Immunoglobulin heavy chain variable region gene utilization by B cell hybridomas derived from rheumatoid synovial tissue.

Rheumatoid arthritis (RA) is a chronic inflammatory disease that primarily affects synovial joints. Activated B lymphocytes and plasma cells are present in the synovial tissue and are thought to contribute to the immunopathology of the rheumatoid joint. To investigate rheumatoid synovial B lymphocytes, we have generated B cell hybridomas from synovial tissue of an RA patient. Here we describe the immunoglobulin VH gene repertoire of eight IgM- and 10 IgG-secreting synovial-derived hybridomas. The VH4 gene family is highly represented (38.5%) in this panel of hybridomas compared with the freque

Superoxide scavenging activity of leukocytes in rheumatoid arthritis and Behçet's diseases.

Superoxide radical anion (O2-) has been suggested to mediate tissue damage associated with chronic inflammatory disorders such as rheumatoid arthritis (RA) and Behçet's disease (BD). Despite that protection from O2- is provided by superoxide dismutase (SOD), there are controversial results about SOD-like activity of polymorhonuclear cells (PMN) in RA and there are no reports about it in BD. In addition, colchicine, a potent inhibitor of phagocytosis, has been used for the treatment of BD, no report about its effect on SOD-like activity of PMN is available. The present study investigates the su

[Anti-stratum corneum antibody in the rat esophagus, anti-epidermal keratin and anti-epidermis autoantibodies in rheumatoid polyarthritis and other rheumatic diseases. Diagnostic value and basic aspects].

Eight immunological parameters were explored in 257 patients with various rheumatic diseases including 107 rheumatoid arthritis (RA). IgG and IgM antibodies (AB) reactive with the Stratum Corneum (SC) of rat oesophagus or with the SC of human skin were assayed by 1/2 quantitative indirect immunofluorescence, IgG and IgM auto AB to epidermal keratins by a specific ELISA, and total serum G and M immunoglobulins by radial immunodiffusion. At the threshold we chose (99 p. cent specificity), IgG anti SC of rat oesophagus, were found in 50 of 107 (46.7 p. cent) RA patients: 52.5 p. cent in sero + an

Reassessing the Kocher Criteria: A Prospective Evaluation of Diagnostic Performance in Pediatric Musculoskeletal Infection.

BACKGROUND: The Kocher criteria are widely used to differentiate septic arthritis (SA) from transient synovitis (TS) in children. However, diagnostic performance has since been proven to be inconsistent. Furthermore, the criteria were originally developed to distinguish SA from TS and have not been validated for differentiating osteomyelitis (OM), which frequently presents with overlapping clinical features. The purpose of this prospective study was to evaluate the diagnostic performance of the Kocher criteria and other clinical variables in differentiating SA from both TS and OM. METHODS: A

[SAPHO syndrome: clinico-rheumatologic and radiologic differentiation and classification of a patient sample of 86 cases].

Synovitis (inflammatory arthritis), acne (pustulosa), pustulosis (psoriasis, palmoplantar pustulosis), hyperostosis (acquired), and ostitis (bland osteomyelitis) are symptoms forming the acronym SAPHO, which is a syndrome of nosologic heterogeneity. All entities forming the SAPHO syndrome are connected by a non-obligate dermatoskeletal association with an aseptic pustulous character. 86 cases were analyzed clinically, radiologically and by histology/histopathology. 31 adult patients showed the typical triad of pustulosis palmo-plantaris (psoriatica, PPP), sterno-costo-clavicular hyperostosis (

A novel cell surface antigen (T305) found in increased frequency on acute leukemia cells and in autoimmune disease states.

Monoclonal antibody T305, prepared by immunizing mice with the T-ALL derived cell line RPMI-8402, immunoprecipitates a single chain glycoprotein with m.w. 160,000 daltons (under reducing conditions) or 180,000 daltons (under nonreducing conditions). In immunofluorescence assays, antibody T305 reacted with a subpopulation of T cells in normal blood (22 +/- 6%), thymus (28 +/- 11%), and lymph node (24 +/- 6%). Increased frequency of T cells reactive with antibody T305 was found in peripheral blood of patients with infectious mononucleosis (greater than 80%), graft-vs-host disease after bone marr

The lung as an autoimmune organ: From mucosal tolerance breach to AI-enabled interception.

