Cardiotonic steroid ouabain stimulates steroidogenesis in Leydig cells via the α3 isoform of the sodium pump.

Cardiotonic steroids such as ouabain are potent inhibitors of the sodium pump and have been widely used for centuries in the treatment of congestive heart failure. In recent decades, however, they have also been identified as hormone-like molecules that trigger signaling cascades of physiological relevance by using the various sodium pump α subunit isoforms as receptors. The murine Leydig cell line MLTC-1 expresses both the ubiquitous, relatively ouabain-insensitive α1 isoform of the sodium pump and the ouabain-sensitive α3 isoform that is normally found in neuronal cells. The physiological re

Lysosomal acid lipase-deficient mice: depletion of white and brown fat, severe hepatosplenomegaly, and shortened life span.

Lysosomal acid lipase (LAL) is essential for the hydrolysis of triglycerides (TG) and cholesteryl esters (CE) in lysosomes. A mouse model created by gene targeting produces no LAL mRNA, protein, or enzyme activity. The lal-/- mice appear normal at birth, survive into adulthood, and are fertile. Massive storage of TG and CE is observed in adult liver, adrenal glands, and small intestine. The age-dependent tissue and gross progression in this mouse model are detailed here. Although lal-/- mice can be bred to give homozygous litters, they die at ages of 7 to 8 months. The lal-/- mice develop enla

The role of magnesium as antithrombotic therapy.

Meta-analysis of previous relatively small clinical trials, comparing intravenous magnesium with placebo in acute myocardial infarction (AMI) patients, mainly without thrombolytic therapy, demonstrated that magnesium reduced in-hospital mortality by 19%, mainly by reducing the incidence of serious arrhythmias and left ventricular heart failure by one quarter. These findings have led us to hypothesize that magnesium treatment inhibits platelet-dependent thrombosis in patients with coronary artery disease (CAD). In a prospective, double blind, and crossover study, we have recently demonstrated t

How adaptive solutions from marine mammal life history could address pressing problems in reproductive biomedicine.

Trends in modern society show an increase in maternal age due to deferred childbearing, and assisted reproductive technologies often make th … ease feeding activities during portions of their reproductive cycle. Strong selective pressures for lipid mobilization to support energetic demands during fasts result in reversible insulin resistance, similar to gestational diabetes mellitus. Second, marine mammals extensively forage as they recover from fasts, engaging in repeated, long-duration dives. This expose …

Liver dysfunction and energy storage mobilization in traíra, Hoplias malabaricus (Teleostei, Erythrinidae) induced by subchronic exposure to toxic cyanobacterial crude extract.

Microcystins (MC) are hepatotoxic for organisms. Liver MC accumulation and structural change are intensely studied, but the functional hepatic enzymes and energy metabolism have received little attention. This study investigated the liver and hepatocyte structures and the activity of key hepatic functional enzymes with emphasis on energetic metabolism changes after subchronic fish exposure to cyanobacterial crude extract (CE) containing MC. The Neotropical erythrinid fish, Hoplias malabaricus, were exposed intraperitoneally to CE containing 100 μg MC-LR eq kg-1 for 30 days and, thereafter, the

Adipose tissue dysfunction in cancer cachexia.

Cancer cachexia is a complex disorder that is driven by inflammation and metabolic imbalances, resulting in extreme weight loss. Adipose tis … nce suggests that, during cancer cachexia, WAT undergoes a browning process, resulting in increased lipid mobilization and energy expenditure. In this review, we have summarized the characteristics of cancer cachexia and WAT browning. Furthermore, this review describes how adipose tissue becomes inflamed in cancer, shedding light on the combinatorial action of multi …

Overview of Bile Acids Signaling and Perspective on the Signal of Ursodeoxycholic Acid, the Most Hydrophilic Bile Acid, in the Heart.

Bile acids (BA) are classically known as an important agent in lipid absorption and cholesterol metabolism. Nowadays, their role in glucose regulation and energy homeostasis are widely reported. BAs are involved in various cellular signaling pathways, such as protein kinase cascades, cyclic AMP (cAMP) synthesis, and calcium mobilization. They are ligands for several nuclear hormone receptors, including farnesoid X-receptor (FXR). Recently, BAs have been shown to bind to muscarinic receptor and Takeda G-protein-coupled receptor 5 (TGR5), both G-protein-coupled receptor (GPCR), independent of th

Age-associated changes in the serum level of apolipoproteins A-I and A-IV and the gene expression as revealed by fasting and refeeding in mice.

Alterations of serum apolipoproteins A-I (apo A-I) and A-IV and their mRNAs in young and old mice by fasting and refeeding were investigated by polyacrylamide gel elecrophoresis and Northern blot, respectively. After fasting for three days, serum apo A-I concentration in young mice (6-9 month-old) was increased about 1.5 fold while that of old animals (25-34 month-old) did not change significantly. Apo A-I mRNA was increased about 3-fold and 1.7-fold in the liver and small intestine of the young mice, respectively. The increase in old animals was not more than 1.5-fold in both tissues. The ser

Lipostatic and ischymetric mechanisms originate dexfenfluramine-induced anorexia.

Rats treated with physiological saline or dexfenfluramine (Isoméride, Laboratoires SERVIER, Neuilly, France) (dF), 1.75, 3.50, and 7.00 mg/kg, were studied in a computer-controlled open-circuit metabolic chamber, in which temperature was regulated at 24 degrees C. Total metabolic rate (TMR), respiratory quotient (RQ), locomotor activity (LA), energy cost of LA, and thus, locomotor free metabolic rate, were scanned at 10-second intervals throughout 22-hour uninterrupted recording sessions. The feeding pattern was also measured in relation to the dF-induced changes in the above parameters. It wa

Inhibition of adenosine 3':k'-monophosphate accumulation white fat acids, lactate, and beta-hydroxybutyrate.

