Regulation of Diacylglycerol Content in Olfactory Neurons Determines Forgetting or Retrieval of Olfactory Memory in Caenorhabditis elegans.

Proper management of memories by forgetting and retrieval is essential for animals to adapt their behavior to changing environments. To elucidate the mechanisms underlying forgetting, we use olfactory learning to an attractive odorant, diacetyl, in Caenorhabditis elegans hermaphrodites as a model. In this learning paradigm, the TIR-1/JNK-1 pathway in AWC sensory neurons accelerates forgetting of the olfactory memory, which is stored as a sensory memory trace in AWA sensory neurons. Our genetic screening revealed that increased neuronal diacylglycerol in the olfactory neuronal circuit, by mutat

Effects of chronic peripheral olfactory loss on functional brain networks.

The effects of sensory loss on central processing in various sensory systems have already been described. The olfactory system holds the special ability to be activated by a sensorimotor act, without the presentation of an odor. In this study, we investigated brain changes related to chronic peripheral smell loss. We included 11 anosmic patients (eight female, three male; mean age, 43.5 years) with smell loss after an infection of the upper respiratory tract (mean disease duration, 4.64 years) and 14 healthy controls (seven female, seven male; mean age, 30.1 years) in a functional magnetic res

An infant with macrocephaly, abnormal neuronal migration and persistent olfactory ventricles.

An asphyxiated male infant was delivered by cesarean section at 38 weeks gestation. The patient was macrocephalic with clinical evidence of fetal immobility syndrome. He died aged two days. Autopsy revealed excessive brain weight, pachygyria, enlarged olfactory tracts and hypoplastic optic nerves. Microscopy revealed evidence of impaired neuronal migration throughout the cortex. The olfactory bulbs showed persistence of the primitive olfactory ventricles and absence of olfactory glomeruli. There was persistence of the fetal connection between lateral and olfactory ventricles. The lateral genic

Induction of tyrosine hydroxylase expression in rat forebrain neurons.

Olfactory nerve input is required for the normal expression of tyrosine hydroxylase (TH) by dopaminergic neurons in the glomerular region of the rodent main olfactory bulb. To determine whether the olfactory nerve exerts a similar influence on neurons in other brain regions, we performed unilateral bulbectomies in rat pups on postnatal day 5-7 and examined the brains 2-6 months later, after the regenerated olfactory nerve had penetrated the forebrain. Tissue was stained for TH, dopamine beta-hydroxylase (DBH) and olfactory marker protein immunoreactivity. We observed novel TH-immunoreactivity

The olfactory-glymphatic syndrome: linking smell dysfunction, cognitive impairment and sleep disturbances in neuropathological disorders.

Olfactory dysfunction is a common feature of several neuropathological disorders - including Alzheimer's and Parkinson's diseases - whose shared profile often includes glymphatic dysfunction, cognitive impairment, and sleep disturbances. Importantly, smell loss frequently precedes cognitive or motor symptoms by years, highlighting its potential as an early clinical marker. This article reviews mechanisms that may link olfactory dysfunction to disease development in a wide range of neuropathological diseases and advances a research-based hypothesis for a shared etiologic pathway underlying thes

Odor Enrichment Increases Hippocampal Neuron Numbers in Mouse.

The hippocampus and olfactory bulb incorporate new neurons migrating from neurogenic regions in the brain. Hippocampal atrophy is evident in numerous neurodegenerative disorders, and altered hippocampal neurogenesis is an early pathological event in Alzheimer's disease. We hypothesized that hippocampal neurogenesis is affected by olfactory stimuli through the neural pathway of olfaction-related memory. In this study, we exposed mice to novel pleasant odors for three weeks and then assessed the number of neurons, non-neuronal cells (mainly glia) and proliferating cells in the hippocampus and ol

Transolfactory neuroinvasion by viruses threatens the human brain.

