First Determination of Pyrethroid Knockdown Resistance Alleles in Human Head Lice (Phthiraptera: Pediculidae) From Chile.

The infestation with the human ectoparasite, Pediculus humanus capitis (De Geer), is a common public health problem affecting schoolchildren worldwide. In Chile, the main active ingredients present in the over-the-counter pediculicides contain pyrethroids. Despite the extended use of these products, there is no evidence of the insecticide resistance status of the head lice geographically located in Chile. The most extended resistant mechanism of pyrethroids consists of the target site insensitivity (Kdr) determined by the presence of mutations linked to insecticide-binding sites in the voltage

The mass use of deltamethrin collars to control and prevent canine visceral leishmaniasis: A field effectiveness study in a highly endemic area.

BACKGROUND: Visceral leishmaniasis (VL) is a zoonosis of great importance. Limitations in current VL control measures compromise efficacy, indicating the need to implement new strategies. The aim of this study was to evaluate the effectiveness of the mass use of deltamethrin-impregnated collars in dogs as a public health measure to control and prevent canine visceral leishmaniasis (CVL). METHODOLOGY: An interventional study was implemented in two endemic areas in the district of Monte Gordo (Bahia-Brazil): an intervention area, in which VL seronegative dogs were collared, and a control area i

Burrowing mayfly Ephemera orientalis (Ephemeroptera: Ephemeridae) as a new test species for pesticide toxicity.

The potential of mayfly Ephemera orientalis McLachlan eggs and first-instar larvae in ecotoxicological testing was investigated. Both stages of E. orientalis showed high tolerance to various environmental variables, such as water temperature, pH, water hardness, and dissolved organic carbon. Toxicological assays were conducted with three insecticides (emamectin benzoate, endosulfan, and cypermethrin), one fungicide (mancozeb), and one herbicide (paraquat dichloride). The two toxicity endpoints for the assay were the 14-day egg median hatching rate (EHC50) in static and renewal exposure systems

Deltamethrin-mediated survival, behavior, and oenocyte morphology of insecticide-susceptible and resistant yellow fever mosquitos (Aedes aegypti).

Insecticide use is the prevailing control tactic for the mosquito Aedes aegypti, a vector of several human viruses, which leads to ever-increasing problems of insecticide resistance in populations of this insect pest species. The underlying mechanisms of insecticide resistance may be linked to the metabolism of insecticides by various cells, including oenocytes. Oenocytes are ectodermal cells responsible for lipid metabolism and detoxification. The goal of this study was to evaluate the sublethal effects of deltamethrin on survival, behavior, and oenocyte structure in the immature mosquitoes o

Identification of QTLs Conferring Resistance to Deltamethrin in Culex pipiens pallens.

Culex pipiens pallens is the most abundant Culex mosquito species in northern China and is an important vector of bancroftian filariasis and West Nile virus. Deltamethrin is an insecticide that is widely used for mosquito control, however resistance to this and other insecticides has become a major challenge in the control of vector-borne diseases that appear to be inherited quantitatively. Furthermore, the genetic basis of insecticide resistance remains poorly understood. In this study, quantitative trait loci (QTL) mapping of resistance to deltamethrin was conducted in F2 intercross segregat

In vitro acaricidal activity of Murraya koenigii (L.) Spreng (Rutaceae) extracts against synthetic pyrethroid-resistant Rhipicephalus (Boophilus) microplus.

Larval packet test was used for detection of resistance status against cypermethrin and deltamethrin, the most commonly used synthetic pyrethroids in Rhipicephalus (Boophilus) microplus collected from Faridkot district, Punjab (India). The slope of mortality, lethal concentration for 50 % (LC50) and resistance levels were determined from the regression graphs of probit mortality of ticks plotted against log values of increasing concentrations of cypermethrin and deltamethrin. Results indicated presence of resistance of levels I and II against cypermethrin (resistance factor (RF) = 2.82) and de

Occupational exposure to pesticides and consequences on male semen and fertility: a review.

