Identification of the C1q gene family and characterization of its transcriptional responses to low-temperature stress and Vibrio parahaemolyticus challenge in the noble scallop Chlamys nobilis.

C1q, a pivotal pattern recognition receptor (PRR) in the innate immune system, plays crucial roles in the defense of aquatic invertebrates against environmental stressors and pathogen infections. However, systematic identification and functional characterization of the C1q gene family in bivalves remain largely unexplored. In this study, leveraging thegenomic and transcriptomic analysis of the noble scallop Chlamys nobilis, we identified a total of 22 C1q family genes, comprising 9 classical C1q genes (designated CnC1q) and 13 C1q tumor necrosis factor-related protein genes (designated CnC1qtn

Investigating the effects of disposable face masks leaching compounds on marine biota: a toxicogenomic approach using stress indices and transcriptomics in tissues of mussel Mytilus galloprovincialis.

The present study provides integrated evidence that Disposable Face Masks (DFMs) leaching process could adversely affect marine bivalves, such as the mussel species Mytilus galloprovincialis. Specifically, cellular (through lysosomal membrane stability, using the Neutral Red Retention Time/NRRT), oxidative (by measuring superoxide anions/O2.-, nitric oxides/NO, and lipid peroxidation, via malondialdehyde/MDA content), and genotoxic (by measuring micronucleus/MN formation) effects were determined in hemolymph/hemocytes of mussels exposed for 96 h to artificial seawater containing DFM leaching c

Polyethylene terephthalate (PET) nanoplastics target carbonic anhydrase and alter physiological functions in Daphnia magna.

Polyethylene terephthalate (PET) nanoplastics are increasingly recognized as relevant contaminants in freshwater ecosystems, yet the molecular mechanisms underlying their sublethal biological effects remain poorly understood. The aim of this study was to investigate whether true-to-life polyethylene terephthalate (PET) nanoplastics, generated from post-consumer PET bottles, affect carbonic anhydrase (CA) activity in Daphnia magna and whether such molecular alteration is associated with physiological dysfunction at the organism level. Label-free PET nanoplastics were generated from post-consume

Synthesis of poly-γ-glutamic acid-crosslinked-pectin-based nanogel and its antioxidant and antibacterial properties.

The poly-γ-glutamic acid (γ-PGA, synthesized by glutamic acid isolated from snail hemolymph) was crosslinked with pectin to synthesize the nanogel composite (γ-PGA-cl-Pect), which was used to evaluate its swelling nature and the efficacy of its antibacterial and antioxidant properties. The average size of the particles' diameter was 173.67 ± 0.73 nm, which indicated the synthesis was effective. The techniques used to analyze the sample both quantitatively and qualitatively were Fourier Transform Infrared Spectroscopy (FTIR), Field Emission Scanning Electron Microscopy (FESEM), Nuclear Magnetic

Disruption of the core intestinal microbiota contributes to fluoride-induced neurotoxicity in the host organisms.

Fluoride is a potential environmental toxic substance associated with dental fluorosis, skeletal fluorosis, and neurotoxic effects. However, the underlying mechanisms remain poorly understood, especially concerning the potential role of fluoride-induced intestinal microbiota dysbiosis in modulating the nervous system via the gut-neuraxis. In this study, an interspecies insect model of fluoride-induced neuronal damage was established to investigate the underlying mechanisms. The results showed markedly elevated malondialdehyde levels, along with reduced glutathione content and decreased catalas

Abnormalities in trehalose homeostasis reduce autophagy, antioxidant and response to air exposure stress in Eriocheir sinensis.

Trehalose, a key carbohydrate in crustaceans and insects, is important in enhancing stress resistance. However, its regulatory mechanisms and functions in Chinese mitten crabs (Eriocheir sinensis) remain unclear. This study was designed to explore the regulation mechanisms of trehalose anabolism by synthesizing dsRNA interference fragments to target trehalose-6-phosphate synthase (TPS) and trehalose hydrolase (TREH) genes, investigating the effects of endogenous trehalose anabolism, antioxidant function, and autophagy in crabs under air exposure stress (AES). First, dsRNAs were synthesized, an

Comparative Transcriptomic Analysis Revealing the Potential Mechanisms of Erythritol-Caused Mortality and Oviposition Inhibition in Drosophila melanogaster.

