BACKGROUND: One of the challenges faced by equine breeders is ensuring delivery of good quality semen doses for artificial insemination when the mare is due to ovulate. Single Layer Centrifugation (SLC) has been shown to select morphologically normal spermatozoa with intact chromatin and good progressive motility from the rest of the ejaculate, and to prolong the life of these selected spermatozoa in vitro. The objective of the present study was a proof of concept, to determine whether fertilizing ability was retained in SLC-selected spermatozoa during prolonged storage.

A bacteriocin produced by Pediococcus acidilactici had an inhibitory and bactericidal effect on Listeria monocytogenes associated with fresh meat. MICs were significantly lower than minimum killing concentrations. When meat was inoculated with L. monocytogenes, the bacteriocin reduced the number of attached bacteria in 2 min by 0.5 to 2.2 log cycles depending upon bacteriocin concentration. Meat treated initially with the bacteriocin resulted in attachment of 1.0 to 2.5 log cycles fewer bacteria than that attained with the control. Investigations of recent outbreaks and sporadic cases of listeriosis in North America and other countries have shown that the infection can be foodborne. Pregnant women and their fetuses, the elderly, and immuno-compromised individuals are at increased risk of listeriosis. Listeria monocytogenes, the causative bacterium, is widespread in nature and is found in many foods. It can grow at refrigerator temperatures and is resistant to adverse conditions and to several antibiotics. The generally low levels of the organism found in foods require procedures to optimize their detection and recovery.

A case of brucellosis due to Brucella abortus, biotype 5, occurred in a bulk milk-tanker driver who collected milk from refrigerated tanks on 23 farms. The organism was isolated from blood cultures by using a serum dextrose broth containing antibiotics. Serological investigations indicate that both abortus and melitensis antigen suspensions should be used in the investigation of cases of suspected brucellosis. Detection of Burkholderia pseudomallei in blood culture is critical to prompt appropriate treatment. Various factors may affect culture positivity. This study evaluated effects of various inocula (0.2-100 CFU/mL), delayed bottle loading periods (12-48 h), and storage temperatures (ambient, 4 and 35 degrees C) for the detection of B. pseudomallei by the BacT/ALERT system. The results suggested that bottles may be preincubated at 35 degrees C for up to 12 h, or held at ambient temperature for up to 24 h, or refrigerated at 4 degrees C for up to 48 h.

This study is the first systematic documentation of the potency of monovalent oral polio vaccine type 3 (mOPV3) kept at ambient temperatures during a polio immunization campaign in Chad.
