A "One-Stone-Three-Birds" Inspired Nanoplatform for Multitargeted Ulcerative Colitis Therapy via Combined Aryl Hydrocarbon Receptor Activation and Reactive Oxygen Species Scavenging.

In the pathogenesis of ulcerative colitis, inflammatory responses, mucosal injury, and gut dysbiosis form a mutually reinforcing network that perpetuates disease progression. Current clinical interventions can rarely modulate these pathological modules simultaneously, underscoring the urgent need for more effective therapeutic strategies. Here, we report a dual-functional nanotherapy that couples aryl hydrocarbon receptor (AhR) activation with on-site reactive oxygen species (ROS) scavenging. An amphiphilic copolymer, HA-TK-LA (HTL), was synthesized by grafting lipoic acid (LA) to hyaluronic a

Integrated metabolomics and gut microbiota analyses reveal the protective effects of matrine in ulcerative colitis.

BACKGROUND: Ulcerative colitis (UC) is a chronic inflammatory bowel disease driven by gut microbial dysbiosis and metabolic dysfunction. Matrine, a natural alkaloid with anti-inflammatory properties, shows therapeutic potential; however, its mechanisms involving the coordinated modulation of bacteria, fungi, and host intestinal luminal metabolism remain unclear. METHODS: We evaluated the therapeutic efficacy of matrine using a dextran sulfate sodium (DSS)-induced murine model of ulcerative colitis. Disease severity was assessed via the disease activity index, colon length, and histopathology.

MiR-29a deficiency disrupts gut microbiota-bile acid homeostasis and impairs the anti-inflammatory effects of lithocholic acid in colitis.

BACKGROUND: Gut microbiota, bile acid metabolism, and their derived metabolites are critical regulators of intestinal inflammation in inflammatory bowel disease (IBD). While microRNA-29a (miR-29a) is a recognized epigenetic regulator of intestinal inflammation, its role in shaping microbiota-driven bile acid metabolism, particularly secondary bile acid production, remains unclear. This study aimed to investigate how miR-29a regulates microbiota-bile acid axis, with an emphasis on lithocholic acid (LCA) and its anti-inflammatory effects in colitis. METHODS: Colitis was induced in wild-type and

Establishment of a long-term secondary lymphedema animal model in the rodent hindlimb.

INTRODUCTION: Secondary lymphedema is a chronic swelling of the extremities caused by physical disruption of the lymphatic system, leading to impaired lymph drainage. It frequently develops in cancer patients after surgical removal of lymph nodes and vessels during tumor resection, when reconnection of lymphatic pathways fails. Current clinical therapies are mainly palliative or conservative, with limited therapeutic effects. Therefore, an animal model that closely mimics the chronic and severe characteristics of secondary lymphedema in patients is required to enable mechanistic and therapeuti

Modulating effects of creatine prevent sex-dependent hippocampal neuroinflammation, neuronal damage, and electrophysiological dysfunction induced by chronic colitis.

Chronic inflammation in inflammatory bowel disease (IBD) disrupts the gut-brain axis, thereby inducing abnormal immune responses in the brain that are increasingly recognized as a source of neurological comorbidities. In the present work, we investigated the structural integrity and electrophysiological function of hippocampal cornu ammonis 1 (CA1) pyramidal neurons in a rat model of ulcerative colitis induced by dextran sulfate sodium administration. Moreover, given the known neuroprotective properties of the nutraceutical creatine, we aimed to determine whether its oral supplementation preve

Quorum-Sensing Molecules Are Elevated in Long-Standing Ulcerative Colitis and Are Linked to the Development of Colitis-Associated Cancer.

BACKGROUND & AIMS: Chronic colonic inflammation can lead to colitis-associated cancer (CAC) in ulcerative colitis (UC) patients. The host-microbiome interface plays a critical role in CAC development. Quorum-sensing molecules (QSMs) are bacterial products that regulate bacterial processes. We investigated whether QSMs are related to risk factors for CAC in UC patients and drive CAC development in mouse models. METHODS: Blood samples from UC patients and mouse models of CAC (azoxymethane-dextran sodium sulfate) were analyzed for 3 bacterial QSM classes: short-chain n-acyl homoserine lactones (

Anti-CD31 antibody preconditioning for enhancement of endothelial cell capture and vascularization: a novel strategy for bioengineering lung scaffolds.

