High environmental concentrations of pharmaceuticals, particularly antibiotics, have been observed worldwide, while antibiotic concentrations in the ng/L to µg/L range may adversely affect biota and contribute to the formation of antibiotic resistance. The underlying causes of high environmental concentrations include, in addition to the low rate of antibiotic' metabolization and the high rate of usage, especially of β-lactam antibiotics, the insufficient removal by conventional wastewater treatment methods. Consequently, alternative methods need to be developed to remove antibiotics from wast

To investigate the effect of ultrasound on the quality of pumpkin juice fermented by yeast, ultrasound (power range 0-400 W, duration 10 min) was used to assist the yeast fermentation of pumpkin juice. The effects of ultrasound-assisted fermentation at different powers on the quality characteristics of pumpkin juice, such as the color, aroma components, carotenoid content, and antioxidant activity, were studied. The optimal combination of fermentation process parameters was determined as follows: a pumpkin juice content of 20 mL, fermentation temperature of 30 °C, fermentation time of 1 day, a

Germline mutations can affect future generations, while somatic mutations cannot. This germline-soma distinction does not seem to make sense for unicellular organisms. We challenge this view, arguing that baker's yeast (Saccharomyces cerevisiae) has a germline. Under aerobic conditions yeast cells use mainly fermentation of glucose to produce ethanol. Only when glucose is exhausted, cells switch to full respiration of the produced ethanol. We hypothesize that only a subset of the cells continue dividing and switch to respiration. A change from exponential to linear growth is consistent with as

Cellulosic biomass represents a promising feedstock for biofuel and biochemical production. However, its recalcitrant structure strongly hinders enzymatic degradation. Cellulosomes are large multienzyme complexes, highly efficient at degrading cellulose. A cellulase in a cellulosome has a dockerin domain that binds to a cohesin module on the CipA (cellulosome integrating protein A). In a native cellulosome all cohesins are identical, so that the cellulase types and their positions in a CipA cannot be controlled. Here, we constructed the largest designer CipA known to date. Using innovative tec

This study demonstrates the potent immunomodulatory effects of yeast-derived vacuoles, highlighting their therapeutic potential in counteracting cyclophosphamide (CPA)-induced immunosuppression. Physicochemical characterization revealed that the vacuoles, with an average particle size of 176.8 nm and a zeta potential of -16.76 mV, exhibit favorable stability and are well-suited for cellular uptake. Vacuole treatment preserved body weight and significantly attenuated CPA-induced weight loss, indicating its protective role under immunosuppressive stress. Additionally, vacuole administration rest

CGSC is a pathogenic basidiomycetous yeast of increasing public health concern due to its ability to cause life-threatening infections in both immunocompromised and immunocompetent individuals. C. gattii species complex (CGSC) is acquired via environmental exposure, particularly through the inhalation of spores from trees, soil, and decaying wood. Infections often manifest in the pulmonary or central nervous system, both of which are associated with high morbidity and mortality. Compounding this threat is the pathogen's expanding geographic range, facilitated in part by climate change, and the

The Ophiostomataceae family encompasses numerous species that exhibit yeast-hyphae dimorphism, including well-known pathogens like the causal agents of Dutch elm disease, blue-staining fungi, and sporotrichosis. This dimorphism is not only observed in pathogenic interactions but also plays a crucial role in symbiotic relationships, such as ambrosia fungi. The ability to switch between these morphological forms may contribute to their adaptability and ecological functions within different environments and host interactions. This study seeks to elucidate the intricate interplay between dimorphis

Lignocellulose (dry plant biomass) is an abundant cheap inedible residue of agriculture and wood industry with great potential as a feedstock for biotechnological processes. Lignocellulosic substrates can serve as valuable resources in fermentation processes, allowing the production of a wide array of chemicals, fuels, and food additives. The main obstacle for cost-effective conversion of lignocellulosic hydrolysates to target products is poor metabolism of the major pentoses, xylose and L-arabinose, which are the second and third most abundant sugars of lignocellulose after glucose. We study

Red yeast rice has been highlighted as a health-food ingredient that reduces serum cholesterol levels. Recently, an increased number of cases of renal impairment induced by a supplement containing red yeast rice have been reported in Japan. A 42-year-old man with no history of chronic kidney disease developed renal dysfunction with proximal tubular damage after supplementation with red yeast rice. An 83-year-old woman with advanced diabetic kidney disease experienced further deterioration of her renal function after supplementation with red yeast rice. We herein report cases of acute kidney in

