Validation of a multiplex RT-PCR assay for screening significant oncogene fusion transcripts in children with acute lymphoblastic leukaemia.

In childhood acute lymphoblastic leukaemia (ALL), cytogenetics play an important role in diagnosis, allocation of treatment and prognosis. Conventional cytogenetic analysis, involving mainly karyotyping in our experience, has not been successful in a large proportion of cases due to inadequate metaphase spreads and poor chromosome morphology. Our aim is to develop a highly sensitive and specific method to screen simultaneously for the four most frequent fusion transcripts resulting from specific chromosomal translocations, namely, both the CML- and ALLtype BCR-ABL transcripts of t(9;22), E2A-P

Multiplex Ligation-Dependent Probe Amplification (MLPA) for the Detection of Copy Number Mutations in the DMD Gene.

The multiplex ligation-dependent probe amplification (MLPA) assay is designed to simultaneously screen all 79 DMD gene exons for copy number variation in Duchenne and Becker muscular dystrophy (DMD/BMD) patients. Large deletions or duplications are found as mutations in >65% of DMD/BMD patients, making the MLPA assay a leading genetic diagnostic tool for patients and their families. This assay requires only standard molecular biology, PCR amplification equipment and access to a capillary electrophoresis instrument for sizing fluorescently labeled amplicons. MLPA is in wide use for copy number

The Multiplex qPCR Assay Enables Simultaneous Detection, Differentiation, and Quantification of Feline Bocavirus-1, -2, and -3 in Clinical Samples.

Feline bocaviruses (FBoVs) are increasingly recognized for their global distribution and potential importance in feline health. Three distinct species-FBoV-1, FBoV-2, and FBoV-3-have been identified; however, the lack of efficient molecular assays has limited the ability to clarify their species distribution, prevalence, and viral loads. In this study, we developed a multiplex quantitative PCR (qPCR) assay targeting the NS1 gene for the simultaneous detection, differentiation, and quantification of all three FBoV species. The assay demonstrated high analytical sensitivity, with limits of detec

Resolving diagnostic complexity: long-read sequencing unmasks a Hong Kongαα/‒α4.2 genotype mimicking a heterozygous ‒α3.7 deletion on multiplex ligation-dependent probe amplification.

INTRODUCTION: Complex structural variants in thalassemia are rare and often undetectable or misdiagnosed by conventional genetic testing methods. We report a case initially misclassified by multiplex ligation-dependent probe amplification (MLPA) as a heterozygous ‒α3.7 deletion that was subsequently correctly identified by third-generation sequencing (TGS) as the Hong Kongαα (HKαα)/‒α4.2 genotype. METHODS: Gap polymerase chain reaction (PCR) was used to screen for 4 common deletional α-thalassemia variants (-SEA, -THAI, ‒α3.7, and ‒α4.2). A PCR-reverse dot blot detected 3 nondeletional mutati

A multiplex bacterial assay for identifying vaginal fluid in various individual states and mixture stains.

Accurate identification of vaginal fluid is imperative for forensic investigations, particularly in resolving sexual assault cases. Body fluid-specific microbiota are considered potential indicators for identifying body fluids. In this study, we selected vaginal core bacteria based on species level and developed a multiplex PCR system for identifying vaginal fluid in various individual states and mixed stains. This system was validated using a set of 350 samples (245 vaginal samples and 105 other body fluid samples). The system demonstrated high sensitivity, stability against inhibitors, and s

Titanium Dioxide Solar Photocatalytic Microbial Inactivation Assessment Utilizing Viability Tests and a Novel Triplex qPCR Assay for Nucleic Acid Degradation Determination.

Photocatalytic oxidation of microorganisms is a powerful alternative to established disinfection approaches, applicable to a variety of water matrices. Bacterial vegetative cells, spores, fungi, and viruses, represent potential biopathogens and photocatalysis targets. Inactivation efficiency is usually evaluated by assessing viability through culture. However, additional inactivation assessment approaches are needed, as some microbes, despite being unculturable, remain metabolically active and pathogenic. Nucleic acid quantification approaches (qPCR) can assess nucleic acid release and degrada

Enhancing One Health Through Wastewater-Based Surveillance: Development and Validation of a Multiplex ddPCR Tool to Differentiate Human and Livestock Contributions.

Wastewater surveillance has emerged as valuable tool for public health, with notable successes in monitoring SARS-CoV-2 and, more recently, antimicrobial resistance (AMR) genes. Traditionally focused on human populations, this approach also holds significant promise for One Health surveillance. However, the extent to which livestock contributes to the microbial and genetic content detected in wastewater, compared to human sources, remains unclear. This gap in knowledge hampers our ability to normalize and accurately interpret wastewater surveillance data, particularly when integrating AMR gene

Diagnostic Accuracy of Multiplex NAAT/PCR and Culture Against Salmonella spp.: A Comparison of Meta-Analytical Methods.

BACKGROUND: Non-typhoidal (NT) Salmonella spp. constitutes a major cause of foodborne illness. Culture is the gold standard, but it is time consuming, whereas multiplex nucleic acid amplification tests (NAATs)/Polymerase Chain Reaction (PCR) offer faster detection with variable reported performance. OBJECTIVES: To compare the diagnostic accuracy of multiplex NAAT/PCR and culture for Salmonella spp. using various statistical models with or without a gold standard assumption. METHODS: A systematic search (PubMed, Web of Science, Scopus; up to April 2024) identified 44 studies (55 comparisons).
