Effects of Mel B arsobal and alpha-difluoromethylornithine on the awakening electroencephalogram of humans with gambiense trypanosomiasis disease: preliminary report.

Five males and three females, at the encephalic stage of sleeping sickness, were submitted to trypanocide therapies. Three of the patients were treated with the Mel B Arsobal drug, the five others with difluoromethylornithine, using different protocols. Awakening electroencephalographic data were obtained prior to treatment and, at regular intervals, during and after treatment. Prior to treatment the awakening tracings showed important abnormalities (slow delta waves were superimposed on theta background rhythms). During treatment (except in one patient treated with Arsobal) recordings returne

Electroencephalogram in developmental delay: specific electroclinical syndromes.

The electroencephalogram (EEG) plays an important role in the evaluation of a child with developmental delay. An EEG is often required to classify seizures in children with developmental delay. Equally important is the role of the EEG in the identification of specific electroclinical syndromes in children who may or may not manifest seizures. Specific electroclinical syndromes include the acquired epileptiform aphasia syndrome, Landau-Kleffner syndrome, and electrical status epilepticus during slow wave sleep. Other clinical situations where the EEG offers diagnostic and prognostic information

Transcallosal evoked potentials in relation to behavior in the rat: effects of atropine, p-chlorophenylalanine, reserpine, scopolamine and trifluoperazine.

Single pulse electrical stimulation of the sensorimotor cortex in waking rats produced an evoked response in the contralateral sensorimotor cortex. The slow wave response consisted of: (1) an early component that was negative at the pial surface and in layer V, and was associated with multiunit discharge; and (2) a late component that was mainly negative at the surface, positive in layer V, and was associated with multiunit suppression. Previous research suggests that the early component represents summed excitatory postsynaptic potentials; the late component summed inhibitory postsynaptic pot

Nutritional modulators of sleep: A narrative review of vitamins, minerals, amino acids, and their neurobiological and chronoepigenetic mechanisms.

This narrative review synthesizes evidence on how vitamins, minerals, and amino acids modulate sleep through neurobiological, circadian, and epigenetic pathways. B-group vitamins (B6, B12, folate) are cofactors in the synthesis of serotonin and gamma-aminobutyric acid (GABA). The lack of these factors increases the excitability of neurons and interferes with sleep. Vitamin D exerts its regulation through its nuclear receptor by controlling the key clock genes and melatonin secretion, and suppresses neuroinflammation. Magnesium and calcium modulate neuronal excitability and melatonin synthesis

Chronic obstructive pulmonary disease and obstructive sleep apnoea-the overlap syndrome.

Chronic obstructive pulmonary disease (COPD) and obstructive sleep apnoea (OSA) are highly prevalent disorders and the co-existence of both disorders, termed the overlap syndrome, affects at least 1% of the adult population. Patients with the overlap syndrome typically experience more pronounced nocturnal oxygen desaturation and there is a high prevalence of pulmonary hypertension in such patients. Recent evidence suggests that the prevalence of each disorder together is higher than might be predicted by simple prevalence statistics, although the evidence is not clear-cut in this regard. Sleep

Fatal familial insomnia: a model disease in sleep physiopathology.

Fatal Familial Insomnia (FFI) is characterized by loss of sleep, oneiric stupor with autonomic/motor hyperactivity and somato-motor abnormalities (pyramidal signs, myoclonus, dysarthria/dysphagia, ataxia). Positon emission tomography (PET) disclosed thalamic hypometabolism and milder involvement of the cortex; neuropathology severe neuronal loss in the thalamic nuclei variably affecting the caudate, gyrus cinguli and fronto-temporal cortices. Genetic analysis disclosed a mutation in the PRNP gene and FFI was transmitted to experimental animals, thus classifying FFI within the prion diseases. R

Effects of antibiotics, minocycline and ampicillin, on human sleep.

