Redescription of Corallobothrium solidum (Cestoda: Proteocephalidea) and erection of a new genus, Essexiella, for tapeworms from channel catfishes (Ictaluridae).

The proteocephalidean tapeworm, Corallobothrium solidum, type species of the genus, is redescribed on the basis of the examination of its type specimens and extensive material recently collected from Malapterurus electricus (type host). Some morphological characteristics of taxonomic importance are reported for the first time, such as the presence of semispherical (U-shaped) sphincters on the external (outer) margin of the suckers, a vaginal sphincter, a well-developed seminal receptacle, and a unique morphology of the eggs. Corallobothrium solidum differs from the 2 remaining species of the g

The nematode Pristionchus pacificus (Nematoda: Diplogastridae) is associated with the oriental beetle Exomala orientalis (Coleoptera: Scarabaeidae) in Japan.

Pristionchus pacificus has been developed as a nematode satellite organism in evolutionary developmental biology. Detailed studies of vulva development revealed multiple differences in genetic and molecular control in P. pacificus compared to the model organism Caenorhabditis elegans. To place evolutionary developmental biology in a comprehensive evolutionary context, such studies have to be complemented with ecology. In recent field studies in western Europe and eastern North America we found 11 Pristionchus species that are closely associated with scarab beetles and the Colorado potato beetl

An historical and genomic view of schistosome conjugal biology with emphasis on sex-specific gene expression.

The genetic programmes associated with the sexual biology of dioecious schistosomes remain a critically important but significantly understudied area of parasitology. Throughout the last four decades, progress has been slow in describing the gross antigenic and proteomic differences linked to sexually mature schistosomes and in characterizing some of the sex-associated transcripts and regulatory mechanisms induced during developmental maturation. These investigations have been severely hindered by the lack of complete EST/genomic information, as well as corresponding post- and functional-genom

Inhibition of p-450 aromatase prevents feminisation and induces protection during cysticercosis.

Cysticercotic male mice undergo an impressive feminisation process, characterised by 200 times increased serum 17beta-estradiol levels while testosterone and dihydrotestosterone are 90% reduced, which results in elevated parasite burden. Administration of Fadrozole (an aromatase inhibitor) in male and female mice suppressed the production of 17beta-estradiol, accompanied with a 70% reduction in parasite burden. This protective effect was associated in male mice with a recovery of the specific cellular immune response. Interleukin-6 (IL-6) serum levels, and its production by splenocytes, was au

A human Ro/SS-A autoantigen is the homologue of calreticulin and is highly homologous with onchocercal RAL-1 antigen and an aplysia "memory molecule".

The Ro/SS-A (Ro) autoantigens consist of at least four immunologically distinct proteins which are recognized by autoantibodies typically found in sera from patients with primary Sjogren's syndrome and in subsets of patients with lupus erythematosus. We recently isolated a 1.9-kb human cDNA clone which encodes one of these Ro autoantigens. Synthetic oligonucleotides corresponding to the human Ro sequence were used to amplify the homologous gene from a murine B cell cDNA library using the polymerase chain reaction. The mouse cDNA-encoded amino acid sequence was found to be 94% homologous to the

Construction of Onchocerca volvulus cDNA libraries and partial characterization of the cDNA for a major antigen.

Adult Onchocerca volvulus were isolated from nodules removed from onchocerciasis patients at four locations--two in the West African Sudan-savanna region (near Bamako, Mali, and Touboro, Cameroon), one in a West African forest region (Kumba, Cameroon) and one near Guatemala City, Guatemala. Four different cDNA expression libraries were constructed in bacteriophage lambda gt11 using poly(A)+ RNA from the adult female worms. Individual cDNA clones of single copy genes were used to compare the genomes of parasites from the different locales and to show that the haploid genome of O. volvulus is 1.

Isolation and characterization of Cu/Zn-superoxide dismutase in Fasciola gigantica.

A full-length complementary DNA (cDNA) encoding Cu/Zn-superoxide dismutase was isolated from Fasciola gigantica that on nucleotide sequencing showed a close homology (98.9%) with Cu/Zn-superoxide dismutase (SOD) of the temperate liver fluke, F. hepatica. Expression of the gene was found in all the three developmental stages of the parasite viz. adult, newly excysted juvenile and metacercaria at transcriptional level by reverse transcription-polymerase chain reaction (RT-PCR) and at the protein level by Western blotting. F. gigantica Cu/Zn-SOD cDNA was cloned and expressed in Escherichia coli.

