High resolution physically based modelling reveals malaria incidence reduction by vector control measures.

Malaria continues to cause over 600,000 deaths annually in sub-Saharan Africa, disproportionately affecting children under five. Despite sustained control efforts, transmission remains highly sensitive to local environmental and climatic variability, underscoring the need for physically grounded models capable of capturing these dynamics. To address this challenge, we developed a high-resolution hybrid modeling framework linking WRF/WRF-Hydro and VECTRI. The framework integrates atmospheric, hydrological, ecological, and intervention processes at 1 km and 50 m resolutions and includes a new co

A multicentric study on understanding the bionomics of Indian malaria vectors across diverse eco-epidemiological settings.

BACKGROUND: India aims to eliminate malaria by 2030; however, a thorough understanding of the current biology and behavior of vector species will facilitate the efforts. Vector species often alter their biting and resting behaviors in response to long-term chemical control measures, posing significant challenges to ongoing vector control interventions. Therefore, it is essential to investigate and update our knowledge of the bionomics of malaria vectors in the current context. METHODS: This study was carried out across 14 districts in eight Indian states between 2021 and 2023, employing vario

The symbiotic Wolbachia in Anopheles and its role in reducing the transmission of Plasmodium: updates and prospects.

Reducing malaria transmission and the prospects for vector control include multi-pronged strategies, such as interrupting the parasite cycle in both vectors and mosquitoes. Effective vector control remains essential to prevent malaria transmission. This is all the more important as problems such as resistance to insecticides and the lack of a highly effective malaria vaccine remain. New generation vector control measures and optimised products are essential to address the public health needs for malaria eradication. Strategies to reduce malaria transmission include the use of insecticide-treat

Field evaluation of drone and AI assisted larval source management in Ghana.

BACKGROUND: Malaria remains a major public health burden in sub-Saharan Africa. In Ghana, in particular, larval source management (LSM) is increasingly recognized as a complementary vector control strategy. This study evaluates a field-adapted LSM approach that integrates drone-based mapping and artificial intelligence (AI)-driven site prioritization to enhance operational efficiency and reduce resource use. METHODS: The intervention replaces conventional manual scouting with aerial mapping conducted one day prior to larvicide application. An AI model analyzes geospatial and morphological fea

Optimizing insecticide deployment strategies to delay quantitative resistance in mosquito populations.

The large-scale use of insecticides remains a cornerstone of malaria vector control, but its long-term effectiveness is undermined by the evolution of quantitative insecticide resistance (qIR) in mosquito populations. We develop and analyze a mathematical model to identify optimal deployment strategies for two insecticides that differ only in their relative efficacy against target mosquito populations. Resistance is represented as a continuous phenotypic trait influencing mosquito fecundity and mortality, and the model accounts for successive deployment periods. Our results show that when muta

Chlorfenapyr-pyrethroid nets for pyrethroid-resistant malaria vectors: efficacy, resistance risks, and policy implications.

The Global Technical Strategy for Malaria 2016-2030 aims to reduce malaria incidence and mortality by 90%, yet widespread pyrethroid resistance among major malaria vectors in sub-Saharan Africa threatens this goal. Thus, the World Health Organization recommends chlorfenapyr-pyrethroid combination nets as a priority intervention where pyrethroid resistance undermines vector control. This systematic review synthesizes evidence on the performance, emerging resistance risks, and policy implications of these next-generation insecticide-treated nets. A structured search of literature from 2010 to 20

Spatio-temporal agent-based modelling of malaria.

Plasmodium falciparum is responsible for the majority of malaria morbidity and mortality each year. Malaria transmission rates vary by location and time of year due to climate and environmental conditions. We show the impact of these factors by developing a stochastic spatiotemporal agent-based malaria model that captures the impact of spatially distributed interventions on malaria transmission. Our model uses spatiotemporal estimates of mosquito climatic suitability and household location data to model the interaction between human and mosquito agents. We apply our model to investigate how st

Variance partitioning reveals contrasting random effect contributions to the density and species composition of malaria-transmitting mosquitoes in western Burkina Faso.

BACKGROUND: Spatial-temporal variation exists in the density and species composition of malaria-carrying mosquitoes, which will in turn influence the transmission of the disease. While there has been extensive research on seasonality and other main drivers of the vector populations, the heterogeneity partitioned as random effects at various spatial-temporal scales is just as important but has not attracted the same attention. METHODS: To investigate the relative contributions of the between-house, between-village and between-year variations, as well as other house-level covariates such as inh

Changes in malaria vector bionomics in Timor Leste between 2009 and 2016; a longitudinal study.

BACKGROUND: Malaria was the leading cause of morbidity in Timor Leste in the past which gradually declined from 223,002 cases in 2006 to 14 cases in 2020, with zero malaria deaths since 2013 leading to malaria-free certification in June 2025. We compare the changes in vector bionomics due to implementation of vector control methods and other related environmental modifications in the country between 2009 and 2016. METHODS: Anopheline adults were collected from four sites using seven standard sampling methods in 2009 and 2016: full- and partial-night indoor human landing catches and outdoor hu

Modelling the impact of mosquito bed net utilization on malaria transmission and evolution of pyrethroid resistance.

