A deep learning-based computational pipeline predicts developmental outcome in retinal organoids.

Retinal organoids have become important models for studying development and disease, yet stochastic heterogeneity in the formation of cell types, tissues, and phenotypes remains a major challenge. This limits our ability to precisely experimentally address the early developmental trajectories towards these outcomes. Here, we utilize deep learning to predict the differentiation path and resulting tissues in retinal organoids well before they become visually discernible. Our approach effectively bypasses the challenge of organoid-related heterogeneity in tissue formation. For this, we acquired a

Functionalized biochar for plasticizer remediation in aquatic systems: synergistic adsorption and advanced oxidation strategies.

Biochar-based treatment strategies have received growing interest; however, no comprehensive review has systematically assessed the mechanisms and performance of functionalized biochar for plasticizer remediation. This review critically compares pristine biochar and its advances in biochar modifications for plasticizer adsorption. Additionally, it unveils how functionalized biochar integrates with advanced oxidation processes (AOPs) to achieve synergistic adsorption and degradation of plasticizers in water. As different studies have used varying initial concentrations, a direct comparison is n

An evaluation of telisotuzumab vedotin for the treatment of non-squamous non-small cell lung cancer.

INTRODUCTION: Advanced non-squamous non-small cell lung cancer (NSCLC) remains associated with substantial mortality despite major advances in targeted therapy and immuno-oncology. c-Met protein overexpression represents a biologically relevant and relatively prevalent phenotype that may define a therapeutically vulnerable population lacking canonical genomic drivers. AREAS COVERED: This review examines the scientific rationale for targeting c-Met protein overexpression and critically evaluates telisotuzumab vedotin (Teliso-V), a c-Met-directed antibody-drug conjugate (ADC) delivering the cyt

Eph receptors in pancreatic cancer: Biological roles and clinical implications.

Pancreatic ductal adenocarcinoma (PDAC) is one of the most aggressive malignancies with a 5-year survival rate of approximately 12%. Treatment options, including surgical resection and chemotherapy, are often ineffective in advanced stages due to late detection. This review examines the role of Erythropoietin-producing human hepatocellular (Eph) receptors in PDAC, emphasizing their potential both as therapeutic targets and biomarkers. Eph receptors, a family of receptor tyrosine kinases, are frequently overexpressed in various cancers and contribute to tumor growth, angiogenesis, and metastasi

Oncolytic human herpesvirus for cancer therapy.

Herpes simplex virus (HSV) is attractive for oncolytic therapy due to its genetic capacity, effect on various tumors, ability to trigger immune responses, and adaptation to the cancer microenvironment. Its oncolytic potential can be enhanced by genetic modifications that help Herpes simplex virus escape adaptive immunity and better direct immune responses. While the virus replicates specifically in cancer cells, it does not harm normal cells. Effects like immunogenic cell death and cytokine release in the tumor microenvironment hinder immune invasion of cancer and improve treatment effectivene

Lung cancer immunotherapy in 2025: where we stand and what comes next?

Lung cancer continues to be the leading cause of cancer-related mortality worldwide, accounting for more deaths than breast, colorectal, and prostate cancers combined. Over the past decade, the introduction of immunotherapy has reshaped treatment paradigms, offering hope for long-term survival in a disease historically associated with dismal outcomes. The incorporation of immune checkpoint inhibitors (ICIs) into the treatment of non-small cell lung cancer (NSCLC) and small cell lung cancer (SCLC) has improved outcomes across early-stage, locally advanced, and metastatic settings. However, only

Organ cross-talk: molecular mechanisms, biological functions, and therapeutic interventions for diseases.

Organ cross-talk, also known as the organ axis or organ interaction network, plays a vital role in maintaining physiological homeostasis and responding to environmental stimuli. This review comprehensively integrates cutting-edge observations in organ communication research, with a particular focus on the brain, heart, and gut-the three core organs that garner the most attention in organ connection studies. The current state of organ interaction network research is clearly presented as a Sankey diagram. For brain-related connections, the interactions among the brain-gut, brain-liver, and brain

Platinum-based therapeutics as emerging multi-modal radiosensitizers in glioblastoma treatment.

Glioblastoma (GBM) is a highly lethal brain cancer with poor patient prognosis and limited treatment options. Standard-of-care therapy involves maximally safe surgical resection followed by radiation therapy (RT) in combination with the alkylating agent temozolomide (TMZ) to treat the residual and brain-invading components of the disease. While RT remains a central part of definitive treatment, its efficacy is limited by intrinsic and acquired radiation resistance, making long-term tumor control and survival rare. While TMZ provides modest anti-tumor benefits, tumor resistance to TMZ and lack

Spatial multi-omics technologies in gastric cancer: applications and advances.

Gastric cancer (GC) is plagued by profound intratumoral heterogeneity and a complex tumor microenvironment (TME), which are the core obstacles to precise diagnosis and treatment. Conventional bulk multi-omics technologies average molecular signals across tissues, thus masking cellular heterogeneity; single-cell multi-omics resolves cellular diversity but dissociates cells from their native spatial context, leading to the loss of critical information on intercellular crosstalk and molecular spatial distribution. These limitations result in an incomplete understanding of GC pathogenesis and TME

Ultra-rare TP53 and KRAS variants predict survival in ICI-treated solid tumours.

