Ventilatory strategies in the prevention and management of bronchopulmonary dysplasia.

Bronchopulmonary dysplasia (BPD) leads to considerable mortality and morbidity in premature infants. Although mechanical ventilation is lifesaving in infants with respiratory distress syndrome (RDS), it may contribute to lung injury and subsequently to BPD. Appropriate ventilatory strategies for reducing BPD include redefining the goals for "adequate gas exchange," using less mechanical ventilation support, refining the methods of mechanical ventilation, and using alternative techniques. Permissive hypercapnia, permissive hypoxemia, minimal peak pressures, rapid rates, early therapeutic contin

Extracellular matrix molecules and their possible roles in the regeneration of frog nervous system.

Recent biochemical and histochemical analyses explored different components of the extracellular matrix (ECM) in the nervous system, and either permissive or non-permissive roles in neuronal development and regeneration were suggested. The aim of this study was to detect the distribution pattern of a few of these molecules in the nervous system of intact frogs and during nerve regeneration. The hyaluronan (HA) and tenascin C reactions were negative in the peripheral nerves, but appeared in their entry zones. In the CNS, different populations of neurons were surrounded with HA and tenascin C-po

Development of substance use and psychiatric comorbidity in an epidemiologic study of white and American Indian young adolescents the Great Smoky Mountains Study.

The progression of substance use and the patterns of comorbidity of substance use and psychiatric disorders are explored prospectively in young adolescents enrolled in the Great Smoky Mountains Study. This study is an epidemiologic study of white and American Indian youths living in rural Southern Appalachia. Results from this study indicate that alcohol use without permission predicts subsequent use of illicit drugs and regular tobacco use. Use of tobacco was not associated with either later alcohol or drug use. Patterns of comorbidity showed strong cross-sectional relationships between subst

Potentiation by steroids of the beta-adrenergic agent-induced stimulation of cyclic AMP in isolated mouse thymocytes.

There is an increasing amount of evidence suggesting that glucocorticoids may modulate the responsiveness of various cell types to beta-adrenergic agents. In some systems, it has been shown, in addition, that steroids potentiate the elevation of cAMP induced by catecholamines. Little is known however of the mechanism underlying steroid action. We have studied this 'permissive action' in isolated thymocytes which have specific receptor sites for both glucocorticoids and beta-adrenergic agents. The glucocorticoid compound dexamethasone did not alter intracellular cAMP level but markedly enhanced

The Big Five Functional Levels of Vascular Calcification.

Vascular calcification is often described either as passive deposition of calcium-phosphate crystals or as an active osteogenic process. Neither view alone explains how mineral formation begins, progresses and becomes self-sustaining within the vascular wall. Here, we organize the main mechanisms of vascular calcification into five functional levels according to their predominant causal role. Level I defines mineral permissiveness through calcium-phosphate pressure and extracellular mineralization inhibitors. Level II determines how nascent mineral is packaged, nucleated and retained through c…

Extracellular Matrix Remodeling as a Mechanobiological Driver of Breast Cancer Aggressiveness: Comparative Oncology, Multi-Omics, and Artificial Intelligence Perspectives.

The extracellular matrix (ECM) is increasingly recognized as an active regulator of breast cancer progression rather than a passive structural scaffold. This narrative review examines how ECM remodeling contributes to tumor aggressiveness through changes in matrix composition, collagen architecture, tissue stiffness, mechanotransduction, stromal permissiveness, immune and metabolic programs, invasion, metastasis and therapeutic response. A structured narrative search of literature published from 1981 to June 2026 was used to support this synthesis. Evidence from breast cancer studies indicates…

Obestatin Treatment Counteracts Muscle Wasting by Reactivation of Autophagy in Duchenne Muscular Dystrophy.

The mechanisms by which muscular dystrophy-related stress is transduced to the autophagic machinery remain poorly characterized. The formula … ing promotes the recovery of physiological skeletal muscle function. Thus, DMD conditions determine permissiveness to the activation of AMPK that sustain autophagy under anabolic conditions stablished by obestatin signaling through mTORC1.

Experimental infection of Oropouche virus in Japanese populations of Aedes albopictus and Culex mosquitoes.

The recent geographical expansion of Oropouche virus (OROV) highlights the critical need to evaluate vector competence in non-endemic region … ections with high viral replication, particularly Ae. albopictus, demonstrating its robust cellular permissiveness. Conversely, OE yielded low infection rates in the thorax and abdomen (0-18.9%) and failed to cause systemic dissemination to secondary tissues (head, wings, and legs). These results demonstrate that the stringent midgut barriers that prevent OROV d …

Upstream Ecological Control of the IAA-Skatole Branch: A pH-Dependent Triple-Lock Framework for Gut-Derived Uremic Toxin Precursors.

