alpha-Phenyl-N-tert-butylnitrone provides protection from dextran sulfate sodium-induced colitis in mice.

Nuclear factor-kappaB (NF-kappaB)-dependent up-regulation of inflammatory cytokines and inducible nitric oxide (iNOS) occurs in inflammatory bowel disease. We investigated the effect of alpha-phenylN-tert-butylnitrone (PBN), a spin-trapping agent that inhibits NF-kappaB activity, on dextran sulfate sodium (DSS)-induced colonic mucosal injury and inflammation in mice. Acute colitis was induced with DSS in female BALB/c mice receiving 0, 0.3, 3, and 30 mg/kg i.p. PBN daily. Colonic mucosal inflammation was evaluated biochemically and histologically. Nitric oxide was evaluated as luminal nitrite/

In vivo production of hydroxyl radical by Enterococcus faecalis colonizing the intestinal tract using aromatic hydroxylation.

Enterococcus faecalis is an intestinal commensal that produces extracellular superoxide (O(2)(*-)) through autoxidation of membrane-associated demethylmenaquinone. To assess free radical production by E. faecalis in vivo, intestinal tracts of rats were colonized using wild-type E. faecalis or a mutant strain with attenuated O(2)(*-) production. Ex vivo electron paramagnetic resonance spin trapping study of colonic contents (mean +/- SD) showed 1.4 +/- 1.5 and 0.094 +/- 0.24 microM 5,5-dimethyl-1-pyrroline-N-oxide-hydroxyl radical adduct/gm stool for rats colonized with wild-type and mutant str

Alterations in cytochrome c oxidase activity and energy metabolites in response to kainic acid-induced status epilepticus.

The effects of kainic acid (KA)-induced limbic seizures have been investigated on cytochrome c oxidase (COx) activity, COx subunit IV mRNA abundance, ATP and phosphocreatine (PCr) levels in amygdala, hippocampus and frontal cortex of rat brain. Rats were killed either 1 h, three days or seven days after the onset of status epilepticus (SE) by CO2 and decapitation for the assay of COx activity and by head-focused microwave for the determination of ATP and PCr. Within 1 h COx activity and COx subunit IV mRNA increased in all brain areas tested between 120% and 130% of control activity, followed

Production of lipid-derived free radicals in L1210 murine leukemia cells is an early oxidative event in the photodynamic action of Photofrin.

Photofrin photosensitization initiates a sequence of oxidative events that begins with singlet oxygen formation and ultimately leads to cell death. We hypothesize that membrane lipid-derived free radical formation is an early event in this process. In the presence of iron and ascorbate, lipid free radicals are generated during cellular photosensitization of L1210 cells as detected by electron paramagnetic resonance spin-trapping techniques. Tocopherol levels decline in an inverse manner to lipid radical formation. Trypan blue dye exclusion by membranes also decreases inversely to lipid radical

N-tert-butyl-alpha-phenylnitrone improves recovery of brain energy state in rats following transient focal ischemia.

Recent results have demonstrated that the spin trapping agent N-tert-butyl-alpha-phenylnitrone (PBN) reduces infarct size due to middle cerebral artery occlusion (MCAO), even when given after ischemia. The objective of the present study was to explore whether PBN influences recovery of energy metabolism. MCAO of 2-hr duration was induced in rats by an intraluminal filament technique. Brains were frozen in situ at the end of ischemia and after 1, 2, and 4 hr of recirculation. PBN was given 1 hr after recirculation. Neocortical focal and perifocal ("penumbra") areas were sampled for analyses of

Free radical-mediated lipid peroxidation in cells: oxidizability is a function of cell lipid bis-allylic hydrogen content.

Oxidizability of lipids in homogeneous solution varies linearly with the extent of their unsaturation. In vitro cellular, as well as in vivo, studies of oxidizability have generally relied upon chemical indicators of peroxidation such as thiobarbituric acid-reactive substances. To examine the oxidizability of lipids in cells, we have measured oxygen uptake and, using electron paramagnetic resonance spin trapping with alpha-(1-oxo-4-pyridyl)-N-tert-butylnitrone (POBN), the real time generation of lipid-derived free radicals. We have used our experimental in vitro cellular lipid modification mod

Bioactivation of N-hydroxy-2-acetylaminofluorenes by N,O-acyltransferase: substituent effects on covalent binding to DNA.