Exposure to pesticides affects many body organs including reproductive system. Disorder of the reproductive system leads to infertility and therefore has been in the center of attention within the recent decades. Pesticides are one of the compounds that might reduce the semen quality in the exposed workers according to current knowledge. Although many underlying mechanisms have been proposed, the mechanisms of action are not clarified yet. The object of the present review was to criticize all the results of studies which evaluated the pesticide effects on male reproductive system. Results indi

Combined toxicity of butachlor, atrazine and λ-cyhalothrin on the earthworm Eisenia fetida by combination index (CI)-isobologram method.

Pesticides in the environment do not appear singly and usually occur as complex mixtures and their combined effect may exhibit toxicity to organisms. The individual and combined toxicities of two herbicides, atrazine and butachlor and an insecticide λ-cyhalothrin have been examined to the earthworm Eisenia fetida, as a non-target terrestrial organism, in artificial soil and filter paper tests. The order of toxicity for the individual pesticides was ranked as atrazine>λ-cyhalothrin>butachlor in both tests. We applied the combination index (CI)-isobologram method which is widely used to study ch

Insecticide resistance allele frequencies in Anopheles gambiae before and after anti-vector interventions in continental Equatorial Guinea.

Anti-malaria interventions that rely on insecticides can be compromised by insecticide-resistance alleles among malaria vectors. We examined frequency changes of resistance alleles at two loci, knockdown resistance (kdr) and acetylcholinesterase-1 (ace-1), which confer resistance to pyrethroids and DDT, and carbamates, respectively. A total of 7,059 Anopheles gambiae sensu stricto mosquitoes were analyzed from multiple sites across continental Equatorial Guinea. A subset of sites included samples collected pre-intervention (2007) and post-intervention (2009-2011). Both L1014S and L1014F resist

Tick control practices in Burkina Faso and acaricide resistance survey in Rhipicephalus (Boophilus) geigyi (Acari: Ixodidae).

Traditional systems account for 95 % of the livestock produced in Burkina Faso. Tick infestation hampers livestock productivity in this area. However, little information exists on tick-control practices used by livestock farmers. We interviewed 60 stockbreeders working in traditional farming systems to obtain the first data on tick-control practices. Sixteen farmers (27 %) did not use conventional practices: seven removed ticks by hand or plastered cattle with dung or engine oil; nine farmers treated cattle with crop pesticides. Forty-four farmers (73 %) used mainly synthetic pyrethroids (SP;

Pyrethroid tolerance in the chewing louse Bovicola (Werneckiella) ocellatus.

Equine pediculosis is a significant health and welfare issue, particularly in elderly and chronically debilitated animals. Currently infestation is controlled predominantly using topically applied pyrethroid insecticides, allowing limited scope for the rotation of drugs and increasing the risk of selection for resistance. Here the insecticidal efficacies of two pyrethroid-based products against the louse Bovicola (Werneckiella) ocellatus collected from donkeys were examined in vitro. The products were cypermethrin (Deosect™, Pfizer Ltd., 5% (w/v) cypermethrin, cutaneous spray) and permethrin (

Persistence of bifenthrin in sandy loam soil as affected by microbial community.

Soil was fortified with bifenthrin at the level of 10 μg g(-1) soil. Soil samples were drawn at regular intervals of 0, 10, 20, 30 and 40 days. For extraction of bifenthrin, soil was extracted with acetone. Clean up was done by liquid-liquid partitioning with dichloromethane after diluting with brine solution. Quantification of bifenthrin residues was done by GC using mega bore column and ECD detector. Recovery of bifenthrin in soil ranged between 92.6 % and 93.8 % at 0.5 and 1.0 μg g(-1). The instrumental limit of detection of bifenthrin was 0.005 μg mL(-1) and LOQ for soil by this method was

Chemical leukoderma.

