Performance of different polygenic risk scores for breast cancer risk prediction: in-depth evaluations across large UK and Australian cohorts.

Polygenic risk scores (PGS) have the potential to support enhanced, risk-based screening for breast cancer. Previous studies for many diseases found that genome-wide PGS (GW-PGS) outperform PGS derived by applying hard GWAS significance thresholds. To support future breast cancer risk predictions, we compared the predictive performance of two existing PGS (including PGS313, a leading hard-thresholding PGS) and five newly developed GW-PGS (applying different methods to recent GWAS). We evaluated the performance of PGS Z-scores and of predicted 5-year absolute breast cancer risks based on age al

Perceived partner substance use, genetic predispositions, and their associations with problematic alcohol use, emotional well-being, and relationship quality.

BACKGROUND: Romantic relationships are important contexts for substance use and emotional well-being. We tested the hypotheses that (i) genetic predispositions for alcohol consumption would be positively associated with partner substance use, (ii) partner substance use would moderate genetic influences on one's own alcohol outcomes, and (iii) partner discordance in substance use would be associated with lower emotional well-being and relationship quality. METHODS: Analyses included 2,357 participants (Mage = 51.4, 58.2% female) from the Collaborative Studies on the Genetics of Alcoholism. Foc

MicroRNA Biogenesis Pathway Gene Variants Are Associated with Prostate Cancer Susceptibility.

Prostate cancer (PrC) is one of the most common malignancies among men worldwide. However, the contribution of genetic variation in microRNA (miRNA) biogenesis pathway genes to PrC susceptibility remains poorly characterized in many ethnically diverse populations. We conducted a case-control study involving 532 PrC patients and 550 controls from the Volga-Ural region of Eurasia to evaluate the association of twenty-one single nucleotide polymorphisms (SNPs) with PrC risk using single-variant and polygenic approaches. Association analyses identified rs595055 in the AGO1 gene as significantly as

Genetic investigation of the association between maternal dietary patterns and offspring ADHD.

In observational studies, an unhealthy dietary pattern during pregnancy is associated with an increased likelihood of offspring ADHD. We investigated whether such associations may be partly attributable to genetic confounding. Polygenic scores (PGSs) for a healthy dietary pattern were calculated for mother, father, and child trios in the COPSAC2010 cohort. Diagnoses and trait scores of ADHD were assessed in the children at age 10. Using trio models, we tested whether the association between maternal genetic liability to a healthy dietary pattern and offspring ADHD could be accounted for by gen

Metabolomic ageing across mental and behavioural disorders.

BACKGROUND: Individuals with mental disorders face excess morbidity and premature mortality. Accelerated ageing has been proposed as a contributing mechanism but population-scale evidence across diverse diagnoses is limited. OBJECTIVE: To examine whether metabolomic ageing differs across mental disorders and whether associations vary by sex, age group and genetic liability. METHODS: Using plasma metabolomic profiles from UK Biobank participants, we applied a metabolomic ageing clock (MileAge) to estimate disorder-specific differences between metabolite-predicted and chronological age. Mental

Evaluating a Polygenic Risk Score for Hepatocellular Carcinoma in Multiethnic Cirrhosis Patients in Texas.

BACKGROUND AND AIMS: Genome-wide association studies (GWAS) that were previously conducted in European and Asian cohorts have identified numerous genetic risk factors for hepatocellular carcinoma (HCC) and its precursors and risk factors. A polygenic risk score (PRS) composed of these key SNPs for HCC may be useful for risk stratification in clinical settings for U.S. patients with cirrhosis. METHODS: Using genetic and clinical data from the Texas HCC Consortium cohort, we evaluated a weighted PRS constructed from 11 independent SNPs previously shown to be associated with advanced liver disea

Joint Associations of Sleep Patterns and Genetic Susceptibility With Dynamic Transitions of Chronic Obstructive Pulmonary Disease and Coronary Heart Disease and the Mediating Role of Inflammatory Biomarkers and Metabolites.

BACKGROUND: Sleep behaviors influence the development of chronic obstructive pulmonary disease (COPD) and coronary heart disease (CHD), but their role in the dynamic transitions of COPD and CHD and their interaction with genetic susceptibility on the comorbidity remain unclear. METHODS: Using UK Biobank data, Cox proportional hazard models and multistate survival models were used to examine the associations of sleep patterns with the risk of dynamic transitions of COPD and CHD, and their interaction with polygenic risk score for comorbidity of COPD and CHD. Four-way decomposition models were

Hematopoietic loss of Y chromosome and risk of symptomatic peripheral artery disease in men.

BACKGROUND: Mosaic loss of chromosome Y (mLOY) is the most common acquired somatic mutation in men and has been linked to multiple age-related diseases, including cardiovascular diseases. However, its association with symptomatic peripheral artery disease (PAD) remains poorly understood. We aimed to investigate the relationship between hematopoietic mLOY and incident symptomatic PAD in a large population-based male cohort. METHODS: We conducted a longitudinal cohort study including 171 698 male participants from the UK Biobank who were free of PAD history. mLOY was assessed using leukocyte-de

Genotype prediction of adult type 2 diabetes from adolescence in a multiracial population.