Gut-derived indole metabolites are implicated in the gut-kidney axis, but the factors controlling the intestinal conversion of indole-3-acetic acid (IAA) to skatole remain incompletely defined. We developed a deterministic, hypothesis-generating framework that represents this conversion as a finite-pool allocation process governed by pH-dependent ecological permissiveness, terminal-conversion capacity, precursor availability, spatial progression, and competing loss. The model separates the available-pool scale from a dimensionless integrated conversion exposure, Ψ. Across 5400 loss-free scenar…

Experimental and ex vivo studies demonstrate neural permissiveness and suggest the capacity for interaction with central nervous system (CNS) tissue. Fostamatinib (R788), a spleen tyrosine kinase inhibitor, sensitizes pancreatic cancer cells to oncolytic vesicular stomatitis virus.

Pancreatic ductal adenocarcinoma (PDAC) is one of the most lethal malignancies with limited treatment options. Oncolytic virus (OV) therapy exploits replication-competent viruses, including vesicular stomatitis virus (VSV), to preferentially infect and kill malignant cells, while sparing non-malignant cells. We previously showed that human PDAC cell lines are highly heterogeneous in their permissiveness to VSV and other OVs. Although ruxolitinib and other JAK1/2 inhibitors can enhance VSV replication in resistant PDAC cell lines, they also increase the susceptibility of non-malignant cells to …

Extracellular Vesicles in Chronic Lymphocytic Leukemia: Tumor Microenvironment Messengers as a Basis for New Targeted Therapies?

In addition to intrinsic genomic and nongenomic alterations, tumor progression is also dependent on the tumor microenvironment (TME, mainly composed of the extracellular matrix (ECM), secreted factors, and bystander immune and stromal cells). In chronic lymphocytic leukemia (CLL), B cells have a defect in cell death; contact with the TME in secondary lymphoid organs dramatically increases the B cells' survival via the activation of various molecular pathways, including the B cell receptor and CD40 signaling. Conversely, CLL cells increase the permissiveness of the TME by inducing changes in th…

Disruption of anthrax toxin receptor 1 in pigs leads to a rare disease phenotype and protection from senecavirus A infection.

Senecavirus A (SVA) is a cause of vesicular disease in pigs, and infection rates are rising within the swine industry. Recently, anthrax tox … acid was decreased in IF compared to WT pigs. Thus, this new ANTXR1 mutation resulted in decreased permissiveness of SVA in pigs. Overall, genetic disruption of ANTXR1 in pigs provides a unique model for GAPO syndrome and prevents circulating SVA infection and clinical symptoms, confirming that ANTXR1 acts as a receptor for the virus.

A quantitative nuclear egress assay to investigate the nucleocytoplasmic capsid release of human cytomegalovirus.

Nuclear egress is a rate-limiting step of herpesviral replication, restricting the nucleocytoplasmic transport of viral capsids. The process … the quantitation of viral nuclear egress improves the characterization of viral mutants, host cell permissiveness and antiviral drug efficacy. In this study, various strains of human cytomegalovirus (HCMV) were used to measure the replication efficiencies in primary human fibroblasts, applying methods of cell fractionation, DNase digestion, sucrose cushions and …

Avian influenza viruses that cause highly virulent infections in humans exhibit distinct replicative properties in contrast to human H1N1 viruses.

Avian influenza viruses present an emerging epidemiological concern as some strains of H5N1 avian influenza can cause severe infections in h … seasonal strain. This explains, in part, the pandemic strain's generally slower growth kinetics and permissiveness to accept mutations causing neuraminidase inhibitor resistance without significant loss in fitness. Our results highlight differential growth properties of H1N1, H5N1 and H7N9 influenza viruses.

Exchanges of genomic domains between poliovirus and other cocirculating species C enteroviruses reveal a high degree of plasticity.

The attenuated Sabin strains contained in the oral poliomyelitis vaccine are genetically unstable, and their circulation in poorly immunized populations can lead to the emergence of pathogenic circulating vaccine-derived polioviruses (cVDPVs). The recombinant nature of most cVDPV genomes and the preferential presence of genomic sequences from certain cocirculating non-polio enteroviruses of species C (EV-Cs) raise questions about the permissiveness of genetic exchanges between EV-Cs and the phenotypic impact of such exchanges. We investigated whether functional constraints limited genetic exch…
