New insights into iNKT cells and their roles in liver diseases.

Natural killer T cells (NKTs) are an important part of the immune system. Since their discovery in the 1990s, researchers have gained deeper insights into the physiology and functions of these cells in many liver diseases. NKT cells are divided into two subsets, type I and type II. Type I NKT cells are also named iNKT cells as they express a semi-invariant T cell-receptor (TCR) α chain. As part of the innate immune system, hepatic iNKT cells interact with hepatocytes, macrophages (Kupffer cells), T cells, and dendritic cells through direct cell-to-cell contact and cytokine secretion, bridging …

The relative importance of blood monocytes and fixed macrophages to the expression of cell-mediated immunity to infection.

Infection in mice with Listeria monocytogenes results in a substantial accumulation of migrant macrophages in the liver. The immigrant cells populate both the infective foci and intervening sinusoids. They have the labeling characteristics of blood monocytes, and their appearance in infective foci in the liver corresponds to the expression of a high level of antimicrobial immunity in this organ. The infected liver acquires additional new macrophages by Kupffer-cell division. The proliferation of these cells, however, is not essential for the expression of immunity in the liver. The results ind

Immune cells in liver regeneration: Current evidence and potential clinical targets.

The livers' ability to regenerate after injury has attracted the investigation of possible therapeutic targets for liver disease. Cells of the immune system are considered fundamental for the initiation, propagation, and termination of liver regeneration as they produce essential signaling molecules, such as cytokines, chemokines, and growth factors. Previous evidence mainly focused on macrophage involvement in liver regeneration, namely Kupffer cells which secrete mitogenic cytokines. However, recent evidence has implicated other immune cell subsets in liver regeneration including platelets, …

Selective or targeted gene/drug delivery for liver tumors: advantages and current status of local delivery.

There are various disorders involving the liver. They include metabolic diseases, hepatitis, liver cirrhosis and cancer, the latter of which may be the most serious. Delivery of therapeutic genes or drugs should be targeted to either one of the following cells in the liver: hepatocytes, Kupffer cells and tumor endothelial cells, or to the tumor cells themselves. To maximize the therapeutic effect and minimize systemic toxicity or nontarget injuries, the sufficient amount or dose of genes or drugs should be specifically delivered to a target, with minimal exposure in their active forms to nonta…

An ultrastructural study of the red pulp of the spleen and the liver in unstable hemoglobin hemolytic anemia.

Electron microscopic observations on the process of red cell destruction in the spleen and liver of a patient with congenital Heinz body hemolytic anemia, associated with a new variant of unstable hemoglobin, are reported. Two major mechanisms of destruction of Heinz body-containing red cells were noted. One was phagocytosis of these cells in toto by cordal macrophages. The other mechanism though less significant quantitatively, was intravascular hemolysis of injured red cells in the splenic microvasculature. In the liver, phagocytosis of damaged red cells by Kupffer cells was rate and there w…

[Basic mechanisms of hepatocellular injury. Role of inflammatory lipid mediators].

The presence of a lesion in the cellular parenchyma is common to a large number of chronic liver diseases, such as viral hepatitides, alcoholic hepatitis, chronic cholestasis and steatohepatitis. Although the pathogenesis may vary according to the etiological agent, a series of mechanisms is common to all. Notable among these mechanisms are Kupffer cell activation and inflammatory cell recruitment, free oxygen radical formation and the development of oxidative stress, cytokine production, mainly TNFa and TGFb, and inflammatory mediator release due to arachidonic acid oxidation through the COX-

3,4,3',4'-tetrachlorobiphenyl-induced effects in the rat liver. II. Electron microscopic autoradiographic localization of 3H-TCB.

Recent results (3) indicate that 200 mg 3,4,3',4'-tetrachlorobiphenyl induces hepatomegaly accompanied by significant decreases in serum and hepatic retinoid content and hepatocyte morphologic alterations of proliferated and vesiculated endoplasmic reticulum and megamitochondria with paracrystalline inclusions. There was also an associated change in the number, size, and distribution of lipid droplets in hepatocytes and fat-storing cells. Electron microscopic autoradiographic techniques were utilized to determine the cellular and subcellular distribution of 3H-3,4,3',4'-tetrachlorobiphenyl (3H

Ischemia-reperfusion of rat liver modulates hepcidin in vivo expression.

The recently identified acute-phase response antimicrobial peptide hepcidin has been postulated to maintain iron homeostasis by modulating iron absorption at both the intestinal and macrophage levels. Hepcidin has also been reported to be responsible for anemia associated with chronic inflammatory diseases, and in anemia in patients with hepatic adenomas. Since Kupffer cells are known to be the primary contributor to early-phase ischemia-reperfusion injury in the liver and iron is known to modulate Kupffer cell production of proinflammatory cytokine and reactive oxygen species, we investigated…

Eplerenone ameliorates the phenotypes of metabolic syndrome with NASH in liver-specific SREBP-1c Tg mice fed high-fat and high-fructose diet.

Because the renin-angiotensin-aldosterone system has been implicated in the development of insulin resistance and promotion of fibrosis in s … nduced TNFα production in the presence and absence of aldosterone were observed in primary-cultured Kupffer cells and bone marrow-derived macrophages. These results indicated that eplerenone prevented the development of NASH and metabolic abnormalities in mice by inhibiting inflammatory responses in both Kupffer cells and macrophages.

Promoter polymorphism of the CD14 endotoxin receptor gene as a risk factor for alcoholic liver disease.

Twin concordance studies indicate that genetic factors influence the individual susceptibility for alcoholic liver disease (ALD). Both clinical and experimental data suggest that Kupffer cell activation by gut-derived endotoxins and other bacterial products is an important pathogenic factor. Activated Kupffer cells release proinflammatory cytokines, a process that is regulated by the CD14 endotoxin receptor (CD14). Recently, a C-->T (-159) polymorphism in the promoter region of the CD14 gene was detected and found to confer increased CD14 expression. In the present study, the association of CD…

A preliminary investigation into the immune cell landscape of schistosome-associated liver fibrosis in humans.

Schistosomiasis is a widespread helminth disease that poses a heavy social and economic burden on people worldwide. Advanced schistosomiasis … lated to Fc gamma R-mediated phagocytosis and antigen processing and presentation. Overall, CAVIN2+ Kupffer cells were the main source of TGFB1, consisting primarily of mononuclear phagocytes in the livers of the SJ group subjects and potentially playing an irreplaceable role in hepatic fibrosis of schistosomiasis.
