Simulated microgravity induces a time-dependent shift from autophagy to apoptosis in osteoblasts, validated by TSPO, ATG12, and BNIP3L expression in vivo.

Emerging evidence indicates that microgravity-induced osteoblast dysfunction is a critical contributor to spaceflight-associated bone loss. This study investigated the temporal dynamics of autophagy-apoptosis crosstalk in MC3T3-E1 osteoblasts under a rotary cell culture system (RCCS)-simulated microgravity. Crucially, time-course analysis (1, 3, 5, 10 days) revealed a biphasic autophagic response: initial enhancement of autophagic flux (LC3-II/Beclin-1 upregulation, p62 degradation) at day 3, exerting cytoprotective effects with reduced apoptosis, subsequently shifting to peak autophagy inhibi

Elevated Coronary Artery Calcium Scores in Astronauts.

INTRODUCTION: We previously published a model of how calcium from resorbing bone can enter and persist in the circulation with consequent precipitation in coronary arteries. The model was based on chronic inflammation as the source of continued bone resorption. We asked whether other forms of resorptive bone loss such as microgravity (disuse) might also be associated with bone calcium accumulation in coronary arteries. METHODS: We analyzed a de-identified database provided by NASA consisting of 52 astronauts who had flown a space mission around two determinations of coronary artery calcium (C

Counteracting inactivity deconditioning with graded hypergravity loading: From clinical neurorehabilitation to space medicine.

BACKGROUND: Neuromuscular and neurodegenerative diseases such as Parkinson's disease result in progressive mobility restrictions, accelerating cardiovascular, muscular, and functional deconditioning. Such changes resemble the multisystem deconditioning that occurs during microgravity exposure, highlighting shared physiological consequences of reduced mechanical loading, even though the underlying mechanisms differ. OBJECTIVE: This study evaluated short-arm human centrifugation (SAHC) training as a whole-body rehabilitation intervention for patients with multiple sclerosis and neurodegenerativ

Longitudinal Analysis of Quantitative Brain MRI in Astronauts Following Microgravity Exposure.

BACKGROUND AND PURPOSE: Assessment of the effects of microgravity on astronauts' brains using microstructural measures by utilizing quantitative MRI, before and after spaceflight would help understand the structural changes. METHODS: Quantitative MRI data sets in 19 astronauts were acquired before and after space missions. Both diffusion tensor metrics and volumetric measures were analyzed in the brain regions involved in the visual function. RESULTS: The fractional anisotropy was reduced in the right posterior thalamic radiations (P = .0009) and remained significant after a false discovery

From the international space station to the clinic: how prolonged unloading may disrupt lumbar spine stability.

BACKGROUND CONTEXT: Prolonged microgravity exposure is associated with localized low back pain and an elevated risk of post-flight disc herniation. Although the mechanisms by which microgravity impairs the spine are unclear, they should be foundational for developing in-flight countermeasures for maintaining astronaut spine health. Because human spine anatomy has adapted to upright posture on Earth, observations of how spaceflight affects the spine should also provide new and potentially important information on spine biomechanics that benefit the general population. PURPOSE: This study compa

Ground-based assessment of JAXA mouse habitat cage unit by mouse phenotypic studies.

The Japan Aerospace Exploration Agency developed the mouse Habitat Cage Unit (HCU) for installation in the Cell Biology Experiment Facility (CBEF) onboard the Japanese Experimental Module ("Kibo") on the International Space Station. The CBEF provides "space-based controls" by generating artificial gravity in the HCU through a centrifuge, enabling a comparison of the biological consequences of microgravity and artificial gravity of 1 g on mice housed in space. Therefore, prior to the space experiment, a ground-based study to validate the habitability of the HCU is necessary to conduct space exp

Differential expression of specific cellular defense proteins in rat hypothalamus under simulated microgravity induced conditions: comparative proteomics.

Microgravity severely halts the structural and functional cerebral capacity of astronauts especially affecting their brains due to the stress produced by cephalic fluid shift. We employed a rat tail suspension model to substantiate simulated microgravity (SM) in brain. In this study, comparative mass spectrometry was applied in order to demonstrate the differential expression of 17 specific cellular defense proteins. Gamma-enolase, peptidyl-prolyl cis-trans isomerase A, glial fibrillary acidic protein, heat shock protein HSP 90-alpha, 10 kDa heat shock protein, mitochondrial, heat shock cognat

Enhancement of osteogenic differentiation and proliferation in human mesenchymal stem cells by a modified low intensity ultrasound stimulation under simulated microgravity.