Chemical leukoderma is defined as an acquired, hypopigmented dermatosis that results from repeated cutaneous application of an agent that destroys epidermal melanocytes in genetically susceptible patients. Chemical leukoderma may develop both at the site of contact with the chemical as well as remotely from the exposure. Avoidance of the causative agent may lead to spontaneous repigmentation, but treatments commonly used in vitiligo, such as narrow-band ultraviolet B phototherapy, PUVA photchemotherapy, or topical immunosuppressants, often are necessary. We present a case of chemical leukoderm

Biological monitoring of 3-phenoxybenzoic acid in urine by an enzyme-linked immunosorbent assay.

An enzyme-linked immunosorbent assay (ELISA) method was employed for determination of the pyrethroid biomarker, 3-phenoxybenzoic acid (3-PBA) in human urine samples. The optimized coating antigen concentration was 0.5 ng/mL with a dilution of 1:4000 for the 3-PBA antibody and 1:6000 for the enzyme conjugate. Urine samples were hydrolyzed with concentrated hydrochloric acid; extracted with dichloromethane and solvent-exchanged into a methanol/buffer solution, prior to analysis in a 96-microwell plate immunoassay. Quantitative recoveries of 3-PBA were obtained for fortified urine samples by ELIS

Prevalence of kdr-like mutations associated with pyrethroid resistance in human head louse populations in Japan.

Pyrethroid insecticides play very important roles in the control of pediculosis, which is caused by human head louse (Pediculus humanus capitis De Geer) worldwide. The development of head louse resistance to pyrethroid pediculicides has become a global issue during the last two decades. Pyrethroid resistance is associated with amino acid substitutions in the alpha-subunit of the para-sodium channel gene; these substitutions are termed kdr-like mutations. The frequencies of four mutations (D11E, M815I, T929I, and L932F) in the sodium channel gene were studied in human head louse colonies collec

Developmental toxicity of bifenthrin in embryo-larval stages of zebrafish.

Bifenthrin (BF) is widely used as a miticide in orchards, nurseries and homes due to its great photostability and insecticidal activity. Recently, extensive research has been conducted on the toxicity of BF in in vitro and in vivo assays. However, no data is so far available regarding the developmental toxicity of BF to fish in early life stages. In this study, the developmental effects of BF were evaluated in embryo-larval zebrafish. At specified stages (24, 48, 72, and 96hpf), spontaneous movement, survival and hatching as well as non-lethal malformation like curved body axis or edema were d

A biological test to quantify pyrethroid in impregnated nets.

First instar larvae of Aedes aegypti (L.) (Diptera: Culicidae) are very susceptible to deltamethrin, which kills all larvae at a very low dose (0.1 microg/l). Thus the sensitivity of this insect to detect that pyrethroid is in parts per billion, as obtained with most common chromatographic methods of analysis. Here we describe a biological test (BT) to quantify deltamethrin in long-lasting insecticidal nets (LLINs) by exposing first instar larvae of Ae. aegypti to serial extracted insecticide solutions from net samples. The deltamethrin concentration in the net was calculated at the doses kill

Scabies: a ubiquitous neglected skin disease.

Scabies has been a scourge among human beings for thousands of years. Its worldwide occurrence with epidemics during war, famine, and overcrowding is responsible for an estimated 300 million people currently infested. Scabies refers to the various skin lesions produced by female mites, and their eggs and scybala that are deposited in the epidermis, leading to delayed-type hypersensitivity reaction. Recent immunological findings such as cross-reactivity with house dust mite allergens and an altered T-helper-1/T-helper-2 pattern contribute to a better understanding of the pathomechanism. Further

Development of methods for determination of the residues of 15 pesticides in medicinal herbs Isatis indigotica Fort. by capillary gas chromatography with electron capture or flame photometric detection.

Two multiresidue methods were developed for the determination of 15 pesticides (organochlorines, organphosphorus compounds, pyrethroids, and fungicides) in medicinal herbs Isatis indigotica Fort. and its formulations. The analytical procedure is based on ultrasonic assisted extraction and liquid-liquid extraction (LLE). After solvents were added, the raw material or granule sample was sonicated in an ultrasonic water bath and then centrifuged, filtered, and cleaned up by LLE. The infusion sample was extracted with petroleum ether by LLE. The pesticide residues were determined by capillary gas
