Anterior cingulate morphology in people at genetic high-risk of schizophrenia.

BACKGROUND: Morphological abnormalities of the anterior cingulate (AC) occur in patients with schizophrenia and in symptomatic high-risk individuals, and may be predictive of subsequent psychosis. We investigated AC sulcal morphology in the Edinburgh High Risk Study cohort to see if such abnormalities are evident and predict psychosis in patients' relatives. We also investigated the association of the cingulate sulcus (CS) and paracingulate sulcus (PCS) variants with intelligence quotient (IQ). PATIENTS AND METHODS: We compared cingulate and paracingulate sulcal anatomy, using reliable standa

N-acetylcysteine normalizes glutamate levels in cocaine-dependent patients: a randomized crossover magnetic resonance spectroscopy study.

Treatment with N-acetylcysteine (NAC) normalizes glutamate (Glu) homeostasis and prevents relapse in drug-dependent animals. However, the effect of NAC on brain Glu levels in substance-dependent humans has not yet been investigated. Proton magnetic resonance spectroscopy ((1)H MRS) was used to investigate Glu changes in the dorsal anterior cingulate cortex (dACC) after a single dose of NAC in cocaine-dependent patients and normal controls. In an open-label, randomized, crossover study, 8 cocaine-dependent patients and 14 healthy controls underwent two scan sessions: one group receiving no comp

Chronic pelvic pain syndrome in men is associated with reduction of relative gray matter volume in the anterior cingulate cortex compared to healthy controls.

PURPOSE: Although chronic pelvic pain syndrome impairs the life of millions of people worldwide, the exact pathomechanisms involved remain to be elucidated. As with other chronic pain syndromes, the central nervous system may have an important role in chronic pelvic pain syndrome. Thus, we assessed brain alterations associated with abnormal pain processing in patients with chronic pelvic pain syndrome. MATERIALS AND METHODS: Using brain morphology assessment applying structural magnetic resonance imaging, we prospectively investigated a consecutive series of 20 men with refractory chronic pel

Reversible brain white matter microstructure changes in heroin addicts: a longitudinal study.

Previous neuroimaging studies have documented the structural damage in heroin addicts. However, little research has detailed the white matter microstructural changes in the human brain as a result of chronic heroin use and importantly, whether such changes can be recovered after short-term abstinence. Decreased fractional anisotropy values in frontal cortex were found in heroin users after 3 days of abstinence in comparison with controls. However, no significant difference was found between these heroin addicts and controls after 1-month abstinence. These results might better our understanding

Common regions of dorsal anterior cingulate and prefrontal-parietal cortices provide attentional control of distracters varying in emotionality and visibility.

Top-down attentional control is necessary to ensure successful task performance in the presence of distracters. Lateral prefrontal cortex, parietal cortex and anterior cingulate cortex have been previously implicated in top-down attentional control. However, it is unclear whether these regions are engaged independent of distracter type or whether, as has been suggested for anterior cingulate cortex, different regions provide attentional control over emotional versus other forms of salient distracter. In the current task, subjects viewed targets that were preceded by distracters that varied in

Voxel-based diffusion tensor analysis reveals aberrant anterior cingulum integrity in posttraumatic stress disorder due to terrorism.

Recent functional neuroimaging work has suggested that interregional functional connectivity between the anterior cingulate cortex (ACC), other limbic, and prefrontal regions may be involved in the pathophysiology of posttraumatic stress disorder (PTSD). However, less attention has been paid to the white matter network. Voxel-based analysis enables an exploration of morphological or functional changes throughout the entire brain. Here we undertook the first application of this technology to diffusion tensor data in patients with PTSD. Participants were 9 victims of the Tokyo subway sarin attac

Prefrontal cortical projections to the striatum in macaque monkeys: evidence for an organization related to prefrontal networks.

The organization of projections from the prefrontal cortex (PFC) to the striatum in relation to previously defined "orbital" and "medial" networks within the PFC were studied in monkeys using anterograde and retrograde tracing techniques. The results indicate that the orbital and medial networks connect to different striatal regions. The ventromedial striatum (the medial caudate nucleus, accumbens nucleus, and ventral putamen) receives input predominantly from the medial PFC (mPFC) and orbital areas 12o, Iai, and 13a, which constitute the "medial" network. More specifically, caudal medial area

Apoptosis in the brains of infants suffering intrauterine cerebral injury.

This study addressed the hypothesis that in human infants severe in utero insults induce a significant proportion of brain cells to undergo apoptosis. Morphologic criteria were used to quantify apoptosis and necrosis in the cingulate gyrus of two groups of infants: six infants who died after severe birth asphyxia with hypoxic-ischemic encephalopathy, and six others who suffered unexpected and apparently sudden intrauterine death at or close to term. The fraction of apoptotic cells was much higher than basal levels determined in animal experiments, and within both groups increased in proportion

Neuroticism and Its Associated Brain Activation in Women With PTSD.

Previous research suggests a diathesis-stress model of posttraumatic stress disorder (PTSD), wherein individuals with high levels of neuroticism who are exposed to traumatic events subsequently develop PTSD. Although studies have established relationships between neuroticism and neurological functioning in various brain regions for healthy and depressed individuals, the specific neural correlates of neuroticism for individuals with PTSD are yet unknown. This relationship is particularly relevant for women, given that their increased risk for PTSD is partially accounted for by their higher base

Navigating toward a novel environment from a route or survey perspective: neural correlates and context-dependent connectivity.

When we move toward a novel environment we may learn it in different ways, i.e., by walking around or studying a map. Both types of learning seem to be very effective in daily life navigation and correspond to two different types of mental representation of space: route and survey representation. In the present study, we investigated the neural basis of route and survey perspectives during learning and retrieval of novel environments. The study was carried out over 5 days, during which participants learned two paths from a different perspective (i.e., route learning and survey learning). Then

Differential roles of EA-TRAPed cells in the anterior cingulate cortex across various intervention times in inflammatory pain.

