Drug-target interaction prediction by integrating heterogeneous information with mutual attention network.

BACKGROUND: Identification of drug-target interactions is an indispensable part of drug discovery. While conventional shallow machine learning and recent deep learning methods based on chemogenomic properties of drugs and target proteins have pushed this prediction performance improvement to a new level, these methods are still difficult to adapt to novel structures. Alternatively, large-scale biological and pharmacological data provide new ways to accelerate drug-target interaction prediction. METHODS: Here, we propose DrugMAN, a deep learning model for predicting drug-target interaction by

Current Perspectives on Severe Drug Eruption.

Adverse drug reactions involving the skin are commonly known as drug eruptions. Severe drug eruption may cause severe cutaneous adverse drug reactions (SCARs), which are considered to be fatal and life-threatening, including Stevens-Johnson syndrome (SJS), toxic epidermal necrolysis (TEN), acute generalized exanthematous pustulosis (AGEP), and drug reaction with eosinophilia and systemic symptoms (DRESS). Although cases are relatively rare, approximately 2% of hospitalized patients are affected by SCARs. There is an incidence of 2 to 7 cases/million per year of SJS/TEN and 1/1000 to 1/10,000 e

Drug interactions with new hepatitis C oral drugs.

INTRODUCTION: Chronic hepatitis C virus (HCV) infection has recently become a curable disease with antiviral therapy. The knowledge of drug interactions using direct-acting antivirals (DAA) may permit maximizing antiviral efficacy and avoiding drug-related toxicities. Ageing in the chronic hepatitis C population, along with added co-morbidities that require other medications, has increased the attention on drug interactions using DAA. AREAS COVERED: This review provides an update of the most clinically significant pharmacokinetic and pharmacodynamic drug interactions occurring between current

The impact of ageing on the barriers to drug delivery.

Generally, we like to see ageing as a process that is happening to people older than ourselves. However the process of ageing impacts on a wide range of functions within the human body. Whilst many of the outcomes of ageing can now be delayed or reduced, age-related changes in cellular, molecular and physiological functionality of tissues and organs can also influence how drugs enter, distribute and are eliminated from the body. Therefore, the changing profile of barriers to drug delivery should be considered if we are to develop more age-appropriate medicines. Changes in the drug dissolution

Trends in drug use among gay and bisexual men in Sydney, Melbourne and Queensland, Australia.

INTRODUCTION AND AIMS: The findings of Australian drug surveys are typically not stratified by sexual orientation, despite the higher prevalence of drug use generally reported among gay and bisexual men. This paper aims to examine trends in drug use among gay and bisexual men in eastern Australia between 2004 and 2011. DESIGN AND METHODS: Data from the cross-sectional, ongoing Gay Community Periodic Surveys (GCPS) were used to analyse drug trends among gay and bisexual men in Sydney, Melbourne and Queensland. Between 2004 and 2011, 45,273 eligible questionnaires were completed. RESULTS: Ther

Development and analysis of a SmPC-based machine-readable dataset of contraindications for clinical decision support and real-world data analysis.

PURPOSE: Unrecognized contraindications pose risks for adverse drug reactions, hospitalizations or death. Clinical decision support systems (CDSS) aim to mitigate medication-related harm, particularly originating from contraindications. However, many CDSS provide limited benefit, as they focus largely on singular risk situations such as drug-drug interactions and often generate alerts of limited clinical relevance. Comprehensive integration of contraindications into CDSS may support more clinically meaningful alerts. The aim of this work was the development and analysis of machine-readable con

Using drug checking data to model the relationship between the prevalence of fentanyl and xylazine to drug overdose deaths in New York City.

BACKGROUND: Drug checking is an increasingly common harm reduction service available across the United States. However, evidence of whether drug-checking data is associated with population-level health measures is limited. METHODS: Beginning in November 2021, the New York City (NYC) Health Department partnered with syringe service programs (SSP) to initiate a drug checking pilot study. SSP participants submitted drug samples for point-of-care drug checking and secondary lab testing. Time series generalized additive modeling (GAM) techniques were used to examine whether there was an associatio

Is drug allergy less prevalent than previously assumed? A 5-year analysis.

BACKGROUND: Rashes are a frequent conundrum in clinical practice as they may be reactive, drug induced or disease specific. Identification of the culprit drug is important as re-exposure may be harmful or even life-threatening and unnecessary avoidance of 'innocent' drugs leads to limitations of treatment options. OBJECTIVE: To objectify the cause of suspected cutaneous drug reactions in a large patient population. METHOD: Over 5years (2006-10), 612 patients with suspected cutaneous drug reactions were evaluated. Histology was assessed. About 200 patients were invited for complete work-up wi

Reporter parasite lines: valuable tools for the study of Plasmodium biology.

The human malaria parasite Plasmodium falciparum causes the most severe form of malaria in endemic regions and is transmitted via mosquito bites. To better understand the biology of this deadly pathogen, a variety of P. falciparum reporter lines have been generated using transgenic approaches to express reporter proteins, such as fluorescent proteins and luciferases. This review discusses the advances in recently generated P. falciparum transgenic reporter lines, which will aid in the investigation of parasite physiology and the discovery of novel antimalarial drugs. Future prospects for the g

Selected hybrid natural products as tubulin modulators.

