Nicotinamide riboside treatment enhances stress sensitivity and modulates hematological dynamics in aged mice.

Nicotinamide riboside (NR) is a precursor to NAD+, a vital molecule for cellular energy metabolism whose levels decline with age. Aging is associated with loss of cognitive and immune function, alterations in hematological parameters, and increased vulnerability to stress. Although NR supplementation can mitigate age-related declines, it remains uncertain whether these positive effects persist when the organism is exposed to chronic physiological stress. We investigated this by exposing aged mice to NR supplementation alongside daily physiological stress and assessed various indices before and

Transcranial alternating current stimulation improves ballistic motor performance in trained and untrained limbs of healthy older adults.

Transcranial alternating current stimulation (tACS) using a combined theta-gamma waveform can improve unilateral ballistic motor performance in the trained limb of older adults. The purpose of this study was to examine the effect of individual (theta) and combined (theta-gamma) tACS waveforms on ballistic motor performance in the trained and untrained contralateral limb (i.e. cross-limb transfer) of older adults. Sixty right-handed healthy older adults (68.9 ± 5.2 years) received either high-definition theta-gamma (6 Hz theta, 75 Hz gamma), theta (6 Hz), or sham tACS over right primary motor c

Irradiation-induced brain senescence accelerates cardiac aging via systemic mechanisms: insights from transcriptomic profiling.

Aging is characterized by a coordinated functional decline across multiple organs. While cell-autonomous mechanisms contribute to local aging phenotypes, the systemic synchronicity of aging suggests a major role for cell non-autonomous drivers. Emerging evidence implicates the hypothalamus-a central regulator of neuroendocrine and homeostatic functions-as a potential source of circulating pro-geronic signals. A hallmark of brain aging is the accumulation of senescent cells, particularly in microglia and brain microvascular endothelial cells, including within the hypothalamus, which contributes

Sensorimotor function as an early marker of cognitive decline and alzheimer's biomarker burden.

Early identification of markers of cognitive decline in cognitively normal older adults is essential for dementia prevention strategies. Sensorimotor measures, such as gait speed, grip strength, and reaction time, may provide sensitive indicators of current and future impairment. This study examined associations between baseline sensorimotor function and cognitive decline in cognitively normal older adults. In this prospective cohort, 246 cognitively normal older adults from the DRIVES Project completed baseline assessments of grip strength, gait speed, simple reaction time, and the Preclinica

Voxel-wise insights into early Alzheimer's disease pathology progression: the association with APOE and memory decline.

Longitudinal investigation of the Apolipoprotein E (APOE) genotype's impact on Alzheimer's disease (AD) biomarker progression, focusing on amyloid beta (Aβ) accumulation and gray matter (GM) atrophy, integrating cognitive decline and baseline levels. Longitudinal florbetapir-PET and T1-weighted MRI data from 100 cognitively normal (CN) and mild cognitive impaired (MCI) participants both with considerable global Aβ accumulation ("high Aβ accumulators") were analyzed using a voxel-wise approach. Associations of APOE genotype and memory decline with Aβ accumulation and GM atrophy were examined se

Targeting angiopoietin like-2 positive senescent cells improves cognitive impairment in adult male but not female atherosclerotic LDLr-/-;hApoB100+/+ mice.

Cellular senescence contributes to cognitive decline in brain diseases. In dyslipidemic and atherosclerotic LDLr-/-;hApoB100+/+ (ATX) mice-a model exhibiting vascular dysfunctions and cognitive impairment-the role of senescence was investigated by targeting angiopoietin-like 2 (angptl2), a senescence marker. Adult ATX mice of both sexes received AAV1-sh-angptl2; cognition was assessed via the Morris Water Maze, cerebrovascular functions were evaluated in vitro and in vivo and hippocampal transcriptomic signatures were analyzed using single-nuclei RNA-sequencing. Sh-angptl2 restored delayed mem

Cognitive and neuroimaging outcome of very prodromal dementia with Lewy bodies.

