Flavobacterium psychrophilum isolates, obtained from ayu, Plecoglossus altivelis, three species of salmonids and two species of cyprinids in Japan, were used in this study. Bacteria were inoculated to serum prepared from ayu or red spotted masu trout (RSMT), Oncorhynchus masou ishikawae, and incubated at 18 °C for 24 h. All isolates (n = 19) from ayu grew well with a 9- to 116-fold increase of CFU in ayu serum, while CFU decreased markedly in RSMT serum. In contrast, isolates (n = 17) from fish species other than ayu exhibited no growth in ayu serum, but some isolates from salmonids survived o

Microplastics are prevalent in aquatic ecosystems, impacting various forms of aquatic life, including fish. In this study, Rainbow trout (Oncorhynchus mykiss) were exposed to two concentrations of microplastics (0 and 500 μg/L) over a 14-day period, during which a comprehensive analysis was conducted to assess the liver accumulation of microplastics and their effects on oxidative stress, the liver response, and transcriptomics. Our findings indicated that microplastics significantly accumulated in the liver and activated the antioxidant system in fish by enhancing the activity of antioxidant e

Parasites manipulate their hosts' phenotype to increase their own fitness. Like any evolutionary adaptation, parasitic manipulations should be costly. Though it is difficult to measure costs of the manipulation directly, they can be evaluated using an indirect approach. For instance, theory suggests that as the parasite infrapopulation grows, the investment of individual parasites in host manipulation decreases, because of cost sharing. Another assumption is that in environments where manipulation does not pay off for the parasite, it can decrease its investment in the manipulation to save res

Cross-boundary nutrient inputs can enhance and sustain populations of organisms in nutrient-poor recipient ecosystems. For example, Pacific salmon (Oncorhynchus spp.) can deliver large amounts of marine-derived nutrients to freshwater ecosystems through their eggs, excretion, or carcasses. This has led to the question of whether nutrients from one generation of salmon can benefit juvenile salmon from subsequent generations. In a study of 12 streams on the central coast of British Columbia, we found that the abundance of juvenile coho salmon was most closely correlated with the abundance of adu

Carotid body glomus cells in mammals contain a plethora of different neurochemicals. Several hypotheses exist to explain their roles in oxygen-chemosensing. In the present study we assessed the distribution of serotonin, acetylcholine and catecholamines in the gills of trout (Oncorhynchus mykiss) and goldfish (Carassius auratus) using immunohistochemistry, and an activity-dependent dye, Texas Red hydrazide (TXR). In fish the putative oxygen sensing cells are neuroepithelial cells (NECs) and the focus in recent studies has been on the role of serotonin in oxygen chemoreception. The NECs of trou

Odontoclasts resorbing teeth are multinucleated cells. Previously, the authors have investigated the distribution of number of nuclei per human odontoclast and showed that the mean number of nuclei per cell is 5.3, the median is 4, and 93.8% of cells have 10 or fewer nuclei. Teleost odontoclasts have features similar to those of mammals; however, the distribution of number of nuclei per cell remains unknown. The present study aimed to examine the distribution of number of nuclei per odontoclast in a teleost fish, Chinook salmon, Oncorhynchus tshawytscha (Walbaum), and to clarify the difference

Northern watersheds are facing rapid hydroclimatic reorganization driven by climate warming, glacier retreat, and intensifying land use. These changes are altering the distribution and accessibility of aquatic habitats, creating both risks and opportunities for cold-adapted species such as Pacific salmon (Oncorhynchus spp.). Effective responses require monitoring approaches capable of detecting contemporary habitat use, as well as emerging or intermittent occupancy in remote and rapidly changing landscapes where conventional monitoring is constrained. Here, we evaluated a species-targeted, qua

Massive algal blooms, including the 2022 Oder disaster linked to the toxic haptophyte Prymnesium parvum, highlight critical gaps in our understanding of waterborne ecotoxicants. Prymnesins are ladder-framed polyether-type compounds produced by P. parvum with well-documented ichthyotoxic properties; however, the cellular mechanisms underlying prymnesin-induced fish cell injury have yet to be resolved at the cellular level. Here, we establish and optimize molecular and cellular bioassays using fluorescence microscopy and flow cytometry to characterize early cell-damage pathways in rainbow trout

Infectious hematopoietic necrosis virus (IHNV) was detected for the first time in Finland in November 2017. The virus was isolated from rainbow trout (Oncorhynchus mykiss) from a sea-cage farm in the Gulf of Bothnia. Sampling was carried out as part of a routine risk-based surveillance program. During inspections based on epidemiological investigations, five other IHNV-positive rainbow trout establishments were detected in 2017-2018. No symptoms indicative of IHN were reported, and only a few macroscopic and histopathological findings were recorded. To assess the pathogenicity of the Finnish i

The impact of hyperlipidemia on fuel selection has never been investigated in fish. This study quantifies how intralipid administration affects i) in vivo mobilization of lipids (lipolytic rate: Ra glycerol) and carbohydrates (hepatic glucose production: Ra glucose) in rainbow trout and ii) key proteins involved in the regulation of fuel metabolism that could explain changes in glycerol and glucose kinetics. Results show that intralipid triples lipolytic rate (from 2.5 ± 0.5 to 7.8 ± 1.1 µmol glycerol kg-1·min-1) and inhibits glucose production by 36% (from 7.3 ± 0.9 to 4.7 ± 0.4 µmol kg-1·min

