Tissue plasminogen activator followed by antioxidant-loaded nanoparticle delivery promotes activation/mobilization of progenitor cells in infarcted rat brain.

Inherent neuronal and circulating progenitor cells play important roles in facilitating neuronal and functional recovery post stroke. However, this endogenous repair process is rather limited, primarily due to unfavorable conditions in the infarcted brain involving reactive oxygen species (ROS)-mediated oxidative stress and inflammation following ischemia/reperfusion injury. We hypothesized that during reperfusion, effective delivery of antioxidants to ischemic brain would create an environment without such oxidative stress and inflammation, thus promoting activation and mobilization of progen

Ontogenic study of the influence of tissue plasminogen activator (t-PA) in neonatal excitotoxic brain insult and the subsequent microglia/macrophage activation.

Intracerebral injections of ibotenic acid in neonatal mice produced white and gray matter lesions that mimic some aspects of the acquired cerebral injuries observed in human newborns (i.e. periventricular leukomalacias in preterm newborns and post-ischemic cortical necrosis in at term infants). We have evaluated the effects of tissue plasminogen activator inactivation (t-PA-/-) on the effects of ibotenic acid (0.01-20 microg), and on F4/80 labeling of microglia/macrophages at different stages. Three ontogenic periods have been identified. In mice injected the day of birth, postnatal (P) day 0,

Increased plasma activity of plasminogen activator inhibitor 1 (PAI-1) in two patients with Klinefelter's syndrome complicated by leg ulcers.

Klinefelter's syndrome is the most frequent major abnormality of sexual differentiation in men with two or more X-chromosomes, and affects one in 500 males. The syndrome is characterized by eunuchoid body proportions, scanty facial and body hair, gynaecomastia, and small firm testes. Leg ulcers, especially in combination with hyperpigmentation, have been reported in association with Klinefelter's syndrome. Thromboembolic processes are also frequently observed. The leg ulcers in patients with Klinefelter's syndrome are usually attributed to venous insufficiency. We describe two patients with Kl

Involvement of superoxide anion and platelet-activating factor in increased tissue-type plasminogen activator during rat gastric microvascular damages.

The role of tissue-type plasminogen activator (t-PA) was investigated in the gastric ulcer formation induced by microvascular derangement. The rat stomach was exposed and repeated electrical stimuli (irritation) were applied on the small arterial wall close to the lesser curvature to induce mucosal ischemia followed by hyperemia. The t-PA activity in the regional blood of the stomach was significantly elevated as early as 5 min after the irritation. Immunohistochemical study using anti-t-PA monoclonal antibody revealed that t-PA was detectable in the endothelial cells of capillaries and collec

Effects of a phorbol ester, a calcium ionophore, and 3',5'-adenosine monophosphate production on hen granulosa cell plasminogen activator activity.

Recent studies conducted in our laboratory have demonstrated that plasminogen activator (PA) is present in granulosa cells collected from the largest preovulatory follicle in the ovary of the domestic hen, and that its activity can be modulated by a variety of hormones in vitro. The present study was conducted to evaluate the intracellular mechanisms involved in the control of hen granulosa cell PA activity through the use of physiological and pharmacological agents. Treatment of granulosa cells with increasing doses (1, 10, and 50 ng/tube) of ovine LH resulted in a significant reduction of PA

Long-term ambient air pollution is not associated with circulating soluble urokinase plasminogen activator receptor.

BACKGROUND: Soluble urokinase plasminogen activator receptor (suPAR) is a stable chronic-inflammation biomarker causally implicated in atherosclerosis, yet its modifiable determinants remain largely uncharacterized. Long-term ambient air pollution is an established cardiovascular risk factor and a biologically plausible determinant of suPAR. METHODS: In this cross-sectional analysis, we tested associations of long-term residential air pollution with circulating suPAR in 5,072 participants of the Multi-Ethnic Study of Atherosclerosis, drawn from six US metropolitan regions and enrolled during

Rational truncation of streptokinase reduces immunogenicity while preserving plasminogen activation: in vitro and in vivo analysis.