Mice deficient for 5-lipoxygenase, but not leukocyte-type 12-lipoxygenase, display altered immune responses during infection with Schistosoma mansoni.

Periovular granuloma formation during Schistosoma mansoni infection is a complex, multifaceted immunologic response. Products of arachidonic acid metabolism have been shown to contribute to this response through studies in which general inhibitors of lipoxygenase function reduce granulomatous inflammation. To determine which lipoxygenases are important for granuloma development in schistosomiasis, wild type mice or mice deficient for 5-lipoxygenase (5-LO) or "leukocyte-type" 12-lipoxygenase (12-LO) were infected with S. mansoni and studied for responses to schistosome eggs and egg antigens. P-glycoprotein-9-mediated multidrug tolerance in Caenorhabditis elegans.

BACKGROUND: The active drug efflux pumps P-glycoproteins (PGPs) are the cornerstones of multidrug resistance in many organisms. In parasitic helminths, resistance to macrocyclic lactones (MLs) has been associated with pgp regulation and structural defects in amphids. In Caenorhabditis elegans, the nuclear hormone receptor (NHR)-8 also influences xenobiotic tolerance by regulating pgp genes. However, the specific contribution of individual transporters and their regulation remain poorly defined. We recently demonstrated that PGP-9 specifically contributes to ivermectin (IVM) tolerance in an IVM

Two Divalent Metal Ions and Conformational Changes Play Roles in the Hammerhead Ribozyme Cleavage Reaction.

The hammerhead ribozyme is a self-cleaving RNA broadly dispersed across all kingdoms of life. Although it was the first of the small, nucleolytic ribozymes discovered, the mechanism by which it catalyzes its reaction remains elusive. The nucleobase of G12 is well positioned to be a general base, but it is unclear if or how this guanine base becomes activated for proton transfer. Metal ions have been implicated in the chemical mechanism, but no interactions between divalent metal ions and the cleavage site have been observed crystallographically. To better understand how this ribozyme functions

Molecular characterization of muscle-parasitizing didymozoids in marine fishes.

Classification and identification of muscle-parasitizing didymozoids found in marine fish is difficult because of their novel parasitism and morphology. Recent sequence analysis has helped, but only seven sequences are available. Therefore, the usefulness of molecular methods for differentiation of muscle-parasitizing didymozoids, as well as genetic differences between the muscle and the other site-parasitizing didymozoids are quite unclear. In the present study, six unidentified didymozoid isolates from the trunk muscles of four marine fish species (Diagramma pictum, Plectorhinchus cinctus, P

Assessing the taxonomic status of an opecoelid trematode from the evileye blaasop, Amblyrhynchote honckenii (Bloch), in South Africa.

The taxonomic status of Pseudoheterolebes cotylophorus (Ozaki 1935) Yamaguti 1959 (Opecoelidae: subfamily Opistholebetinae) has been a subject of taxonomic confusion since its initial description by Ozaki in 1935 as Opistholebes cotylophorus Ozaki 1935 from diodontid fish in Japan. The species' current recognised classification dates back to Yamaguti's reclassification of O. cotylophorus as Pseudoheterolebes cotylophorus; however, doubts persist regarding this classification and the validity of this species. The species was reported by Bray in 1986 from off the coast of South Africa, based on

Host-parasite cross-species regulation by Schistosoma japonicum EV-miRNA sja-miR-2a-3p limits hepatic fibrosis via BCL2 suppression in hepatic stellate cells.

Schistosomiasis remains an important zoonotic parasitic disease, and hepatic fibrosis is a major cause of chronic pathology following Schistosoma japonicum infection. In schistosomiasis, hepatic fibrosis is the major cause of severe pathological damage. However, during the long-term co-evolution and coexistence between Schistosoma parasites and their hosts, the parasite can modulate the host hepatic fibrotic process through extracellular vesicle (EV)-mediated cross-species communication. In this study, we identified an EV-derived microRNA from Schistosoma japonicum, sja-miR-2a-3p, that suppres

MORPHOLOGICAL OBSERVATION AND DETAILED MOLECULAR CHARACTERIZATION OF FISCHOEDERIUS ELONGATUS (DIGENEA: GASTROTHYLACIDAE) FROM THE RUMEN OF DOMESTIC CATTLE IN CAMBODIA.

