Intracellular infection of Cutibacterium acnes in macrophages of extensive peri-implantitis lesions: A clinical case series.

Cutibacterium acnes is a facultative anaerobic, gram-positive rod, and a commensal bacterium of the body surface including oral cavity. A causal relationship between C. acnes and chronic granulomatous diseases, such as sarcoidosis and orthopedic implant-associated infections, has been previously reported. Typically, C. acnes has been observed inside macrophages, allowing evasion of host immunity, and triggering a persistent inflammatory response. However, such findings have not been reported in peri-implantitis lesions. In this case series, we collected inflamed tissues from extensive peri-imp

Recent advances in understanding inflammatory acne: Deciphering the relationship between Cutibacterium acnes and Th17 inflammatory pathway.

Acne vulgaris is a common chronic inflammatory skin disease of the pilosebaceous units. Four factors contribute to acne: hyperseborrhea and dysseborrhea, follicular hyperkeratinisation, skin microbiome dysbiosis and local immuno-inflammation. Recent key studies have highlighted a better understanding of the important role of Cutibacterium acnes (C. acnes) in the development of acne. Three major findings in the last decade include: (1) the ability of C. acnes to self-organize in a biofilm associated with a more virulent activity, (2) the loss of the C. acnes phylotype diversity and (3) the cent

Dynamic evaluation of pathological changes in a mouse acne model by optical imaging technology.

Acne vulgaris is a disorder of the pilosebaceous unit that is primarily caused by hyperseborrhoea, colonization with Propionibacterium acnes, hyperkeratosis and an inflammatory response. Existing pharmacodynamic assessment methods primarily focus on a single causative factor at a certain time point, making it difficult to assess multiple factors simultaneously in real time. Therefore, it is crucial to establish a dynamic and nondestructive method for the assessment of acne in vivo. This study utilized four-dimensional optical imaging techniques to assess the pathogenic factors and pathological…

The potential role for phage therapy for genetic modification of cutaneous diseases.

Antimicrobial resistance has become increasingly common across the globe, claiming more than 33,000 lives annually in Europe and 35,000 lives in the United States alone. The problem lies in trying to find potential solutions capable of tackling resistance and being able to fight infections that may resist various antimicrobials. Since Alexander Fleming's discovery in 1928, every antimicrobial synthesized in the past 70 years has developed at least one or more strains of resistant bacteria. One particular alternative to antimicrobials has brought hope to many in the scientific community: the ba

Anti-acne Treatment using Nanotechnology based on Novel Drug Delivery System and Patents on Acne Formulations: A Review.

BACKGROUND: Acne is one of the most infectious diseases related to human skin. This disease of acne is of higher prevalence in adults. The main causative organism involved in the acne vulgaris is Propionibacterium acne, which leads to sebum production and follicular hyperkeratinization. METHODS: The scientific data shows inflammatory mediators of genetic factors, stress, physiological factor, androgens and hormonal changes; all these factors play a major part in the pathophysiology of acne. Several drugs are currently available for the treatment of Acne like bactericidal, antibiotic and sever…

The use of light in the treatment of acne vulgaris-a review.

Topical and oral antibiotic therapy is also a popular method of treatment. The effectiveness of this method is limited by the increasing resistance of bacteria to antibiotics. Over the decades since the introduction of antibiotics to treat acne, the resistance levels of bacteria have changed. This defense mechanism is developed evolutionarily. Modifications of antibiotic receptor sites, alteration of drug influx/efflux, or enzymatic degradation are common mechanisms used by bacteria to initiate and strengthen internal antibiotic resistance. The basic chromophores used in light therapy are hemo

Pathogenesis of acne vulgaris: what's new, what's interesting and what may be clinically relevant.

Acne vulgaris is the most common skin disorder seen in dermatology and primary care offices today with significant associated morbidity. The pathogenesis of acne is complex and multifactorial, and there continues to be an influx of new information to increase our understanding of this chronic disease. Recent advances in acne pathogenesis will be discussed, including theories regarding the sequence of events in acne formation, the functions of P. acnes, TLR involvement and role of the sebaceous gland and factors influencing sebum production.

Optimization of the microbiological quality control validation of corneal medium using a clinical C. acnes isolate.

The microbiological quality of corneal tissue for transplantation must be monitored in a validated procedure complying with the European Pharmacopoeia (Ph. Eur.). An automated culture system is used to detect test strains in the presence of the medium. The Ph. Eur. mentioned C. acnes ATCC 11827 strain has allegedly been isolated from skin. This strain is known to display variable growth in routine matrix validation. As the Ph. Eur. also opens the use of clinical isolates, this study analyzed the use of clinical isolates for matrix validation. We compared the time-to-detection (TTD) in absence a

The Evolving Microbial Paradigm in Acne.

