Uterine fibroid polygenic risk score (PRS) associates and predicts risk for uterine fibroid.

Uterine fibroids (UF) are common pelvic tumors in women, heritable, and genome-wide association studies (GWAS) have identified ~ 30 loci associated with increased risk in UF. Using summary statistics from a previously published UF GWAS performed in a non-Hispanic European Ancestry (NHW) female subset from the Electronic Medical Records and Genomics (eMERGE) Network, we constructed a polygenic risk score (PRS) for UF. UF-PRS was developed using PRSice and optimized in the separate clinical population of BioVU. PRS was validated using parallel methods of 10-fold cross-validation logistic regress

Pathway Phenotypes Underpinning Depression, Anxiety, and Chronic Fatigue Symptoms Due to Acute Rheumatoid Arthritis: A Precision Nomothetic Psychiatry Analysis.

Rheumatoid arthritis (RA) is a chronic inflammatory and autoimmune disorder which affects the joints in the wrists, fingers, and knees. RA is often associated with depressive and anxiety symptoms as well as chronic fatigue syndrome (CFS)-like symptoms. This paper examines the association between depressive symptoms (measured with the Beck Depression Inventory, BDI), anxiety (Hamilton Anxiety Rating Scale, HAMA), CFS-like (Fibro-fatigue Scale) symptoms and immune-inflammatory, autoimmune, and endogenous opioid system (EOS) markers, and lactosylcer-amide (CD17) in RA. The serum biomarkers were a

Fate or coincidence: do COPD and major depression share genetic risk factors?

Major depressive disorder (MDD) is a common comorbidity in chronic obstructive pulmonary disease (COPD), affecting up to 57% of patients with COPD. Although the comorbidity of COPD and MDD is well established, the causal relationship between these two diseases is unclear. A large-scale electronic health record clinical biobank and genome-wide association study summary statistics for MDD and lung function traits were used to investigate potential shared underlying genetic susceptibility between COPD and MDD. Linkage disequilibrium score regression was used to estimate genetic correlation betwee

In (deficit) schizophrenia, a general cognitive decline partly mediates the effects of neuro-immune and neuro-oxidative toxicity on the symptomatome and quality of life.

BACKGROUND: Schizophrenia and deficit schizophrenia are accompanied by neurocognitive impairments. The aim of this study was to examine whether a general factor underpins impairments in key Cambridge Neuropsychological Test Automated Battery (CANTAB) probes, verbal fluency test (VFT), world list memory (WLM), True Recall, and mini mental state examination (MMSE). METHODS: We recruited 80 patients with schizophrenia and 40 healthy controls. All patients were assessed using CANTAB tests, namely paired-association learning, rapid visual information processing, spatial working memory, one touch s

Polygenic Risk of Psychiatric Disorders Exhibits Cross-trait Associations in Electronic Health Record Data From European Ancestry Individuals.

BACKGROUND: Prediction of disease risk is a key component of precision medicine. Common traits such as psychiatric disorders have a complex polygenic architecture, making the identification of a single risk predictor difficult. Polygenic risk scores (PRSs) denoting the sum of an individual's genetic liability for a disorder are a promising biomarker for psychiatric disorders, but they require evaluation in a clinical setting. METHODS: We developed PRSs for 6 psychiatric disorders (schizophrenia, bipolar disorder, major depressive disorder, cross disorder, attention-deficit/hyperactivity disor

Multiomics Analyses Identify Genes and Pathways Relevant to Essential Tremor.

INTRODUCTION: The genetic factors and molecular mechanisms predisposing to essential tremor (ET) remains largely unknown. OBJECTIVE: The objective of this study was to identify pathways and genes relevant to ET by integrating multiomics approaches. METHODS: Case-control RNA sequencing of 2 cerebellar regions was done for 64 samples. A phenome-wide association study (pheWAS) of the differentially expressed genes was conducted, and a genome-wide gene association study (GWGAS) was done to identify pathways overlapping with the transcriptomic data. Finally, a transcriptome-wide association study

Rare Variants in the Gene ALPL That Cause Hypophosphatasia Are Strongly Associated With Ovarian and Uterine Disorders.