Fischoederius elongatus (Poirier, 1883) is a common rumen fluke found in Asia and Africa. Nucleotide sequence data for F. elongatus have been reported but are limited to certain ribosomal and mitochondrial DNA regions. High diversity in the nucleotide sequences of the mitochondrial cytochrome c oxidase subunit 1 (COI) haplotypes has been reported in some Asian countries. Some researchers have advocated the possibility of cryptic species within F. elongatus based on the high genetic diversity within the species as well as the genetic distance within and between sister species. However, the numb

The phylogenetic position of the unusual diplostomoidean genus Schwartzitrema Vigueras, 1941 (Digenea, Diplostomoidea) and description of 2 new species from Neotropics.

Schwartzitrema Vigueras, 1941, is a small, rarely reported genus of diplostomoideans that parasitize avian definitive hosts, predominantly anhingas, on several continents. No DNA sequences were previously available for any Schwartzitrema spp. In the present study, we describe adults of 2 new Schwartzitrema spp. from anhingas in Brazil. We also provide description of metacercaria of one of the new species from Argentina. Schwartzitrema macrotesticulata n. sp. lacks a clear separation between its testes, appearing almost as a contiguous, irregularly shaped, lobular mass. A highly characteristic

High-throughput sequencing of Strongyloides stercoralis - a fatal disseminated infection in a dog.

The rhabditid nematode Strongyloides stercoralis is known worldwide as the causative agent of strongyloidiasis in humans. In addition to public health concerns, S. stercoralis also infects dogs, which represent a possible reservoir for potentially zoonotic transmissions. We describe the first confirmed case of fatal disseminated infection in a dog in the Czech Republic. The microscopic and histological results were supported by a complex genotyping approach. Using high-throughput sequencing of the hypervariable region (HVR-IV) of 18S rDNA and Sanger sequencing of the partial cytochrome c oxida

Anantrum gallopintoi sp. nov. (Bothriocephalidae Blanchard, 1849), a cestode parasite of the shorthead lizardfish Synodus scituliceps (Synodontidae) from the Pacific coast of Costa Rica.

A new species of bothriocephalid cestode in the genus Anantrum is described from the intestine of the shorthead lizardfish Synodus scituliceps from the north Pacific coast of Costa Rica. The new species is described based on an integrative taxonomic approach that includes the use of light and scanning electron microscopy, 28S rDNA sequencing, and phylogenetic analysis. Anantrum gallopintoi sp. nov. is the third known member of this genus and can be distinguished from A. tortum (Linton, 1905) and A. histocephalum Jensen & Heckmann, 1977 by a combination of morphological and ecological traits an

Possible transmission of Strongyloides fuelleborni between working Southern pig-tailed macaques (Macaca nemestrina) and their owners in Southern Thailand: Molecular identification and diversity.

Human strongyloidiasis is caused by Strongyloides stercoralis, S. fuelleborni fuelleborni and Strongyloides f. kellyi. Strongyloides fuelleborni is a soil-transmitted nematode parasite typically infecting non-human primates. The southern pig-tailed macaque (Macaca nemestrina) is distributed throughout the southern part of Thailand and could be a source of zoonotic transmission of this nematode. Here, we extracted DNA from Strongyloides speciescultured from the feces of southern pig-tailed macaques and their owners. Using PCR and sequencing of the extracted DNA, we compared the nucleotide seque

The first determination of Trichuris sp. from roe deer by amplification and sequenation of the ITS1-5.8S-ITS2 segment of ribosomal DNA.

Trichuris nematodes were isolated from roe deer (Capreolus capreolus). At first, nematodes were determined using morphological and biometrical methods. Subsequently genomic DNA was isolated and the ITS1-5.8S-ITS2 segment from ribosomal DNA (RNA) was amplified and sequenced using PCR techniques. With u sing morphological and biometrical methods, female nematodes were identified as Trichuris globulosa, and the only male was identified as Trichuris ovis. Functional analysis of putative operons in Brugia malayi.

Operons are a common mode of gene organization in Caenorhabditis elegans. Similar gene arrangements suggest that functional operons may exist in Brugia malayi. To definitively test this hypothesis, a bicistronic reporter vector consisting of an upstream firefly luciferase gene and a downstream renilla luciferase gene was constructed. The genome was then surveyed to identify 15 gene pairs that were likely to represent operons. Two of four domains upstream of the 5' gene from these clusters exhibited promoter activity. When constructs replicating the promoter and intergenic arrangement found in
