Induction of multiple matrix metalloproteinases in human dermal and synovial fibroblasts by Staphylococcus aureus: implications in the pathogenesis of septic arthritis and other soft tissue infections.

Infections of body tissue by Staphylococcus aureus are quickly followed by degradation of connective tissue. Patients with rheumatoid arthritis are more prone to S. aureus-mediated septic arthritis. Various types of collagen form the major structural matrix of different connective tissues of the body. These different collagens are degraded by specific matrix metalloproteinases (MMPs) produced by fibroblasts, other connective tissue cells, and inflammatory cells that are induced by interleukin-1 (IL-1) and tumor necrosis factor (TNF). To determine the host's contribution in the joint destructio

Characterization of novel macrophage cell lines derived from goat and sheep blood and their susceptibility to ruminant virus infections.

Macrophages are present in all vertebrate tissues and play a key role in both the prevention and pathogenesis of infectious diseases. In vitro cultures of macrophages provide a valuable tool for studying the innate immune functions of macrophages and macrophage-pathogen interactions in specific animal species. We previously demonstrated that porcine macrophages were effectively immortalized by transferring the simian virus 40 large T antigen (SV40LT) gene and porcine telomerase reverse transcriptase (pTERT) gene using lentiviral vectors. In the present study, we investigated whether this immor

Linear B Cell Epitopes Derived from the Multifunctional Surface Lipoprotein BBK32 as Targets for the Serodiagnosis of Lyme Disease.

BBK32 is a multifunctional surface lipoprotein expressed by Borrelia burgdorferisensu lato, the causative agent of Lyme disease. Previous studies suggested that BBK32 could be a sensitive antigen target of new, more effective, serodiagnostic assays for the laboratory diagnosis of Lyme disease. However, nonspecific antibody binding to full-length BBK32 has hampered its use as a target in clinical assays. Specificity can be improved by the use of peptides composed of linear B cell epitopes that are unique to B. burgdorferi, eliminating cross-reactive epitopes that bind to antibodies generated by

[Clinico-epidemiologic characteristics of Lyme disease treated at the Infectious Disease in Novy Sad 1993-1998].

INTRODUCTION: Borrelia burgdorferi, the etiological agent of Lyme disease, is transmitted by the bite of Ixodes ricinus, registered in all parts of Yugoslavia. Vectors are very active in spring and early summer and the disease has a seasonal distribution. Generally speaking, there are three defined stages of the disease, but some of them can be misdiagnosed or really absent. Serological analysis of Lyme disease is very difficult to interpret, especially in later stages, so confirmation by immunoblot assays is recommended. The aim of this study was to present some epidemiologic and clinical cha

The role of anti-mutated citrullinated vimentin antibodies in the diagnosis of early rheumatoid arthritis.

OBJECTIVE: Anti-mutated citrullinated vimentin (MCV) antibodies have been reported as a fairly sensitive serological marker of rheumatoid arthritis (RA). We evaluated the diagnostic value of anti-MCV in a large cohort of Chinese patients with early RA. METHODS: One hundred seventy patients with early RA (<1 yr duration), 66 with other rheumatic diseases, 10 with infectious diseases, and 60 healthy individuals were included in our study. Serum anti-MCV and second-generation anti-cyclic citrullinated peptide antibodies (anti-CCP2) were measured by ELISA, and rheumatoid factor (RF) was measured

Acute respiratory deterioration in rheumatoid arthritis-associated interstitial lung disease associated with pulmonary Mycobacterium abscessus infection: a case report.

Nontuberculous mycobacteria (NTM) are important pathogens in patients with chronic lung diseases, including rheumatoid arthritis-associated interstitial lung disease (RA-ILD). Although acute exacerbations of ILD associated with NTM infection have been reported sporadically, cases involving pulmonary Mycobacterium abscessus infection remain extremely limited. We describe a fatal case of acute respiratory deterioration in a patient with RA-ILD following pulmonary infection with M. abscessus. A 76-year-old man with longstanding RA-ILD developed pulmonary NTM disease initially caused by Mycobacter

Autoimmune disease-associated lymphomas: research progress and review.

Autoimmune diseases (ADs) are strongly associated with a significantly increased risk of lymphoma, with the standardised incidence ratio (SIR) markedly elevated in certain conditions, most notably in Sjögren's disease (SjD) where an SIR as high as 18.8 has been reported. The risk is particularly prominent for diffuse large B-cell lymphoma (DLBCL) and mucosa-associated lymphoid tissue (MALT) lymphoma. This review systematically elucidates the epidemiological features, pathological mechanisms, risk factors, and therapeutic strategies of ADs-associated lymphomas. Epidemiological studies have conf

Rare variants in genes related to inborn errors of immunity in patients with rheumatoid arthritis and secondary immunodeficiency.

OBJECTIVES: Treatment of rheumatoid arthritis (RA) relies on immunomodulatory drugs that can compromise immunity, inducing secondary immunodeficiency (SID). However, features of SID, such as hypogammaglobulinaemia and severe/recurrent infections, occur in only a minority of patients, suggesting a potential underlying genetic predisposition. Given the expanding spectrum of genes implicated in inborn errors of immunity (IEIs) and the fact that IEIs can present with rheumatic manifestations, we hypothesised that a subset of patients with RA with SID harbour IEI-related variants. METHODS: We scre

A new quantum-inspired pattern based on Goldner-Harary graph for automated alzheimer's disease detection.

OBJECTIVES: Periodic fever, aphthous stomatitis, pharyngitis, and cervical adenopathy (PFAPA) syndrome is a non-Mendelian autoinflammatory disorder until now considered to be specifically limited to paediatric age. Recently, an increasing number of reports seems to suggest that PFAPA syndrome, diagnosed by the Marshall criteria revised by Thomas et al., can also affect adults. METHODS: The Marshall/Thomas criteria have been applied to 989 adult patients presenting for recurrent fever episodes: all patients enrolled were reviewed for demographic, clinical, and therapeutic data. The development, validation and application of an indirect enzyme-linked immunosorbent assay (ELISA) for the detection of antibodies to Streptococcus dysgalactiae subspecies dysgalactiae in lamb and ewe sera.

BACKGROUND: Neonatal infectious arthritis (NIA) is a bacterial disease of lambs caused by Streptococcus dysgalactiae sub species dysgalactiae (SDSD). NIA negatively impacts animal welfare; whilst its control entails widespread antibiotic use. The aims of this study are to develop, validate and apply an indirect, ELISA assay to inform SDSD vaccine development and epidemiology studies. METHODS: The antigen (inactivated isolate of SDSD) was incubated with sera from SDSD infected sheep, negative control sheep and serum from sheep flocks with outbreaks of SDSD NIA. SDSD antibody was detected with

Neutrophil Extracellular Traps (NETs) in health and disease.

Neutrophil extracellular traps (NETs) are web-like structures composed of DNA, histones, and antimicrobial proteins that extend the defensive repertoire of neutrophils beyond classical phagocytosis and degranulation. Initially considered solely antimicrobial, NETs are now recognized as dynamic regulators of immunity, inflammation, and tissue remodeling. Their formation is orchestrated by the generation of reactive oxygen species, neutrophil elastase-mediated chromatin remodeling, and peptidyl arginine deiminase 4-driven histone citrullination. At the same time, clearance involves DNase activit
