Neural underpinnings of verbal memory encoding and recognition impairments in individuals with clinically stable mood or schizophrenia spectrum disorders.

BACKGROUND: Verbal memory impairment is prevalent in mood and schizophrenia spectrum disorders and contributes to poor functional outcomes. The neural correlates of these memory difficulties remain unclear due to a scarcity of neuroimaging studies assessing verbal memory and low ecological validity of current paradigms. The aim of this study was to examine neurocircuitry abnormalities across mood and schizophrenia spectrum disorders using a new verbal memory task simulating everyday grocery shopping. METHODS: Fifty-seven cognitively impaired individuals with clinically stable mood or schizoph

A Novel Intracellular Domain Missense Variant in GRM7 Associated with Early-Onset Neurodevelopmental Encephalopathy.

INTRODUCTION: Biallelic pathogenic variants in the GRM7 gene, which encodes the metabotropic glutamate receptor 7 (mGlu7), have been associated with early-onset neurodevelopmental disorders characterized by epilepsy, developmental delay, and white matter abnormalities. However, reported cases predominantly involve variants affecting the extracellular or transmembrane domains, and the full phenotypic spectrum remains unclear. CASE PRESENTATION: We present the case of a male patient exhibiting early-onset focal epilepsy, global developmental delay, and neuroimaging evidence of corpus callosum t

Baseline mood and non-motor symptom burden are associated with cognitive progression in Parkinson's disease: an interpretable follow-up cohort analysis with separate neuroimaging, molecular, and digital analyses in independent samples.

Clinically useful artificial intelligence and machine-learning studies in healthcare require interpretable features, internal validation, and explicit boundaries between primary inference and external context. Among 1,612 baseline Parkinson's Progression Markers Initiative (PPMI) participants, 1,439 had evaluable post-baseline cognition and contributed 5,909 records through Year 5. Composite cognitive progression occurred in 720 participants (50.0%). Each standard deviation increase in baseline mood/non-motor burden was associated with higher odds of progression (adjusted odds ratio 1.37, 95%

Concussion pathophysiology: From biomechanical insult to clinical phenotype - or is injury truly the beginning?

Mild traumatic brain injury (mTBI) is associated with substantial morbidity worldwide. Emerging evidence demonstrates that both impact- and blast-related mTBI produce diffuse microstructural and functional alterations, e.g., diffuse axonal injury, astroglial and microglial activation, cerebrovascular dysfunction, and neurometabolic disturbance. Rotational acceleration in impact injury preferentially induces white matter shear. Blast overpressure exerts prominent effects at cerebrovascular interfaces, particularly within perivascular spaces (PVS). These biomechanical differences may yield disti

Multimodal diffusion MRI biomarkers of white matter alterations and clinical impairment in mTBI.

INTRODUCTION: Mild traumatic brain injury (mTBI), which includes concussion, often results in subtle white matter abnormalities that are not detectable with conventional neuroimaging. This study investigated microstructural brain changes in individuals with subacute mTBI using advanced diffusion MRI (dMRI) techniques, including multi-shell diffusion tensor imaging (DTI), free-water corrected DTI (fw-DTI), diffusion kurtosis imaging (DKI), and neurite orientation dispersion and density imaging (NODDI). METHODS: Twenty individuals with mTBI and 24 healthy controls (HCs) underwent dMRI and cogni

Experience-dependent rapid structural changes in the human brain: A systematic review.

Structural brain plasticity has traditionally been viewed as a gradual process developing over days to weeks of training or experience. Recent neuroimaging evidence, however, demonstrates that measurable structural changes can occur within hours of behavioral or physiological manipulation, a phenomenon referred to as rapid structural plasticity. This systematic review synthesizes evidence for such short-interval (<2 h) structural alterations in gray matter across magnetic resonance imaging (MRI) modalities in humans. Thirty studies met inclusion criteria, assessing diffusion-weighted magnetic

Multimodal imaging reveals resilient memory networks in carriers of pathogenic ARID1B variants.

The phenotypic spectrum associated with pathogenic ARID1B variants is remarkably broad, ranging from classic Coffin-Siris syndrome to non-syndromic intellectual disability and autism spectrum disorders. While speech delay, motor impairments and learning difficulties are well documented, brain imaging investigations remain scarce in this population. We combined multimodal neuroimaging and neuropsychological assessments in 12 patients carrying pathogenic ARID1B variants (age = 13.8 ± 4.7 years) and 34 age-matched healthy controls. Whole-brain voxel-wise analyses included arterial spin labelling

Peripheral inflammation is associated with emotional dysregulation, suicide attempts, and reduced microstructural integrity of white matter in young adults with borderline personality disorder.

BACKGROUND: Borderline personality disorder (BPD) is a psychiatric condition characterized by functional impairment and high rates of suicide attempts. Although its pathophysiology is still unclear, emerging evidence suggests a role of peripheral inflammation. This study investigates the association between peripheral inflammatory markers and clinical, behavioural, and neuroimaging features in individuals with BPD. METHODS: Seventy-nine BPD patients and 26 controls underwent a multidimensional clinical evaluation, including emotional dysregulation and suicide attempt assessment, and blood sam

Predictive Models for Motor Outcomes From Deep Brain Stimulation in Parkinson's Disease: A Systematic Review.

Deep brain stimulation (DBS) is an effective treatment for Parkinson's disease, but the extent of improvement in motor symptoms varies. A tool which accurately predicts patient outcomes based on data available pre-surgery would be useful for clinical decision-making and patient expectation management. Such data includes clinical and cognitive measures, neuroimaging, kinematics, functional connectivity and genetics. In this systematic review, we assess predictive models of motor outcomes from DBS. We searched the databases Web of Science, PubMed and Scopus for primary research articles that tes

Emerging diagnostic biomarkers and therapeutic targets in Alzheimer's disease.