Succinic acid is an important C4 platform compound that serves as a raw material for the production of 1,4-butanediol, tetrahydrofuran, and biodegradable plastics such as polybutylene succinate (PBS). Compared to the traditional petrochemical-based route that uses maleic anhydride as a raw material, the microbial fermentation method for producing succinic acid offers more sustainable economic value and environmental friendliness. Yeasts with good acid tolerance can achieve low-pH fermentation of succinic acid, significantly reducing the cost of product extraction. Therefore, constructing high-…

Alzheimer's disease (AD) is an age-related, multifaceted neurological disorder associated with accumulation of aggregated proteins (amyloid Aβ and hyperphosphorylated tau), loss of synapses and neurons, and alterations in microglia. AD was recognized by the World Health Organization as a global public health priority. The pursuit of a better understanding of AD forced researchers to pay attention to well-defined single-celled yeasts. Yeasts, despite obvious limitations in application to neuroscience, show high preservation of basic biological processes with all eukaryotic organisms and offer g…

Demand for low- or non-alcoholic beers has been growing in recent years. Thus, research is increasingly focusing on non-Saccharomyces species that typically are only able to consume the simple sugars in wort, and therefore have a limited production of alcohol. In this project, new species and strains of non-conventional yeasts were sampled and identified from Finnish forest environments. From this wild yeast collection, a number of Mrakia gelida strains were selected for small-scale fermentation tests and compared with a reference strain, the low-alcohol brewing yeast Saccharomycodes ludwigii.…

Candia auris is an emerging human pathogenic yeast; yet, despite phenotypic attributes and genomic evidence suggesting that it probably emerged from a natural reservoir, we know nothing about the environmental phase of its life cycle and the transmission pathways associated with it. The thermotolerant characteristics of C. auris have been hypothesised to be an environmental adaptation to increasing temperatures due to global warming (which may have facilitated its ability to tolerate the mammalian thermal barrier that is considered a protective strategy for humans against colonisation by envir

Numerous studies have demonstrated that people with type 2 diabetes mellitus (associated with IAPP peptide aggregation) show an increased incidence of Alzheimer's disease (associated with Aβ aggregation), but the mechanism responsible for this correlation is presently unknown. Here, we applied a yeast-based model to study the interactions of IAPP with PrP (associated with TSEs) and with the Aβ42 peptide. We demonstrated that fluorescently tagged IAPP forms detergent-resistant aggregates in yeast cells. Using the FRET approach, we showed that IAPP and Aβ aggregates co-localize and physically in

The fission yeast Schizosaccharomyces pombe is a popular model organism in molecular biology and cell physiology. With its ease of genetic manipulation and growth, supported by in-depth functional annotations in the PomBase database and genome-wide metabolic models,S. pombe is an attractive option for synthetic biology applications. However,S. pombe currently lacks modular tools for generating genetic circuits with more than 1 transcriptional unit. We developed a toolkit to address this gap. Adapted from the MoClo-YTK plasmid kit for Saccharomyces cerevisiae and using the same modular cloning

Patients with the fatal disorder Transthyretin Amyloidosis (ATTR) experience polyneuropathy through the progressive destruction of peripheral nervous tissue. In these patients, the transthyretin (TTR) protein dissociates from its functional tetrameric structure, misfolds, and aggregates into extracellular amyloid deposits that are associated with disease progression. These aggregates form large fibrillar structures as well as shorter oligomeric aggregates that are suspected to be cytotoxic. Several studies have shown that these extracellular TTR aggregates enter the cell and accumulate intrace

Ergosterol is a specific sterol component of yeast and fungal membranes. Its biosynthesis is one of the most effective targets for antifungal treatments. However, the emergent resistance to multiple sterol-based antifungal drugs emphasizes the need for new therapeutic approaches. The allylamine terbinafine, which selectively inhibits squalene epoxidase Erg1 within the ergosterol biosynthetic pathway, is mainly used to treat dermatomycoses, whereas its effectiveness in other fungal infections is limited. Given that ergosterol biosynthesis depends on iron as an essential cofactor, in this report

There is growing interest in yeast selection for industrial fermentation applications since it is a factor that protects a wine's identity. Although it is strenuous evaluating the oenological characteristics of yeasts in selection processes, in many cases the most riveting yeasts produce some undesirable organoleptic characteristics in wine. The aim of the present work is to improve an industrial yeast strain by reducing its hydrogen sulfide (H2S) production. To accomplish this, two different improvement approaches were used on said yeast: hybridization by mass mating and adaptive laboratory e…