Long-lasting Insecticide Nets (LLINs) are central to malaria prevention, but their effectiveness is threatened by the increasing pyrethroid resistance and low bed net utilization in malaria-endemic regions globally. This study formulates a genotype-specific compartmental model accounting for malaria transmission. It incorporates the blocking and insecticidal killing effects of LLINs, and pyrethroid-induced fitness costs, to evaluate the impact of LLIN utilization on malaria and resistance dynamics. Reproductive numbers with and without LLINs were derived, and sensitivity analysis performed. Th

Insecticide resistant Anopheles from Ethiopia but not Burkina Faso show a microbiota composition shift upon insecticide exposure.

BACKGROUND: Malaria remains a key contributor to mortality and morbidity across Africa, with the highest burden in children under 5. Insecticide-based vector control tools, which target the adult Anopheles mosquitoes, are the most efficacious tool in disease prevention. Due to the widespread use of these interventions, insecticide resistance to the most used classes of insecticides is now pervasive across Africa. Understanding the underlying mechanisms contributing to this phenotype is necessary to both track the spread of resistance and to design new tools to overcome it. METHODS: Here, we c

Meta-analysis on the entomological effects of differentially treated ITNs in a multi-site experimental hut study in sub-Saharan Africa.

BACKGROUND: Restricting the placement of active ingredients (AIs) to specific panels on insecticide-treated nets (ITNs) has the potential to reduce the amount of AI required to treat a net. If the restricted placement of the AIs can exploit mosquito behaviour, particularly where they interact with the bed net interface, and not impact the net's effectiveness, then the reduction in AI could result in cost reductions. METHODS: Nine individual experimental hut trials were conducted to compare the efficacy of three different partially-treated net relative to fully treated nets; roof-only treated

First large-scale assessment of pyrethroid resistance in Anopheles darlingi (Diptera: Culicidae) in Brazil (2021-2024): a crucial step in informing decision-making in malaria control.

BACKGROUND: Malaria continues to pose a significant public health threat in northern Brazil. Current control strategies for Anopheles darlingi, the primary malaria vector in the Amazon region, depend on long-lasting insecticidal nets (LLINs) and indoor residual spraying (IRS) with pyrethroid insecticides. Despite decades of insecticide use, there are very few records of pyrethroid resistance in this mosquito species in Brazil, likely due to a lack of investigations, underscoring the urgent need for further actions. OBJECTIVES: To assess the susceptibility of An. darlingi from all malaria-ende

Climate and human-driven changes in malaria vector distribution in China: implications for Anopheles species management.

BACKGROUND: Malaria, transmitted by Anopheles mosquitoes, remains a major global health threat. While China achieved malaria-free certification in 2021, the country remains vulnerable to imported cases, a risk exacerbated by climate change and human activities. Accurate predictions of malaria vector distributions are essential for effective prevention and management. RESULTS: We employed the Bayesian additive regression trees (BART) model to assess the current distribution of Anopheles sinensis and Anopheles lesteri in China, incorporating recent occurrence data alongside climatic, soil, huma

Early evening outdoor biting by malaria-infected Anopheles arabiensis vectors threatens malaria elimination efforts in Zanzibar.

BACKGROUND: The Zanzibar Malaria Elimination Programme relies on insecticide-treated nets as the principal vector control method, supplemented by reactive focal indoor residual spraying. Despite the success, local malaria transmission persists, and the underlying reasons for sustained transmission remain unclear, yet critical to optimizing vector control for elimination. Entomological characterization of transmission dynamics was conducted to identify the gaps with existing interventions and opportunities for complementary interventions. METHODS: Adult malaria vectors were collected monthly f

Elevating larval source management as a key strategy for controlling malaria and other vector-borne diseases in Africa.

Larval source management (LSM) has a long history of advocacy and successes but is rarely adopted where funds are limited. The World Health Organization (WHO) guidelines on malaria prevention recommend the use of LSM as a supplementary intervention to the core vector control methods (insecticide-treated nets and indoor residual spraying), arguing that its feasibility in many settings can be limited by larval habitats being numerous, transient, and difficult to find or treat. Another key argument is that there is insufficient high-quality evidence for its effectiveness to support wide-scale imp

What is the optimal In2Care stations density to achieve Aedes aegypti population reduction in a dengue endemic setting?

BACKGROUND: Autodissemination traps are among the most innovative strategies for suppressing mosquito vector populations. These traps are particularly effective against Aedes aegypti due to the species' skip oviposition behavior, where eggs from a single clutch are distributed across multiple breeding sites. Evaluating the efficacy of different densities of In2Care stations under large-scale field conditions is crucial for understanding their potential impact on Ae. aegypti populations. METHODOLOGY/PRINCIPAL FINDINGS: A total of 3,250 In2Care stations were deployed in Goiânia, the 10th larges
