Gastrointestinal nematode (GIN) infections significantly impact the health of ruminants. The indiscriminate use of anthelmintics contributes to the selection of drug-resistant parasites. Compounds containing selenium (Se) and tellurium (Te) have emerged as promising alternatives. This study aimed to evaluate the anthelmintic activity of diphenyl diselenide (PhSe)2, phenylselenenyl chloride (4-Cl-PhSe)2, and ammonium trichloro (dioxoethylene-O,O') tellurate (AS-101) on eggs and larvae (L3) of GIN. The compounds were tested using egg-hatch and larval-migration inhibition tests. Additionally, the

Candida albicans biofilms present a major clinical challenge due to their resistance to conventional antifungal therapies. Sophorolipids (SLs), glycolipid biosurfactants produced by Starmerella riodocensis, have emerged as promising antifungal agents. This study evaluates SLs produced using soybean meal, a low-cost agro-industrial substrate, for antibiofilm activity and synergistic potential with amphotericin B (AmB) and itraconazole (ITZ), two widely used antifungals. Both crude and purified SLs reduced biofilm formation and metabolic activity, with purified SLs showing superior efficacy. SLs

Functional precision oncology (FPO) enables individualized therapy selection using patient-derived tumor models, yet the concordance of distinct ex vivo testing strategies remains unclear. Here, we compare two orthogonal drug sensitivity platforms across molecularly characterized models spanning diverse pediatric (n = 13) and adult (n = 6) cancers. A rapid ATP-based assay quantifies viability within three days, whereas a long-term dynamic image-based platform captures microtumor dynamics over two weeks, incorporating pharmacokinetic features. Up to 50 drugs were profiled across both platforms,

The integration of ICIs has reshaped therapeutic strategies in head and neck squamous cell carcinoma (HNSCC). ICIs are established in recurrent/metastatic (R/M) HNSCC and have been extensively evaluated in resectable and locoregionally advanced (LA) disease, without success until the recent KEYNOTE-689 and NIVOPOSTOP studies. These pivotal trials demonstrate that perioperative and adjuvant ICIs can improve clinical outcomes and change clinical practice. Their implementation in resectable HNSCC may raise several unanswered questions, including optimal sequencing with surgery and radiotherapy, p

Trastuzumab deruxtecan (T-DXd), a HER2-directed antibody-drug conjugate (ADC), has revolutionized the treatment landscape for HER2-positive and HER2-low breast cancer, offering unprecedented improvements in progression-free survival (PFS) and overall survival (OS). However, the emergence of resistance limits its long-term efficacy and highlights the need for rational post-progression strategies. This review summarizes current understanding of resistance mechanisms to T-DXd, including dynamic HER2 expression loss, dysregulation of DNA damage repair, tumor microenvironment remodeling, and immune

Device- and mucosa-associated candidiasis is difficult to cure because Candida biofilms shield cells from antifungals, leading to relapse and device failure. Standard treatment decisions are still largely guided by planktonic susceptibility tests, which poorly predict the drug exposure needed to clear mature biofilms. Here we synthesize evidence that natural-compound adjuvants can dismantle key biofilm defenses and outline design rules to rationalize biofilm-aware combination therapy. Across Candida albicans, non-albicans species and Candida auris, the most reproducible adjuvant effects fell i

Acute myeloid leukemia (AML) is a highly aggressive malignancy defined by significant biological diversity and variable patient outcomes. A key subset of AML is driven by abnormalities that lead to the overexpression of the oncogenic transcription factors HOXA9 and MEIS1. These abnormalities include KMT2A (formerly MLL) rearrangements and NPM1 mutations, as well as other rare lesions such as NUP98 rearrangements. This review focuses on the biology of the KMT2A, NPM1, and HOX/MEIS1 pathways, dissecting their molecular mechanisms of leukemogenesis. A central theme is the role of the scaffolding

Hematological malignancies, including multiple myeloma (MM), leukemia, and lymphoma, represent a major global health burden, accounting for approximately 6.6% of all cancer cases and contributing to significant mortality. The evolutionary conserved WNT/β-catenin signaling pathway is a critical regulator of normal hematopoietic stem cell homeostasis, and its dysregulation is a hallmark of various hematological malignancies. Aberrant activation through mutations, overexpression of ligands, or disruption of the destruction complex drives uncontrolled proliferation, impaired differentiation, and t