CONTEXT: Mutations in alkaline phosphatase (AlkP), liver/bone/kidney (ALPL), which encodes tissue-nonspecific isozyme AlkP, cause hypophosphatasia (HPP). HPP is suspected by a low-serum AlkP. We hypothesized that some patients with bone or dental disease have undiagnosed HPP, caused by ALPL variants. OBJECTIVE: Our objective was to discover the prevalence of these gene variants in the Vanderbilt University DNA Biobank (BioVU) and to assess phenotypic associations. DESIGN: We identified subjects in BioVU, a repository of DNA, that had at least one of three known, rare HPP disease-causing vari

Comorbidity classes and associated impairment, demographics and 9/11-exposures in 8,236 children and adolescents.

The extensive comorbidity of psychiatric disorders in children and adolescents leads to clinical heterogeneity, and is an often-overlooked issue in etiopathogenic and treatment studies in developmental psychopathology. In a representative sample (N=8236) of New York City public school students assessed six months after 9/11, latent class analysis was applied to 48 symptoms across seven disorders: posttraumatic stress, agoraphobia, separation anxiety, panic disorder, generalized anxiety (GAD), major depression (MDD) and conduct disorder (CD). Our objective was to identify classes defined by hom

In pursuit of neurophenotypes: The consequences of having autism and a big brain.

A consensus has emerged that despite common core features, autism spectrum disorder (ASD) has multiple etiologies and various genetic and biological characteristics. The fact that there are likely to be subtypes of ASD has complicated attempts to develop effective therapies. The UC Davis MIND Institute Autism Phenome Project is a longitudinal, multidisciplinary analysis of children with autism and age-matched typically developing controls; nearly 400 families are participating in this study. The overarching goal is to gather sufficient biological, medical, and behavioral data to allow definiti

Phenotypic and imaging features of FLNA-negative patients with bilateral periventricular nodular heterotopia and epilepsy.

PURPOSE: Periventricular nodular heterotopia (PVNH) is a malformation of cortical development due to impaired neuronal migration resulting in the formation of nodular masses of neurons and glial cells in close proximity to the ventricular walls. We report the clinical characteristics of the largest case series of FLNA-negative patients with seizures and bilateral periventricular heterotopia. METHODS: Participants were recruited through the Epilepsy Phenome/Genome Project (EPGP), a multicenter collaborative effort to collect detailed phenotypic data and DNA on a large number of individuals wit

Myeloid dendritic cells frequencies are increased in children with autism spectrum disorder and associated with amygdala volume and repetitive behaviors.

The pathophysiology of autism spectrum disorder (ASD) is not yet known; however, studies suggest that dysfunction of the immune system affects many children with ASD. Increasing evidence points to dysfunction of the innate immune system including activation of microglia and perivascular macrophages, increases in inflammatory cytokines/chemokines in brain tissue and CSF, and abnormal peripheral monocyte cell function. Dendritic cells are major players in innate immunity and have important functions in the phagocytosis of pathogens or debris, antigen presentation, activation of naïve T cells, in

Mouse inbred strain differences in ethanol drinking to intoxication.

Recently, we described a simple procedure, Drinking in the Dark (DID), in which C57BL/6J mice self-administer ethanol to a blood ethanol concentration (BEC) above 1 mg/ml. The test consists of replacing the water with 20% ethanol in the home cage for 4 h early during the dark phase of the light/dark cycle. Three experiments were conducted to explore this high ethanol drinking model further. In experiment 1, a microanalysis of C57BL/6J behavior showed that the pattern of ethanol drinking was different from routine water intake. In experiment 2, drinking impaired performance of C57BL/6J on the a

A sodium-HIF1α axis coordinates immune metabolic reprogramming and mitochondrial remodeling in salt-sensitive hypertension.

Salt-sensitivity of blood pressure (SSBP) is associated with immune-metabolic dysfunction, yet the mechanism that coordinates sodium exposure, mitochondrial remodeling, and blood pressure response remains undefined. Phenome-wide and laboratory-value association studies (PheWAS and LabWAS) in the All of Us Research Program identified fluid, electrolyte, and acid-base balance disorders and renal phenotypes as the strongest disease associations. At the same time, hypertension was linked to reduced serum potassium, chloride, and eGFR, corroborating the centrality of renal-electrolyte physiology in

Genome-wide meta-analysis dissects the genetic basis and health implications of circulating taurine.