The clinical utility of streptokinase, a widely used and cost-effective thrombolytic agent, has gained importance post COVID-19 due to increase in cardiovascular diseases globally. But the strong humoral immunogenicity exhibited by it, restricts its usage. Building upon our prior work of developing truncated streptokinase variants and their plasminogen activation study, we report here the results of immunogenicity studies for identification of variants with reduced immunogenicity. A panel of ten rationally truncated and recombinantly expressed streptokinase fragments were screened for antibody

Sex- and stressor-dependent effects of metformin on cardiac fibrosis in plasminogen activator inhibitor-1 deficiency.

Heart failure arises when persistent cardiac stress drives maladaptive remodeling and fibrosis. Plasminogen activator inhibitor-1 (SERPINE1/PAI-1) acts as a repressor of cardiac fibrosis (CF). While complete human PAI-1 deficiency is rare, individuals carrying loss-of-function PAI-1 mutations and genetic loss of PAI-1 in mice accelerates CF following cardiac stress and with age. Here, we used this model to investigate sex-specific mechanisms underlying CF in PAI-1 deficiency and an effective intervention to prevent its progression. Our transcriptomic profiling identified Serpine1/SERPINE1-posi

Soluble Urokinase Plasminogen Activator Receptor Predicts Survival and Hepatic Decompensation in Advanced Hepatocellular Carcinoma.

BACKGROUND AND AIMS: The introduction of immune checkpoint inhibitor (ICI) based therapies has significantly improved the prognosis of patients with unresectable hepatocellular carcinoma (HCC). However, the variable treatment response and the uncertain benefit in patients with advanced liver cirrhosis emphasise the urgent need for prognostic and predictive biomarkers guiding patient selection. The soluble urokinase plasminogen activator receptor (suPAR) is strongly associated with inflammation, liver cirrhosis and various types of cancer. In this study, we investigated suPAR as a potential nov

Cardiac Rupture and Death Following Administration of Recombinant Tissue-Type Plasminogen Activator (rt-PA).

Administration of recombinant tissue-type plasminogen activator (rt-PA) in the hyperacute phase of ischemic stroke is fraught with various pitfalls. An 81-year-old man had a stroke with aphasia and right incomplete paralysis, and CT angiography showed occlusion of the left internal carotid artery (ICA) within two hours of onset. The stroke neurologist administered rt-PA and started endovascular treatment, but during the treatment, the patient went into cardiopulmonary arrest without warning and died despite resuscitation. Autopsy revealed that the cause of death was cardiac rupture due to suba

Enhancing Thrombolysis Safety in Post-Acute Ischemic Stroke with Tissue Plasminogen Activator-Associated Microparticles.

Recombinant tissue-type plasminogen activator (tPA) is the only approved thrombolytic drug for acute ischemic stroke, a condition associated with severe disabilities and high mortality. However, when the blood-brain barrier (BBB) is damaged, tPA can exacerbate cerebral injury and increase the risk of hemorrhagic transformation, limiting its use to a small subset of patients. To address this challenge and minimize extravascular accumulation, we combined tPA with micrometer-sized particles (DPN). We then tested their safety and neuroprotective effects. After a 1 h transient occlusion of the midd

Association of plasma soluble urokinase plasminogen activator receptor concentrations and migraine with aura: a REFORM study.

Soluble urokinase plasminogen activator receptor (suPAR) has garnered attention as a potential blood-based biomarker for low-grade chronic inflammation. However, its specific association with migraine, including its subtypes, remains to be elucidated. We sought to examine the association of plasma suPAR levels with migraine and its subtypes. In this single-centre, cross-sectional study, plasma was collected at a single time point in adults with migraine and sex-matched healthy controls from October 2020 to June 2022. The quantification of plasma suPAR levels was performed in a blinded fashion

The relationship between the urokinase-type plasminogen activator gene Pro141Leu polymorphism and patients with prostate cancer.