BACKGROUND: The analgesic effects of multiple electroacupuncture (EA) sessions and single EA sessions differ significantly in pain management. Area 24b (A24b) of the anterior cingulate cortex (ACC) is crucial in pain processing. EA relieves pain by targeting and modulating the neuronal activity within this subregion. However, whether the cumulative effect of EA antinociception is connected to A24b mechanisms has remained unclear. METHODS: In our study, we used the Complete Freund's Adjuvant (CFA) model to induce inflammatory pain and the Spared Nerve Injury (SNI) model to induce neuropathic p

Asymmetry of the sulcal pattern of the anterior cingulate cortex modulates delay discounting.

Delay discounting (DD), the preference for smaller immediate over larger delayed rewards, is associated with impulsivity and cognitive control, representing a transdiagnostic process across different neuropsychiatric disorders. While previous neuroimaging studies have mainly focused on the functional neural substrates of DD, their anatomical correlates have received less attention. Based on accumulating evidence about the role of cortical sulci morphology as an indirect marker of fetal neurodevelopment, we examined the relationship between DD and the sulcal pattern of the anterior cingulate co

Topiramate and anterior cingulate cortex high-frequency stimulation mitigate aggression and promote sociability in socially isolated male mice.

Excessive aggression induced by social isolation (SI) has been closely linked to dysfunction in prefrontal circuits, particularly the anterior cingulate cortex (ACC). Topiramate, an antiepileptic drug known to enhance GABAergic and inhibit glutamatergic transmission, has demonstrated antiaggressive effects. In our previous work, we showed that topiramate's antiaggressive effects were associated with reduced neuronal death, improved neuronal morphology, and attenuated neuroinflammation in the ACC following SI. However, its impact on ACC neuronal activity remains poorly understood. In this study

Increased Cerebral Blood Flow in the Anterior Cingulate Cortex and Plasma Noradrenaline Level During Cardiac Pacemaker Stimulation.

BACKGROUND: Chest symptoms, such as angina and palpitation, are common complaints in patients with cardiovascular diseases, but few studies have addressed how cardiac afferent information is processed through the brain. METHODS AND RESULTS: We recruited 10 patients (mean age 74.7±1.9 years; 9 men) with cardiac pacemaker implantation. The patients underwent brain H215O positron emission tomography (PET) followed by blood sampling during right ventricular pacing of sham (1.5 V) and stimulation (7.5-8 V) conditions with a 10min interval. A voxel-wise analysis of the brain PET images identified t

The neural substrates responsible for reward sensitivity association with delay discounting: left dorsal anterior cingulate cortex connectivity with right precuneus.

Delay discounting refers to the tendency to devalue rewards as the delay to their receipt increases. Reward sensitivity, defined as an individual's responsiveness to rewarding stimuli, has been suggested to influence delay discounting behavior. However, the neural mechanisms underlying this association remain largely unexplored. In the present study, we investigated the neural basis of the relationship between reward sensitivity and delay discounting using voxel-based morphometry (VBM) and resting-state functional connectivity (RSFC) analyses. Behaviorally, reward sensitivity was positively co

Experience-Dependent Plasticity to Visual Sequences in Mouse Anterior Cingulate Cortex Reflects Familiarity.

Anterior cingulate cortex (ACC) is a prefrontal area implicated in functions including cognitive control, attention, and prediction. Mouse ACC receives input from the visual system and uses visual information to direct behavior. While extensive work has described experience-dependent plasticity in mouse V1, less is known about how ACC itself adapts to visual experience. Our previous work demonstrated that visual sequences, presented across days, can drive plasticity in the timing of visually evoked responses in mouse ACC. However, it is not known whether this plasticity ("sequence plasticity")

Differential brain regional activity and connectivity of anterior cingulate cortex between major depressive disorder with and without suicidal ideation under resting state.

BACKGROUND: Major depressive disorder (MDD) is closely associated with suicide, which often begins with suicidal ideation (SI). However, the underlying neural mechanisms remain unclear. METHODS: We included 73 MDD patients with SI (MDD-SI), 44 MDD patients without SI (MDD-NSI) and 78 healthy controls (HCs), then compared the amplitude of low-frequency fluctuations (ALFF), functional connectivity (FC), and effective connectivity (EC) differences across groups and analyzed their relationship with SI severity. FC and EC analyses used brain regions with ALFF differences between MDD-SI and MDD-NSI

Durability of clinical benefit with Stanford Neuromodulation Therapy (SNT) in treatment-resistant depression.

BACKGROUND: Depression, the leading cause of disability worldwide, is a chronic disease characterized by a relapsing-remitting course. Acute treatments such as electroconvulsive therapy, ketamine, and repetitive transcranial magnetic stimulation are often effective at initiating remission, but relapse to a major depressive episode is common without ongoing interventions. Stanford Neuromodulation Therapy (SNT) produces high rates of remission after five days of acute treatment; however, the duration of this remission following a single course of SNT in people suffering from treatment-resistant

Sex differences in distributed error-related neural activation in problem-drinking young adults.

BACKGROUND: Detecting and responding to errors is central to goal-directed behavior and cognitive control and is thought to be supported by a network of structures that includes the anterior cingulate cortex and anterior insula. Sex differences in the maturational timing of cognitive control systems create differential periods of vulnerability for psychiatric conditions, such as substance use disorders. METHODS: We examined sex differences in error-related activation across an array of distributed brain regions during a Go/No-Go task in young adults with problem alcohol use (N=69; 34 females;