Recellularization of acellular lung scaffolds is a promising technique for the generation of bioengineered lungs. However, at the present time, pre-clinical evaluation of bioengineered lungs shows graft failure likely due to thrombus formation and bleeding from bronchi. This is believed to be due to the impairment of barrier function, incomplete vascular endothelial cell coverage, and limited endothelial cell function. Therefore, efficient recellularization of the vasculature region of the lung scaffold with functional endothelial cells is an important goal. Coating of decellularized tissues,

Harnessing distinct bioactive profile of yak casein: A comparative study on hydrogels for infected wound healing.

Infected wounds impair tissue regeneration through persistent bacterial colonization, excessive oxidative stress, and prolonged inflammation. While casein-based hydrogels have emerged as promising wound dressings, investigations have predominantly focused on bovine milk casein (B-CA). Yak milk casein (Y-CA) differs markedly from B-CA in monomer composition, which will affect their bioactivity. In our study, composite hydrogels were fabricated using Y-CA or B-CA as the protein matrix, oxidized dextran (ODex) as a chemical crosslinker, and tannic acid (TA). This yields dual-crosslinked Y-CA@TA o

Double emulsions enable in situ generation of permeation enhancers for oral delivery of macromolecules.

Oral administration of peptide therapeutics is limited by gastrointestinal degradation and poor epithelial permeability. Here, water-in-oil-in-water (W/O/W) double emulsions produced by microfluidics were designed to co-entrap octreotide and medium-chain triglycerides, enabling digestion-triggered generation of permeation-enhancing medium chain fatty acids. Synthesis parameters were systematically optimized to obtain stable, monodisperse droplets with defined core-shell morphology. The emulsions comprised an inner aqueous phase containing the payload, encapsulated within a Miglyol® 812 N oil p

Plasma Regulates Homeostatic Pulmonary Endothelial Signaling to Mitigate Vascular Leak Following Polytrauma and Hemorrhagic Shock.

INTRODUCTION: In hemorrhagic shock (HS), plasma resuscitation preserves vascular integrity and protects against trauma-induced coagulopathy and organ injury. Despite demonstrated clinical benefit, the endothelial mechanisms underlying plasma resuscitation remain incompletely defined. This study investigated endothelial-specific responses to plasma resuscitation to identify targetable pathways that promote vascular repair after traumatic injury. METHODS: A murine model of severe polytrauma-HS (PT/HS) with demonstrable vascular endotheliopathy by 24 hours was used to compare pulmonary vascular

Sophoraflavanone G alleviates ulcerative colitis in mice by reshaping the gut microbiome and restoring the Th17/Treg balance through the PI3K/AKT/NF-κB signaling pathway.

Ulcerative colitis (UC) is characterized by diarrhea, bloody stool and a feeling of incomplete defecation. Sophoraflavanone G (SFG) is one of the main chemical components of Sophora flavescens Ait and possesses various pharmacological activities. This study aims to investigate the therapeutic efficacy of naturally derived SFG in alleviating dextran sulfate sodium (DSS)-induced UC and the underlying mechanisms. A mouse model of DSS-induced UC was created and treated with SFG, antibiotics (ABX) cocktail, or the AKT agonist SC79. Clinical symptoms and signs of UC were assessed. Expression levels

PKM2 preconditioning protects endothelial cells from pyroptosis and BBB disruption via NRF2/TRX/TXNIP signaling in neonatal hypoxic-ischemic brain injury.

BACKGROUND: Neonatal hypoxic-ischemic brain damage (HIBD) is a leading cause of neurological deficits and death in neonates. In HIBD, the death of endothelial cells and disruption of the blood-brain barrier (BBB) are closely related to the severity of brain damage and long-term clinical outcomes. There is increasing evidence that a glycolytic enzyme, pyruvate kinase M2 (PKM2), is essential for managing metabolic processes in endothelial cells, but its role (and underlying molecular mechanism) in hypoxic-ischemic (HI)-associated endothelial cell metabolism, cell survival, and BBB function remai

Targeting SIRT3 dysfunction with a sprayable honokiol hydrogel for diabetic burn wound repair.

BACKGROUND: Diabetic burn wound healing is impaired by persistent mitochondrial oxidative stress, chronic inflammation, bacterial infection, and defective tissue regeneration. Although honokiol possesses antioxidant and anti-inflammatory properties, its therapeutic potential is limited by poor local retention. Moreover, the therapeutic significance of restoring mitochondrial sirtuin 3 (SIRT3) signaling in diabetic wound repair remains largely unexplored. This study developed a sprayable honokiol-loaded catechol-chitosan hydrogel as a mechanism-guided therapeutic strategy for diabetic burn woun

The Interaction Between Iron Overload and Lead Exposure on Bone Metabolism.