BACKGROUND: Understanding the risk for type 2 diabetes (T2D) early in the life course is important for prevention. Whether genetic information improves prediction models for diabetes from adolescence into adulthood is unknown. METHODS: With the use of data from 1030 participants in the Bogalusa Heart Study aged 12 to 18 followed into middle adulthood, we built Cox models for incident T2D with risk factors assessed in adolescence (demographics, family history, physical examination, and routine biomarkers). Models with and without a 38 single-nucleotide polymorphism diabetes genotype score were

Predisposing Factors in Acute-on-Chronic Liver Failure.

Acute-on-chronic liver failure (ACLF) is a syndrome with high short-term mortality in patients with chronic liver disease. The definition of ACLF has been addressed recently in many publications, and despite regional differences the number and severity of organ failures are decisive for the presence and severity of ACLF. However, the predisposition of patients to develop ACLF has not been addressed in prospective studies. Several predisposing factors for ACLF have been mentioned, but still clear-cut analysis and the sequential processes have not been explored. One reason is that different fact

Exclusion of COL2A1 and VDR as developmental dysplasia of the hip genes.

Developmental dysplasia of the hip (DDH) is a spectrum of disorders affecting the proximal femur and/or acetabulum leading to an abnormal formation of the hip. Genetic factors are involved in the etiology of DDH. Early recognition of DDH affords the best results from treatment and a better knowledge of the genetics of DDH could enhance early diagnosis. Variants in the Type II collagen (COL2A1) and vitamin D receptor (VDR) genes have been associated with patients with osteoarthritis of the hip secondary to DDH, suggesting these genes could contribute to DDH. To see whether there was linkage bet

Existence of a genetic risk factor on chromosome 5q in Italian coeliac disease families.

Coeliac disease (CD) is a malabsorptive disorder of the small intestine resulting from ingestion of gluten. The HLA risk factors involved in CD are well known but do not explain the whole genetic susceptibility. Several regions of potential linkage on chromosomes 3q, 5q, 10q, 11q, 15q and 19q have already been reported in the literature. These six regions were analyzed with the Maximum Lod Score method on a dense set of markers. A new sample of 89 Italian sibpairs was available for study. There was no evidence for linkage for any of the regions tested, except for chromosome 5q. For this region

Polygenic Susceptibility in Peripartum, Alcohol-Induced, and Cancer Therapy-Related Cardiomyopathies.

IMPORTANCE: Rare monogenic variants linked to nonischemic dilated cardiomyopathy (DCM) are enriched among individuals with secondary cardiomyopathies, such as peripartum (PPCM), alcohol-induced (ACM), and cancer therapy-related (CCM) cardiomyopathies. However, it remains unclear whether a polygenic predisposition to DCM also contributes to these conditions. OBJECTIVE: To assess the association of a DCM polygenic score with PPCM, ACM, and CCM, and to evaluate the contributions of monogenic and polygenic susceptibilities to these secondary cardiomyopathies. DESIGN, SETTING, AND PARTICIPANTS: T

Exploration of Genetic Overlap of Brain Phenotypes With Schizophrenia: Different Methods Provide Complementary Insights.

BACKGROUND: Genetic studies have shown associations between genetic risk for schizophrenia and brain imaging phenotypes. However, prior studies focused on a single neuroimaging modality and/or employed methods that do not fully elucidate the shared genetic architecture between them, limiting our understanding of their complex genetic relationship. STUDY DESIGN: We used genome-wide association study summary statistics for schizophrenia alongside 37 brain measurements, selected based on adequate SNP-based heritability and representing structural, microstructural, and functional brain features d

Damaging missense variants in innate immunity genes are associated with earlier age of breast cancer onset in BRCA1 185delAG carriers.

BACKGROUND: Penetrance of breast cancer (BC) among women who carry pathogenic variants (PVs) in BRCA1 is incomplete, and the age at BC diagnosis varies considerably, even among carriers of the same PV, suggesting the involvement of genetic and non-genetic risk modifying factors. Polygenic Risk Score (PRS) models based on common sequence variants account for less than 10% of the total risk variability among BRCA1 PV carriers, indicating that further genetic modifiers remain to be identified. METHODS: Here, for the first time, we applied whole-exome sequencing for this challenge, investigating

MIR218 polygenic risk score is associated with cognitive function and neurochemical metabolites among patients with depressed bipolar disorders.

BACKGROUNDS: Evidence from animal and population studies has consistently revealed that microRNA 218 (MIR218) is involved in susceptibility to depression and cognitive functions. Nevertheless, few studies have evaluated the association between MIR218 and clinical features in patients with depressed bipolar disorder (BD). METHODS: A total of 66 patients with depressed BD and 49 healthy controls (HCs) were recruited for this study. MIR218 polygenic risk score (PRS) was used to assess the addictive effects of the MIR218 regulated genes. We compared the MIR218 PRS between patients with depressed

Evaluating genomic polygenic risk scores for childhood acute lymphoblastic leukemia in Latinos.

The utility of polygenic risk score (PRS) models has not been comprehensively evaluated for childhood acute lymphoblastic leukemia (ALL), the most common type of cancer in children. Previous PRS models for ALL were based on significant loci observed in genome-wide association studies (GWASs), even though genomic PRS models have been shown to improve prediction performance for a number of complex diseases. In the United States, Latino (LAT) children have the highest risk of ALL, but the transferability of PRS models to LAT children has not been studied. In this study, we constructed and evaluat