BACKGROUND: The prognostic relevance of ultra-rare TP53 and KRAS variants in advanced solid tumours treated with immune-checkpoint inhibitors (ICI) is unclear. METHODS: We performed a retrospective cohort study using the Memorial Sloan Kettering Cancer Center MSK-IMPACT clinical-genomic dataset (cBioPortal study ID: tmb_mskcc_2018). TP53 and KRAS alterations were stratified by rarity (ultra-rare vs common) and analysed for overall survival (OS). Kaplan-Meier analyses and multivariable Cox proportional hazards models were fitted, adjusting for tumour type, tumour mutational burden (TMB), tumou

Microvascular and stromal normalization combined with nano-immunotherapy: A synergistic strategy to overcome drug delivery barriers in solid tumors.

Solid tumors like advanced-stage prostate carcinoma pose significant hindrances to therapeutic approaches due to a rich tumor microenvironment (TME) that establishes strong physical and immune resistance to treatment. The TME is defined by a dense, fibrotic stroma and non-functional vasculature, which produce high mechanical stress, high interstitial fluid pressure (IFP), and hypoxia. These properties critically inhibit therapeutic agent penetration and suppress anti-tumor immunity. This review details the mechanistic foundation of such hindrances and discusses the nascent paradigm of TME norm

Ultrahigh-molecular-weight tadpole polymers with statistically symmetric head-to-tail geometry: the key topological intermediate bridging linear and cyclic architectures.

Polymer topology profoundly influences material properties by regulating chain arrangement, conformational entropy, and interactions, yet understanding topological entanglement constraints like threading and knotting remains a fundamental challenge in polymer science, partly due to the scarcity of structurally precise, ultrahigh-molar-mass ring-containing polymers as model systems. Herein, we address this synthetic challenge with a dual-terminal intramolecular cyclization strategy to synthesize tadpole polymers, the key architectural intermediate bridging linear and monocyclic topologies. This

Blindly Trusting the Stage: The Therapeutic Consequences of Clinical Staging Inaccuracy in Gastric Cancer.

BACKGROUND: This study aimed to quantify clinical staging accuracy rates and evaluated the consequences if treatment decisions were based solely on clinical stages. PATIENTS AND METHODS: This retrospective study included 931 patients with nonmetastatic gastric cancer who underwent upfront radical gastrectomy, allowing the postoperative pathological stage (pTNM) to serve as the definitive gold standard. We compared concordance rates for T, N, and overall stages, and simulated guideline-directed treatment decisions on the basis of Chinese Society of Clinical Oncology (CSCO) criteria to calculat

From Assembly to Application: A Comprehensive Review of Metal-Hydrogen-Bonded Organic Frameworks.

Metal-hydrogen-bonded organic frameworks (M-HOFs) have emerged as novel crystalline porous materials that integrate robust metal coordination units with extensive hydrogen-bonded networks, enabling architectures that offer both structural stability and tunable functionality. Extensive hydrogen bonding confers structural integrity, while incorporation of metal ions enhances chemical and thermal stability and introduces catalytic and sensing properties. Their tunable porosity, diverse topologies, and mild synthesis conditions render them highly desirable for applications in chemical separation,

Novel Liquid Biopsy in Gastrointestinal Cancers.

Gastrointestinal (GI) cancers represent a significant global health burden, being among the leading causes of cancer-related deaths. The prognosis for patients remains unsatisfactory, largely because most cancers are detected at advanced stages. Traditional diagnostic methods, such as radiological and histopathological examinations and serum tumor markers like AFP, CEA, CA-125, and CA-199, possess limitations in sensitivity and specificity, particularly for early screening. A major drawback of tissue biopsy is its inability to fully capture the inherent heterogeneity within tumors, as mutation

Self-Assembling Enveloped Virus-Mimicking Particle for Extrahepatic Targeting mRNA Delivery.

Precise and efficient delivery of messenger RNA (mRNA) to extrahepatic tissues remains a major challenge in advancing mRNA-based therapeutics. Although enveloped viruses and virus-like particles (VLPs) serve as natural RNA delivery systems with varied extrahepatic targeting capabilities, their clinical translation is often hampered by bottlenecks in tunability, safety, immunogenicity, and scalability. Here, we developed a bottom-up self-assembling enveloped virus-mimicking particle (EVMP) composed of highly simplified yet high-performance virus-mimicking peptide (VMP) and tissue-targeting enve

Cornering: A Geometric Framework for Surgical Dissection in Minimally Invasive Surgery.

BackgroundPrecise tissue dissection is a fundamental component of minimally invasive surgery (MIS), yet it is often taught implicitly. While triangulation and traction-countertraction are widely recognized as geometric principles for exposure and transection, the geometric conditions that enable the dynamic advancement of dissection along tissue interfaces have not been clearly articulated.MethodsWe performed a schematic geometric analysis of surgical dissection focusing on the relationships among traction direction, opposing tissue surfaces, and the dissecting edge. Based on this analysis, we

The isolated perfused lung: a bridge between in vitro and in vivo studies of inhaled medicinal products.

Understanding the complex relationship between drug deposition, pulmonary pharmacokinetics, and clinical efficacy is essential to developing effective inhaled therapies. Harnessing formulation physicochemical properties can improve aerodynamic properties and enhance lung retention, yet predictive in vitro and in silico models remain essential for advanced formulation development. A range of in vitro lung epithelial models has been explored to study transepithelial transport. However, these systems often lack physiological complexity. Advanced platforms such as organoids and lung-on-a-chip mode
