Hertwig's epithelial root sheath, enamel matrix proteins, and initiation of cementogenesis in porcine teeth.

OBJECTIVES: The aim of this study was to analyze the association between Hertwig's epithelial root sheath (HERS) cells, enamel matrix proteins (EMPs), and cementogenesis. MATERIAL AND METHODS: Porcine teeth were examined at the beginning of root formation by light and transmission electron microscopy. Colloidal gold immunocytochemistry was used to analyze the protein expression of amelogenin and ameloblastin. RESULTS: Before and during disintegration of HERS, its cells displayed the cytologic features of protein synthesis and secretion. While some cells assumed an ameloblast-like phenotype,

Influence of aging on tooth eruption: experimental canine mandibular allograft.

PURPOSE: Aging is clinically related to tooth eruption; however, there are no known studies that have elucidated the relationship. We examined whether tooth eruption would occur normally in a mature subject. MATERIALS AND METHODS: Using vascularized composite tissue mandibular transplantation, we extracted portions of immature mandibles including the tooth germs from young beagle dogs and placed them into unrelated immature and mature beagle dogs. We then examined eruption of the lower first molar in the grafted mandibular bone and compared the results clinically, radiographically, and histol

The developmental biology of cementum.

In conclusion, we have reviewed an extensive literature on early cementogenesis and performed a detailed morphological and molecular analysis to illustrate and verify key issues in the current debate about epithelial and mesenchymal contributions to root cementum. We have demonstrated that prior to cementogenesis, Hertwig's epithelial root sheath disintegrates and dental follicle cells penetrate the epithelial layer to invade the root surface. Our studies confirmed that HERS became disrupted or disintegrated prior to cementum deposition. We visualized how mesenchymal cells from the dental foll

The relationship between dental age, bone age and chronological age in 54 children with short familial stature.

Patients with short familial stature (SFS) have a short body height, but their bone age is normal. The aim of this research was to assess dental maturity in 54 patients with SFS, aged from 4 to 15 1/2 years. Bone age was judged from radiographs of the left wrist, and dental age from orthopantomograms. Dental age was retarded in relation to chronological age and to bone age.

Microradiography and fluorescence microscopy of bone remodeling on the basal crypt of permanent mandibular premolars in dogs during eruption.

Alveolar bone of erupting teeth was studied in order to define the types of calcified tissues deposited as well as the rate of tooth growth. The third (P3) and fourth (P4) mandibular premolars of 30 dogs aged 12-24 weeks were analyzed by microradiography and microscopy in fluorescent and ordinary light. The bone plate separating P3 and P4 from the mandibular canal presented a complex arrangement of lamellar and woven bone, and even of chondroid tissue. During the pre-eruptive phase, this plate shifted towards the base of the mandible by means of selective resorption and apposition activities.

The severity of dental fluorosis in children exposed to water with a high fluoride content for various periods of time.

Inhabitants of the Ikeno district of Japan were accidentally exposed to drinking water containing 7.8 ppm fluoride (F) for 12 years, after which water with 0.2 ppm F was substituted. Dental examinations of local inhabitants revealed that only children aged seven years or less at the introduction or aged 11 months or more at the removal of the high-F water had fluorosis. Regular inspections were made of the 86 children between those age limits. The severity of fluorosis in three tooth types (first permanent molars, upper central incisors, and first premolars) was assessed and related to the per

Fluoride therapy in clinical practice.

Research into the use of fluorides is dynamic and ongoing. As a consequence, anyone interested in clinical application must be up to date in the current literature. A number of other trace elements have also been found to have impact, particularly on the disease of dental caries. Therefore, to treat oral disease effectively with these products, it is important that the clinician keep current with published studies. Fluorides are, at present, a major therapeutic measure in the treatment of both dental caries and periodontal disease. However, fluorides will only reduce disease, not eliminate it.

Traumatized primary anterior teeth. Prognosis related to calcific reactions in the pulp cavity.

The frequency of a complicating pulp necrosis and the process of the physiologic root resorption were studied in traumatized primary teeth exhibiting partial or total pulp obliteration. The material comprised 88 incisors in 72 children aged 0.7--5.7 years (mean 2.9 years) at the time of injury. Trauma had resulted either in subluxation (25 teeth), or luxation (13 teeth), whereas the type of injury was unknown in 50 teeth. All cases were observed until eruption of the permanent incisors. Forty-four teeth initially displayed a reversible greyish color. The ultimate finding observed in all teeth

LncRNA TP53TG1 promotes the growth and osteo/dentinogenic differentiation of dental pulp stem cells by activating the Smad3 and JNK1/2 pathway.

