunction in the pathogenesis of intervertebral disc herniation: a mitochondrial related genome-wide Mendelian randomization analysis.

BACKGROUND: As a degenerative disease, the pathophysiology of intervertebral disc herniation (IDH) closely related to mitochondrial dysfunction. However, the specific molecular mechanisms involved have yet to be precisely established. METHODS: Here, we employed a two-sample, two-step Mendelian Randomization (MR) approach, along with summary-Data-Based MR, genetic colocalization, full phenomenon association analysis, and GO and KEGG enrichment analysis to investigate the genetical effects of mitochondrial dysfunction on IDH. RESULTS: From the intercross between mitochondrial-related genes and e

Associations of non-alcoholic fatty liver disease with arrhythmic disorders and electrocardiogram traits: A Mendelian randomization study.

INTRODUCTION: Observational studies indicate a link between arrhythmias, electrocardiogram (ECG) characteristics, and non-alcoholic fatty liver disease (NAFLD). However, it is uncertain whether NAFLD acts merely as a risk marker or directly contributes to the development and progression of these arrhythmic disorders. Mendelian randomization (MR) was employed to investigate the causal relationship of NAFLD on arrhythmias and ECG traits. METHODS: A two-sample Mendelian randomization (MR) analysis was performed to examine the causal relationship of genetically predicted NAFLD (imaging and biopsy

Detecting potential causal relationship between inflammatory bowel disease and rosacea using bi-directional Mendelian randomization.

The association between rosacea and inflammatory bowel disease (IBD) has been studied in previous observational studies. It is unclear, however, whether the association is causal or not. Independent genetic variants for IBD were chosen as instruments from published Genome-wide association studies (GWAS) studies involving 38,155 cases with an IBD diagnosis and 48,485 controls in order to investigate the causal effect of IBD on rosacea. Summarized data for rosacea were gathered from various GWAS studies that included 1195 cases and 211,139 controls without rosacea. Reverse-direction Mendelian ra

Convergent imaging and genetic signatures of gray matter atrophy in Parkinson's disease.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by widespread structural brain alterations, yet the specific patterns of brain atrophy and their underlying genetic mechanisms remain incompletely understood. Here, we integrated large-scale neuroimaging meta-analysis with population-scale imaging genetics to systematically characterize the genetic architecture linking gray matter volume (GMV) abnormalities to PD. We first performed a meta-analysis of structural MRI studies comprising 3212 patients with PD and 2056 controls, identifying robust patterns of GMV re

Prospects of Mendelian randomization in hepatology: a comprehensive literature review with practice guidance.

Mendelian randomization (MR), a powerful statistical tool for causal inference, has been widely applied in various fields of medical research, even extending to economics and psychology. In hepatology, MR has been utilized to identify risk factors and potential therapeutic targets for liver diseases, including metabolic dysfunction-associated steatotic liver disease, cholestatic and autoimmune liver diseases, and hepatobiliary cancer. MR can provide evidence of causation via associations between genetic variants, modifiable exposures and liver disease occurrence or outcomes, using large existi

Association analysis of rare and common variants with multiple traits based on variable reduction method.

Pleiotropy, the effect of one variant on multiple traits, is widespread in complex diseases. Joint analysis of multiple traits can improve statistical power to detect genetic variants and uncover the underlying genetic mechanism. Currently, a large number of existing methods target one common variant or only rare variants. Increasing evidence shows that complex diseases are caused by common and rare variants. Here we propose a region-based method to test both rare and common variant associated multiple traits based on variable reduction method (abbreviated as MULVR). However, in the presence o

Retinal ciliopathies through the lens of Bardet-Biedl Syndrome: Past, present and future.

The primary cilium is a highly specialized and evolutionary conserved organelle in eukaryotes that plays a significant role in cell signaling and trafficking. Over the past few decades tremendous progress has been made in understanding the physiology of cilia and the underlying pathomechanisms of various ciliopathies. Syndromic ciliopathies consist of a group of disorders caused by ciliary dysfunction or abnormal ciliogenesis. These disorders have multiorgan involvement in addition to retinal degeneration underscoring the ubiquitous distribution of primary cilia in different cell types. Genoty

Pleiotropic effect of common PHOX2B variants in Hirschsprung disease and neuroblastoma.

Hirschsprung disease (HSCR) is a heterogeneous congenital disorder that affects the enteric nervous system, while neuroblastoma is an embryonal tumor of the sympathetic nervous system. Familial cases of both HSCR and neuroblastoma appear to be functionally linked to PHOX2B, which plays a key role in the development of neural crest derivatives. However, the association between common PHOX2B variants and disease risk is contested. Additionally, large-scale examination for pleiotropy or shared genetic susceptibility in sporadic HSCR and neuroblastoma cases lacks theoretical support. Here, we repo

Carney complex and the familial lentiginosis syndromes: link to inherited neoplasias and developmental disorders, and genetic loci.

Lentigines, synonymous but not identical to freckles or ephelides, are common skin lesions. In a small number of patients, however, these lesions constitute part of genetic syndromes that are associated with inherited forms of neoplasias or other pathologic processes of the cardiovascular, endocrine, and gastrointestinal systems. The familial lentiginoses, as these syndromes are collectively known, include Carney complex, the LEOPARD and Peutz-Jeghers syndromes, and the newly described 'syndrome of arterial dissections with lentiginosis'; isolated, familial lentiginosis has also been described

A two-sample Mendelian randomization study of the causal relationship between respiratory diseases, gastric cancer risk, and Helicobacter pylori infection.

BACKGROUND: Gastric cancer is a major global burden, yet evidence linking respiratory diseases to gastric cancer is limited. This study examined whether genetic susceptibility to respiratory diseases including asthma, chronic obstructive pulmonary disease (COPD), idiopathic pulmonary fibrosis (IPF), and lung diseases due to external agents (LDEA) was associated with gastric cancer and Helicobacter pylori infection, given shared immune and inflammatory pathways. METHODS: We conducted a two-sample Mendelian randomization analysis using genetic variants as instrumental variables to assess causal
