GIGANTEA as a central integrator of circadian timing, stress physiology, and developmental trade-offs.

GIGANTEA (GI) has emerged as a context-dependent regulatory hub that translates circadian timing into developmental and stress-responsive outputs. Although first characterized for its role in photoperiodic flowering, GI is now recognized as a multifunctional scaffold that integrates light cues, metabolic status, hormone signaling, proteostasis, and chromatin regulation. Here, we synthesize recent mechanistic and conceptual advances to show how GI-centered networks operate across molecular, physiological, and systems levels. We first examine the structural and biochemical features that enable G

The impact of the six pillars of lifestyle medicine on the biology of skin aging.

Skin aging arises from the interaction between intrinsic genetic programs and cumulative environmental and behavioral exposures collectively defined as the exposome. Increasing evidence indicates that core molecular pathways driving skin aging, including mitochondrial dysfunction, oxidative stress, chronic inflammation, cellular senescence, and extracellular matrix remodeling are responsive to modifiable lifestyle factors. This narrative review examines the mechanistic and translational evidence linking the six pillars of Lifestyle Medicine; nutrition, physical activity, stress regulation, sle

Vascular Deletion of HDAC6 Ameliorates Diabetic Retinal Microangiopathy and Diabetic Retinopathy in an Experimental Model of Type 1 Diabetes.

Diabetic retinopathy (DR) is a leading cause of vision loss among working-age adults and is characterized by progressive retinal microvascular dysfunction driven by hyperglycemia-induced inflammation and oxidative stress. Histone deacetylase 6 (HDAC6), a cytoplasmic deacetylase involved in multiple cellular processes, has been implicated in vascular pathology; however, its specific role in retinal endothelial cells remains unclear. In this study, we investigated whether endothelial-specific deletion of HDAC6 protects against diabetic retinal injury. Endothelial HDAC6 knockout mice were generat

Seven days of warm water immersion modulate baseline fluid-regulatory hormones without altering acute multisystem physiological stress responses in older males.

Older adults exhibit heightened vulnerability to heat-related morbidity, yet it remains unclear whether short-term heat acclimation modifies integrative physiological stress responses in aging. We tested whether 7 days of passive heat acclimation via warm water immersion alters basal concentrations and acute exercise-heat stress responses of biomarkers reflecting fluid regulation, gastrointestinal epithelial integrity, renal stress, and inflammation in older males. Twelve habitually active older males (median [interquartile range] age: 68 [64-73] yr; [Formula: see text]: 34.1 [29.4-36.1] mL O2

Photobiomodulation restores blood-brain barrier integrity after hypoxia via endothelial von Willebrand factor modulation in a humanised tricellular transwell model.

A functional blood-brain barrier (BBB) is essential for CNS homeostasis, and its disruption is an early feature of both acute brain injury and chronic neurodegenerative disorders. Hypoxia induces BBB breakdown by triggering endothelial dysfunction, oxidative stress, metabolic dysregulation and thrombo-inflammatory signalling that compromise barrier integrity. However, strategies that restore BBB function remain limited. Here, we investigated whether photobiomodulation (PBM), a non-invasive light therapy, can rescue BBB dysfunction following acute hypoxic stress. Using a multicellular in vitro

What do we know about the long-term consequences of stress on ageing and the progression of age-related neurodegenerative disorders?

The aim of this paper is to review evidences that stressful events throughout life can have a long-term impact on ageing and the progression of Alzheimer's disease. As early as the prenatal or neonatal period, stress can alter the rate of cognitive decline and neurodegenerative changes in the brain in a stressor-dependent manner, with prenatal restraint and maternal separation usually causing damage to the brain, whereas neonatal handling was found protective. The occurrence of negative outcomes of early stress can, however, be reversed by subsequent events known to be beneficial to the ageing

AT1 and AT2 Receptors in the Prelimbic Cortex Modulate the Cardiovascular Response Evoked by Acute Exposure to Restraint Stress in Rats.

The prelimbic cortex (PL) is an important structure in the neural pathway integrating stress responses. Brain angiotensin is involved in cardiovascular control and modulation of stress responses. Blockade of angiotensin receptors has been reported to reduce stress responses. Acute restraint stress (ARS) is a stress model, which evokes sustained blood pressure increase, tachycardia, and reduction in tail temperature. We therefore hypothesized that PL locally generated angiotensin and angiotensin receptors modulate stress autonomic responses. To test this hypothesis, we microinjected an angioten

Identification of heat-tolerant germplasm and elucidation of heat tolerance mechanisms in polyploid rice.