The effects of two kinds of antibiotics, minocycline (MNC) and ampicillin (AB-PC), on human sleep were investigated on 19 healthy male students to test for a relationship between human sleep and protein synthesis. These drugs and placebos were capsulated identically in appearance and were given to the subjects using the single blind method. MNC has been proven to prevent protein synthesis whereas AB-PC does not inhibit protein synthesis, and both antibiotics are commonly used in clinical practice. With the administration of a single dose of 200 mg of MNC, an apparent decrease in slow wave slee

Sleep architecture changes during a trek from 1400 to 5000 m in the Nepal Himalaya.

The aim of this study was to examine sleep architecture at high altitude and its relationship to periodic breathing during incremental increases in altitude. Nineteen normal, sea level-dwelling volunteers were studied at sea level and five altitudes in the Nepal Himalaya. Morning arterial blood gases and overnight polysomnography were performed in 14 subjects at altitudes: 0, 1400, 3500, 3900, 4200 and 5000 m above sea level. Subjects became progressively more hypoxic, hypocapnic and alkalinic with increasing altitude. As expected, sleep architecture was affected by increasing altitude. While

Nontraumatic "spindle coma": clinical, EEG, and prognostic features.

"Spindle coma" denotes the association of alteration of consciousness with electroencephalographic (EEG) activity resembling slow-wave sleep. Clinical, EEG, and pathologic data were reviewed for 36 patients with this syndrome, who were divided into two groups, based on the level of consciousness at the time of the initial neurologic evaluation. Fifteen patients were stuporous or semicomatose; none exhibited abnormalities of brainstem motor function, and 14 survived. Twenty-one patients were comatose; 20 had abnormalities of brainstem motor function, and only 4 survived. The outcome of nontraum

Characterization of sleep disorders in patients with E200K familial Creutzfeldt-Jakob disease.

The largest cluster of E200K familial Creutzfeldt-Jakob disease (fCJD) which occurs is in Jews of Libyan origin in Israel. Insomnia is a very common early complaint in those patients and may even be the presenting symptom. The aim of this study was to assess and characterize sleep pathology in E200K fCJD patients. To do so, sleep studies of 10 consecutive fCJD patients were compared with those of 39 age and gender-matched controls. All patients presented pathological sleep characterized by fragmentation of sleep, loss of sleep spindles and reduced REM sleep amount. Respiration was characterize

Neurophysiological aspects of neonatal seizures.

Recently, amplitude-integrated EEG (aEEG) has been increasingly used and proved useful in neonatal intensive care units (NICU) for the management of neonatal seizures. It does not replace, but is supplementary to standard EEG. This article reviews some of findings obtained with standard EEGs, and tries to interpret them with recent findings in the field of basic science. Seizures mainly occur in active-REM sleep in neonates. This is in sharp contrast to those in older children and adults, in whom epileptic seizures occur mainly in NREM sleep. This may be explained by neurotransmitter effects o

Transient intracranial pressure elevations (B waves) are associated with sleep apnea.

BACKGROUND: Repetitive transient intracranial pressure waveform elevations up to 50 mmHg (ICP B-waves) are often used to define pathological conditions and determine indications for ICP-reducing treatment. We recently showed that nocturnal transient ICP elevations are present in patients without structural brain lesions or hydrocephalus in whom they are associated with sleep apnea. However, whether this signifies a general association between ICP macropatterns and sleep apnea remains unknown. METHODS: We included 34 patients with hydrocephalus, or idiopathic intracranial hypertension (IIH), w

Improvement of intensive care unit sound environment and analyses of consequences on sleep: an experimental study.

OBJECTIVE: Uninterrupted sleep is of vital importance for restoration and regaining health. In intensive care units (ICUs) where recovering and healing is crucial, patients' sleep often is fragmented and disturbed due to noise from activities from oneself, other patients, and alarms. The aim of our study was to explore if sleep could be improved by modifying the sound environment in a way that is practically feasible in ICUs. METHODS: We studied the effects of originally recorded ICU noise and peak reduced ICU noise on sleep in healthy male participants. Sleep was registered with polysomnogra

Biomechanics-based active control of bedding support properties and its influence on sleep.