Viral neuroinvasion via the olfactory system has been investigated in a variety of virus-animal models by scientists in many fields including virologists, pathologists, and neurologists. In humans, herpes simplex virus type 1 (HSV-1), human herpesvirus 6 (HHV-6), Borna disease virus, rabies virus, and influenza A virus have been shown to take the olfactory route for neuroinvasion based on forensic and post-mortem specimens. This article briefly summarizes the anatomy, physiology, and immunology of the olfactory system and presents a battery of neurovirulent viruses that may threaten the human

The diversified function and potential therapy of ectopic olfactory receptors in non-olfactory tissues.

Olfactory receptors (ORs) are mainly distributed in olfactory neurons and play a key role in detecting volatile odorants, eventually resulting in the production of smell perception. Recently, it is also reported that ORs are expressed in non-olfactory tissues including heart, lung, sperm, skin, and cancerous tissues. Interestingly, ectopic ORs are associated with the development of diseases in non-olfactory tissues. For instance, ectopic ORs initiate the hypoxic ventilatory responses and maintain the oxygen homeostasis of breathing in the carotid body when oxygen levels decline. Ectopic ORs in

Olfactory combinatorial coding supports risk-reward decision-making in Caenorhabditis elegans.

Olfactory-driven behaviors are essential for animal survival, but mechanisms for decoding olfactory inputs remain poorly understood. We have used whole-network Ca2+ imaging to study olfactory coding in Caenorhabditis elegans. We show that the odorant 1-octanol is encoded combinatorially in the periphery as both an attractant and a repellent. These inputs are integrated centrally, and their relative strengths determine the sensitivity and valence of the behavioral response through modulation of locomotory reversals and speed. The balance of these pathways also dictates the activity of the locom

Identification of chemosensory genes and antennal sensilla in Nassophasis sp. (Coleoptera: Rhynchophorinae).

Phytophagous insects rely on plant volatiles to select and locate hosts for feeding or reproduction and their olfactory system is essential for detecting plant volatiles. The stem-boring pest, Nassophasis sp. damages Dendrobium and causes economic losses. Currently, there are no effective methods for its control. However, understanding the morphological and molecular basis of its olfactory system may identify new pathways for their management and control. In this study, we observed the stemborer's antennal sensilla using scanning electron microscopy, and transcriptome sequencing was undertaken

Behavioral and neurobiological implications of kairomones for rodents: an updated review.

Like many other vertebrates, rodents communicate via pheromones, which favors intraspecies communication. In contrast, kairomones are semiochemicals involved in interspecific communication, facilitating information between organisms of different species but advantageous for the receiver. Kairomones induce behavioral, physiological, and endocrinological changes in rodents, and have been proven to activate specific neuronal pathways in one or multiple components of the olfactory system (the main olfactory system, accessory olfactory system, and Gruenberg ganglion). The sophisticated olfactory ne

Chlamydia pneumoniae can infect the central nervous system via the olfactory and trigeminal nerves and contributes to Alzheimer's disease risk.

Chlamydia pneumoniae is a respiratory tract pathogen but can also infect the central nervous system (CNS). Recently, the link between C. pneumoniae CNS infection and late-onset dementia has become increasingly evident. In mice, CNS infection has been shown to occur weeks to months after intranasal inoculation. By isolating live C. pneumoniae from tissues and using immunohistochemistry, we show that C. pneumoniae can infect the olfactory and trigeminal nerves, olfactory bulb and brain within 72 h in mice. C. pneumoniae infection also resulted in dysregulation of key pathways involved in Alzheim

Perforated Patch-clamp Recording of Mouse Olfactory Sensory Neurons in Intact Neuroepithelium: Functional Analysis of Neurons Expressing an Identified Odorant Receptor.

Analyzing the physiological responses of olfactory sensory neurons (OSN) when stimulated with specific ligands is critical to understand the basis of olfactory-driven behaviors and their modulation. These coding properties depend heavily on the initial interaction between odor molecules and the olfactory receptor (OR) expressed in the OSNs. The identity, specificity and ligand spectrum of the expressed OR are critical. The probability to find the ligand of the OR expressed in an OSN chosen randomly within the epithelium is very low. To address this challenge, this protocol uses genetically tag

Multifaceted sublethal effects of neurotoxic insecticides on olfactory-mediated orientation in the spotted wing drosophila.