N-Acetoxyarylamines are reactive metabolites that lead to arylamine adduct formation with biological macromolecules. A series of 7-substituted-N-hydroxy-2-acetylaminofluorenes were converted to reactive N-acetoxyarylamines by enzymatic N,O-acyltransfer in the presence of DNA. The N-arylhydroxamic acid substrates that contained electronegative 7-substituents formed greater amounts of DNA adducts than either the unsubstituted compound (N-OH-AAF) or those analogs that contained electron-donating groups in the 7-position. Glutathione did not decrease the rates of DNA adduct formation, but other nu

Decreased plasma superoxide scavenging activity in immunological disorders--carboxyethylgermanium sesquioxide (Ge-132) as a promoter of prednisolone.

We investigated so-called superoxide scavenging activity (SSA) of plasma in patients with several immunological disorders, such as rheumatoid arthritis (RA), systemic lupus erythematosus (SLE), polymyo-dermatomyositis (PM), progressive systemic sclerosis (PSS), myasthenia gravis (MG) and autoimmune thyroid disease (AT), using the electron paramagnetic resonance/spin trapping technique. Since carboxyethylgermanium sesquioxide, Ge-132, has been reported to modulate both the immune response and leukocyte functions, we have studied in vivo effect of Ge-132 on plasma SSA and other laboratory parame

Water concentrations across lipid bilayers by HYSCORE spectroscopy of nitroxides.

Structural organization of cellular membranes and membrane proteins is governed by the hydration water. Distribution of water molecules across lipid bilayers is highly heterogeneous due to the amphiphilic nature of this barrier, consisting of a 3- to 4-nm-thick hydrophobic core and a hydrophilic region formed by lipid polar head groups. Hyperfine sublevel correlation (HYSCORE) spectroscopy is capable of selective detection of hyperfine interactions originating from water molecules H-bonded directly to the N‒O⋅ group of the nitroxide. Here, we describe a high-spatial-resolution method to measur

ESR evidence for in vivo formation of free radicals in tissue of mice exposed to single-walled carbon nanotubes.

Nanomaterials are being utilized in an increasing variety of manufactured goods. Because of their unique physicochemical, electrical, mechanical, and thermal properties, single-walled carbon nanotubes (SWCNTs) have found numerous applications in the electronics, aerospace, chemical, polymer, and pharmaceutical industries. Previously, we have reported that pharyngeal exposure of C57BL/6 mice to SWCNTs caused dose-dependent formation of granulomatous bronchial interstitial pneumonia, fibrosis, oxidative stress, acute inflammatory/cytokine responses, and a decrease in pulmonary function. Metal-organic framework-based Z-scheme heterostructure enables oxygen-independent sonodynamic immunotherapy through tumor-associated macrophage reprogramming.

Sonodynamic therapy (SDT) provides spatially controlled tumor treatment, but its efficacy in hypoxic tumors is limited by oxygen-dependent reactive oxygen species (ROS) generation and tumor-associated macrophage (TAM)-mediated immunosuppression. Here, we developed a hyaluronic acid (HA)-modified, metal-organic framework (MOF)-based CuO₂@FeTCPP Z-scheme nanosonosensitizer to combine hypoxia-tolerant hydroxyl radical (•OH) generation with macrophage reprogramming. Band-edge analysis, interfacial X-ray photoelectron spectroscopy, and spin-trapping and scavenging experiments provided complementary

Exploring the Chemical Doping of a Water-Soluble Cylindrical Micelle-Forming Conjugated Polyelectrolyte.

The chemical doping of water-soluble conjugated polyelectrolytes (CPEs) offers a promising pathway for the direct printing of semiconducting polymer films by using environmentally friendly solvents. In this study, we explored the chemical doping of the cationic cylindrical micelle-forming CPE poly(cyclopentadithiophene-alt-thiophene) (PCT-NBr) in aqueous solution using two Fe(III)-halide dopants, FeCl3 and FeBr3. Treatment with nonoxidizing salts (KCl and KBr) showed that polymer micelles preferentially interact with Br- ions over Cl- ions, resulting in a more rigid micelle and spectroscopic e

Tungsten disulfide-zeolitic imidazolate framework-8 nanoplatform loaded with gadolinium and conjugated with folic acid-PEG for bimodal CT/MRI imaging and 15 MV X-ray radiosensitization of glioblastoma and prostate cancer cells.