Heat stress has become one of the major abiotic stresses limiting rice growth and development. Polyploid rice represents an important approach in rice breeding and may contribute to heat tolerance improvement; however, the processes by which polyploid rice responds to heat stress remain mainly ambiguous. In this study, we evaluated the heat tolerance of 40 neo-tetraploid rice lines at the seedling stage. Among them, six lines displayed high heat tolerance (HD441-HD446), and 16 lines showed low heat tolerance (HD465-HD480). Cytological observations of the two lines, HD441 and HD477, which exhib

Epitranscriptomic m6A reader HNRNPC amplifies oxidative stress-autophagy dysregulation and aggravates placental dysfunction in preeclampsia.

Preeclampsia (PE) is a placenta-driven hypertensive disorder characterized by oxidative stress, mitochondrial dysfunction, and abnormal autophagy. However, how these stress-response programs are coordinated at the post-transcriptional level remains incompletely understood. We integrated transcriptomic, proteomic, and untargeted metabolomic profiling of placentas from PE patients and normotensive controls, followed by validation in an independent clinical cohort. Mechanistic roles were interrogated using hypoxia-treated HTR-8/SVneo trophoblasts and an L-NAME-induced PE-like mouse model. Autopha

Role for HtrA in stress induction and virulence potential in Listeria monocytogenes.

In silico analysis of the Listeria monocytogenes genome revealed lmo0292, a gene predicted to encode a HtrA-like serine protease. A stable insertion mutant was constructed, revealing a requirement for htrA in the listerial response to heat, acid, and penicillin stress. Transcriptional analysis revealed that htrA is not induced in response to heat shock but is induced in response to low pH and penicillin G stress. Furthermore, htrA expression was shown to be dependent upon the LisRK two-component sensor-kinase, a system known to respond to changes in integrity of the cell envelope. In addition,

GDF15 as a sentinel of metabolic integrity: a narrative review of stress signaling, multisystem pathophysiology, and therapeutic potential.

Growth differentiation factor 15 (GDF15) is a stress responsive cytokine belonging to the transforming growth factor-β (TGF-β) superfamily, serving as a sentinel of metabolic integrity. Its expression is induced by multiple convergent stress pathways, including the integrated stress response (ISR), endoplasmic reticulum (ER) stress, oxidative stress, inflammation, and hypoxia. GDF15 is secreted by a wide range of organs, such as adipose tissue, skeletal muscle, liver, heart, kidney, immune cells, and the central nervous system, with the relative contribution of each tissue varying across diffe…

Molecular and physiological responses of black rockfish (Sebastes schlegelii) to short- and medium-term ocean acidification.

Ocean acidification (OA) is one of the greatest threats to marine species, with widespread impacts on their physiological functions. However, the adaptive capacities of many marine species to OA and the underlying mechanisms remain unclear. In this study, we investigated the effects of short-term (4 days) and medium-term (30 days) CO2 exposure (pH 8.0, 7.6, and 7.3) on black rockfish (Sebastes schlegelii), focusing on histopathological changes in gill tissues, ion transport biomarkers, oxidative stress indicators, and transcriptomic responses. The results showed that both short-term and medium

Dysregulation of mitochondrial bioenergetics, RNA processing, and cellular stress responses in FUS-ALS: a multi-dataset transcriptomic perspective.

BACKGROUND: Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative disorder characterized by motor neuron (MN) degeneration. Mutations in the FUS gene, particularly the aggressive P525L variant, disrupt nuclear localization of the FUS RNA-binding protein, resulting in cytoplasmic accumulation, altered RNA metabolism, and toxic aggregation. Astrocytes also contribute to disease progression through non-cell-autonomous mechanisms. OBJECTIVE: To identify shared and cell-type-specific molecular mechanisms underlying FUS-associated ALS through integrated transcriptomic analysis of h

Cystine/Glutamate Xc- Antiporter Induction Compensates for Transsulfuration Pathway Repression by 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) to Ensure Cysteine for Hepatic Glutathione Biosynthesis.

Exposure to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) has been associated with the induction of oxidative stress and the progression of steatosis to steatohepatitis with fibrosis. It also disrupts metabolic pathways including one-carbon metabolism (OCM) and the transsulfuration pathway with possible consequences on glutathione (GSH) levels. In this study, complementary RNAseq and metabolomics data were integrated to examine the hepatic transsulfuration pathway and glutathione biosynthesis in mice following treatment with TCDD every 4 days for 28 days. TCDD dose-dependently repressed hepatic c

Transcriptomic analysis of response to high-temperature stress in cotton.

High temperature stress (HT) is a major abiotic factor limiting global crop production. Cotton (Gossypium hirsutum L.) is particularly susceptible to heat stress, yet the underlying molecular mechanisms remain poorly understood. Here, we integrated physiological, transcriptomic, and functional analyses to elucidate the response of cotton material ZhongMianSuo087 (ZMS087) to HT. Field-based assays revealed that heat stress compromised cotton productivity by impairing pollen viability, reducing chlorophyll biosynthesis and leaf area, and altering biomass partitioning, leading to significant yiel