Erythritol has shown excellent insecticidal performance against a wide range of insect species, but the molecular mechanism by which it causes insect mortality and sterility is not fully understood. The mortality and sterility of Drosophila melanogaster were assessed after feeding with 1M erythritol for 72 h and 96 h, and gene expression profiles were further compared through RNA sequencing. Enrichment analysis of GO and KEGG revealed that expressions of the adipokinetic hormone gene (Akh), amylase gene (Amyrel), α-glucosidase gene (Mal-B1/2, Mal-A1-4, Mal-A7/8), and triglyceride lipase gene (

Identification and Physicochemical Properties of the Novel Hemolysin(s) From Oral Secretions of Helicoverpa armigera (Lepidoptera: Noctuidae).

Hemolysins cause the lysis of invading organisms, representing major humoral immunity used by invertebrates. Hemolysins have been discovered in hemolymph of Helicoverpa armigera larvae as immune factors. As oral immunity is great important to clear general pathogens, we presumed that hemolysins may be present in oral secretions (OS). To confirm this hypothesis, we conducted four testing methods to identify hemolysin(s) in larval OS of H. armigera, and analyzed physicochemical properties of the hemolysin in comparison with hemolytic melittin of Apis mellifera (L.) (Hymenoptera: Apidae) venom. W

The Heat Shock Protein 60 and Pap1 Participate in the Sporothrixschenckii-Host Interaction.

Sporothrixschenckii is one of the etiological agents of sporotrichosis, a worldwide-distributed subcutaneous mycosis. Its cell wall contains a glycoconjugate composed of rhamnose, mannose, glucuronic acid, and proteins, named peptidorhamnomannan, which harbors important Sporothrix-specific immunogenic epitopes. Although the peptidorhamnomannan carbohydrate moiety has been extensively studied, thus far, little is known about the protein core. Here, using LC-MS/MS, we analyzed the S.schenckii peptidorhamnomannan peptide fraction and generated mass signals of 325 proteins, most of them likely to

Interspecific competition between two endoparasitoids Cotesia vestalis (Hymenoptera: Braconidae) and Oomyzus sokolowskii (Hymenoptera: Eulophidae).

Two endoparasitoids, Cotesia vestalis and Oomyzus sokolowskii, parasitize the same host, larvae of Plutella xylostella. These two species have evolved different parasitization strategies. O. sokolowskii expresses a single factor, venom, and exerts virtually no detrimental effects on the development of its host. C. vestalis, on the other hand, injects polydnavirus (PDV) and venom during oviposition, and teratocytes are released into the host's hemolymph after egg hatching. Parasitization suppresses host immune reactions and redirects its developmental program. Because both these species parasit

The hydroxyproline motif of male sex peptide elicits the innate immune response in Drosophila females.

Seminal fluid elicits a variety of physiological and behavioral changes in insect females. In Drosophila melanogaster females, sex peptide (SP) is the major seminal agent eliciting oviposition and reduction of receptivity. But SP also has many other effects; for example, it stimulates food intake, egg production, ovulation, juvenile hormone production and antimicrobial peptide synthesis. Thus, SP very probably has several receptors. To identify putative targets and signaling cascades, we studied the genome-wide regulation of genes by microarray analysis of RNA isolated from females after matin

Endemicity of spotted fever group rickettsiae in Connecticut.

To compare rickettsial infectivity and seropositivity rates against spotted fever group (SFG) rickettsiae, ticks and wild mammals were collected from three areas where Rickettsia rickettsii was thought to be enzootic in Connecticut during 1978-1979, and from four additional sites (with no reported human cases) between 1976 and 1979. Of the 1,001 Dermacentor variabilis examined by the hemolymph test, 59 (5.9%) contained rickettsia-like organisms; direct immunofluorescence tests verified the presence of SFT rickettsiae in 14 specimens. Prevalence of rickettsiae-infected ticks at Newtown, an area

A novel antibody treatment reduces deformed wing virus loads in the western honey bee (Apis mellifera).

