Abnormal RNA metabolism is one of the core mechanisms in tumorigenesis. DDX23, a member of the family of DEAD-box protein, characterized as an ATP-dependent RNA helicase, facilitates cancer progression due to its abnormal expression. However, systematic studies examining DDX23 across the pan-cancer spectrum remain scarce. We utilized databases like TCGA and GEO to thoroughly examine the expression pattern of DDX23 in our study, its prognostic value, diagnostic efficacy, and immune-related properties across multiple cancers. We foresee possible regulatory routes by building a network of protein

Mosquito-borne arboviruses, including dengue virus (DENV), Zika virus (ZIKV), and chikungunya virus (CHIKV), represent a growing global health burden, yet no specific antiviral therapies are currently available for most of these infections. This unmet need highlights the importance of identifying novel antiviral compounds from biologically relevant sources such as plant-derived natural products. In this study, a bioactivity-guided approach was applied to methanolic extracts from eight Solanaceae species to identify compounds with antiviral activity against DENV-2 in Vero cells. Five extracts s

BACKGROUND: Singleton-Merten syndrome (SGMRT) is a rare immunogenetic disorder that variably features juvenile open-angle glaucoma (JOAG), psoriasiform skin rash, aortic calcifications and skeletal and dental dysplasia. Few families have been described and the genotypic and phenotypic spectrum is poorly defined, with variants in DDX58 (DExD/H-box helicase 58) being one of two identified causes, classified as SGMRT2.
METHODS: Families underwent deep systemic phenotyping and exome sequencing. Functional characterisation with in vitro luciferase assays and in vivo interferon signature using bulk

Triple-negative breast cancer (TNBC) is a clinically aggressive subtype lacking targeted therapies, often characterized by hyperactivation of the Wnt/β-catenin signaling pathway and an enriched population of cancer stem cells (CSCs). Here, we identify the DEAD-box RNA helicase DP103 as a novel modulator of Wnt/β-catenin signaling in TNBC, acting independently of its canonical helicase function. DP103 expression correlates with increased phosphorylation of LRP6 and nuclear β-catenin accumulation, enhancing Wnt transcriptional activity. Mechanistically, DP103 physically interacts with GSK3β to f

We present a comprehensive overview of the dependency of several Old World alphaviruses for the host protein G3BP. Based on their replication ability in G3BP-deleted cells, Old World alphaviruses can be categorized into two groups, being either resistant or sensitive to G3BP deletion. We observed that all sensitive viruses have an Arg residue at the P4 position of the cleavage site between the nonstructural protein P1 (nsP1) and nsP2 regions of the replicase precursor polyprotein (1/2 site), while a different residue is found at this site in viruses resistant to G3BP deletion. Swapping this re

RNA-binding proteins (RBPs) govern mRNA fate and critically regulate spermatogenesis. The poly(rC)-binding protein 2 (PCBP2), a testis-enriched RBP, remains uncharacterized in spermatogenesis. In this study, we identified and validated PCBP2 downregulation in testicular tissues from patients with non-obstructive azoospermia (NOA) by integrating bulk and single-cell RNA sequencing (scRNA-seq) datasets. To investigate its function, we generated mice with a germline-specific ablation of Pcbp2. These mice exhibited delayed meiotic prophase and impaired spermiogenesis. Specifically, we observed a r

In recent years, Zika virus infections have spread geographically across various territories, including French Polynesia, the South Pacific, and South America, with a significant impact on Brazil. Severe Zika virus infections can lead to neurological complications, such as Guillain-Barré syndrome in adults. Furthermore, infection during pregnancy can result in microcephaly and other congenital brain malformations in the fetus. Aedes aegypti cohabits with humans and is the principal vector responsible for transmitting Zika virus to humans. Recent advances have elucidated the molecules involved

Stress granules (SGs) fulfill a pivotal role in host defense mechanisms, by sequestering both mRNA and protein via the process of liquid-liquid phase separation (LLPS). In this study, we showed that perillaldehyde (PAE), a natural occurring compound, bound directly to the core protein of SGs, Ras GTPase-activating protein-binding protein 1/2 (G3BP1/2), thereby inducing the assembly of SGs through the LLPS of G3BP/RNA complexes in vitro. Moreover, in Parkinson's disease (PD) models using Caenorhabditis elegans (C. elegans) and mice, PAE administration prompted SG formation, enhanced eIF2α phosp

Zika virus (ZIKV) has rapidly emerged as a global public health concern. Viral NS2B-NS3 protease processes viral polyprotein and is essential for the virus replication, making it an attractive antiviral drug target. We report crystal structures at 1.58-angstrom resolution of the unlinked NS2B-NS3 protease from ZIKV as free enzyme and bound to a peptide reversely oriented at the active site. The unlinked NS2B-NS3 protease adopts a closed conformation in which NS2B engages NS3 to form an empty substrate-binding site. A second protease in the same crystal binds to the residues K14K15G16E17 from t