Adult stem cells can differentiate into multiple lineages depending on their exposure to differing biochemical and biomechanical inductive factors. Lack of mechanical signals due to disuse can inhibit osteogenesis and induce adipogenesis of mesenchymal stem cells (MSCs). Long-term bed rest due to both brain/spinal cord injury and space travel can lead to disuse osteoporosis that is in part caused by a reduced number of osteoblasts. Thus, it is essential to provide proper mechanical stimulation for cellular viability and osteogenesis, particularly under disuse conditions. The objective of this

Sympathetic neural influence on bone metabolism in microgravity (Review).

Bone loss is one of the most important complications for astronauts who are exposed to long-term microgravity in space and also for bedridden elderly people. Recent studies have indicated that the sympathetic nervous system plays a role in bone metabolism. This paper reviews findings concerning with sympathetic influences on bone metabolism to hypothesize the mechanism how sympathetic neural functions are related to bone loss in microgravity. Animal studies have suggested that leptin stimulates hypothalamus increasing sympathetic outflow to bone and enhances bone resorption through noradrenali

[Neck-deep immersion effects on gaze fixation by primates].

Effects of load deprivation on gaze fixation were studies in two primates (Macaca mulatta) immersed neck-deep twice with a two-week interval. Gaze fixation was traced using electrooculography and measurement of head turn velocity in 5 hours since the beginning of immersion. Decrease of the maximum head velocity and vestibuloocular reflex gain in both animals were significantly higher in the second compared with the first period of immersion and the results of previous investigations with three primates repeatedly immersed chest-deep. During the second neck-deep immersion, loss of head velocity

Orthostatic response in rats after hindlimb unloading: effect of transcranial electrical stimulation.

INTRODUCTION: Orthostatic hypotension is a commonly observed phenomenon after exposure to microgravity and in various forms of autonomic failure. It has been suggested that insufficient activation of supraspinal structures responsible for descending sympathetic drive could play a significant role in this disorder. We examined the effect of transcranial electrical stimulation (TES) of autonomic nuclei within the brain on the orthostatic hypotension induced by exposure to simulated microgravity using a hindlimb unloading model. METHODS: There were 20 male Wistar rats that were suspended by thei

Size and cell number of the utricle in kinetotically swimming fish: a parabolic aircraft flight study.

Humans taking part in parabolic aircraft flights (PAFs) may suffer from space motion sickness (SMS, a kinetosis). Since it has been repeatedly shown earlier that some fish of a given batch also reveal a kinetotic behavior during PAFs (especially so-called spinning movements and looping responses) and due to the homology of the vestibular apparatus among all vertebrates, fish can be used as model systems to investigate the origin of susceptibility to motion sickness. Therefore, we examined the utricular maculae (they are responsible for the internalization of gravity in teleosteans) of fish swi

Is HSP70 upregulation crucial for cellular proliferative response in simulated microgravity?

Astronauts are susceptible to a variety of conditions such as motion sickness, muscular atrophy, bone demineralization and cardiovascular deconditioning. These findings suggest that the adaptation to the absence of gravity is due, at least in part, to the effects exerted by microgravity at the cellular level. Indeed, a number of studies have indicated that gravity affects mammalian cell growth and differentiation through the modulation of gene expression. We have characterized the behaviour of endothelial cells and of the human monocytic cell line U937 cultured in the NASA-developed bioreactor

The effects of a change in gravity on the dynamics of prehension.

Investigating cyclic vertical arm movements with an instrumented hand-held load in an airplane undergoing parabolic flight profiles allowed us to determine how humans modulate their grip force when the gravitational and the inertial components of the load force are varied independently. Eight subjects participated in this study; four had already experienced parabolic flights and four had not. The subjects were asked to move the load up and down continuously at three different gravitational conditions (1 g, 1.8 g, and 0 g). At 1 g, the grip force precisely anticipated the fluctuations in the lo

Effects of clinostat-microgravity on bone and calcium metabolism in rats.

Decreases in bone minerals and tissue volume after space flight have been observed in humans and animals, with a variety of results. Such data obtained from space flight experiments have given unsatisfactory results due to short periods of space flight and differences in age, body weights, and strain of animals used. Therefore, ground-based animal models have been developed in order to elucidate changes in bone affected by space flight. For example, a tail-suspended rat model has been established to study the effects of microgravity on bones by producing hind limb unloading. However, problems

Effect of head-down-tilt bed rest and hypovolemia on dynamic regulation of heart rate and blood pressure.

Adaptation to head-down-tilt bed rest leads to an apparent abnormality of baroreflex regulation of cardiac period. We hypothesized that this "deconditioning response" could primarily be a result of hypovolemia, rather than a unique adaptation of the autonomic nervous system to bed rest. To test this hypothesis, nine healthy subjects underwent 2 wk of -6 degrees head-down bed rest. One year later, five of these same subjects underwent acute hypovolemia with furosemide to produce the same reductions in plasma volume observed after bed rest. We took advantage of power spectral and transfer functi