TP53TG1 is a long non-coding RNA related to the TP53 gene, which plays an important role in various biological processes such as tumorigenesis, cell cycle regulation, and DNA damage repair. In recent years, researchers have begun to explore the role of TP53TG1 in dental pulp biology, especially its potential impact on pulpitis and other pulp-related diseases. However, the role of TP53TG1 in human dental pulp stem cells (hDPSCs) remains unclear. In this study, we obtained TP53TG1 knockdown dental pulp stem cells by plasmid transfection to determine the biological role of TP53TG1 in DPSCs. We fo

Progressive tooth pattern changes in Cilk1-deficient mice depending on Hedgehog signaling.

Primary cilia function as critical sensory organelles that mediate multiple signaling pathways, including the Hedgehog (Hh) pathway, which is essential for organ patterning and morphogenesis. Disruptions in Hh signaling have been implicated in supernumerary tooth formation and molar fusion in mutant mice. Cilk1, a highly conserved serine/threonine-protein kinase localized within primary cilia, plays a critical role in ciliary transport. Loss of Cilk1 results in severe ciliopathy phenotypes, including polydactyly, edema, and cleft palate. However, the role of Cilk1 in tooth development remains

Root resorption of primary molars and dental development of premolars in children with Osteogenesis Imperfecta medicated with bisphosphonates, grouped according to age and gender.

BACKGROUND: Osteogenesis imperfecta (OI) is an inherited disorder characterized by bone fragility and skeletal alterations. The administration of bisphosphonates (BPs) to patients with OI reduces pain, thereby improving their quality of life. The main mechanism of action of BPs is the inhibition of osteoclast action. In the oral cavity of children with OI during growth and development, physiological processes that require the function of osteoclasts occur. The aim of this investigation was to study the dental development of premolars and the root resorption of primary molars in children with O

Tooth Development Associated with Mutations in Hereditary Vitamin D-Resistant Rickets.

Hereditary vitamin D-resistant rickets (HVDRR) is a rare genetic disorder caused by mutations at the level of the vitamin D receptor ( VDR) gene. The disease is characterized by refractory hypocalcemia, elevated serum levels of 1,25-dihydroxy-vitamin D, retarded growth, sparse body hair (sometimes alopecia), premature tooth loss, enlarged pulp chambers, thin dentine, and hypoplastic enamel. The aims of this study were 1) to document the dental development of children with HVDRR in association with the mutation type within the VDR and 2) to evaluate the association between dental development an

New bone formation at the site of autotransplanted developing mandibular canines: a case report.

Until recently, adequate bone at the recipient site has been a determinant for tooth autotransplantation. This article presents the transplantation of developing mandibular canines to a suboptimal position because of inadequate bone at the recipient sites. Five years later, after eruption and orthodontic alignment, all criteria for successful healing were fulfilled. In addition, new bone that was not present at the time of surgery was identified at the roots of the transplanted teeth, implying that transplanted developing teeth can stimulate bone to grow along their roots.

Replantation of an avulsed permanent maxillary incisor with an immature apex: report of a case.

A case is reported of the replantation of a maxillary incisor with an immature apex following a traumatic avulsion. A 14-month follow-up clinical examination revealed the patient to be asymptomatic, the tooth to be still functional, and a recall radiograph showed no evidence of renewed periradicular breakdown. The indications for, and limitations of, replantation of an avulsed permanent incisor with an immature apex are discussed.

Genes affecting tooth morphogenesis.

The development of dentition is a fascinating process that encompasses a complex series of epithelial-mesenchymal interactions involving growth factors, transcription factors, signal receptors and other soluble morphogens. It is not surprising that such a complex process is prone to disturbances and may result in tooth agenesis. Initial discoveries indicating that the homeo-domain protein MSX1 and the paired-domain transcription factor PAX9 are causative genes in tooth morphogenesis were made in mice. Both genes are co-expressed in dental mesenchyme and either one, when homozygously deleted, r