Neurotoxic insecticides are recognized and classified by their primary mode of action, but they frequently exhibit secondary targets and downstream consequences that are often neglected. Hazards to arthropod physiology and behavior can sublethally occur through environmentally relevant pesticide residues and/or by contact with non-targeted developmental stages. Drosophila suzukii is a key invasive pest of soft-skinned fruits and provides a suitable model to study how neurotoxicants interfere with olfactory-mediated orientation, a key process underlying host detection and oviposition. However,

Optimized Electrode Placements for Non-invasive Electrical Stimulation of the Olfactory Bulb and Olfactory Mucosa.

The olfactory system is known to be dysfunctional in the early stages of Parkinson's disease (PD) and Alzheimer's disease (AD). It is also shown that intact olfactory function can be a key role player for regaining consciousness after brain injuries. Modulation of the olfactory regions has been attempted successfully with electrical stimulation over the years, either directly (transethmoidally, intraoperatively, internasally, etc.) or indirectly through the vagus nerve. We sought to develop a means of delivering optimized electrical stimulation to the olfactory region in a non-invasive fashion

Mechanical constraints on cell plasticity: insights from the olfactory epithelium.

The olfactory epithelium (OE) is a unique model to study how mechanical forces and tissue architecture regulate stem cell plasticity. In vertebrate, its lifelong neurogenesis and regenerative capacity depend on horizontal basal cells and globose basal cells, whose plasticity is gated by niche architecture and morphogenetic cues. While molecular pathways in OE development and regeneration are well-characterized, the role of physical forces, from basal niche stiffness to lamellar folding, remains unexplored. This review integrates single-cell analyses, developmental studies, and mechanobiology t

Changes in expression of the low affinity receptor for neurotrophins, p75NGFR, in the regenerating olfactory system.

We have disrupted the integrity of the rat olfactory neuroepithelium using intranasally applied TX-100, a procedure known to reversibly eliminate the sensory neuron input from the neuroeithelium to the olfactory bulb [Margolis et al. (1974) Denervation in the primary olfactory pathway of mice: biochemical and morphological effects. Brain Res. 81, 469-483]. One week after TX-100 exposure, we observed a disruption of the pseudo-stratified organization of the neuroepithelium which was accompanied by a 60% reduction in neuroepithelial width, compared to saline-treated controls. Full recovery of th

A sex comparison of the anatomy and function of the main olfactory bulb-medial amygdala projection in mice.

We previously reported that some main olfactory bulb (MOB) mitral/tufted (M/T) cells send a direct projection to the "vomeronasal" amygdala in female mice and selectively respond to volatile male mouse urinary odors. We asked whether MOB M/T cells that project to the vomeronasal amygdala exist in male mice and whether there is a sexually dimorphic response of these neurons to volatile male urinary pheromones. Gonadectomized male and female mice received bilateral injections of the retrograde tracer, Cholera toxin-B (CTb) into the medial amygdala (Me), which is part of the vomeronasal amygdala.

Functional classification and central nervous projections of olfactory receptor neurons housed in antennal trichoid sensilla of female yellow fever mosquitoes, Aedes aegypti.

Mosquitoes are highly dependent on their olfactory system for, e.g. host location and identification of nectar-feeding and oviposition sites. Odours are detected by olfactory receptor neurons (ORNs) housed in hair-shaped structures, sensilla, on the antennae and maxillary palps. In order to unravel the function of the olfactory system in the yellow fever vector, Aedes aegypti, we performed single-sensillum recordings from trichoid sensilla on female antennae. These sensilla are divided into four distinct morphological types. Based on the response to a set of 16 odour compounds, we identified 1

Olfactory impairment in motor neuron disease: a pilot study.

The ability to identify smells was tested in nine males and six females with motor neuron disease (MND) of varying severity, using the University of Pennsylvania Smell Identification Test (UPSIT). The olfactory impairment found in MND patients compared with age and sex matched controls is statistically significant at the 0.005 level. The relationship with Parkinson's disease, with Alzheimer's dementia and the possible aetiological implications of this new aspect of the MND are discussed.
