Chiglitazar prevents diabetes-induced skeletal muscle loss by enhancing myogenic differentiation through the MEK/ERK pathway.

Long-term diabetes markedly impairs skeletal muscle mass and function, contributing to adverse clinical outcomes. Chiglitazar (CHI), a pan-PPAR agonist, effectively reduces blood glucose levels and suppresses lipid synthesis. This study examined the protective effects of CHI against muscle loss in type 2 diabetes (T2D) using clinical data and an in vitro model involving C2C12 myoblasts cultured under high-glucose (HG) conditions. In vivo efficacy was evaluated in db/db mice. Clinical analyses revealed that CHI improves muscle quality and strength. In vitro, HG conditions inhibited myotube fusi…

Embryological approach to philtral hair sparsity in bilateral cleft lip deformities.

OBJECTIVE: In bilateral cleft lip cases, surgeons have long known that the medial nasal prominence or prolabium is devoid of muscle. Despite … erentiation potential are arrested on each side of the cleft. Therefore, a reduced concentration of myoblasts and myocytes as well as hormone receptors in the medial process could hinders the normal physiological development of the different layers of the philtral skin and especially the skin appendages. CONCLUSION: Philtral hair sparsity in patients with …

The histone code reader Spin1 controls skeletal muscle development.

While several studies correlated increased expression of the histone code reader Spin1 with tumor formation or growth, little is known about physiological functions of the protein. We generated Spin1M5 mice with ablation of Spin1 in myoblast precursors using the Myf5-Cre deleter strain. Most Spin1M5 mice die shortly after birth displaying severe sarcomere disorganization and necrosis. Surviving Spin1M5 mice are growth-retarded and exhibit the most prominent defects in soleus, tibialis anterior, and diaphragm muscle. Transcriptome analyses of limb muscle at embryonic day (E) 15.5, E16.5, and at

An evaluation of leukaemia inhibitory factor as a potential therapeutic agent in the treatment of muscle disease.

The exogenous delivery of growth factors and cytokines is a potential therapeutic strategy to alleviate the degenerative effects of primary inherited myopathies such as Duchenne muscular dystrophy. The mdx mouse diaphragm is a model for examining the progressive degeneration of dystrophic muscle. We have delivered leukaemia inhibitory factor to the mdx diaphragm using slow release alginate gels. Previous studies have reported an improvement in the histology of mdx diaphragms after delivery of leukaemia inhibitory factor in a similar manner, but little attention has been paid to the mechanism b

C2C12-Derived ApoVs Promote Skeletal Muscle Development and Ameliorate Age-Related Muscle Loss Through Igf1r/PI3K/AKT/mTOR Pathway.

BACKGROUND: Apoptosis coincides with the differentiation of skeletal myoblasts, and numerous studies have shown that the apoptotic activity is required for myogenic differentiation. Although the role of apoptosis in skeletal muscle differentiation has been well documented, its mechanism is largely unknown. METHODS: Apoptotic extracellular vesicles (apoVs) were extracted from differentiated C2C12 cells or STS-treated undifferentiated C2C12 myoblasts. C2C12 myoblasts, 8-week-old male mice or 15-month-old male mice were used as in vitro and in vivo models, respectively. These models were treated…

Comprehensive analysis of miRNA expression, secretion profile, and functional network during myogenesis in response to spaceflight.

MicroRNAs (miRNAs) are key regulators of skeletal muscle development and homeostasis. As secretory factors, previous studies have identified and validated a space flight-associated circulating microRNA (miRNA) signature in response to space flight. However, the muscle-derived miRNAs are not clear. Here, we comprehensively analyzed the variations of miRNA during mouse myoblast myogenesis in response to space flight, including expression and secretion. We validated some miRNA expression by qRT-PCR analysis in the limb muscle of a disuse-induced atrophy mouse model, mimicking the unloading follow

Restoration of myogenesis in ALS-myocytes through miR-26a-5p-mediated Smad4 inhibition and its impact on motor neuron development.

Amyotrophic lateral sclerosis (ALS) is the most common adult-onset paralytic disorder, characterized primarily by a progressive loss of moto … ine signaling in association with miR-26a-5p that, secreted from wild-type (WT) and captured by ALS myoblasts, enhances ALS-related myoblast differentiation by repressing Smad4-related signals. Moreover, miR-26a-5p and -431-5p work in a paracrine way ameliorating motoneuron differentiation. These findings emphasize the need to better understand intercellula …

Duchenne muscular dystrophy cell culture models created by CRISPR/Cas9 gene editing and their application in drug screening.

Gene editing methods are an attractive therapeutic option for Duchenne muscular dystrophy, and they have an immediate application in the generation of research models. To generate myoblast cultures that could be useful in in vitro drug screening, we have optimised a CRISPR/Cas9 gene edition protocol. We have successfully used it in wild type immortalised myoblasts to delete exon 52 of the dystrophin gene, modelling a common Duchenne muscular dystrophy mutation; and in patient's immortalised cultures we have deleted an inhibitory microRNA target region of the utrophin UTR, leading to utrophin u…

Phospholipase A(2) enhances the endothelial cell detachment effect of a snake venom metalloproteinase in the absence of catalysis.

Microvessel disruption leading to hemorrhage stands among the most dangerous consequences of envenomings by snakes of the family Viperidae. A PIII metalloproteinase (SVMP), balteragin, purified from the venom of the snake Bothrops alternatus, displays a potent hemorrhagic effect, and a moderate myotoxicity in vivo. Previous studies described the ability of this SVMP to induce the detachment of C2C12 myoblasts in culture, without causing cytolysis. Surprisingly, a purified acidic phospholipase A2 (PLA2) from the same venom was found to increase this detaching activity of the SVMP on myoblasts. …

Radicicol enhances the regeneration of skeletal muscle injured by crotoxin via decrease of NF-kB activation.

This study evaluated cellular and molecular effects of radicicol, a heat shock protein (HSP) inducer, on the regeneration of skeletal muscle injured by crotoxin, the main toxin isolated from Crotalus durissus terrificus venom. Regenerating muscles treated with radicicol had decreased NF-kB activation. Differentiating myoblasts treated with radicicol showed reduced number of NF-kB positive nuclei and increased fusion index. The results suggest that radicicol enhances regeneration of muscle by attenuating NF-kB activation and increasing myogenic differentiation.

Spheroid-based generation of 3D myogenic tissues using transgene- and sorting-free differentiation of hiPSCs.

Human induced pluripotent stem cells (hiPSCs) represent a clinically relevant cell source for skeletal muscle tissue engineering, yet transgene-free maturation remains challenging. Moreover, the potential of spheroids as building blocks for muscle engineering remains poorly defined. This study evaluated whether hiPSC-derived myogenic spheroids enhance biofabrication outcomes in an in vitro bio-artificial muscle (BAM) system compared to a conventional single cell approach.
Methods. CD56+ hiPSC-derived myoblasts were generated using a transgene-free protocol and differentiated toward 2D myot…