Taurine is a semiessential micronutrient critical to many biological processes. However, the genetic variation and the relationships between circulating taurine and human health are unclear. Here, we conducted the largest genome-wide meta-analysis to date, integrating summary statistics from five publicly available independent genome-wide association studies (GWAS) on circulating taurine levels in 36,490 European individuals. We employed LiftOver to harmonize genomic coordinates across the summary statistics derived from five large European cohorts with plasma taurine measurements, followed by

Therapeutic targets for atherosclerotic coronary artery disease: druggable genome-wide Mendelian randomization analysis.

BACKGROUND AND AIMS: Despite advances in treating atherosclerotic coronary artery disease (ACAD), the risk of major adverse cardiovascular events remains high. We aimed to identify potential therapeutic targets for ACAD by integrating genome-wide association studies (GWAS) and Mendelian randomization (MR) analyses. METHODS: Leveraging druggable genome data, cis-eQTL/cis-pQTL from blood and coronary artery tissue, alongside GWAS summary data for coronary atherosclerosis, we performed drug-target MR to identify causal genes for ACAD at both transcript and protein levels. Phenome-wide associatio

MKRN1 as a prioritized drug target for postpartum depression: evidence from druggable proteome profiling and multi-layer validation.

Postpartum depression (PPD) is a significant global health concern affecting women, yet effective and innovative therapeutic targets remain limited. Although genome-wide association studies (GWAS) have identified genetic risk loci, their underlying mechanisms and translational potential remain poorly understood. Therefore, we integrated PPD GWAS data with protein quantitative trait loci from two independent datasets to identify risk genes through proteome-wide association studies (PWAS). Validation was performed using colocalization analysis and Mendelian randomization (MR). To assess the safe

High-content imaging driven algal phenotypes enable precise multilevel discrimination of heavy metal stress.

Conventional water quality monitoring based on fixed chemical thresholds often fails to capture the integrated biological effects of pollutant mixtures and sublethal stress. To address this limitation, we developed a high-content imaging (HCI)-based pipeline that converts phenotypic changes in algal cells into multidimensional quantitative features for toxicity assessment. A heavy metal stress model was established by exposing Chlorella vulgaris to copper, nickel, and zinc across concentrations ranging from regulatory standards to toxic levels. An integrated HCI analysis workflow was applied t

Shear stress-induced endothelial HEG1 signalling regulates vascular tone and blood pressure.

BACKGROUND AND AIMS: Endothelial cells (ECs) sense flow shear stress for vasodilation, a crucial mechanism for maintaining systemic blood pressure (BP). Impaired shear stress signalling contributes to endothelial dysfunction and hypertension. Heart development protein with EGF-like domain 1 (HEG1), a flow-sensitive, endothelial-derived protein, is inversely associated with cardiovascular risks. This study aimed to elucidate the role of endothelial HEG1 in BP regulation and the underlying mechanisms. METHODS: Phenome-wide association study, computational fluid dynamics analysis, single-cell RN

Cold antagonizes salt-induced jasmonate signaling to reconfigure the defense responses of pepper (Capsicum annuum L.).

Combined cold and salt stress antagonizes jasmonate signaling via MYC2 suppression, reallocating resources to flavonoid-based defense and revealing a strategic target for crop multi-stress resilience. Plants are frequently exposed to a combination of abiotic stresses, but the mechanisms governing their non-additive interactions remain largely unclear. In this study, we integrated phenomic, transcriptomic, and metabolomic analyses to elucidate the interplay between cold and salt stress in pepper (Capsicum annuum L.). Unlike individual stresses, which led to progressive damage, the combined stre

Deep Learning for Analysis of Bone Marrow Adiposity: Breakthroughs from Recent Large-Scale Analyses in the UK Biobank.

PURPOSE OF REVIEW: Bone marrow adipose tissue (BMAT) is a significant fat depot with distinct skeletal, haematological and metabolic roles. It increases with ageing, osteoporosis, metabolic disease, and cancer, and is emerging as a biomarker for fracture risk. Quantification of bone marrow adiposity (BMA) has relied on MRI, proton spectroscopy, computed tomography, or histology, but large-scale studies have been limited by requiring labour-intensive analysis. Deep learning (DL) now enables scalable, automated BMA measurement, with greatest progress in MRI. This review highlights recent advance