BACKGROUND: Osteoporosis (OP) is a common metabolic bone disease characterized by chronic bone loss and structural deterioration of bone tissue, and it is closely associated with environmental exposure to heavy metals. Although lead (Pb) and iron (Fe) are known to affect bone metabolism, the impact of iron overload under fixed lead exposure, along with the underlying metabolic mechanisms, remains unclear. This study aimed to investigate the effects of combined iron and lead exposure on bone mineral homeostasis and metabolic remodeling. METHODS: Male C57BL/6 mice were exposed for 8 weeks to le

Alkaline sphingomyelinase (ENPP7) attenuates DSS-induced colitis by modulating FOXO1-mediated antioxidative stress responses.

BACKGROUND: Alkaline sphingomyelinase (alk-SMase), also known as ectonucleotide pyrophosphatase/phosphodiesterase 7 (ENPP7), is an intestinal enzyme involved in sphingolipid metabolism and has been implicated in the regulation of inflammation. However, its role in intestinal inflammation and the underlying mechanisms remain unclear. This study aimed to investigate the role of ENPP7 in dextran sulfate sodium (DSS)-induced colitis, with a particular focus on oxidative stress and FOXO1-related signaling pathways. METHODS: ENPP7 knockout (KO) and wild-type (WT) mice were used to establish a DSS-in

Engineering IL-10-Overexpressing MSCs via a Non-Viral PEG-PEI Nanoplatform for Potent Therapy of Inflammatory Bowel Disease.

Genetic engineering of therapeutic cells is a key strategy to enhance cell-based therapies, yet current gene delivery methods-viral vectors, electroporation, and commercial non-viral reagents-are limited by safety concerns, high cost, operational complexity, cytotoxicity, and poor scalability. We developed a safe, efficient, low-cost, and scalable non-viral gene delivery platform using a polyethylene glycol-polyethyleneimine (PEG-PEI) copolymer to engineer mesenchymal stromal cells (MSCs) for inflammatory bowel disease (IBD) treatment. The PEG-PEI copolymer was synthesized via covalent conjuga

Dioscin suppresses colitis-associated colorectal tumorigenesis by inhibiting PPARγ-mediated fatty acid oxidation in TAMs.

BACKGROUND: Chronic inflammation plays a key role in colorectal carcinogenesis, where tumor-associated macrophages (TAMs) critically promote tumor progression. Metabolic reprogramming of TAMs, particularly through the modulation of fatty acid oxidation (FAO), represents a promising therapeutic strategy; however, effective agents remain to be identified. Although the natural compound dioscin exhibits anti-tumor properties, its ability to regulate TAM plasticity and metabolism in colitis-associated colorectal cancer (CAC) remains unclear. METHODS: A mouse model of CAC was established using azox

Chemical characterization of aconite polysaccharide and its ameliorative effect on ulcerative colitis by regulating gut microbiota and THDCA-driven TGR5/cAMP/PKA/NLRP3 pathway.

Aconite shows potential to improve ulcerative colitis (UC). However, the specific impact and mechanisms of aconite polysaccharides (APs) on UC remain poorly understood. This study aims to elucidate the chemical structures of APs, among which one was further evaluated for its effects on dextran sulfate sodium (DSS)-induced UC, and the underlying mechanism was explored with a focus on gut microbiota (GM). Four distinct APs were isolated, among which aconite polysaccharide A-1 (APA-1) was the predominant one. APA-1 is characterized as a glucan composed of →4)-α-glucose (Glc)-(1→, with residues su

In vitro models of blood-spinal cord barrier in spinal cord injury: A meta-analysis of model design, performance, and physiological relevance.

BACKGROUND: In vitro models of the blood-spinal cord barrier (BSCB) are widely utilized for developing therapeutics against neurological diseases such as spinal cord injury (SCI). However, high variability among existing in vitro BSCB models severely limits their predictive power for preclinical research, highlighting the need for a comprehensive synthesis of current model characteristics and performance. METHODS: We first reviewed in vitro BSCB models under SCI conditions and conducted a comprehensive meta-analysis of mainstream Transwell-based models. The synthesis systematically analyzed c
