Memantine Promotes Bactericidal Effect of Neutrophils Against Infection with Pseudomonas aeruginosa and Its Drug-Resistant Strain, by Improving Reactive Oxygen Species Generation.

Pseudomonas aeruginosa is an opportunistic pathogen, which usually presents multiple antibiotic resistance. Host-directed therapy involves modulating the host defense system and the interplay between innate and adaptive immunity is a new strategy for designing anti-infection drugs. Memantine (MEM), a drug used to treat Alzheimer's disease, has a good inhibitory effect on neonatal mice with Escherichia coli-associated bacteremia and meningitis; however, the inhibitory effect and mechanisms of MEM against P. aeruginosa infection remain unclear. Here, we investigated whether MEM could inhibit P. …

Memantine Displays Antimicrobial Activity by Enhancing Escherichia coli Pathogen-Induced Formation of Neutrophil Extracellular Traps.

Bacterial infection remains one of the leading causes of death worldwide due to the continuous rise of multiple antibiotic-resistant bacteria. Focusing solely on bacteria as the drug targets is a major limitation inherent in the conventional antibiotic therapy. Recently, host-directed therapies have become such an innovative approach to modulate the host defense system and the interplay of innate and adaptive immunity. Our previous studies showed that memantine (MEM), an α7 nAChR antagonist, could efficiently block multi-drug resistant Escherichia coli-caused bacteremia and meningitis in a mou

Nanoparticulate drug delivery systems for the treatment of neglected tropical protozoan diseases.

Neglected Tropical Diseases (NTDs) comprise of a group of seventeen infectious conditions endemic in many developing countries. Among these diseases are three of protozoan origin, namely leishmaniasis, Chagas disease, and African trypanosomiasis, caused by the parasites Leishmania spp., Trypanosoma cruzi, and Trypanosoma brucei respectively. These diseases have their own unique challenges which are associated with the development of effective prevention and treatment methods. Collectively, these parasitic diseases cause more deaths worldwide than all other NTDs combined. Moreover, many current

Mechanism and efficacy of sub-50-nm tenfibgen nanocapsules for cancer cell-directed delivery of anti-CK2 RNAi to primary and metastatic squamous cell carcinoma.

Improved survival for patients with head and neck cancers (HNC) with recurrent and metastatic disease warrants that cancer therapy is specific, with protected delivery of the therapeutic agent to primary and metastatic cancer cells. A further objective should be that downregulation of the intracellular therapy target leads to cell death without compensation by an alternate pathway. To address these goals, we report the utilization of a sub-50-nm tenfibgen (s50-TBG) nanocapsule that delivers RNAi oligonucleotides directed against the essential survival signal protein kinase CK2 (RNAi-CK2) in a

Host-directed broad-spectrum immunotherapeutic strategy for respiratory infections: Heat-killed Caulobacter crescentus (HKCC) as an innate-immune based biotherapeutic/postbiotic.

Respiratory infections are among the leading causes of illness and death. The lack or limited effectiveness of vaccines for many respiratory pathogens underscores the urgent need for alternative, broadly protective strategies. While adaptive immunity is commonly used in vaccine development, the therapeutic potential of activating and enhancing innate immunity remains underutilized. Innate immune-focused interventions could offer rapid, pathogen-independent protection, bridging the gap until pathogen-specific responses develop. Therefore, exploring new broad-spectrum innate immune-targeting imm

Inhibition of virally induced TFEB proteasomal degradation as a host-centric therapeutic approach for coronaviral infection.

The endolysosomal pathway plays an evolutionarily conserved role in pathogen clearance, and viruses have evolved complex mechanisms to evade this host defense system. Here, we describe a previously unidentified aspect of coronaviral infection, whereby the master transcriptional activator of lysosomal homeostasis-TFEB-is targeted for proteasomal-mediated degradation upon viral infection. Through mass spectrometry analysis and an unbiased small interfering RNA screen, we identify that TFEB protein stability is coordinately regulated by the E3 ubiquitin ligase subunit DCAF7 and the PAK2 kinase. Seeking innovative concepts in development of antiviral drug combinations.