Autoimmune diseases have traditionally been conceptualized as systemic disorders that secondarily involve the lung. This roadmap review proposes a complementary framework in which the lung is considered an autoimmune organ: a mucosal site where tolerance may be breached, a vulnerable target of systemic autoimmunity, and a sentinel from which future disease trajectories can be inferred. Across connective tissue diseases, the lung is also a major determinant of prognosis through airway disease, interstitial lung disease, progressive fibrosis, pulmonary vascular involvement and acute alveolar inj

Neutrophils as critical orchestrators of chronic inflammation.

Neutrophils are the first key effector innate immune cells recruited toward inflammatory sites. Through the release of neutrophilic extracellular traps (NETs), the production of reactive oxygen species (ROS), degranulation and phagocytosis, neutrophils play a central role in the rapid elimination of invading pathogens. Recently, increasing attention has been given to the role of neutrophils in chronic inflammation, challenging the dichotomy between innate and adaptive immune responses. In chronic inflammatory conditions, neutrophils generally display a hyperinflammatory phenotype via dysregula

Isorhapontigenin attenuates allergic asthma by reducing airway inflammation and oxidative stress.

Gnetum cleistostachyum has been used in traditional Chinese medicine to treat arthritis and bronchitis. Isorhapontigenin, a naturally occurring stilbenoid structurally related to resveratrol, is isolated from G. cleistostachyum and has been reported to exhibit anti-inflammatory and antioxidant properties. Here, we examined whether isorhapontigenin can ameliorate airway inflammation, oxidative stress, and airway hyperresponsiveness (AHR) in an ovalbumin (OVA)-induced mouse model of allergic asthma. Female BALB/c mice were sensitized and challenged with OVA and subsequently administered isorhapo

Cardiovascular disease incidence among aging patients with rheumatoid arthritis: Results from the Canadian Longitudinal Study on Aging (CLSA).

BACKGROUND AND AIMS: Rheumatoid arthritis (RA) is associated with a higher risk of cardiovascular disease (CVD). This study examined CVD incidence among middle-aged and older Canadians with and without RA and identified the role of potential risk factors for CVD in this population. METHODS AND RESULTS: Data were obtained from the Canadian Longitudinal Study on Aging (CLSA) using three time points between 2011 and 2021. Incidence rate ratios (IRR) for CVD were calculated to compare CVD incidence among RA and non-RA individuals. Cox proportional hazards regression models identified the potentia

Cold Atmospheric Plasma as a Potential Disease-Modifying Therapy for Osteoarthritis.

Osteoarthritis (OA) is a disabling joint disease characterised by cartilage degradation, synovial inflammation, and subchondral bone remodelling. Furthermore, catabolic inflammatory processes as well as dysregulated cellular signalling and oxidative stress are central to OA pathogenesis. Despite its growing global burden, currently available therapies primarily provide symptomatic relief and fail to target underlying molecular mechanisms and halt disease progression. Cold atmospheric plasma (CAP), a partially ionised, non-thermal gas that generates controlled reactive oxygen and nitrogen speci

Preclinical characterization of AZD1163, a first-in-class anti-PAD2/4 bispecific antibody for the treatment of rheumatoid arthritis.

Anti-citrullinated protein autoantibodies promote inflammation and joint tissue injury and define a poor prognostic group of patients with rheumatoid arthritis (RA). Citrullinated autoantigens that drive this autoimmune response are generated by peptidylarginine deiminase (PAD) enzymes, which are predominately expressed and released by neutrophils and monocytes. Accordingly, blocking the enzymatic activity of PADs to curb the generation of citrullinated autoantigens that drive autoimmunity and tissue injury may provide therapeutic benefit. Herein, we developed a high affinity, bispecific, anti

Efficacy and safety of anakinra for acute calcium pyrophosphate deposition disease flare: a systematic review and meta-analysis.

OBJECTIVE: Calcium pyrophosphate deposition disease (CPPD) is a common inflammatory arthritis among older adults. Comorbidities often limit conventional treatment of acute CPPD flare. Anakinra, an interleukin-1 inhibitor, is a promising alternative. We aimed to evaluate the efficacy and safety of anakinra in acute CPPD flares. METHODS: We systematically searched studies from MEDLINE, EMBASE, and the Cochrane Database up until August 2024. The primary outcome was a positive treatment response between Day 3 and Day 5 after anakinra initiation. Secondary outcomes included changes in pain ratings
