Liver development in a rat model of fetal alcohol syndrome.

The inhibitory effects of alcohol on hepatic growth in adults raises the possibility that the liver may be involved in fetal alcohol syndrome (FAS) in infants. To test this hypothesis, pregnant Sprague-Dawley rats were fed liquid diets containing either ethanol as 36% of the total calories, or were allowed ad libitum feeding of a control liquid diet (controls) throughout pregnancy. Other dams were exposed to the ethanol diet only during the first or last half of pregnancy. Pups delivered of dams exposed to the various diets (N = 40-45/group) were killed at 1, 3, 7, and 14 days of age. In addit

Hydrogen sulfide donor NaHS inhibits formaldehyde-induced epithelial-mesenchymal transition in human lung epithelial cells via activating TGF-β1/Smad2/3 and MAPKs signaling pathways.

Formaldehyde (FA) long term exposure leads to abnormal pulmonary function and small airway obstruction of the patients. Hydrogen sulfide (H2S) is one of the recognized gaseous transmitters involved in a wide range of cellular functions. It is unknown the involvement of H2S in FA-induced lung injury. The purpose of this study is to investigate the therapeutic potential and mechanism of H2S on FA-induced epithelial-mesenchymal transition (EMT) of human lung epithelial cells. The cell viability of Beas2B and A549 cells after FA treatment were assessed using MTT assay. The endogenous H2S was visua

Folic Acid-Conjugated Albumin-Stabilized Gold-Terbium Nanoparticles for SPCCT/MRI Imaging: An in vitro Cellular and Phantom Study.

INTRODUCTION: Triple-negative breast cancer (TNBC) remains challenging to image because of its biological heterogeneity and the limitations of single-modality imaging. Multifunctional nanoparticles may provide complementary contrast for spectral photon-counting CT (SPCCT) and MRI, potentially improving multimodal tumor imaging. METHODS: Folic acid-conjugated, albumin-stabilized gold-terbium nanoparticles (BSA@Au/Tb-FA) were synthesized and characterized using TEM, XRD, FTIR, XPS, and DLS. In vitro cellular uptake, cytotoxicity, and hemocompatibility were evaluated using MDA-MB-231 cells using

Metabolic and biochemical profiling reveals phenotypic heterogeneity in Zucker diabetic fatty rats.

BACKGROUND: Zucker diabetic fatty (ZDF) rats exhibit significant phenotypic variability despite genetic uniformity, yet a comprehensive characterization of these divergent phenotypes remains limited. METHODS: Male ZDF (fa/fa) rats (4 months, n = 22) and ZDF (fa/+) lean controls (n = 10) were maintained on standard chow for 6 months. Based on metabolic trajectory, ZDF (fa/fa) rats were stratified into three phenotypes: obese normoglycemic (O, n = 8), diabetic with cachexia (D, n = 8), and diabetic without cachexia (D-C, n = 6). Comprehensive assessments included body weight, glycemic control,

Multifunctional antibacterial micelle with targeting ability and lipase-responsibility for therapy of methicillin-resistant Staphylococcus aureus infection.

Bacterial infections frequently trigger severe inflammatory cascades accompanied by oxidative stress responses, which collectively impair tissue regeneration and exacerbate disease progression. Consequently, there is a pressing clinical need to develop therapeutic strategies capable of simultaneously eradicating pathogenic bacteria and modulating the infection microenvironment. The infected tissue microenvironment is characterized by macrophage infiltration and upregulation of folate receptor expression on their cellular surfaces. Concurrently, pathogenic bacteria actively secrete abundant lip

Next-Generation Multimodality of Nanomedicine Therapy: Size and Structure Dependence of Folic Acid Conjugated Copolymers Actively Target Cancer Cells in Disabling Cell Division and Inducing Apoptosis.

Nanomedicine as a multimodality treatment of cancer utilizes the advantages of nanodelivery systems of drugs. They are superior to the clinical administration of different therapeutic agents in several aspects, including simultaneous delivery of drugs to the active site, precise ratio control of the loading drugs and overcoming multidrug resistance. The role of nanopolymer size and structural shape on the internalization process and subsequent intracellular toxicity is limited. Here, the size and shape dependent mechanism of a functionalized copolymer was investigated using folic acid (FA) cov

Hypo-Expression of Flice-Inhibitory Protein and Activation of the Caspase-8 Apoptotic Pathways in the Death-Inducing Signaling Complex Due to Ischemia Induced by the Compression of the Asphyxiogenic Tool on the Skin in Hanging Cases.

The FLICE-inhibitory protein (c-FLIPL) (55 kDa) is expressed in numerous tissues and most abundantly in the kidney, skeletal muscles and heart. The c-FLIPL has a region of homology with caspase-8 at the carboxy-terminal end which allows the molecule to assume a tertiary structure similar to that of caspases-8 and -10. Consequently, c-FLIPL acts as a negative inhibitor of caspase-8, preventing the processing and subsequent release of the pro-apoptotic molecule active form. The c-FLIP plays as an inhibitor of apoptosis induced by a variety of agents, such as tumor necrosis factor (TNF), T cell r

Tumor-targeting photodynamic therapy based on folate-modified polydopamine nanoparticles.

