Gustatory hedonic value: potential function for forebrain control of brainstem taste processing.

Among well-nourished populations, eating beyond homeostatic needs when presented with caloric-dense palatable food evidences the assertion t … nd the ventral forebrain. This review discusses the current understanding of centrifugal control of taste processing in subcortical gustatory nuclei and the potential role of such modulation in hedonic responding.

Masking mechanisms of bitter taste of drugs studied with ion selective electrodes.

The masking mechanisms of the bitter taste of propantheline bromide (PB) and oxyphenonium (OB) bromide by native and modified cyclodextrins, saccharides, surfactants, organic acids, nonionic and anionic polymers, and other compounds were investigated with ion selective electrodes. The intensity of the bitter taste for a mixed solution of cyclodextrin with PB or OB was quantitatively explained from the observed electromotive force with the following assumptions: the complex and the masking agent do not have any tastes and the bitter taste is independent of other tastes. Sodium dodecyl sulfate r…

Preparation, characterization and taste-masking properties of polyvinylacetal diethylaminoacetate microspheres containing trimebutine.

The objectives of this study were to produce acid soluble, polyvinylacetal diethylaminoacetate (AEA) microspheres containing trimebutine (as maleate), using a water-in-oil-in-water (w/o/w) emulsion solvent evaporation method, to characterize their in-vitro release properties, and to evaluate the taste-masking potential of this formulation in human volunteers. The pH of the external aqueous phase was the critical factor in achieving a high loading efficiency for trimebutine in the microencapsulation process; nearly 90% (w/w) loading efficiency was obtained at above pH 10. Trimebutine was comple…

Desert toads discriminate salt taste with chemosensory function of the ventral skin.

Toads obtain water by absorption across their skin. When dehydrated, desert toads exhibit stereotyped hydration behavior in which they press … amiloride, a blocker for Na+ channels known to be responsible for epithelial ion transport and salt taste transduction. In behavioral experiments, avoidance of a NaCl solution was also reduced by adding amiloride to the solution, suggesting involvement of amiloride-sensitive Na+ channels for detecting the hyperosmotic salt solution. Neural tracing with …

Nicotine and ethanol interaction on conditioned taste aversions induced by both drugs.

The present study was designed to explore the interactive effects of nicotine and ethanol in the pretreatment and preexposure conditioned taste aversion (CTA) paradigm. The first experiment examined the effects of ethanol pretreatment on a nicotine induced CTA. The second experiment examined the effects of nicotine pretreatment on an ethanol CTA. The results of these two experiments revealed an asymmetrical interaction between ethanol and nicotine. Although nicotine pretreatment blocked an ethanol induced CTA, ethanol pretreatment merely attenuated a nicotine-induced CTA. These findings demons…

Amphetamine-induced modification of quinine palatability: analysis by the taste reactivity test.

The effects of low doses of d-amphetamine (0.25-0.5 mg/kg, IP) on taste reactions elicited by quinine solutions in a 5-10-min taste reactivity test were assessed in a series of three experiments. Amphetamine consistently suppressed aversive reactions elicited by quinine solutions. The results suggest that amphetamine, like morphine, attenuates the aversiveness of the taste of quinine solution.

Co-evolution of glucose-6-phosphate dehydrogenase deficiency and quinine taste sensitivity.

We hypothesized that 'quinine' taste sensitivity functions to regulate the intake of bitter-tasting, naturally occurring antimalarial substances of plant origin, and that this genetic trait has co-evolved with the G6PD locus. This hypothesis was tested by evaluating taste sensitivity to quinine sulphate and sodium chloride among 17 G6PD-deficient and 25 G6PD-normal African American subjects 14-40 years of age. There was no significant difference in mean 'quinine' taste sensitivity between the two groups, although there was a trend towards greater 'quinine' taste acuity among the G6PD-deficient…

Palatal taste buds in man: topographical arrangement in islands of keratinized epithelium.

In an attempt to find and reconstruct the exact anatomical correlate of taste sensitivity in the human soft palate, a novel approach was used to examine the mucosal surface in conjunction with serial tissue sectioning. Six human subjects, 15 to 29 years of age, 3 females and 3 males, served to take precision impressions of the hard and the anterior soft palate, using non-commercial trays and a self-curing resin. From these impressions, replicas were cast in Epon and subsequently sputter-coated with gold and examined in a scanning electron microscope. In addition, one biopsy was taken from one …

HPLC-purified human satietin does not produce conditioned taste aversion in rats.

Human satietin is thought to be an endogenous glycoprotein that can suppress food intake and body weight. However, it was also found to be aversive when rats infused intracerebroventricularly (ICV) with human satietin were subjected to a two-bottle taste aversion test. More recently, the human satietin previously thought homogenous was separated by HPLC into two Peaks, denoted as A and B. In the present study, male Sprague-Dawley rats were fitted with chronic third ventricle cannulas and presented with fluid for 1 h/day, while food was given ad lib. After training, the rats were ICV infused wi…

Conditioned taste reactivity in rats after phenylpropanolamine, d-amphetamine or lithium chloride.