Peptide multi-receptor agonists have advanced obesity treatment, yet challenges remain in achieving maximal weight loss and metabolic control, especially in patients with obesity and type 2 diabetes. Here we demonstrate enhanced metabolic benefits of a combination therapy with retatrutide, a unimolecular GLP-1R/GIPR/GCGR tri-agonist, and cagrilintide, an AMLNR/CALCR co-agonist, in diet-induced obese male rats. Daily co-administration produces dose-dependent reductions in body weight and food intake that exceed both equimolar monotherapies and matched-dose comparator combinations incorporating

Treatment of extensively drug-resistant (XDR) organisms is complicated by pharmacodynamic and pharmacokinetic (PK) limitations of current antimicrobial agents. Prostatitis is an infection that is historically difficult to treat with beta-lactams given poor tissue penetration as compared to fluoroquinolones and sulfamethoxazole-trimethoprim. Cefiderocol is a novel cephalosporin that is often used to treat a wide variety of gram-negative bacteria that have extensive resistance to other antibiotics. Cefiderocol remains stable against many β-lactamases and shows enhanced bacterial cell entry as a

Treating bipolar depression that comes with obsessive, looping thoughts can be tricky because standard antidepressants often help only a little and can sometimes push the mood too high. In this report, we followed a 32-year-old man whose low mood and health-related ruminations lingered despite mood stabilisers (valproate, low-dose aripiprazole, pregabalin, and Deanxit). Hoping to copy some of ketamine's brain-circuit effects, he tried an over-the-counter mix of dextromethorphan, piracetam, and L-glutamine; the result was modest at best. Ten days after he added 600 mg of the herbal CYP2D6 inhib

Autophagy is a lysosome-dependent recycling process that maintains cellular homeostasis and helps cells adapt to therapeutic stress. In oral squamous cell carcinoma (OSCC), dysregulated autophagy may promote chemoresistance by supporting metabolic adaptation, removing damaged cellular components, and limiting treatment-induced cell death. Its effects are nevertheless context dependent, as autophagy can also interact with apoptosis, ferroptosis, and other cytotoxic pathways. This review summarizes molecular mechanisms linking autophagy to OSCC chemoresistance, focusing on non-coding RNAs, p53/T

Pathogenic xenobiotics are ubiquitous in the environment. Environmental carcinogens induce mutations leading to tumors; pathogens relying on environmental media transmission cause infections; exposure to chemical or biological pollutants triggers toxicant poisoning. The mechanisms of xenobiotics work by abnormal expression of genes. Since their inception, genome-wide CRISPR/Cas9 knockout libraries have been rapidly applied in vitro to dissect the mechanism of diseases. Genome-wide screening of CRISPR/Cas9 knockout libraries has helped to identify genes essential for tumor survival, combination

Design of modular, transferable protein assemblies has broad applicability and in structural biology could help with the ever-troublesome crystallization bottleneck, including finding robustly behaved protein crystals for rapidly characterizing ligands or drug candidates or generating multiple polymorphs to illuminate diverse conformations. Nanobodies as crystallization chaperones are well-established but still unreliable, as we show here. Instead, we show an exemplar of how robust crystallization behavior can be engineered by exploring many combinations (>200) of nanobody surface mutations ov

Premature deaths account for about half of all fatalities in developing countries. In low- and middle-income countries, cardiovascular diseases have consistently been the primary cause of premature mortality for men and women during the past three decades. Current evidence indicates that fixed-dose combination therapy, the so-called Polypill, effectively reduces the burden of cardiovascular diseases, with greater benefits observed in combinations that include aspirin. Polypill demonstrates high adherence and an acceptable safety profile, with adverse drug events being comparable between the gr

Most clinically used antibiotics exert their effects by targeting essential intracellular components of bacterial cells. Therefore, enhancing their ability to traverse the bacterial envelope is crucial for restoring or improving therapeutic efficacy. We investigated the potential of outer membrane (OM)-disrupting agents-EDTA, NV716, colistin, and squalamine-to potentiate antibiotic activity against the multi-drug-resistant pathogen Pseudomonas aeruginosa. Our objective was to assess the therapeutic value of this strategy while also delineating its limitations by comparing responses across anti

Triple-negative breast cancer (TNBC) represents a formidable subtype with a grim prognosis. This study aims to pinpoint the molecular targets of Glycosyltransferases (GTs) in TNBC, with the goal of improving prognostic accuracy and boosting the effectiveness of immune therapies. Using publicly available datasets, we combined differentially expressed and correlated genes based on AUCell scores from single-cell sequencing. These were then subjected to enrichment analysis and utilized for constructing a risk model. A total of 780 genes were identified as being closely associated with GTs. Using 1