The gills are a critical organ and the primary site of colonization with the pathogen Aeromonas salmonicida. They are covered by protective mucus, mainly comprised by the highly glycosylated mucins. The aim of this study was to characterize the rainbow trout (Oncorhynchus mykiss) gill mucus and mucin production during health and colonization with A. salmonicida. Ex vivo imaging of gill mucus demonstrated that it was heterogenous in its glycosylation and organized in strands. Metabolic labelling time-course experiments in vivo revealed that the metabolic label was incorporated into mucins in th

Residues of human pharmaceuticals are widely detected in surface waters and can be taken up by and bioaccumulate in aquatic organisms, especially fish. One of the key challenges in assessing the bioaccumulation potential of ionizable organic compounds, such as the pharmaceuticals, is the lack of empirical data for biotransformation. In the present study, we assessed the in vitro intrinsic clearances (CLINT) of twelve pharmaceuticals, individually and some additionally as mixtures, in rainbow trout (Oncorhynchus mykiss) liver S9 fractions (RT-S9) adhering to the OECD test guidance 319B. The tes

Exposure to mercury (Hg) through fish consumption poses significant environmental and public health risks, given its status as one of the top ten hazardous chemicals. Aquaculture is expanding, driving a surge in demand for sustainable aquafeeds. Tuna byproducts, which are rich in protein, offer potential for aquafeed production, yet their use is challenged by the high content of heavy metals, particularly Hg. However, these byproducts also contain elevated levels of selenium (Se), which may counteract Hg adverse effects. This study examines the fate of dietary Hg and Se in an aquaculture model

Flavobacterium columnare, which causes columnaris disease, is one of the costliest pathogens in the freshwater fish-farming industry. The virulence mechanisms of F. columnare are not well understood and current methods to control columnaris outbreaks are inadequate. Iron is an essential nutrient needed for metabolic processes and is often required for bacterial virulence. F. columnare produces siderophores that bind ferric iron for transport into the cell. The genes needed for siderophore production have been identified, but other components involved in F. columnare iron uptake have not been s

Fishes may exploit environmental vortices to save in the cost of locomotion. Previous work has investigated fish refuging behind a single cylinder in current, a behavior termed the Kármán gait. However, current-swept habitats often contain aggregations of physical objects, and it is unclear how the complex hydrodynamics shed from multiple structures affect refuging in fish. To begin to address this, we investigated how the flow fields produced by two D-shaped cylinders arranged in tandem affect the ability of rainbow trout (Oncorhynchus mykiss) to Kármán gait. We altered the spacing of the two

Most studies of virulence of infection focus on pairwise host-parasite interactions. However, hosts are almost universally co-infected by several parasite strains and/or genotypes of the same or different species. While theory predicts that co-infection favours more virulent parasite genotypes through intensified competition for host resources, knowledge of the effects of genotype by genotype (G × G) interactions between unrelated parasite species on virulence of co-infection is limited. Here, we tested such a relationship by challenging rainbow trout with replicated bacterial strains and fluk

The pharmacokinetic-pharmacodynamic predictor of antimicrobial activity for tetracyclines is reported to be the area under the concentration-time curve at steady state (AUC(ss)) divided by the minimal inhibitory concentration of the targeted pathogen. Here, we estimate AUC(ss) values for oxytetracycline (OTC) in serum of rainbow trout Oncorhynchus mykiss by using a destructive sampling study design. Seventy-two rainbow trout were fed OTC-medicated feed at 74.7 +/- 1.5 mg/kg (mean +/- SD) body weight (BW) by oral gavage for 10 consecutive days. Serum was collected from nine fish at 1, 3, 6, 8,

Proliferative kidney disease is a parasitic infection of salmonid fishes caused by Tetracapsuloides bryosalmonae. The main target organ of the parasite in the fish is the kidney. To investigate the influence of water temperature on the disease in fish, rainbow trout Oncorhynchus mykiss infected with T bryosalmonae were kept at 12 degrees C and 18 degrees C. The number of parasites, the type and degree of lesions in the kidney and the mortality rate was evaluated from infection until full development of disease. While mortality stayed low at 12 degrees C, it reached 77% at 18 degrees C. At 12 d

Knowledge of the uptake mechanisms and metabolism of metals is essential for understanding the factors governing metal toxicity, discerning means by which acclimation and homeostasis may be achieved and characterising interactions between the metal of interest and other environmental moieties. Zinc is both an important aquatic contaminant and a vital micronutrient. The physiological characterisation of dietary zinc absorption in fish has, therefore, important implications for environmental protection and aquaculture. The present study aimed to elucidate the mechanism of intestinal zinc uptake

Together with cAMP, calcium ions play an important role in the regulation of melatonin synthesis in the pineal organ of all vertebrate species, irrespective of the conspicuous phylogenetic transformation of the melatonin-producing cell, the pinealocyte. Here we address the question how the intracellular concentration of free calcium ions [Ca2+]i is regulated in directly light-sensitive trout pinealocytes and in rat pinealocytes which have lost the direct light sensitivity and respond to norepinephrine. Isolated pinealocytes identified by the S-antigen immunoreaction were investigated by means