This review discusses the microbiology of acne vulgaris, a chronic inflammatory condition of the pilosebaceous unit that affects most adolescents and can persist into adulthood. The current standard of care consists largely of antibacterial interventions, based on the traditional view of Cutibacterium acnes as a pathogen. Alternative treatments are suggested by the "comedo switch" hypothesis, which attributes acne to aberrant differentiation of LRIG1+ sebaceous progenitor cells. While there is strong evidence to support this idea, it does not explain the efficacy of antibacterial interventions

Ultrasound-triggered interfacial engineering-based microneedle for bacterial infection acne treatment.

Acne is an inflammatory skin disease mainly caused by Propionibacterium acnes, which can cause local inflammatory reactions and develop into chronic inflammatory diseases in severe cases. To avoid the use of antibiotics and to effectively treat the site of acne, we report a sodium hyaluronate microneedle patch that mediates the transdermal delivery of ultrasound-responsive nanoparticles for the effective treatment of acne. The patch contains nanoparticles formed by zinc porphyrin-based metal-organic framework and zinc oxide (ZnTCPP@ZnO). We demonstrated activated oxygen-mediated killing of P. …

Identification of natural inhibitors to inhibit C. acnes lipase through docking and simulation studies.

Acne vulgaris is a common skin disease affecting 80-90% of teenagers worldwide. C. acnes producing lipases are the main virulence factor that catalyzes sebum lipid into free fatty acid that is used for C. acnes growth. Recently, computational biology and bioinformatics play a significant role in drug discovery programs and the identification of novel lead(s). In this study, potential inhibitors against the C. acnes lipase have been identified via cost-effective computational investigations. Molecular docking, MD simulations, and binding affinity analysis have been performed between the active

Cutibacterium acnes Pneumonia in an Immunocompromised Patient: A Case Report and Review of the Literature.

Cutibacterium acnes (C. acnes), known previously as Proprionebacterium acnes, is a common skin and mucous membrane colonizing bacteria. On rare occasions it has been associated with invasive infections in both immunocompetent and immunosuppressed patients. We describe the case of a 55-year-old female with a history of Sjogren's syndrome and psoriatic arthritis on TNF-alpha inhibitor therapy with etanercept. She presented with signs and symptoms of a thoracic infection with pulmonary nodules, hilar adenopathy, and bronchiectasis on thoracic computed tomography. She was diagnosed with C. acnes p

The Importance of Recognizing Yeast Acne Malessezia versus Propionbacterium.

We all know that acne vulgaris ' is very common. That is why it is called vulgaris which means common. We are wanting to present however that yeast acne is also common. In fact, yeast may be found on the skin in 75-98% of healthy people. Under particular biological conditions, homeostasis of the skin microbiome fails and growth of the fungi exceeds ideal levels. When this happens, "yeast acne" can occur. We feel that the incidence of yeast acne has been increasing in prevalence in our clinic, especially in young adult males, and so we wrote this paper in order to increase awareness.

Acne vulgaris.

Acne vulgaris is one of the most common dermatological diseases, and caused by a combination of pathogenetic factors including follicular hyperkeratosis, seborrhea, colonization with Propionibacteriumacnes and inflammatory phenomena. Various medical treatments address these causative factors, but they are not always well tolerated. In the last decades, there has been increased interest in laser and light-based treatments. These optical devices mainly target Propionibacteriumacnes by activating porphyrins produced by the bacterium, whereas some treatments directly lead to the destruction of seb…

The pathogenesis of acne.

Acne vulgaris is a multifactorial disease of as yet incompletely elucidated etiology and pathogenesis. The following have been identified as the most significant factors: follicular hyperkeratosis, increased sebum secretion, Propionibacterium (P.) acnes, and inflammation. Increased sebum production and follicular hyperkeratosis result in the development of microcomedones, and changes in follicular milieu in intensive growth of P. acnes. P. acnes secretes several proinflammatory products, which play an important role in the development of inflammation. These include lipases, proteases, hyaluron…

Acne vulgaris and malignancy.

Seventy patients with lymphomas and leukaemias were interviewed. Details of past acne vulgaris were recorded and a clinical examination performed. A history of acne was less common in the patients than in matched controls. In addition, fewer patients had received treatment prescribed by a medical practitioner for their acne than had the controls. Examination revealed fewer patients with acne. These results suggest a reduced incidence of acne or its more severe forms in patients who have developed lymphomas and leukemias, and provide some support for the hypothesis that increased exposure or re