Modulators of microtubule dynamics have received increasing attention because of their potential to stop cancer growth. Although it belongs to the category of complex protein-protein interactions (PPIs), which are generally considered difficult to modulate through small molecules, the use of microtubule is considered a well-validated target. There are a number of bioactive natural products and related compounds that are currently in use as drugs or in clinical trials as next generation anti-cancer agents. The present review article is focused on two such bioactive natural products, epothilone

The epidemiology of alcohol, drug, and mental disorders among homeless persons.

This article describes recent research on the prevalence of alcohol, drug, and mental (ADM) disorders and the characteristics of homeless substance abusers and persons with mental illness. Methodological problems in homelessness research are reviewed, particularly in relation to definitions of homelessness and sampling- and case-ascertainment methods. Prevalence rates of ADM disorders are much higher in homeless groups than in the general population. As is true of homeless people in general, homeless substance abusers and mentally ill persons are characterized by extreme poverty; underutilizat

Human disease-drug network based on genomic expression profiles.

BACKGROUND: Drug repositioning offers the possibility of faster development times and reduced risks in drug discovery. With the rapid development of high-throughput technologies and ever-increasing accumulation of whole genome-level datasets, an increasing number of diseases and drugs can be comprehensively characterized by the changes they induce in gene expression, protein, metabolites and phenotypes. METHODOLOGY/PRINCIPAL FINDINGS: We performed a systematic, large-scale analysis of genomic expression profiles of human diseases and drugs to create a disease-drug network. A network of 170,02

DNA Nanostructures for Overcoming Cancer Drug Resistance: Mechanisms, Therapeutic Strategies, and Translational Challenges.

Cancer drug resistance, a major cause of chemotherapy failure, arises from multiple interconnected barriers, including reduced intracellular drug accumulation, enhanced drug efflux, adaptive survival signaling, increased DNA repair, and redox or metabolic remodeling. To address these limitations, recent advances in programmable DNA nanostructures have provided new opportunities for constructing multifunctional nanotherapeutic platforms. This review examines how the structural characteristics of DNA nanostructures influence drug delivery, molecular regulation, and resistance modulation, and ana

ProphDR: An Interpretable Deep Learning Model for Predicting Cancer Drug Response via Multi-Omics and Cross-Attention Mechanisms.

Predicting cancer drug responses (CDRs) accurately remains a significant challenge due to the complexity of tumor biology and the limitations of existing "black-box" machine learning models. To address this, we propose ProphDR, an interpretable deep learning framework that integrates multiomics data and drug structural information using a hierarchical attention mechanism. ProphDR incorporates a Criss-Cross Gene-level Multiomics Integration (CGMI) module to capture gene-level features and a cross-attention (CA) module to model drug-gene interactions. Evaluated on datasets from GDSC and CCLE, Pr

Novel explainable deep learning based drug sensitivity prediction for early treatment of breast cancer.

High-level screening technologies have generated a vast volume of drug-sensitivity data for a panel of cancer cell lines and hundreds of chemicals. By identifying molecular genetic factors of drug sensitivity and developing novel anticancer medicines, computational approaches to analysing these data can assist in the development of anticancer therapies. Conventional deep learning models lack the ability to select the best imputation strategy or to handle missing values. This may compromise the dataset’s originality and introduce data sensitivity issues. To address these issues, we introduce th

Optimal control of multiple myeloma assuming drug resistance and off-target effects.

Multiple myeloma (MM) is a plasma cell cancer that occurs in the bone marrow. A leading treatment for MM is the monoclonal antibody Daratumumab, targeting the CD38 receptor, which is highly overexpressed in myeloma cells. In this work we model drug resistance via loss of CD38 expression, which is a proposed mechanism of resistance to Daratumumab treatment. We develop an ODE model that includes drug resistance via two mechanisms: a direct effect in which CD38 expression is lost without cell death in response to Daratumumab, and an indirect effect in which CD38 expression switches on and off in

High Risk, Low Key: The New Face of Drug Use.

In today's rapidly evolving substance use landscape, the traditional image of drug consumption is being replaced by a subtler, more socially accepted aesthetic. High Risk, Low Key: The New Face of Drug Use explores how modern intoxicants, from THC-infused beverages to microdosed psilocybin and nitrous oxide canisters, are increasingly marketed as wellness products rather than recreational drugs. This shift, driven by cultural, technological, and mental health trends, has led to the normalization and concealment of daily drug use, especially among youth. Cannabis, once emblematic of rebellion,

Targeting CAFs-Mediated Stromal Signaling in a Patient-Derived Organotypic Colorectal Tumor Model.

Colorectal cancer, a significant cause of cancer-related mortality, often exhibits drug resistance, highlighting the need for improved tumor models to advance personalized drug testing and precision therapy. We generated organoids from primary colorectal cancer cells cultured through the conditional reprogramming technique, establishing a framework to perform short-term drug testing studies on patient-derived cells. To model interactions with stromal cells in the tumor microenvironment, we combined cancer cell organoids with carcinoma-associated fibroblasts, a cell type implicated in disease p