The cognitive and neuroimaging evolution over the course of dementia with Lewy bodies (DLB) from prodromal stage - Pro-DLB (subjective (SCI) to mild cognitive impairment (MCI)) - is poorly understood. The aim of this study was to analyze from 5-year longitudinal data the trajectories of Pro-DLB patients. The "Lewy- MEMENTO" prospective clinical cohort recruited 773 patients for either SCI or MCI. The Pro-DLB group was compared to a group with prodromal Alzheimer's disease (Pro-AD), a group with "prodromal DLB and AD" (Pro-DLB + AD), and a group without prodromal DLB and AD (no symptom [NS]). Self-testing for symptoms and signs of prodromal alpha-synucleinopathies: results from the Tasmanian ISLAND Sleep Study.

Isolated rapid eye movement (REM) sleep behavior disorder (iRBD) may occur at least 10 years before a clinical diagnosis of an α-synucleinopathy, such as dementia with Lewy bodies (DLB), Parkinson's disease (PD) or multiple system atrophy (MSA). The extent to which people with iRBD manifest subtle features of these conditions is an area of active research and whether these changes can be measured remotely in the community is an important aim. Better characterisation may aid early detection and monitoring for people at risk of neurodegeneration. The aim of this study was to investigate remote t

Necroptosis induced by MLKL overexpression in liver triggers cellular senescence and leads to chronic inflammation and fibrosis.

Cellular senescence and necroptosis are two cell fates, which trigger an inflammatory response and increase with age, that have been proposed to play a role in inflammaging. In this study, we performed the first study to directly test the possible interaction between necroptosis and cellular senescence. Using a novel Mlkl-KI mouse model, we were able to specifically induce (~ 4-fold) the overexpression of MLKL, the necroptotic executioner, in hepatocytes (hMlkl-KI mice). The overexpression of MLKL led to increased necroptosis and cell damage/death in liver, as shown by increased levels of MLKL

High-definition tDCS of the motor cortex affects prefrontal and primary motor activity differently in young and older adults.

The ability to suppress irrelevant or distracting inputs that interfere with goal-driven behavior is known to decline with increasing age. Although noninvasive stimulation of the motor cortices has been shown to modulate age-related changes in motor activity, the findings remain preliminary, and it is unknown whether this effect extends beyond the motor cortex. In this study, 125 healthy adults, categorized into young (20-35 years) and older groups (55-72 years) underwent three visits (i.e., anodal, cathodal, and sham). During each visit, they received 20 min of high-definition transcranial di

Plasma lipidomic fingerprinting enables high-accuracy biomarker discovery for Alzheimer's disease: a targeted LC-MRM/MS approach.

Dysregulation of lipid metabolism is increasingly recognized as a key factor in the pathogenesis of Alzheimer's disease (AD). Unfortunately, an accurate lipidomic fingerprints in AD patients' biofluids remains challenging. A comprehensive analysis of plasma samples from 26 patients with AD and 30 healthy individuals was performed using untargeted and targeted lipidomics techniques with strict lipid annotation criteria. By monitoring characteristic fragments per precursor, we achieved precise lipid characterization and quantification for approximately 270 lipid species. Multivariate statistical

An mTOR inhibitor discovery system using drug-sensitized yeast.

Inhibition of the target of rapamycin (TOR/mTOR) protein kinase by the drug rapamycin extends lifespan and health span across diverse species. However, rapamycin has potential off-target and side effects that warrant the discovery of additional TOR inhibitors. TOR was initially discovered in Saccharomyces cerevisiae (yeast) which contains two TOR paralogs, TOR1 and TOR2. Yeast lacking functional Tor1 are viable but are hypersensitive to growth inhibition by TORC1 inhibitors, which is a property of yeast that can be exploited to identify TOR inhibitors. Additionally, yeast lacking FK506-sensiti

Age-independent anti-angiogenic therapy for choroidal neovascularization by targeting secretogranin III.