Antiviral drugs are crucial for managing viral infections, but current treatment options remain limited, particularly for emerging viruses. These drugs can be classified based on their chemical composition, including neutralizing antibodies (nAbs), recombinant human receptors (rhRs), antiviral CRISPR/Cas systems, interferons, antiviral peptides (APs), antiviral nucleic acid polymers, and small molecules. Some of these agents target viral factors, host factors, or both. A major challenge for virus-targeted treatments is their narrow-spectrum effectiveness and the potential for drug resistance,

Recapitulating hepatitis E virus-host interactions and facilitating antiviral drug discovery in human liver-derived organoids.

Hepatotropic viruses naturally have narrow host and tissue tropisms, challenging the development of robust experimental models. The advent of organoid technology provides a unique opportunity for moving the field forward. Here, we demonstrate that three-dimensional cultured organoids from fetal and adult human liver with cholangiocyte or hepatocyte phenotype support hepatitis E virus (HEV) replication. Inoculation with infectious HEV particles demonstrates that human liver–derived organoids support the full life cycle of HEV infection. By directing organoids toward polarized monolayers in a tr

Antiviral Strategies Targeting Enteroviruses: Current Advances and Future Directions.

Enteroviruses, a diverse genus within the Picornaviridae family, are responsible for a wide range of human infections, including hand, foot, and mouth disease, respiratory disease, aseptic meningitis, encephalitis, myocarditis, and acute flaccid paralysis. Despite their substantial global health burden and the frequent emergence of outbreaks, no specific antiviral therapies are currently approved for clinical use against non-polio enteroviruses. This review provides a comprehensive overview of the current landscape of antiviral strategies targeting enteroviruses, including direct-acting antivi

A single mycobacterial ligand organizes multi-receptor signaling to reprogram macrophage lipid metabolism.

How innate immune receptors integrate signals from complex microbial ligands remains poorly understood, yet this integration may offer new avenues for host-directed therapies. Here, we show that the architecture of a single pathogen-derived component can organize the coordinated engagement of multiple pattern-recognition receptors to reprogram host cell behavior. We find that the mycobacterial lipoglycan mannose-capped lipoarabinomannan (ManLAM) uses distinct structural features to engage two pattern-recognition receptors, Toll-like receptor 2 (TLR2) and Dectin-2, thereby driving macrophage li

Residual HIV activity and host immunometabolic remodeling during antiretroviral therapy: implications for cardiovascular-kidney-metabolic risk.

Antiretroviral therapy (ART) suppresses productive HIV replication but does not eliminate viral persistence, residual transcription, or host immune perturbation. Low-level viremia (LLV), generally referring to detectable plasma HIV RNA below the threshold for virologic failure, has traditionally been interpreted in relation to assay variability, incomplete ART exposure, resistance, or impending rebound. Persistent or recurrent LLV may signal residual HIV activity or residual antigenic exposure, but it is not a direct measure of these processes and should not be equated with ongoing productive

Rational modulation of immune mechanisms synergizes the anti-tumor effects of targeted radiation therapy in pre-clinical models.

Immunotherapy has revolutionized cancer treatment, offering new hope for many patients. However, while some individuals show remarkable responses, the overall success rate remains limited. This has spurred interest in combination therapies, particularly with established treatments like radiation therapy (RT), to improve outcomes. RT is a cornerstone of cancer therapy and known to influence the immune landscape, yet a systematic characterization of its effects on tumor-infiltrating leukocytes (TILs) and a rationale-based therapy is still lacking. In this study, we employed a diverse set of pre-

Comparative innate immune responses across major RNA and DNA viral infections: Mechanisms, immunopathology, and therapeutic perspectives.

Innate immunity is the first defense against viral infections, limiting viral replication and initiating adaptive immunity. Viral pathogens are recognized by pattern recognition receptors (PRRs), including Toll-like receptors (TLRs), (RIG-I-like receptors) RLRs, and the (cyclic GMP-AMP synthase) cGAS-STING pathway, which trigger interferon production and antiviral responses. This review compares innate immune responses to major RNA viruses (influenza, SARS-CoV-2, HIV) and DNA viruses (HSV, HBV, CMV). While these viruses activate similar pathways, they differ in interferon dynamics, inflammator

Familial 3M Syndrome - as an Example of Diagnostic Difficulties in Rare Genetic Syndromes.

