Biopsy of muscle from 13 cases of human polymyositis permitted an ultrastructural study in which attention was directed to the cellular population of the inflammatory reaction. The constant feature was the presence of small and large lymphocytes, and cells of the latter type were frequently in intimate contact with the surface of the muscle fibres. Macrophages were seen both within and outside degenerating muscle fibres. Plasma cells were seen in 2 cases. Filaments bearing some resemblance to virus inclusions were seen in one case. The implications of the cellular reaction in human polymyositi

Contraction of both striated and smooth muscles is regulated by intracellular Ca2+ signals. The mechanisms of contraction following the Ca2+ signal are significantly different between striated and smooth muscles. The Ca2+-dependent reversible phosphorylation of the 20 kDa myosin light chain plays an essential role in regulating smooth muscle contraction. The phosphorylation theory of smooth muscle contraction was proposed in the mid-1970s. In the late 1980s, quantitative evaluation of cytosolic Ca2+ concentrations and contraction revealed that the contraction was not always associated with cha

Advanced glycation end products (AGEs) are consumed in massive amounts with daily diets, among which high molecular weight protein-bound AGEs, such as Myofibrillar protein-bound Nε-(carboxymethyl)lysine (MP-CML), resist gastrointestinal absorption. This study investigated MP-CML metabolism by human gut microbiota using an in vitro colonic fermentation model. MP-CML digest was separately incubated with faecal suspensions from six volunteers (F1-F3, M1-M3). The F3 sample exhibited the highest activity, reducing CML from 146.54 ± 1.45 μg/mL to 68.62 ± 2.06 μg/mL after 72 h, corresponding to a rat

The N-terminal extension of human cardiac troponin T (TnT), which modulates myofilament Ca2+ sensitivity, contains several hypertrophic cardiomyopathy (HCM)-causing mutations including S69F. However, the functional consequence of S69F mutation is unknown. The human analog of S69F in rat TnT is L71F (TnTL71F). Because the functional consequences due to structural changes in the N-terminal extension are influenced by the type of myosin heavy chain (MHC) isoform, we hypothesized that the TnTL71F-mediated effect would be differently modulated by α- and β-MHC isoforms. TnTL71F and wild-type rat TnT

Nemaline myopathy, the most common congenital myopathy, is characterized by mutations in genes encoding myofilament proteins such as skeletal α-actin. These mutations are thought to ultimately lead to skeletal muscle weakness. Interestingly, some of the mutations appear to be more potent in males than in females. The underlying mechanisms remain obscure but may be related to sex-specific differences in the myofilament function of both limb and respiratory muscles. To verify this, in the present study, we used skeletal muscles (tibialis anterior and diaphragm) from a transgenic mouse model harb

The diagnosis of an inflammatory myopathy is often established based on basic histologic studies. Additional immunohistochemical studies are sometimes required to support the diagnosis and the classification of inflammatory myopathies. Staining for major histocompatibility complex 1 (MHC1) often shows increased sarcolemmal labeling in inflammatory myopathies. Endomysial capillary staining C5b-9 (membrane attack complex) is a feature that is reported as frequently associated with dermatomyositis. Immunohistochemical staining for C4d is widely used for various applications including the assessme

The current study reports the first family with confirmed myofibrillar myopathy MFM in the Middle East and the third family worldwide. This study highlights the importance of considering MFM in young patients presenting with idiopathic cardiomyopathy, arrhythmia or atrioventricular block in the Gulf states. This is the first report that presented 2 different types of cardiomyopathy and 2 different indications of permanent pacemaker placement in the same generation of a family with MFM. This report studies a Qatari family consisting of one brother and 3 sisters. The brother had restrictive card

BACKGROUND: Anesthetic-induced malignant hyperthermia (MH) in humans and pigs is associated with dramatic alterations in cardiac function. However, it remains controversial as to whether MH-associated cardiac symptoms represent a primary difference of myocardium or a secondary alteration consequent to increases in the hyperthermic stress. Here the authors describe changes in myosin isoform expression in the hearts of MH-susceptible pigs with and without prior exposure to halothane.
METHODS: One group of pigs was diagnosed as MH susceptible by halothane challenge and Hal-1843 nucleotide examin

A histochemical study of cytochrome c oxidase (CCO) was performed in the muscles from eight patients with full-blown zidovudine myopathy. All patients had ragged-red fibres (total cumulative count: 160) and myofilamentous changes, that predominated in type 1 fibres and included diffuse or punch-out myofibrillar loss (75 affected fibres) and constant cytoplasmic body formation (106 affected fibres). Inflammatory infiltrates were present in four out of eight patients. A partial CCO deficiency (22-47% of fibres; both types 1 and 2 affected) was detected in all cases, and contrasted with the norma

Attention has focused on possible defects at the level of the contractile proteins in myocardial hypertrophy and failure. In small mammals such as the rat and rabbit, myofibrillar and myosin ATPases are depressed in situations of decreased contractility. Converse increases in enzymic activity together with enhanced contractility are seen in the hyperthyroid state. The molecular basis for these changes in enzymic activity is now known to result from transitions in three heavy chain isoenzymes of myosin, V1, V2 and V3 which possess different ATPase activities. Transitions in isoenzyme compositio

A 25-year-old woman with a non-familial congenital nonprogressive myopathy was found to have atypical core-like lesions in type 1 muscle fibers. Typical core lesions (approximately 13 micrometers in diameter) and smaller, PAS positive (4.1 micrometers in diameter) atypical core were associated with a predominant type 1 fibre myopathy. A specific deficiency of fructose 1, 6-diphosphatase was found with normal values for nine other muscle glycolytic and mitochondrial marker enzymes. The data provide evidence for a specific muscle enzyme deficiency in a patient with atypical central core disease.
