Vaginal drug delivery for the localised treatment of cervical cancer.

Cervical cancer is usually treated by surgery, with the more advanced cancers requiring adjuvant chemotherapy or radiotherapy. The location of the cervix makes it easily accessible through the vagina for the localised delivery of chemotherapeutic drugs. Localised delivery has the advantage of direct delivery to the site of action resulting in a lower dose having to be required and a reduction in systemic side effects. This approach would be advantageous for fertility sparing surgery, whereby localised delivery could be used to reduce tumour size allowing for a much smaller tumour to be removed

Cellulose and Nanocellulose Emulsions in Biomedical Applications: From Fundamental Mechanisms to Therapeutic Translation.

Poor aqueous solubility remains one of the most persistent challenges in pharmaceutical development, limiting clinical translation and requiring innovative formulation strategies; approximately 40% of newly discovered pharmaceutical compounds are affected, underscoring the scale of the problem. Emulsion-based delivery systems overcome this limitation by maintaining drugs in a dissolved state, increasing absorption surface area, and enabling controlled release; however, conventional emulsions face thermodynamic instability and coalescence challenges. Cellulose and nanocellulose-cellulose nanocr

Recent advances in biotin-based therapeutic agents for cancer therapy.

Biotin receptors, as biomarkers for cancer cells, are overexpressed in various tumor types. Compared to other vitamin receptors, such as folate receptors and vitamin B12 receptors, biotin receptor-based targeting strategies exhibit superior specificity and broader potential in treating aggressive cancers, including ovarian cancer, leukemia, colon cancer, breast cancer, kidney cancer, and lung cancer. These strategies promote biotin transport via receptor-mediated endocytosis, which is triggered upon ligand binding. Biotin, as the ligand of the biotin receptor, can be conjugated to anti-cancer

Enzyme/pH-sensitive nanoparticles based on poly(β-L-malic acid) for drug delivery with enhanced endocytosis.

Nanoparticles (NPs) derived from branched copolymers of poly (β-L-malic acid) (PMLA) have been extensively investigated for drug delivery due to their high density of pendant carboxyl groups. This abundant functional group availability enhances their potential as effective drug delivery systems; however, the strong negative charge of PMLA poses a challenge in its uptake by cancer cells due to electrostatic repulsion. In this study, we developed novel enzyme- and pH-sensitive nanoparticles (EP-NPs) based on PMLA, demonstrating tumor-specific behavior and selective activation within tumor tissue…

A QbD optimization of a pH-responsive Eudragit S100-chitosan nanoformulation for the co-delivery of pentoxifylline and simvastatin in colorectal cancer therapy.

Colorectal cancer (CRC) presents a significant global health challenge, where the efficacy of conventional treatments is often hampered by systemic toxicity, poor bioavailability, and drug resistance. Drug repurposing and nanotechnology offer promising avenues to overcome these limitations. While pH-responsive Eudragit S100-chitosan hybrid systems are established as robust pH-responsive oral delivery carriers, the present study introduces a novel therapeutic strategy by combining these platforms with the co-delivery of the repurposed drugs pentoxifylline (PTX) and simvastatin (SIM) for synergi

Polymeric microcontainers improve oral bioavailability of furosemide.

Microcontainers with an inner diameter of 223 μm are fabricated using the polymer SU-8, and evaluated in vitro, in situ and in vivo for their application as an advanced oral drug delivery system for the poorly water soluble drug furosemide. An amorphous sodium salt of furosemide (ASSF) is filled into the microcontainers followed by applying a lid using Eudragit L100. It is possible to control the drug release in vitro, and in vitro absorption studies show that the microcontainers are not a hindrance for absorption of ASSF. In situ perfusion studies in rats are performed with ASSF-filled microc

Biodistribution assessment of cationic pullulan nanogel, a nasal vaccine delivery system, in mice and non-human primates.

Cationic cholesteryl-group-bearing pullulan nanogel (cCHP-nanogel) is an effective drug-delivery system for nasal vaccines. However, cCHP-nanogel-based nasal vaccines might access the central nervous system due to its close proximity via the olfactory bulb in the nasal cavity. Using real-time quantitative tracking of the nanogel-based nasal botulinum neurotoxin and pneumococcal vaccines, we previously confirmed the lack of deposition of vaccine antigen in the cerebrum or olfactory bulbs of mice and non-human primates (NHPs), rhesus macaques. Here, we used positron emission tomography to invest

Engineering Diselenide-IR780 Homodimeric Nanoassemblies with Enhanced Photodynamic and Immunotherapeutic Effects for Triple-Negative Breast Cancer Treatment.

Photodynamic therapy (PDT) has emerged as an efficient approach for non-invasive cancer treatment. However, organic small-molecule photosensitizers are often associated with defects in hydrophobicity, poor photostability, and aggregation-caused quenching, which limit their application. Usually, the carrier-assisted drug delivery system is a common strategy to solve the above obstacles, but additional carrier material could increase the risk of potential biological toxicity. The carrier-free drug delivery system with easy preparation and high drug-loading capability is proposed subsequently as

A Selective Bis-Catecholate Siderophore-Ampicillin Conjugate Expands Activity against Multidrug-Resistant Pseudomonas aeruginosa.

Antimicrobial resistance is a global public health crisis, with multidrug-resistant P. aeruginosa among the most critical threats. The high cost and slow pace of novel antibiotic development, coupled with rapid resistance evolution, demand innovative strategies. Here, we present a Trojan horse approach using siderophore-antibiotic conjugates (SACs) to hijack bacterial iron uptake systems and deliver β-lactam payloads across the Gram-negative outer membrane. We designed a series of ampicillin conjugates with mono-catecholate, bis-catecholate, and catecholate-hydroxamate siderophores. These conj