Follicular lymphoma (FL) outcomes are influenced by host immune activity. CD20-directed therapy plus programmed cell death 1 inhibition (PD-1i) increases T-cell tumor killing and natural killer cell antibody-dependent cell cytotoxicity. Mounting evidence supports immune priming using PD-1i before cancer directed agents. Our multicenter, phase 2 1st FLOR study enrolled 39 patients with previously untreated advanced-stage FL to receive 4 cycles of nivolumab (240 mg), then 4 cycles of 2-weekly nivolumab plus rituximab 375 mg/m2 (induction), then 1 year of monthly nivolumab (480 mg) plus 2 years o

Development of a PROTAC-Based Targeting Strategy Provides a Mechanistically Unique Mode of Anti-Cytomegalovirus Activity.

Human cytomegalovirus (HCMV) is a major pathogenic herpesvirus that is prevalent worldwide and it is associated with a variety of clinical symptoms. Current antiviral therapy options do not fully satisfy the medical needs; thus, improved drug classes and drug-targeting strategies are required. In particular, host-directed antivirals, including pharmaceutical kinase inhibitors, might help improve the drug qualities. Here, we focused on utilizing PROteolysis TArgeting Chimeras (PROTACs), i.e., hetero-bifunctional molecules containing two elements, namely a target-binding molecule and a proteolys

FDA-Approved Amoxapine Effectively Promotes Macrophage Control of Mycobacteria by Inducing Autophagy.

Antibiotic resistance poses a significant hurdle in combating global public health crises, prompting the development of novel therapeutics. Strategies to enhance the intracellular killing of mycobacteria by targeting host defense mechanisms offer numerous beneficial effects, which include reducing cytotoxicity caused by current lengthy anti-tubercular treatment regimens and slowing or circumventing the development of multidrug-resistant strains. The intracellular pathogen Mycobacterium tuberculosis infects macrophages and exploits host machinery to survive and multiply. Using a cell-based scre

A drug repurposing screen identifies antiviral compounds against Puumala Orthohantavirus.

Hantaviruses are zoonotic negative-sense RNA viruses that cause haemorrhagic fever with renal syndrome (HFRS) and hantavirus pulmonary syndrome (HPS), yet no approved antiviral therapies are available. To identify host-directed modulators of hantavirus infection we performed a drug repurposing screen using live Puumala virus (PUUV). We identified and validated 70 drugs with antiviral activity in A549 cells and primary human endothelial cells. Functional clustering confirmed the known infection-inhibitory effect of several groups of compounds, including inhibitors of heat shock proteins, mTOR p

A Case of Cryptococcal Meningitis in an Immunocompetent Individual.

Cryptococcal meningitis is classically associated with immunocompromised hosts but can also occur in individuals without identifiable immune dysfunction. Its nonspecific and often indolent presentation may delay diagnosis, particularly when early neuroimaging suggests alternative etiologies. We report a case of a 49-year-old immunocompetent man who presented with progressive headaches, sensory changes such as transient hearing loss, visual changes, and balance issues. Initial imaging showed a hyperdense area on CT, which was interpreted as a possible cerebrovascular accident, delaying recognit

Rare Cutaneous Presentation of Disseminated Cryptococcus gattii Infection in an Immunocompetent Male: A Case Report.

Cryptococcosis is a systemic mycosis caused by Cryptococcus neoformans and Cryptococcus gattii, affecting mainly immunocompromised and immunocompetent hosts, respectively. Cutaneous involvement may indicate secondary hematogenous dissemination from systemic disease or, less frequently, primary cutaneous cryptococcosis resulting from direct inoculation without systemic infection. Although cutaneous cryptococcosis can present with diverse morphologies, cutaneous involvement is uncommon in C. gattii infection and is more frequently associated with C. neoformans. We present the case of a 56-year-o

Host Epigenetic Alterations and Hepatitis B Virus-Associated Hepatocellular Carcinoma.

Hepatocellular carcinoma (HCC) is the most frequent primary malignancy of the liver and a leading cause of cancer-related deaths worldwide. Although much progress has been made in HCC drug development in recent years, treatment options remain limited. The major cause of HCC is chronic hepatitis B virus (HBV) infection. Despite the existence of a vaccine, more than 250 million individuals are chronically infected by HBV. Current antiviral therapies can repress viral replication but to date there is no cure for chronic hepatitis B. Of note, inhibition of viral replication reduces but does not el