That an aversive property of phenylpropanolamine (PPA) in part contributes to its anorexic capacity is suggested by the demonstration of conditioned taste avoidance to PPA doses ranging from 10-40 mg/kg. In order to further evaluate the putative aversive property of PPA, the present experiment compared the effects of PPA on multiple measures of aversion (chin rubs, gaping) in the taste reactivity (TRT) paradigm with those produced by the classic agent lithium chloride and by amphetamine. Male rats were infused via an intraoral cannula with 0.5 M sucrose followed by injection with either vehicl…

Taste aversion involving central opioid antagonism is potentiated in morphine-dependent rats.

A sensitive taste conditioning test was used to measure the aversive effect of a single intraventricular (i.c.v.) or subcutaneous (s.c) injection of an opioid antagonist that readily crosses the blood brain barrier (naltrexone), and one of two that do not (methylnaltrexone and diallylnormorphinium). This was done in drug-naive rats and in rats implanted 5 days earlier with a pellet containing 75 mg morphine. It was found that the morphine exposure had no significant effect on the dose-response curve of the taste aversion produced by s.c. methylnaltrexone and s.c. diallynormorphinium but reduce…

Alterations in brain aldehyde dehydrogenase activity modify ethanol-induced conditioned taste aversion.

The role of peripherally and centrally acting acetaldehyde in ethanol-induced conditioned taste aversion (CTA) was investigated using various enzyme manipulations. Cyanamide, an aldehyde dehydrogenase inhibitor (ALDH) elevates blood acetaldehyde levels in the presence of ethanol. Concurrent administration with 4-methylpyrazole (4MP), an alcohol dehydrogenase inhibitor, prevents peripheral accumulation of acetaldehyde by cyanamide. Under both treatment conditions brain and liver ALDH activity is inhibited. Water-deprived rats were pretreated 4 hr prior to fluid presentation with intraperitoneal…

Off-vertical rotation produces conditioned taste aversion and suppressed drinking in mice.

The effects of off-vertical rotation upon the intake of tap water immediately after rotation, and upon conditioned taste aversion, were assessed in mice with the tilt of the rotation axis varying from 5 to 20 degrees from the earth-vertical. Conditioned taste aversion occurred in all mice that were rotated, but the intake of tap water was suppressed only in mice that were rotated at 15 or 20 degrees of tilt. The greater suppression of tap water intake and the stronger conditioned aversion in the mouse as the angle of tilt was increased in this experiment are consistent with predictions from si…

Food sensitivities, taste changes, aphthous ulcers and atopic symptoms in allergic disease.

Abnormalities in taste and buccal sensation, and production of abdominal symptoms, are associated with ingestion of specific foods by atopic individuals as shown by Food Sensitivity Analysis. Of 61 atopic patients all had abnormalities in buccal sensation and up to 87% related nausea or other abdominal symptoms to ingestion of specific foods. Eighty-two percent had positive skin tests to foods, 56% had a history of aphthous ulcers and 18% could associate development of the ulcers with ingestion of specific foods. Application of the responsible allergen to the ulcer base produced severe pain in…

A Multimodal Integration Framework for Taste and Odor Prediction and Flavor Molecule Discovery.

Data-driven molecular property prediction is increasingly used to accelerate molecular discovery and computer-aided design. In flavor science, accurate prediction of molecular taste and odor attributes could facilitate the screening and rational design of candidate flavor molecules. However, this task remains challenging because structure-flavor relationships are complex, single molecular representations may provide incomplete property-relevant information, and the available labels are highly imbalanced. To address these challenges, we implemented and evaluated Uni-Flavor, a multimodal predict…

Electrochemical Sensors as a Tool for Taste Perception in Pharmaceutical Products: Advances and Perspectives.

Taste masking in pharmaceutical products is a complex and subjective process that requires reliable evaluation methods. This review focuses on the electronic tongue (e-tongue), an emerging sensor-based technology designed to mimic human taste perception without the need for human panels. E-tongue systems provide objective data to support the development of palatable formulations. In this review, we discuss the principles, types of e-tongue devices, data processing approaches, and their applications in pharmaceutical research. By comparing e-tongue performance with human taste assessment, we hi…

GPRC6A as a novel kokumi receptor responsible for enhanced taste preferences by ornithine.

The concept of 'kokumi', which refers to an enhanced and more delicious flavor of food, has recently generated considerable interest in food science. However, kokumi has not been well studied in gustatory physiology, and the underlying neuroscientific mechanisms remain largely unexplored. Our previous research demonstrated that ornithine (L-ornithine), which is abundant in shijimi clams, enhanced taste preferences in mice. The present study aimed to build on these findings and investigate the mechanisms responsible for kokumi in rats. In two-bottle preference tests, the addition of ornithine, …