Alzheimer's disease (AD) is a progressive neurodegenerative disease and is the most common cause of dementia in the world. Cognitive decline, memory problems, behavioral disturbances, and, finally, loss of functional independence characterize it. AD has risen and continues to increase globally, especially among aging people. A team of researchers at the University of Alberta says that we need faster, better diagnosis and treatment of AD. It elaborates on the progressive understanding of AD pathophysiology, biomarkers, emergent targets, and the latest delivery solution. The paper covers and dis

Resolving the mystery of Chinese developmental dyslexia: in search of predictors and early markers.

Developmental dyslexia is a specific learning disability for which early identification and intervention are crucial. In alphabetic languages, phonological awareness and letter naming are well-established early markers of reading disorders. For developmental dyslexia in Chinese, a language that differs from alphabetic languages in multiple aspects, little is known about its early markers. Here, we report findings from a 2.5-year longitudinal behavioral study and a functional neuroimaging study designed to identify the neural underpinnings of predictors of Chinese reading disability. In Experim…

Beyond the role in motor function, entopeduncular nucleus, a critical neural hub in psychiatric disorders and sleep regulation.

BACKGROUND: The entopeduncular nucleus (EP) is a major output nucleus of the basal ganglia and plays a critical role in integrating motor, emotional, and behavioral processes. Although traditionally linked to motor control, accumulating evidence indicates that the EP is deeply involved in psychiatric disorders, sleep regulation, addiction, and mood-related behaviors. As the rodent homolog of the human internal Globus pallidus (GP) the EP represents a key translational structure bridging preclinical and clinical neuroscience. METHODS: This narrative review was conducted following established n

Neurophysiological Characteristics Associated with Driving Abilities in Older Adults: A Scoping Review.

With population aging, motor vehicle accidents involving older drivers have increased. Age-related cognitive decline affects driving performance; however, the underlying neural mechanisms remain unclear. This scoping review explored neurophysiological characteristics associated with driving in older adults, including those at risk of dementia. Following PRISMA-ScR guidelines, we searched PubMed, Scopus, and CINAHL for studies examining driving-related neurophysiological measures in older adults aged ≥60 years. Twelve studies were included. Findings converge on load-dependent neural compensatio

Sleep Oscillations Across Cortical, Subcortical and Cerebellar Structures in Magnetoencephalography.

Sleep involves widespread changes in neural activity, with distinctive oscillatory patterns emerging across frequency bands and brain regions. Characterising these dynamics is essential for understanding their functional roles in health and their disruption in sleep-related disorders. However, most work on healthy humans has used techniques with limited temporal or spatial resolution, focusing mainly on the cerebral cortex. Growing evidence suggests that subcortical and cerebellar structures contribute to sleep dynamics, yet these regions remain largely unexplored in human neuroimaging due to

Utilizing stimulation-evoked hemodynamic activity to predict antidepressant response to intermittent theta-burst stimulation in adults with major depression.

BACKGROUND: The instantaneous neural response to prefrontal theta burst stimulation (TBS) may serve as predictive marker for antidepressant treatment success. This study aimed to (1) assess whether baseline theta burst stimulation (TBS)-induced prefrontal hemodynamic responses can predict treatment outcome of four weeks of TBS in adults with major depressive disorder (MDD); (2) assess the test-retest reliability of TBS-induced hemodynamic responses. METHODS: Forty-four MDD participants were recruited and underwent two consecutive-day concurrent TBS/functional near-infrared spectroscopy (fNIRS

Real-world anti-amyloid therapy beyond traditional symptomatic populations: a longitudinal case series.

INTRODUCTION: Anti-amyloid monoclonal antibodies are approved as disease-modifying therapies for early Alzheimer's disease (AD), but real-world evidence remains limited, particularly among individuals treated during preclinical or minimally symptomatic stages. This study characterized clinical and biomarker trajectories among anti-amyloid therapy recipients across disease stages. METHOD: We conducted a longitudinal retrospective case series of three biomarker-positive individuals from the Alzheimer's Disease Neuroimaging Initiative who received lecanemab or donanemab. Regionally heterogeneous gamma alterations and their diagnostic value in early-onset Alzheimer's disease.

Growing evidence links amyloid accumulation to altered gamma activity, positioning gamma oscillations as a promising biomarker for Alzheimer's pathology. We examined resting state gamma activity in early-onset Alzheimer's disease (EOAD) patients confirmed by CSF biomarkers. We aim to provide a holistic view of gamma activity patterns in EOAD by integrating structural and functional neuroimaging. In this cross-sectional study, we compared gamma power and coherence-total gamma (30-48 Hz), and sub-bands (gamma-1: 30-35 Hz; gamma-2: 35-40 Hz; gamma-3: 40-48 Hz)- alongside gray matter volumes in EO…

Multimodal temporal dissociation in anti-GABAA receptor encephalitis: functional recovery precedes structural resolution-a case report.

BACKGROUND: Anti-γ-aminobutyric acid A receptor (GABAA receptor) encephalitis is a rare but severe subtype of autoimmune encephalitis (AE), typically characterized by refractory seizures and rapidly progressive neurological dysfunction. Although structural abnormalities on magnetic resonance imaging (MRI) are frequently observed, the temporal relationships among clinical manifestations, electrophysiological changes, immune activity, and neuroimaging findings remain insufficiently characterized. CASE PRESENTATION: We report a 61-year-old man with anti-GABAA receptor encephalitis who underwent