Glioblastoma multiforme and castration-resistant prostate cancer remain difficult to image and radiosensitize. We evaluated WS2@ZIF-8-Gd-FA-PEG, a folate-functionalized nanoplatform combining CT/MRI phantom contrast with 15 MV X-ray radiosensitization. The corrected conjugate was identified as FA-PEG-COOH, and surface functionalization was supported by 1H NMR, molecular-mass analysis, UV-visible spectroscopy, zeta potential, FTIR and XPS. In vitro BBB transport reached 8.46 ± 0.55% at 24 h for the targeted formulation versus 4.72 ± 0.31% for the non-targeted formulation, while barrier-integrit

The role of NO, H2O2, and non-NO/H2O2 mechanisms in acetylcholine (ACh)-induced dilation of human arterioles in the absence and presence of coronary artery disease.

This study defines the mechanisms of vasodilation to acetylcholine (ACh) in arterioles from patients with and without coronary artery disease (CAD). Human adipose arterioles (HAA) dissected from discarded surgical samples were cannulated and pressurized at 60 mmHg for measurement of diameter changes by videomicroscopy. No difference in baseline dose response to ACh was observed between non-CAD and CAD patients. L-NAME, NO synthase inhibitor, reduced dilation in both groups but to a greater extent in non-CAD. Peg-CAT, H2O2 scavenger, attenuated response to ACh in non-CAD but not in CAD. Inhibit

Dynamics of nitrergic system activation in the rat brain provoked by experimentally induced seizures.

Epilepsy is a pathophysiological condition displaying a highly diverse phenotype. Consequently, comprehending the mechanisms underlying seizures necessitates moving beyond a simplistic model focused on the imbalance between the classical excitatory and inhibitory neurotransmitter systems. Nitric oxide (NO), a nonclassical and multifunctional gaseous neurotransmitter, has the potential to exert a profound influence on epileptic reactivity. Unfortunately, numerous studies have not provided clear answers about its involvement in the pathophysiology of epilepsy. The objective of our study was to d

Ligand-Based Radical Reactivity of Metal Thiosemicarbazones Prompts the Identification of Platinum(II)-Based Cytoprotectants.

CuATSM, a copper(II) complex of a bis(thiosemicarbazone) of diacetyl, prevents oxidative cell death and acts as a neuroprotectant in vivo, prompting its evaluation to treat amyotrophic lateral sclerosis and other neurodegenerative conditions in the clinic. We recently demonstrated that CuATSM functions as a potent radical-trapping antioxidant (RTA), inhibiting lipid peroxidation and associated ferroptotic cell death by a noncanonical mechanism based on radical addition to the ligand backbone. Herein we report our investigations of the generality of this reactivity, which include studies of cor

Understanding the Oxidative Properties of Nickel Oxyhydroxide in Alcohol Oxidation Reactions.

The NiOOH electrode is commonly used in electrochemical alcohol oxidations. Yet understanding the reaction mechanism is far from trivial. In many cases, the difficulty lies in the decoupling of the overlapping influence of chemical and electrochemical factors that not only govern the reaction pathway but also the crystal structure of the in situ formed oxyhydroxide. Here, we use a different approach to understand this system: we start with synthesizing pure forms of the two oxyhydroxides, β-NiOOH and γ-NiOOH. Then, using the oxidative dehydrogenation of three typical alcohols as the model reac

A potential method to improve the in vitro cytotoxicity of half-sandwich Os(ii) complexes against A2780 cells.

The [Os(η6-pcym)(dpa)(VP)]PF6 (1-VP) complex contains the histone deacetylase (HDAC) inhibitor valproate (2-propylpentanoate; VP) as a monodentate O-donor ligand and shows ca. 3-fold higher in vitro cytotoxicity against A2780 human ovarian carcinoma cells than its chlorido analogue [Os(η6-pcym)(dpa)Cl]PF6 (1-Cl); pcym = 1-methyl-4-(propan-2-yl)benzene (p-cymene), dpa = 2,2'-dipyridylamine. The complex 1-VP showed promising selectivity towards the A2780 ovarian carcinoma cell line (IC50 = 20.9 μM) over normal human hepatocytes (IC50 > 200.0 μM). Moreover, the complex 1-VP was found to be inacti

Innovative DNA-Targeted Metallo-prodrug Strategy Combining Histone Deacetylase Inhibition with Oxidative Stress.

Cancer remains a global health challenge. There is an urgent need to develop innovative therapeutics that can overcome the shortcomings of existing cancer therapies. DNA enzymes involved in nucleic acid compaction and organization are an attractive cancer drug target for therapeutic exploitation. In this work, a family of Cu(II) prodrugs containing suberoylanilide hydroxamic acid (SAHA), a well-established histone deacetylase inhibitor (HDACi) and clinically approved cancer drug, and phenanthrene ligands as DNA intercalative components have been rationally developed.