Recent studies reported that anti-angiogenic drugs targeting vascular endothelial growth factor (VEGF) alleviate choroidal neovascularization (CNV) in young but not aged animals. We recently developed a disease-targeted anti-angiogenic therapy against secretogranin III (Scg3), which selectively binds to diseased but not healthy vessels in young mice. Herein, using a unique in vivo ligand binding assay, we predicted and confirmed that Scg3 selectively binds CNV vessels in both young and aged mice. In contrast, VEGF with minimal increased binding to CNV vessels exhibited an age-dependent decline

Comparative phenotyping of C57BL/6J substrains reveals distinctive patterns of cardiac aging.

Research in aging often refers to animal models, particularly C57BL/6J (B6J) mice, considered gold standard. However, B6J mice are distributed by different suppliers, which results in divers substrains exhibiting notable phenotypic differences. To ensure a suitable phenotype of cardiac aging, we performed heart analyses of young (5 months) and old B6J mice (24 months) from two substrains: B6JRj (Janvier) and B6JCrl mice (Charles River). In hearts of both substrains, myocardial fibrosis increased with age; however, only in old B6JRj mice cardiac hypertrophy associated with a decreased ejection

Age differences in spontaneous cerebrovascular reactivity at rest.

Deficits in cerebral blood flow (CBF) and cerebrovascular reactivity (CVR) may indicate vulnerability to vascular dysfunction and cognitive impairment. Prior findings on age-related change in CVR are mixed, and no studies to date have investigated age group differences in spontaneous CVR (sCVR) at rest. The present study compared whole brain and regional sCVR at rest between 24 healthy younger adults (ages 18-34) and 163 cognitively unimpaired older adults (ages 55-87). Relative to younger adults, older adults had deficits in both whole brain and regional sCVR at rest. Among the regions examin

Center frequency as optimal frequency of visual stimulation for spreading entrained gamma rhythms to other target brain regions in cognitively normal older adults.

Gamma entrainment using 40 Hz sensory stimulation has shown promise in AD mouse models, but inconsistent results in AD patients, possibly due to interspecies and interindividual differences in center frequency (CF). This study aimed to investigate whether gamma rhythms entrained by visual stimulation at an individual's CF can spread better than those at other frequency conditions in older adults. We entrained gamma rhythms in 32 cognitively normal older participants using light flickering at 32 Hz, 34 Hz, 36 Hz, 38 Hz, and 40 Hz. We identified each individual's CF among these five frequencies

Association of chronic pain and analgesic use with the risks of Parkinson's and Alzheimer's disease.

Chronic pain and analgesic use are linked to increased Parkinson's disease (PD) and dementia risk, but the effects of each individually and when both are present remain unclear. This study examined whether chronic pain and analgesic use, separately and together, are associated with the incident risk of these conditions. A total of 355,709 participants (mean age 56.51, 48.4% male) free of parkinsonism and dementia at baseline were included. Chronic musculoskeletal pain lasting ≥ 3 months and regular use of analgesics, including paracetamol, non-steroidal anti-inflammatory drugs (NSAIDs), opioid

Long term cognitive outcome of prodromal and mild dementia with Lewy bodies: a cohort study.

The cognitive evolution of early dementia with Lewy bodies (DLB) is less well known than that of Alzheimer's disease (AD). During dementia, DLB progresses like AD. The aim of this study was to analyze the long-term cognitive decline of early DLB. Participants were recruited for either mild cognitive impairment or mild dementia with a suspicion of DLB or AD, or as healthy older subjects (AlphaLewyMA study, NCT01876459, 2013). Using beta regression, we compared the slope of the Mini-Mental State Examination (MMSE) score of 110 DLB patients (DLB group), 57 AD patients (AD group), 19 DLB and AD pa

Association between periodontal disease and intrinsic capacity in older adults: insights from NHANES 2009-2014.

Periodontal disease (PD) is a chronic inflammatory condition linked to systemic health issues, yet its relationship with intrinsic capacity (IC)-a key determinant of healthy aging-remained underexplored. The present study investigated the associations between moderate/severe PD and IC domains in older adults in a cross-sectional analysis from the National Health and Nutrition Examination Survey (NHANES) 2009-2014. From the 30,468 NHANES subjects, we included participants aged ≥ 60 years with available PD and IC data. IC was assessed across five domains: locomotion, cognition, psychological wel
