Thrombosis is the key driver for ischemic events, including stroke, which is the leading cause of global mortality. However, current antithrombotic therapies carry substantial bleeding risks. Targeting protease-activated receptor-1 (PAR-1), a thrombin-activated receptor central to thrombus growth, represents a promising antithrombotic strategy for safer intervention.we mined the venom gland transcriptome of Conus spp., applied in silico Furin cleavage prediction, and synthesized and screened the anti-PAR-1 activity of a series of conotoxin-derived peptides.Cb-26 exhibited the strongest but rev

Visualizing nitric oxide (NO) dynamics during macrophage polarization is critical for understanding immune regulation yet remains challenging due to the poor photostability and specificity of existing probes. Herein, we engineered a macrophage-targeted ratiometric nanoprobe (ManNAc-UCNPs) to monitor rapid NO fluctuations during immune plasticity. The probe integrates a photostable core-shell-shell upconversion nanoparticle (UCNP) donor with an NO-responsive dye acceptor via a self-calibrating Luminescence Resonance Energy Transfer (LRET) mechanism. Surface modification with N-Acetyl-D-mannosam

This research presents a new ratiometric fluorescent probe designed for Cu²⁺ detection, utilizing a dual mechanism of intramolecular charge transfer (ICT) and fluorescence resonance energy transfer (FRET). In this probe, coumarin acts as the energy donor while naphthalimide serves as the acceptor, with a 2-picolinic ester moiety functioning as the Cu²⁺ binding site. In the absence of Cu²⁺, the 2-picolinic ester effectively suppresses electron transfer, inhibiting both ICT and FRET, resulting in strong blue fluorescence (λem = 478 nm) from the coumarin moiety. However, upon Cu²⁺ binding, the Cu

A direct Z-scheme ZnIn2S4/polypyrrole (PPy) heterojunction with wide near-infrared (NIR) response range was established by in situ electropolymerization of pyrrole monomers on ZnIn2S4 nanosheets surface, which was integrated into photoelectrochemical (PEC) aptasensing system for sensitive determination of di(2-ethylhexyl)phthalate (DEHP) based on hemin monomer-mediated biocatalytic precipitation effect. Driven by interfacial electric field, NIR-response ZnIn2S4/PPy heterojunctions exhibited effective charge carrier separation, thereby enhancing initial photocurrent response. The hemin monomers

During the conversion of fibroblasts into induced pluripotent stem cells (iPSCs), cellular metabolism shifts from oxidative phosphorylation toward glycolysis; however, how functional and structural mitochondrial adaptations are coordinated remains incompletely understood. Here, we examined selected aspects of mitochondrial function, targeted gene expression, and ultrastructure in fibroblasts and iPSCs derived from healthy donors and patients with osteogenesis imperfecta (OI). Targeted gene expression analysis was performed in all four cell types, while functional and ultrastructural assays foc

Ventricular assist devices (VADs) are increasingly used in pediatric patients with end-stage heart failure as a bridge to heart transplantation (HT). Thromboembolic stroke remains one of the most devastating complications and may jeopardize transplant candidacy. We report a 13-year-old male with dilated cardiomyopathy supported with an Impella 5.5 left VAD who developed an acute ischemic stroke due to left middle cerebral artery occlusion while waiting for transplant. He underwent emergent mechanical endovascular thrombectomy with good immediate neurological improvement. Given high panel react

Kidney transplantation is considered the gold standard treatment for patients with end-stage kidney disease. Historically, outcomes in kidney transplantation have been focused on reducing rates of rejection as the dominant cause of graft loss. However, managing the risk of rejection with infection continues to be a delicate balancing act for transplant physicians. It has long been recognised that viruses are an important cause of morbidity and mortality in immunosuppressed patients with significant implications for kidney graft function and patient outcomes worldwide. This is a review article

Molecular excitonic spins have garnered significant interest for quantum technologies because they can be initialized into addressable, multilevel quantum states through spin-selective intersystem crossing or singlet fission. However, excitonic spin coherence is difficult to maintain above liquid helium temperatures due to typical crystal packings, which promote decoherence through exciton hopping between magnetically inequivalent sites. Here, we engineer donor-acceptor cocrystals where molecular packing in isolated π-stacks of magnetically equivalent molecules suppresses hopping-induced decoh

Obesity is a known risk factor for diseases of the pancreas, including diabetes, pancreatic cancer and pancreatitis, but mechanisms remain unclear. To elucidate how obesity impacts pancreatic immune homeostasis, we performed spatial, transcriptomic and functional profiling of human pancreatic immune cells from obese and non-obese organ donors. Obesity was associated with higher density of tissue resident memory T-cells (TRM) in the exocrine pancreas which display high cytotoxic functions and aggregated around macrophages. Single cell sequencing of pancreatic macrophages revealed two main subse

Solar-energy-based bio- and photocatalytic hybrid systems offer a sustainable approach to reducing carbon emissions by converting carbon dioxide (CO2) into valuable chemicals and fuels, such as carbon monoxide (CO), formate, or methanol. A key strategy involves coenzyme-dependent oxidoreductases, necessitating efficient and cost-effective coenzyme regeneration methods using economical energy sources. Herein, the oxidized form of methylviologen dichloride (MV2+) is reduced to MV•+ under visible light using triethanolamine (TEOA) as an electron donor and conjugated-polymer-based water-processabl