UNLABELLED: The deformed wing virus (Iflavirus aladeformis) (DWV) is a key driver of colony loss in the western honey bee (Apis mellifera). Here, we demonstrate that orally delivered anti-DWV antibodies can act systemically to reduce DWV loads in naturally infected honey bees. Immunoglobulin Y (IgY) was produced in adult chickens against two DWV proteins, harvested from their eggs, and fed to bees in a sucrose solution. An enzyme-linked immunosorbent assay demonstrated that orally delivered anti-DWV IgY migrated to the hemolymph. We next assessed the ability of orally delivered anti-DWV IgY to

Lead optimization of antifungal peptides with 3D NMR structures analysis.

Antimicrobial peptides are key components of the innate immune response in most multicellular organisms. These molecules are considered as one of the most innovative class of anti-infective agents that have been discovered over the last two decades, and therefore, as a source of inspiration for novel drug design. Insect cystine-rich antimicrobial peptides with the CS alpha beta scaffold (an alpha-helix linked to a beta-sheet by two disulfide bridges) represent particularly attractive templates for the development of systemic agents owing to their remarkable resistance to protease degradation.

Characterization of a plasma-derived protein fraction from Magallana gigas and its modulatory effects on lipopolysaccharide-induced inflammatory responses in HT-29 cells.

A plasma-derived protein fraction (MG-1) was partially purified from the hemolymph of the Pacific oyster, Magallana gigas, and its potential modulatory effects on lipopolysaccharide (LPS)-induced inflammatory responses were evaluated in HT-29 intestinal epithelial cells. SDS-PAGE analysis revealed two major protein bands, MG-11 and MG-12, which shared identical peptide sequences based on LC-MS/MS analysis and may represent different molecular forms of the same protein. The reconstructed MG-12 sequence consisted of 192 amino acid residues and exhibited similarity to previously reported cavortin

Oil ingestion and genotoxic biomarkers in three swimming crab species (Decapoda, Portunidae) from tropical estuaries affected by oil spills.

The 2019 oil spill along the Brazilian coast represents one of the largest marine contamination events in the region, with evidence of long-term persistence and re-emergence of residues in subsequent years. This study primarily reports the ingestion of oil residues by three swimming crab species (Callinectes danae, C. exasperatus, and C. marginatus) from tropical estuaries in northeastern Brazil following oil resurgences recorded in 2022. Specimens were collected in three estuaries (Atapuz, Suape, and Carneiros) between 2022 and 2023. Oil-like fragments retrieved from stomach contents were cha

Dietary vitamin C modulates antioxidant capacity, immune responses and muscle hydroxyproline concentration in juvenile Pacific white shrimp (Penaeus vannamei).

Vitamin C (VC) plays a crucial role in regulating redox homeostasis; however, its biochemical responses in Penaeus vannamei to graded dietary levels, particularly in relation to immune, antioxidant defense and structural protein metabolism remain insufficiently characterized. This study therefore investigated the physiological and biochemical modulations and VC requirement of P. vannamei by evaluating the effects of graded vitamin C concentrations (5.99-157.78 mg/kg) on growth, immune responses, hepatopancreas vitamin C concentration and muscle hydroxyproline and collagen concentrations. Life-stage-resolved toxicological assessment and environmental relevance of triclosan in the freshwater gastropod Biomphalaria glabrata.

TCS (triclosan) is frequently detected in freshwater compartments, including surface water, sediments, biosolids, and pore water; however, its life-stage-specific toxicity in gastropod mollusks remains insufficiently characterized. To address this gap, this study aimed to determine the life-stage-dependent lethal and sublethal toxicity of TCS in Biomphalaria glabrata and identify environmentally relevant endpoints for gastropod ecotoxicology and life-stage-aware ecotoxicological assessment. We applied a life-stage-resolved framework covering embryonic development, newly hatched juveniles, acut

An upside to chronic inflammation? Starvation reconfigures immune pathways in Manduca sexta, producing chronic inflammation that may help defend against bacterial infection.

Most animals face periods of food scarcity, and pathogens are ever-present. Therefore, selection will favour animals that can maintain immune defense during starvation, despite declining resources. Using the caterpillar Manduca sexta as a model system, we show how two days of starvation reconfigures the immune system. Metabolomic and proteomic analysis of the plasma showed that starvation shifted the immune system towards a more 'pro-inflammatory' state, with increased activity in the phenoloxidase/eumelanin pathways relative to controls. Melanization occurred more rapidly in starved caterpill