The large increase in cyclic AMP accumulation by rat white fat cells seen in the presence of lipolytic agents plus methylxanthines and adenosine deaminase was markedly inhibited by lactate. However, lipolysis was unaffected by lactate. Octanoate, hexanoate, heptanoate, and beta-hydroxybutyrate inhibited both cyclic AMP accumulation and lipolysis by rat fat cells. The mechanism by which these acids inhibit lipolysis differs from that for long chain fatty acids such as oleate. Oleate directly inhibited triglyceride lipase activity of homogenized rat adipose tissue. In contrast, octanoate, beta-h

Inferred Mobility-Resolved Resistome Architecture Suggests Recurrent Co-Resistance Modules on a Conserved Chromosomal Backbone in Multidrug-Resistant Escherichia coli from Intensive Swine Production in Hungary.

Background: Multidrug-resistant (MDR) Escherichia coli in intensive pig production represents a persistent animal health and One Health concern. Here, we integrated quantitative phenotypic susceptibility data with whole-genome sequencing (WGS) to characterize the resistome and its inferred genomic context (chromosomal vs. plasmid-predicted contigs and mobile genetic element (MGE)-proximal regions) in swine-associated MDR E. coli from Hungary. Methods: A total of 203 E. coli isolates from large-scale pig farms were tested by broth microdilution. Based on resistance-oriented screening from an ex

Intramuscular Fat as a Biomarker of Muscle Health: Strengths, Limitations and Open Challenges.

Intramuscular fat (IMF), also known as myosteatosis, refers to the accumulation of lipids within and around skeletal muscle fibres. Excess IMF is increasingly recognised as a sign of poor muscle quality, linked to reductions in strength, power and mobility, and associated with adverse outcomes in metabolic disease, osteoarthritis, cardiovascular diseases and cancer. It is important to note that an increase in intramuscular fat is not always pathological. These associations and the ongoing uncertainty have sparked growing interest in IMF as a biomarker of muscle health beyond muscle mass and cr

Influenza A Virus Binding to α,2-3- and α,2-8-Linked Sialo-Gangliosides Reconstituted in Phase-Separated Vesicles.

We investigated the binding of influenza A viruses (IAVs) to gangliosides containing either α,2-3- or α,2-8-linked sialic acid residues, or a combination of both. The respective gangliosides were reconstituted into phase-separated giant unilamellar vesicles to mimic the cellular membrane environment. Dual-color fluorescence imaging of influenza particles interacting with these vesicles revealed that the order of virus binding density and the lateral mobility of bound particles does not correlate with (i) the intrinsic affinity of viral hemagglutinin (HA) for specific sialic acid conformations

Synergistic thermosonication combined with germinants and licorice extract improves microbial inactivation, physicochemical quality, and volatile profile stability of ready-to-eat chicken breasts.

This study evaluated thermosonication combined with germinants and licorice extract (GTL) in the "germination-inactivation-inhibition" strategy to control spore contamination and preserve the quality of ready-to-eat (RTE) chicken breasts. The microbial counts, color, pH, texture, lipid oxidation, and volatile profile of RTE chicken breasts were assessed. The result demonstrated that the GTL treatment significantly reduced the total viable counts (TVC) and Clostridium counts, with TVC remaining below 4 log CFU/g and Clostridium counts lower than in the control (4.42 vs. 7.31 log CFU/g) by day 6

Green HPTLC-densitometry for simultaneous determination of antibiotic residues in milk with greenness assessment and application to real samples.

A validated HPTLC densitometric approach was employed for the simultaneous determination of Cefazolin (CFZ), Sulfadimidine (SDD), and Marbofloxacin (MFC) residues in milk. The method utilizes a mobile phase composed of ethyl acetate, methanol, and triethylamine in a 50:30:1 v/v/v ratio, with detection carried out at 270 nm. The chromatographic conditions were carefully optimized, and the retardation factor (Rf) values for CFZ, MFC, and SDD were 0.29, 0.58, and 0.73, respectively. Sample preparation included protein precipitation with acetonitrile, followed by dispersive solid-phase extraction

Resolving Spectral Complexity in 4D Lipidomics Using a Two-Dimensional Deconvolution Framework.

The application of data-independent acquisition (DIA) in 4D lipidomics has been constrained by spectral interference due to fragment ion overlap, a bottleneck that existing one-dimensional deconvolution methods fail to fully resolve. Here, we overcome this limitation by introducing a two-dimensional liquid chromatography-ion mobility (LC-IM) deconvolution framework that unlocks the full potential of 4D lipidomics. By mathematically modeling the orthogonal LC-IM separation dimensions, our method reconstructs high-quality MS/MS spectra from highly complex DIA data, effectively disentangling co-e

Mitochondrial Dysfunctions in Human Primary Coenzyme Q10 Deficiencies.

Coenzyme Q10 (CoQ10) is an essential lipid-soluble molecule that plays a central role in mitochondrial energy production as a mobile electron carrier. In addition to its bioenergetic function, CoQ10 participates in antioxidant defense, redox homeostasis, lipid and nucleotide metabolism, and mitochondrial quality control. Primary CoQ10 deficiencies are rare inherited mitochondrial disorders caused by pathogenic variants in nuclear genes involved in CoQ10 biosynthesis. These defects lead to reduced CoQ10 levels and impaired mitochondrial functions. Clinically, primary CoQ10 deficiencies display