OBJECTIVE: Although many genes associated with prostate cancer have been identified, there are still many cases of prostate cancer whose genetics have not been identified. The urokinase-type plasminogen activator gene is thought to play a key role in cancer invasion and metastasis. The Pro141Leu polymorphism may be involved in the development of prostate cancer, although genetic evidence is lacking. METHODS: We used the real time polymerase chain reaction based TaqMan assay to investigate whether the urokinase-type plasminogen activator gene Pro141Leu polymorphism is associated with prostate

Targeting Plasminogen Activator Inhibitor-1 with a Novel Small Molecule Inhibitor Attenuates Lung Fibrosis.

Fibrotic lung diseases are associated with significant morbidity and mortality, and few therapies have been FDA-approved for patients with these conditions. Therefore, developing effective anti-fibrotic treatments represents an unmet clinical need. Plasminogen activator inhibitor 1 (PAI-1) is an attractive therapeutic target as its expression is up-regulated in the context of fibrotic lung disease, and a causal role for PAI-1 in lung fibrogenesis has been established in complementary animal models. Here, we study the efficacy of a novel small molecule PAI-1 inhibitor, MDI-2517, to attenuate lu

Soluble urokinase-type plasminogen activator receptor improves early risk stratification in cardiogenic shock.

AIMS: Soluble urokinase-type plasminogen activator receptor (suPAR) is a biomarker reflecting the level of immune activation. It has been shown to have prognostic value in acute coronary syndrome and heart failure as well as in critical illness. Considering the complex pathophysiology of cardiogenic shock (CS), we hypothesized suPAR might have prognostic properties in CS as well. The aim of this study was to assess the kinetics and prognostic utility of suPAR in CS. METHODS AND RESULTS: SuPAR levels were determined in serial plasma samples (0-96 h) from 161 CS patients in the prospective, obs

The Plasminogen Activator System, Glucocorticoid, and Mineralocorticoid Receptors in the Primate Endometrium During Artificial Menstrual Cycles.

As a key mechanism in fibrinolysis and tissue remodeling, the plasminogen activator system has been suggested in the process of endometrial … ndometrium with their patterns of expression following similar fluctuations to urokinase and tissue plasminogen activators particularly within the endometrial vasculature. These proteins are present in endometrial vasculature in high levels during the proliferative phase, decreasing levels during the secretory phase followed by rising levels in the menstrual phase. Precision targeting of the plasminogen activator inhibitor-1 mechanism increases efficacy of fibrinolytic therapy in empyema.

Plasminogen activator inhibitor-1 (PAI-1) is an endogenous irreversible inhibitor of tissue-type (tPA) and urokinase (uPA) plasminogen activators. PAI-1-targeted fibrinolytic therapy (PAI-1-TFT) is designed to decrease the therapeutic dose of tPA and uPA, attenuating the risk of bleeding and other complications. Docking site peptide (DSP) mimics the part of the PAI-1 reactive center loop that interacts with plasminogen activators, thereby affecting the PAI-1 mechanism. We used DSP for PAI-1-TFT in two rabbit models: chemically induced pleural injury and Streptococcus pneumoniae induced empyema…

Relationship between plasminogen activator inhibitor-1 gene alterations and fibrosis in peritoneal dialysis patients.

Peritoneal fibrosis (PF) is a pathological change that occurs mostly long-term peritoneal dialysis (PD) patients, as a result of triggering the inflammatory response. Plasminogen activator inhibitor-1 (PAI-1) is an important molecule featured in the development of fibrosis. It has been shown in literature that PAI-1 gene alterations are associated with fibrosis in many tissues and organs. However, PAI-1 gene alterations in long-term PD patients have not yet been investigated. In this study, PAI-1 4G/5G polymorphism was examined by reverse hybridization, and all coding exons of the PAI-1 gene w