BACKGROUND: Photodynamic therapy (PDT), a clinical anticancer therapeutic modality, has a long history in clinical cancer treatments since the 1970s. However, PDT has not been widely used largely because of metabolic problems and off-target phototoxicities of the current clinical photosensitizers. PURPOSE: The objective of the study is to develop a high-efficiency and high-specificity carrier to precisely deliver photosensitizers to tumor sites, aiming at addressing metabolic problems, as well as the systemic damages current clinical photosensitizers are known to cause. METHODS: We synthesiz

The expression of microRNAs and exposure to environmental contaminants related to human health: a review.

Environmental contaminants exposure may lead to detrimental changes to the microRNAs (miRNAs) expression resulting in several health effects. miRNAs, small non-coding RNAs that regulate gene expression, have multiple transcript targets and thereby regulate several signalling molecules. Even a minor alteration in the abundance of one miRNA can have deep effects on global gene expression. Altered patterns of miRNAs can be responsible for changes linked to various health outcomes, suggesting that specific miRNAs are activated in pathophysiological processes. In this review, we provide an overview

Brain death induces apoptosis in donor liver of the rat.

BACKGROUND: A difference in short- and long-term function between living-related and cadaveric donor organs is consistently shown in kidney- and liver-transplant studies. We hypothesize that this is caused by induction of apoptosis and inflammation of the potential graft because of the phase of brain death (BD) in the cadaveric donor that predisposes for additional transplant injury. Previously, we have shown inflammation in the liver of brain-dead donors by increased expression of cell adhesion molecules and influx of leukocytes. The key inflammatory mediator in inflammation is tumor necrosis

Regulation of apoptosis through cysteine oxidation: implications for fibrotic lung disease.

Tissue fibrosis is believed to be a manifestation of dysregulated repair following injury, in association with impaired reepithelialization, and aberrant myofibroblast activation and proliferation. Numerous pathways have been linked to the pathogenesis of fibrotic lung disease, including the death receptor Fas, which contributes to apoptosis of lung epithelial cells. A redox imbalance also has been implicated in disease pathogenesis, although mechanistic details whereby oxidative changes intersect with profibrotic signaling pathways remain elusive. Oxidation of cysteines in proteins, such as S

Rapamycin-loaded nanostructured lipid carrier modified with folic acid intended for breast cancer therapy.

Breast cancer stands as the most common form of malignancy among women globally, and it showcases commendable rates of cure when detected in early-stage and non-metastatic conditions. To overcome drug resistance and side effects observed in conventional chemotherapy, the present study aims to deliver rapamycin (RAP), a mTOR protein inhibitor, into a nanostructured lipid carrier (NLC) functionalized with folic acid for promoting active targeting to breast cancer cells. In the first step, the synthesis of 1,2-distearoyl-sn-glycero-3-phosphatidylethanolamine-N-[amino(polyethylene glycol)-2000] (a

Ferulic Acid Interferes with Radioactive Intestinal Injury Through the DJ-1-Nrf2 and Sirt1-NF-κB-NLRP3 Pathways.

Radiation-induced intestinal injury is a common complication of radiotherapy for abdominal and pelvic malignancies. Due to its rapid proliferation, the small intestine is particularly sensitive to radiation, making it a critical factor limiting treatment. Ferulic acid (FA), a derivative of cinnamic acid, exhibits antioxidant, anti-inflammatory, and anti-radiation properties. In this study, we established a mouse model of radiation-induced intestinal injury using a dose of 11 Gy at a rate of 96.62 cGy/min. Our findings indicate that FA's protective effects against radiation-induced intestinal i

Ferulic Acid Improves Synaptic Plasticity and Cognitive Impairments by Alleviating the PP2B/DARPP-32/PP1 Axis-Mediated STEP Increase and Aβ Burden in Alzheimer's Disease.

The burden of Alzheimer's disease, the most prevalent neurodegenerative disease, is increasing exponentially due to the increase in the elderly population worldwide. Synaptic plasticity is the basis of learning and memory, but it is impaired in AD. Uncovering the disease's underlying molecular pathogenic mechanisms involving synaptic plasticity could lead to the identification of targets for better disease management. Using primary neurons treated with Aβ and APP/PS1 animal models, we evaluated the effect of the phenolic compound ferulic acid (FA) on synaptic dysregulations. Aβ led to synaptic

Distinct Trypanosoma cruzi isolates induce activation and apoptosis of human neutrophils.

Neutrophils are critical players in the first line of defense against pathogens and in the activation of subsequent cellular responses. We aimed to determine the effects of the interaction of Trypanosoma cruzi with human neutrophils, using isolates of the two major discrete type units (DTUs) associated with Chagas' disease in Latin America (clone Col1.7G2 and Y strain, DTU I and II, respectively). Thus, we used CFSE-stained trypomastigotes to measure neutrophil-T. cruzi interaction, neutrophil activation, cytokine expression and death, after infection with Col1.7G2 and Y strain. Our results sh

Folate receptor-α targeted delivery of berberine via folate-functionalized bovine serum albumin nanocarriers to enhance intracellular internalization and suppress 3D spheroid formation as a pioneering therapeutic modality for glioblastoma.

Glioblastoma multiforme, the most aggressive brain malignancy, exhibits poor prognosis and intrinsic resistance to standard therapies, often limited by systemic toxicity and suboptimal efficacy. Berberine, a bioactive isoquinoline alkaloid, exhibits potent anti-glioma activity; however, its clinical utility is hindered by rapid metabolism, low bioavailability, and poor penetration across the blood-brain barrier. To overcome these challenges, we designed folic acid-functionalized BSA nanoparticles for targeted and sustained delivery of berberine to glioma cells. Folic acid modification facilita