Proper body support plays an import role in the recuperation of our body during sleep. Therefore, this study uses an automatically adapting bedding system that optimises spinal alignment throughout the night by altering the stiffness of eight comfort zones. The aim is to investigate the influence of such a dynamic sleep environment on objective and subjective sleep parameters. The bedding system contains 165 sensors that measure mattress indentation. It also includes eight actuators that control the comfort zones. Based on the measured mattress indentation, body movements and posture changes a

Potentiating effect of diltiazem on pentobarbital-induced hypnosis is augmented by serotonergic system: the TMN and VLPO as key elements in the pathway.

To investigate the mechanism by which L-type Ca+ channel blockers exerted potentiating effects on pentobarbital-induced hypnosis, the present study was undertaken to determine if the interaction of diltiazem and serotonergic system influences the architecture of pentobarbital sleep in rats and examined c-Fos expression in the ventrolateral preoptic nucleus (VLPO) and the tuberomammillary nucleus (TMN). The polysomnogram consisting of EEG and EMG was recorded for analyzing sleep architecture. The results showed that diltiazem (2.0 and 5.0 mg/kg, p.o.) increased both total pentobarbital sleep an

Long-term polysomnographic study of the efficacy and safety of zolpidem in elderly psychiatric in-patients with insomnia.

The effects of 20 mg zolipidem were studied in an open, polysomnographically-monitored 179 day trial in 14 elderly psychiatric patients suffering from severe insomnia. After a placebo run-in of 7 days, zolpidem was given for 179 days followed by a 30-day wash-out period. Polysomnographic recordings (PSG) were performed just before active treatment; 30, 90 and 179 days into the treatment period; and at the end of the wash-out period. Statistically significant improvements in total sleep time, sleep efficiency and percentage of rapid eye movement sleep were observed after 30 days, all of which w

Electrocortical spectrum power effects of different classes of neuroleptics in rats.

In freely moving rats the effects on behaviour and ECoG spectrum power of different classes of neuroleptics (phenothiazines, butyrophenones and benzamides) after systemic and intraventricular administration were studied. Chlorpromazine and haloperidol given systemically or microinjected into the third cerebral ventricle produced behavioural and ECoG slow-wave sleep (SWS) accompanied by a significant increase in ECoG total voltage power as well as in the lower frequency bands; haloperidol was found to be more powerful in inducing behavioural and SWS. In contrast, equimolar doses of 1-sulpiride

[Chronological change of EEG findings in a case of pyridoxine dependency seizures].

A patient of pyridoxine dependent seizures was reported. He was born at 34 weeks' gestation and weighted 2,760 g. Apgar scores were 6 and 9 at 1 and 5 minutes, respectively. He showed the first seizure 2 hours after his birth. Phenobarbital, phenytoin, sodium valproate, diazepam and clonazepam were not effective. Pyridoxal phosphate (50 mg) was given intravenously, resulting in suppression of convulsions. However, muscle tonus was severely depressed. In EEG, a discontinuous pattern was found in quiet and indeterminate sleep on the 2nd day of life. At 5th week multifocal spikes were found, and

Sleep-neuroinflammation interactions in Alzheimer's disease: a systematic review of biomarker pathways.

Although sleep disturbances are increasingly associated with Alzheimer's disease (AD), the mechanistic interplay between sleep physiology, neuroinflammation and AD-related pathology is not fully understood. The question of whether disrupted sleep contributes to neurodegeneration or merely reflects early pathological processes is still being debated. In accordance with PRISMA-2020 guidelines, we conducted a systematic review to synthesize the current evidence linking objectively measured sleep alterations, glial activation and AD-related outcomes.