BACKGROUND: Hereditary spastic paraplegia is a neurodegenerative phenotype characterized by a progressive loss of corticospinal motor tract function. In a majority of affected individuals the pathogenesis remains undetermined.
METHODS: We identified a series of patients with a phenotype of nonsyndromic spastic paraplegia in whom no diagnosis had been reached before exome sequencing. We measured the expression of interferon stimulated genes (ISGs) in peripheral blood from these patients.
RESULTS: Five patients from four families with previously unexplained spastic paraplegia were identified w

BACKGROUND: The Thr allele at the non-synonymous single-nucleotide polymorphism (nsSNP) Thr946Ala in the IFIH1 gene confers risk for Type 1 diabetes (T1D). The SNP is embedded in a 236 kb linkage disequilibrium (LD) block that includes four genes: IFIH1, GCA, FAP and KCNH7. The absence of common nsSNPs in the other genes makes the IFIH1 SNP the strongest functional candidate, but it could be merely a marker of association, due to LD with a variant regulating expression levels of IFIH1 or neighboring genes.
METHODOLOGY/PRINCIPAL FINDINGS: We investigated the effect of the T1D-associated variat

Alkhurma virus (ALKV) is a recently discovered class-4 flavivirus that was responsible for several cases of severe haemorrhagic fever in humans in Saudi Arabia. It has been shown for other flaviviruses that processing of the viral polyprotein is partly due to the virus-encoded NS2B/NS3 trypsin-like serine protease. As the viral proteinase plays a critical role in the virus replication cycle, it represents one of the main targets for antiviral therapy against members of the Flavivirus genus. We report here on the identification of the ALKV NS2B and NS3 domains and the expression and purificatio

There are two types of infection caused by pathogenic microorganisms, intracellular infection and intercellular infection. Infection of pathogenic leptospira is an intercellular infection. The immunological reaction of host to intercellular infection is unique. The potential immunogen of an expressed protein should meet three criteria: it can be degraded (by antigen-present cells in the host); it should have antigenic epitope which can be recognized by specific antibodies and have at least one epitope that can be recognized by an MHC II protein and T cell receptor. In this study we report the

We report the first case in Western Europe of a person presenting with dermatomyositis associated with melanoma differentiation antigen 5 antibodies. She sequentially developed severe mucocutaneous erythematous and itchy lesions of the face, scalp, neck, knees and recurrent aphthae. In addition she presented painful, periungual, edematous digital lesions and small ulcers with digital necrosis. Her rapidly evolving, near-fatal interstitial lung disease responded to high-dose intravenous cyclophosphamide. However, her recurrent mucocutaneous manifestations improved only after rituximab administr

PURPOSE OF REVIEW: Type 1 diabetes (T1D) results from interplay between genetic predisposition, immune system, and environmental factors. Epidemiological and experimental data strongly suggest a role for enteroviruses in the development of T1D, but a lot of controversies and unanswered questions remained. This review focuses on issues that are fueling debate.
RECENT FINDINGS: Beyond HLA genes, which provide genetic susceptibility for T1D, other loci have been identified to be associated with the disease. There is a link between T1D and single-nucleotide polymorphisms (SNPs) in the interferon-

The role of innate immune receptors, specifically RNA sensors, in non-pathogen-driven lung fibrosis remains unclear. Here, we demonstrate that the cytosolic RNA sensor MDA5 could translocate to the cell surface upon bleomycin-induced damage. MDA5-deficient mice exhibited exacerbated lung fibrosis with increased IL-17A⁺ γδ T cells and neutrophils, which is independent of the canonical MAVS pathway and type I interferon signaling. Phenotypically, MDA5-deficient lung CD11b⁺ macrophages, which resemble human SPP1⁺ macrophages, showed elevated expression of CCR2 ligands. Mechanistically, cell-surfa

The efficient transmission of SARS-CoV-2 caused the COVID-19 pandemic, which affected millions of people around the globe. Despite extensive efforts, specific therapeutic interventions and preventive measures against COVID-19 and its consequences, such as long COVID, have not yet been identified due to the lack of a comprehensive knowledge of the SARS-CoV-2 biology. Therefore, a deeper understanding of the sophisticated strategies employed by SARS-CoV-2 to bypass the host antiviral defense systems is needed. One of these strategies is the inhibition of the Ras GTPase-activating protein-binding

West Nile virus (WNV) nonstructural protein 3 (NS3) harbors the viral triphosphatase and helicase for viral RNA synthesis and, together with NS2B, constitutes the protease responsible for polyprotein processing. NS3 is a soluble protein, but it is localized to specialized compartments at the rough endoplasmic reticulum (RER), where its enzymatic functions are essential for virus replication. However, the mechanistic details behind the recruitment of NS3 from the cytoplasm to the RER have not yet been fully elucidated. In this study, we employed immunofluorescence and biochemical assays to demo
