The generation of regulatory B cells by helminth parasites.

The activation of B cells by pathogens to elicit antibody production is a central facet of immunity. Pathogens also evoke the expansion of B cells that can function in the regulation of immunity. Different pathogens have been described with the capacity to drive such regulatory B (Breg) cells. One group of pathogens, parasitic helminths, has been used experimentally to identify and explore immunological mechanisms of Breg cells. Several species of helminths have demonstrated the capacity to expand Breg cell populations in mice, while the presence of Breg cells in humans infected with certain h

Unveiling the intracellular survival gene kit of trypanosomatid parasites.

Trypanosomatids are unicellular protozoans of medical and economical relevance since they are the etiologic agents of infectious diseases in humans as well as livestock. Whereas Trypanosoma cruzi and different species of Leishmania are obligate intracellular parasites, Trypanosoma brucei and other trypanosomatids develop extracellularly throughout their entire life cycle. After their genomes have been sequenced, various comparative genomic studies aimed at identifying sequences involved with host cell invasion and intracellular survival have been described. However, for only a handful of genes…

Sitting at the window to the world--ocular parasites.

Parasitic infections cause significant ophthalmic disease, both in developing countries and in the Western world. The parasitic infections Acanthamoeba keratitis, ocular toxoplasmosis, and ocular toxocariasis are responsible for a significant proportion of ocular pathology. Especially in light of the recent increase of immunocompromised (i.e. using immunosuppressants or HIV) and aged populations, parasitic infections of the eye are rising in number. This reviews aims to describe the pathogenesis, symptoms, diagnosis and management of infection, as well as preventative measures for these three

Aminoacyl-tRNA synthetases as drug targets in eukaryotic parasites.

Aminoacyl-tRNA synthetases are central enzymes in protein translation, providing the charged tRNAs needed for appropriate construction of peptide chains. These enzymes have long been pursued as drug targets in bacteria and fungi, but the past decade has seen considerable research on aminoacyl-tRNA synthetases in eukaryotic parasites. Existing inhibitors of bacterial tRNA synthetases have been adapted for parasite use, novel inhibitors have been developed against parasite enzymes, and tRNA synthetases have been identified as the targets for compounds in use or development as antiparasitic drugs…

Cessation of a salmon decline with control of parasites.

The resilience of coastal social-ecological systems may depend on adaptive responses to aquaculture disease outbreaks that can threaten wild … izootics of wild salmon without significantly increasing the annual number of treatments. Levels of parasites on wild juvenile salmon significantly influence the growth rate of affected salmon populations, suggesting that these changes in management have had positive outcomes for wild salmon populations. These adaptive changes have not occurred through form …

Thioredoxin glutathione reductase-dependent redox networks in platyhelminth parasites.

SIGNIFICANCE: Platyhelminth parasites cause chronic infections that are a major cause of disability, mortality, and economic losses in developing countries. Maintaining redox homeostasis is a major adaptive problem faced by parasites and its disruption can shift the biochemical balance toward the host. Platyhelminth parasites possess a streamlined thiol-based redox system in which a single enzyme, thioredoxin glutathione reductase (TGR), a fusion of a glutaredoxin (Grx) domain to canonical thioredoxin reductase (TR) domains, supplies electrons to oxidized glutathione (GSSG) and thioredoxin (Tr…

Apicoplast-targeting antibacterials inhibit the growth of Babesia parasites.

The apicoplast housekeeping machinery, specifically apicoplast DNA replication, transcription, and translation, was targeted by ciprofloxacin, thiostrepton, and rifampin, respectively, in the in vitro cultures of four Babesia species. Furthermore, the in vivo effect of thiostrepton on the growth cycle of Babesia microti in BALB/c mice was evaluated. The drugs caused significant inhibition of growth from an initial parasitemia of 1% for Babesia bovis, with 50% inhibitory concentrations (IC(50)s) of 8.3, 11.5, 12, and 126.6 μM for ciprofloxacin, thiostrepton, rifampin, and clindamycin, respectiv

[Algorithm for the coproscopic diagnosis of human intestinal parasites].

The purpose of the study was to elaborate a detection algorithm for human intestinal helminth eggs. There is a broad spectrum ofcoproscopic methods recommended for the detection of Opisthorchis eggs in man and animals; these include Fulleborn's method, formalin-ether method, Goryachev's, Katoh's, Kalantaryan's, Shcherbovich's, and Kotelnikov-Varenichev methods. Combined coproscopic methods are significantly more effective in detecting the causative agents of enteric parasitoses than is Katoh's method. Among the considered coproscopic techniques for the diagnosis of human ascariasis, it is most

Environmental constraints guide migration of malaria parasites during transmission.

Migrating cells are guided in complex environments mainly by chemotaxis or structural cues presented by the surrounding tissue. During transmission of malaria, parasite motility in the skin is important for Plasmodium sporozoites to reach the blood circulation. Here we show that sporozoite migration varies in different skin environments the parasite encounters at the arbitrary sites of the mosquito bite. In order to systematically examine how sporozoite migration depends on the structure of the environment, we studied it in micro-fabricated obstacle arrays. The trajectories observed in vivo an

[The distribution of intestinal parasites among children in Denizli].

Intestinal parasitic disease appears to be an important national health problem in our country. Parasitic infection distribution is related to the socioeconomic status and environmental factors. The aim of this study was to determine the distribution of intestinal parasites among children in the 1-15 age group in the Denizli province. Parasitic infection was observed in 256 (10.2%) out of 2518 children. The highest rate of 31.4% was that of Giardia intestinalis. The rates of Enterobius vermicularis and Blastocystis hominis were found to be 29.6% and 14% respectively.

Protein disulfide isomerase assisted protein folding in malaria parasites.

In eukaryotes, the formation of protein disulfide bonds among cysteine residues is mediated by protein disulfide isomerases and occurs in the highly oxidised environment of the endoplasmic reticulum. This process is poorly understood in malaria parasites. In this paper, we report the gene isolation, sequence and phylogenetic comparisons, protein structure and thioredoxin-domain analyses of nine protein disulfide isomerases-like molecules from five species of malaria parasites including Plasmodium falciparum and Plasmodium vivax (human), Plasmodium knowlesi (simian) and Plasmodium berghei and P…