Sonodynamic therapy (SDT) emerges as a promising noninvasive modality for deep tumors but is hindered by the hypoxic tumor microenvironment and glutathione (GSH)-mediated reactive oxygen species (ROS) scavenging. Herein, we report a GSH-responsive nanoplatform fabricated from an mPEG-b-PMNC copolymer, enabling spatiotemporal codelivery of nitric oxide (NO) donors and the sonosensitizer chlorin e6 (Ce6) via self-assembled micelles. Upon exposure to elevated intracellular GSH, the micelles disintegrate to release NO and Ce6 selectively. The released NO attenuates hypoxia through downregulation o

Chronic wounds are a worldwide medical challenge due to the complex and multifaceted etiologies, including bacterial infection, persistent inflammation, and impaired angiogenesis. Developing a comprehensive strategy integrating antibiosis and anti-inflammation to promote revascularization and accelerate wound healing is highly desirable. Nevertheless, current therapeutic methods still face two major challenges: 1) how to combat bacterial drug resistance, 2) how to achieve spatiotemporal control over bacterial elimination and inflammation reduction. To address these issues, a novel H2S donor fu

SEC24D is a key component of the Coat Protein Complex II, which plays a critical role in the selective sorting and transport of cargo proteins from the endoplasmic reticulum. This function is particularly essential for the secretion of extracellular matrix proteins, including collagens. Biallelic pathogenic variants in SEC24D have been associated with Cole-Carpenter Syndrome 2, a rare skeletal dysplasia characterized by craniofacial abnormalities and recurrent fractures. We reported a 12-year-old male patient presenting with recurrent bone fractures, severe skeletal deformities, limb shortenin

This case report describes an unexpected intraoperative discovery during secondary revision surgery for residual unilateral cleft lip deformity in an 18-year-old female. Despite a consistent patient history of a prior "autologous costal cartilage rhinoplasty," surgical exploration revealed an undocumented silicone dorsal nasal implant and ectopic costal cartilage within the alveolar cleft defect, the first known report of such concomitant findings. The ectopic cartilage, representing a biologically unsound approach to alveolar reconstruction, was excised entirely to establish a viable bony rec

A novel diphenylamine- and cyano-substituted [5]helicene derivative (M111) was rationally designed, synthesized, and characterized as a fluorescence-based sensor for methanol detection. The molecule features a rigid π-conjugated helical backbone with a donor-acceptor (D-A) architecture, enabling efficient intramolecular charge transfer (ICT) in the excited state. Photophysical studies in solvents of varying polarities revealed pronounced solvatochromic behavior, including bathochromic shifts and fluorescence quenching in polar media, particularly methanol. In dichloromethane-methanol mixtures,

Lipoprotein glomerulopathy (LPG) is an ultrarare kidney disorder caused by pathogenic variants in the APOE gene. Although kidney biopsy presents typical findings, such as dilated capillary loops containing lipoprotein thrombi, definitive diagnosis requires molecular genetic analysis of APOE. There is no specific treatment for the disease, and, in the scenario of a disorder with glomerular lipoprotein deposition, it may recur after kidney transplantation. Herein we reported 4 cases of post-transplant recurrence of LPG in Brazilian patients, including 1 case of early relapse (in the first year f

The use of robotic surgery in transplantation is increasing; however, robotic liver transplantation (RLT) remains a challenging undertaking. To our knowledge, this is a report of the first RLT in North America and the first RLT using a whole graft from a deceased donor in the world. This paper describes the preparation leading to the RLT and the surgical technique of the operation. The operation was performed in a 62-year-old man with hepatitis C cirrhosis and hepatocellular carcinoma with a native Model for End-Stage Liver Disease score of 10. The total console time for the operation was 8 ho

Genetic variation in host immunity impacts the disproportionate burden of infectious diseases that can be experienced by First Nations peoples. Polymorphic human leukocyte antigen (HLA) class I and killer cell immunoglobulin-like receptors (KIRs) are key regulators of natural killer (NK) cells, which mediate early infection control. How this variation impacts their responses across populations is unclear. We show that HLA-A∗24:02 became the dominant ligand for inhibitory KIR3DL1 in First Nations peoples across Oceania, through positive natural selection. We identify KIR3DL1∗114, widespread acr

Nitriles and isonitriles are important σ-donor ligands in coordination chemistry. Isonitriles also function in low-valent complexes as π-acceptor ligands similar to CO. Herein we present the unusual behavior of the highly reducing, high-spin iron(I) complex [Fe(hmds)2]- toward these compound classes. Rare examples of side-on coordination of nitriles to the metal center are observed. Insights gained by 57Fe Mössbauer spectroscopy as well as DFT and CASSCF calculations give an interplay between the resonance structures of not only an iron(I) π-complex and an iron(III) metallacycle but also point

In this study, we present an innovative "click-to-release" strategy for the design of highly specific H2Sn bioorthogonal probes that undergo a specific click reaction with H2Sn and release fluorophores by a following rearrangement. A library of cyclooctyne derivatives was established and successfully demonstrated the availability of the release strategy. Then, a model probe CM-CT was synthesized, which can achieve effective fluorophore release (>80%) in the presence of a H2Sn donor. To further validate the application of this class of probes, a new probe QN-RHO-CT based on Rhodamine 110 was de
