The hOCT1 SNPs M420del and M408V alter imatinib uptake and M420del modifies clinical outcome in imatinib-treated chronic myeloid leukemia.

Although the prognosis of chronic myeloid leukemia (CML) patients treated with imatinib is good, many fail to develop an optimal response or lose one. This heterogeneity could be attributed to the presence of human organic cation transporter-1 (hOCT1) single nucleotide polymorphisms (SNPs). In the present study, we analyzed the effect of 23 hOCT1 SNPs on imatinib treatment outcome in newly diagnosed CML patients using MassARRAY sequencing and pyrosequencing. The only SNP associated with outcome was M420del (rs35191146), with patients with the M420del demonstrating an increased probability of i

Levosimendan Can Improve the Level of B-Type Natriuretic Peptide and the Left Ventricular Ejection Fraction of Patients with Advanced Heart Failure: A Meta-analysis of Randomized Controlled Trials.

BACKGROUND AND AIMS: Levosimendan, a calcium (Ca2+)-sensitizing cardiotonic agent, is mainly used in patients with advanced heart failure. However, no research could explain how levosimendan reduces the mortality in advanced heart failure patients. We aim to illustrate the efficacy of levosimendan through clinical indexes. METHODS: We searched PubMed, Embase, and CENTRAL from 1994 to August 2019 to compare the efficacy of levosimendan infusion for the treatment of advanced heart failure with that of other agents (placebo, dobutamine, furosemide, and prostaglandin E1). Levels of B-type natriur

Sodium oxybate for the maintenance of abstinence in alcohol-dependent patients: An international, multicenter, randomized, double-blind, placebo-controlled trial.

BACKGROUND: Sodium oxybate (SMO) has been shown to be effective in the maintenance of abstinence (MoA) in alcohol-dependent patients in a series of small randomized controlled trials (RCTs). These results needed to be confirmed by a large trial investigating the treatment effect and its sustainability after medication discontinuation. AIMS: To confirm the SMO effect on (sustained) MoA in detoxified alcohol-dependent patients. METHODS: Large double-blind, randomized, placebo-controlled trial in detoxified adult alcohol-dependent outpatients (80% men) from 11 sites in four European countries.

Treatment of MDA5-positive dermatomyositis complicated by gangrenous cholecystitis with tofacitinib.

BACKGROUND: Dermatomyositis is a rare idiopathic inflammatory disease with diverse presentations that can have varying degrees of cutaneous and systemic involvement. This phenotypic heterogeneity makes DM a therapeutic challenge. Some therapeutic drugs, such as hormones and immunosuppressants, have poor therapeutic effects. In recent years, tofacitinib has been reported to be effective in the treatment of dermatomyositis. CASE PRESENTATION: We report a case of anti-MDA5 antibody-positive dermatomyositis that was relieved after treatment with tofacitinib, during which gallbladder gangrene and

Efficacy of slow repetitive transcranial magnetic stimulation in the treatment of resistant auditory hallucinations in schizophrenia: a meta-analysis.

OBJECTIVE: Slow repetitive transcranial magnetic stimulation (rTMS), at a frequency of 1 Hz, has been proposed as a treatment for auditory hallucinations. Several studies have now been reported regarding the efficacy of TMS treatment, but results were inconsistent. Therefore, meta-analytic integration of the published trials is needed to evaluate the prospects of this new treatment. DATA SOURCES: A literature search was conducted using PubMed and Web of Science for the years 1966 until February 2006. We used the search terms transcranial magnetic stimulation, TMS, rTMS, and hallucination*. S

Omics Data and Their Integrative Analysis to Support Stratified Medicine in Neurodegenerative Diseases.

Molecular and clinical heterogeneity is increasingly recognized as a common characteristic of neurodegenerative diseases (NDs), such as Alzheimer's disease, Parkinson's disease and amyotrophic lateral sclerosis. This heterogeneity makes difficult the development of early diagnosis and effective treatment approaches, as well as the design and testing of new drugs. As such, the stratification of patients into meaningful disease subgroups, with clinical and biological relevance, may improve disease management and the development of effective treatments. To this end, omics technologies-such as gen

Efficacy of Transcranial Alternating Current Stimulation for Major Depressive Disorder and the Role of Intensity: A Systematic Review and Exploratory Meta-Analysis.

BACKGROUND: Current treatments for major depressive disorder (MDD) are limited by efficacy and adverse effects. Transcranial alternating current stimulation (tACS) has emerged as a promising alternative. However, there is still a lack of systematic evaluation of its effectiveness. Recent studies suggest that currents greater than 7 milliamperes (mA) may be necessary to significantly alter potentials in deep brain regions associated with depression. We conducted a systematic review and exploratory meta-analysis to assess the overall efficacy and safety of tACS for MDD and to explore potential a

Lymphopenia and mortality in adult sepsis: A systematic review and focused meta-analysis of prognostic evidence.

BACKGROUND: Lymphopenia is common in sepsis, but published studies use heterogeneous exposure definitions and time points. We evaluated its prognostic association with mortality using a focused meta-analysis of comparable lymphopenia phenotypes and used additional evidence to contextualize biologic and infectious-source interpretation. METHODS: PubMed and Embase were searched from inception to April 1, 2026. Studies directly estimating lymphopenia-related mortality associations were assessed for quantitative synthesis. Pooling was restricted to comparable threshold-based, persistent, or basel

Insufficiency of peripheral blood as a substitute tissue for detecting EGFR mutations in lung cancer: a meta-analysis.

The detection of epidermal growth factor receptor (EGFR) mutations in non-small cell lung cancer tissues is necessary for effective treatment with EGFR tyrosine kinase inhibitors. However, tumor tissues may not be available in all situations. Studies have evaluated the potential use of serum or plasma for detecting the EGFR mutation status, but the results have been inconclusive. Here, a meta-analysis was performed to determine whether blood samples could serve as substitutes for tissue specimens in detecting the EGFR mutation status. Databases, including PubMed and Embase, were searched for r

Inclusion of historical control data in the analysis of a current trial for zero-inflated count models.

Historical control information can be used to improve the efficiency and precision of treatment-effect estimation in a current trial. However, this benefit depends on the similarity between historical and current controls as well as the methods used to borrow information from the historical data. This study extends the meta-analytic-predictive (MAP) framework to zero-inflated count models. The MAP approach is a Bayesian method for borrowing information from multiple historical controls under exchangeability assumptions for model parameters. We assess the benefits of allowing exchangeability ac

G-computation of average treatment effects on the treated and the untreated.

BACKGROUND: Average treatment effects on the treated (ATT) and the untreated (ATU) are useful when there is interest in: the evaluation of the effects of treatments or interventions on those who received them, the presence of treatment heterogeneity, or the projection of potential outcomes in a target (sub-) population. In this paper we illustrate the steps for estimating ATT and ATU using g-computation implemented via Monte Carlo simulation. METHODS: To obtain marginal effect estimates for ATT and ATU we used a three-step approach: fitting a model for the outcome, generating potential outcom
